콩 추출물 투여가 흰쥐 혈청의 항산화효소 활성에 미치는 영향 (Activities of Antioxidant Enzymes in Serum of Rats Feeding Soybean Extract)

  • 본 시험은 콩 추출물의 흰쥐 투여에 따른 항산화 효소의 활성을 분석하고, 추출물의 원료인 콩 품종간, 추출물의 투여량 및 지방식이조성에 따른 효소활성도를 측정하였다. 시험결과를 요약하면 아래와 같다. 1. 콩추출물을 투여한 처리구에서 SOD와 GSHpx의 활성이 대조구에 비하여 높게 나타났으나, 통계적 유의성은 인정되지 않았다. 2. 지방식이 및 콜레스테롤을 흰쥐에 투여한 결과 콩추출물 섭취에 따른 항산화계 효소 SOD와 GSHpx의 활성은 검정콩 1호가 다원콩 및 황금콩에 비하여 높았다. 3. 지방식이조성 및 추출물의 투여량에 따른 항산화계 효소 활성도는 통계적으로 유의성이 인정되지 않았다. 4. 각 지방식이에 따른 콩추출물의 SOD와 GSHpx활성도에서 품종과 콩추출물 농도간에서는 상호작용효과가 인정되었다. 5. GSHpx 활성도에서 품종, 콩추출물 농도 및 지방식이간에서 상호작용효과가 인정되었다.

성장기의 쥐와 늙은 쥐 조직의 Glutathione Peroxidase 활성에 대한 Methionine 투여의 효과 (Effect of Methionine Supplementation on Glutathione Peroxidase Activity in Young and Old Murine Tissues)

  • 자라나는 과정의 젊은 흰쥐와 14개월 된 늙은 흰쥐, 생쥐들을 대상으로 Met 투여에 따른 간, 혈장, 신장, 비장 등의 glutathione peroxidase(GSHPx) 효소 변화를 측정 비교하였다. 성장기의 쥐에 있어서 Se을 Selenite 형태로 투여했을 때 0.5%의 식이 Met 공급은 같은 Se을 Se-Met 형태를 공급했을 때 보다 효소 활성이 증가하였다. 그러나, 14개월 된 늙은 쥐에서는 Met 투여에 대한 효소 활성은 큰 변화가 없었다. 일반적으로 식이 Se 증가는 효소 활성과 거의 정비례하나 1ppm Se 수준 이상에서는 완만하게 증가 또는 거의 정체하는 상태를 보이지만, 늙은 쥐에서는 Se의 식이 수준이 2ppm이 되어도 여전히 효소 활성이 증가하는 경향을 보였다. 성장기의 생쥐 실험에서도 2ppm Se 수준에서의0.3%와 0.8%의 식이 Met 투여는 효소 활성에 큰 영향이 없었다. 그러나 실험식이 투여 18일 경엔 4일째 보다 Met 투여군이 4.2배의 증가를 보인 반면 비투여군은 2.5배의 증가에 머물렀다. 18일경 이후 효소 활성이 감소하여 42일재는 반으로 감소함을 보였다. Se의 생체 활성은 특히 식이 Se 수준이 낮을 때 Met에 의해 증가되는 폭이 컸으며 Se 수준이 높을수록 상대적으로 감소함을 보였다. 위의 제반 결과에서 Met의 요구량이 Se-Met에 의해 대체되고 있음을 짐작할 수 있다. 그러므로 식이 Met 수준 상태에 따라 효소의 활성이 달라질 수 있다는 것을 시사한다.

Evaluation of a Dietary Organic Selenium Supplement at Different Dietary Protein Concentrations on Growth Performance, Body Composition and Antioxidative Status of Broilers Reared under Heat Stress

  • Three hundred chicks were randomly assigned among four treatments to evaluate the effect of an organic selenium supplement at various levels of dietary protein. Two levels of supplemental selenium (0 and 0.3 mg/kg) from zinc-L-selenomethionine were tested at two levels of dietary protein (normal and reduced) in a completely randomized design with $2{\times}2$ factorial layout. The experiment lasted up to 49 d of age during which all birds were exposed to $31{\pm}1^{\circ}C$. The effects of selenium or its interaction with CP on growth performance and carcass characteristics were not significant. However, feeding the reduced-CP diet decreased weight gain in the starting period and increased liver and abdominal fat weights relative to body weight. Ferric reducing ability of plasma (FRAP) was not significantly affected by dietary CP and Se or their interaction though FRAP values were numerically higher in the Se-supplemented group. Dietary CP content did not affect the activity of plasma glutathione peroxidase (GSHPx), though Se significantly elevated plasma GSHPx activity. The interaction of CP and Se was not significant for FRAP and plasma GSHPx activity.

해양오염의 진단을 위한 생화학적 오염지표에 관한 연구 II. 황해산 넙치(Paralichthys olivaceus)의 산소라디칼 및 제거효소의 변화 (Study on Biochemical Pollutant Markers for Diagnosis of Marine Pollution II. Changes in Oxygen Radicals and Their Scavenger Enzymes of the Flounder(Paralichthys olivaceus) in the Yellow Sea)

  • 해양오염의 진단을 위한 생화학적 오염지표 설정의 기초 연구의 일환으로, 오염이 심각한 서해(또는 황해)산 넙치(Paralichthys olivaceus)의 혈액 및 뇌중의 활성산소종 및 그들의 제거효소의 활성을 분석 평가하였다. 이들 넙치의 혈청중의 단백질의 함량은 해역이나 양식산의 자연산사이에는 뚜렷한 차이를 발견할 수 없었지만, 서해안 양식산 넙치의 뇌 및 근육중의 단백질 함량이 동해안의 포항의 자연산 넙치(대조군)의 단백질 함량에 비해 각각 30-45%(brain) 및 25-45%(muscle)나 유의적으로 감소하였다(p<0.001). 서해안의 양식 및 자연산 넙치의 혈청중의 과산화지질(MDA)의 량은 대조굼으로 사용한 동해안 포항의 자연산 넙치 대비 각각 30-80% 및 125-145%나 현저히 높았다. 서해안의 양식산 넙치의 혈청중의 히드록시 라디칼의 생성량은 동해안 포항의 자연산 넙치 대비 각각15-30%정도나 오히려 낮았을 뿐만 아니라 서해안의 자연산 넙치도 격포를 제외하고 영광의 자연산 넙치의 히 시 라디칼의 생설양도 약 20%나 낮았다. 서해안의 양식산 넙치의 혈청중의 SOD의 활성은 동해안 포항의 자연산 넙치 대비 15-35%정도나 오히려 낮았을 뿐만 아니라 서해안의 자연산 넙치도 영광은 35%나 낮았지만, 단지 격포만이 SOD의 활성이 35% 정도 높았을 뿐이다. 서해안의 양식산 넙치의 혈청중의 GSHPx의 활성은 동해안 포항의 자연산 넙치 대비 15-25%정도나 유의적으로 높았지만, 서해안의 자연산 넙치의 GSGPx의 활성은 어느 곳이나 낮았다. 이상의 실험결과에서 볼 때 뇌나 근육중의 단백질의 함량이나 뇌중의 GSHPx의 활상, 그리고 혈청중의 과산회지질의 함량이 넙치의 오염지표로서의 가능성을 검토할 가치가 있다고 판단된다.

Effect of Maternal Selenium Nutrition on pulmonary Selenium, Glutathione Peroxidase, and Phospholipid Levels in Neonatal Rats

  • The present study was designed to determine if prenatal and postnatal Se nutriture affects Se concentration, glutathione peroxidase(GSHPx) activity and phospholipid distribution of the neonatal rat lung. Female SD rats were bred and fed a semipurified Se-deficient(0.04ppm, Se-) or a Se-adequate(0.5ppm, Se+) diet through pregnancy and lactation. On d 2 of lactation, maternal dietary Se had no significant effect on pulmonary Se concentration of pups. On d 16 of lactation, mean milk Se concentration in Se- dams was significantly lower than that in Se+ dams. Milk Se concentration was reflected on lung Se concentration and GSHPx activity of d 16 pups, which were dramatically decreased in Se- pups. In addition, pulmonary disaturated phosphatidyl choline/total phosphatidyl choline ratio was also significantly decreased in Se- pups, implying impaired function of pulmonary surfactant. These data indicate that adequate Se nutrition is important in the maturation of neonatal rat lungs.

Silymarin Modulates Cisplatin-Induced Oxidative Stress and Hepatotoxicity in Rats

  • Cisplatin (CDDP) is a widely used anticancer drug, but at high dose, it can produce undesirable side effects such as hepatotoxicity. Because silymrin has been used to treat liver disorders, the protective effect of silymarin on CDDP -induced hepatotoxicity was evaluated in rats. Hepatotoxicity was determined by changes in serum alanine aminotransferase [ALT] and aspartate aminotransferase [AST], nitric oxide [NO] levels, albumin and calcium levels, and superoxide dismutase [SOD], glutathione peroxidase [GSHPx] activities, glutathione content, malondialdehyde [MDA] and nitric oxide [NO] levels in liver tissue of rats. Male albino rats were divided into four groups, 10 rats in each. In the control group, rats were injected i.p. with 0.2 ml of propylene glycol in saline 75/25 (v/v) for 5 consecutive days [Silymarin was dissolved in 0.2 ml of propylene glycol in saline 75/25 v/v]. The second group were injected with CDDP (7.5 mg /kg, I.P.), whereas animals in the third group were i.p. injected with silymarin at a dose of 100 mg/kg/day for 5 consecutive days. The Fourth group received a daily i.p. injection of silymarin (100 mg/kg/day for 5 days) 1 hr before a single i.p. injection of CDDP (7.5 mg/kg). CDDP hepatotoxicity was manifested biochemically by an increase in serum ALT and AST, elevation of MDA and NO in liver tissues as well as a decrease in GSH and the activities of antioxidant enzymes, including SOD, GSHPx in liver tissues. In addition, marked decrease in serum NO, albumin and calcium levels were observed. Serum ALT, AST, liver NO level, MDA was found to decreased in the combination group in comparison with the CDDP group. The activities of SOD, GSHPx, GSH and serum NO were lower in CDDP group than both the control and CDDP pretreated with silymarin groups. The results obtained suggested that silymarin significantly attenuated the hepatotoxicity as an indirect target of CDDP in an animal model of CDDP-induced nephrotoxicity.

Effect of iron and selenium status on glutathione peroxidase activity and lipid peroxidation in rats

  • The combined effects of iron and selenium status on glutathione peroxidase (GSHPx) activity, cytochrome P-450 activity, and lipid peroxidation in the liver and intestinal mucosa of rats were investigated. In experiment one, four experimental groups (+Se+Fe, -Se+Fe, +Se++Fe, -Se++Fe) were manipulated for 3 weeks with intramuscular administration of irondextran (++Fe) and/or normal diet (+Fe) and deionized water (-Se) and/or selenium-supplemented deionized water (+Se). In experiment two, 2% dietary carbonyl iron (instead of the parenteral administration) was fed for 3 weeks to rats. Body weight of rats was significantly decreased in both parenterally and orally iron-overloaded groups (p<0.01), regardless of Se supplement. Serum iron was significantly increased in parenterally iron-overloaded groups but it was marginally increased in orally iron-overloaded groups. There was no significant difference in hemoglobin content among experimental groups in either experiment one or two. Total iron in the small intestine, intestinal mucosa, and livers was significantly high in both parenterally and orally iron-overloaded rats, regardless of selenium status. In the liver and intestine, GSHPx activity was significantly higher in all selenium-supplemented groups, compared to Se-deficient groups (p<0.01) and lipid peroxidation was significantly enhanced in both parenterally and orally iron-overloaded groups, compared to iron-adequate groups. There was no significant difference in cytochrome P-450 activity in the livers between groups in both experiment one and two. These results indicated that GSHPx activity in liver and intestinal mucosa was depended on selenium status, regardless of iron status, and iron-overload enhances lipid peroxidation in liver and intestinal mucosa by increasing the tissue iron content.

나이에 따른 흰쥐의 혈액, 간, 뇌조직의 철분함량, 산화 스트레스 지표에 대한 비타민 C 와 비타민 E공급의 역할 (The Role of Vitamin C and Vitamin E Supplementation on Iron Contents and Biomarkers of Oxidative Stress in Blood, Liver and Brain of Aging Rats)

  • The purpose of this study was to investigate the effect of vitamin C and vitamin E supplementation on the iron contents and oxidative stress of the rats. Rats were fed 18g ascorbic acid and 300IU $\alpha$-tocopherol/kg diet, respectively. Rats were sacrificed at 1, 3, 5 and 7 month of age. The blood, liver and brain were selected for the quantitation of iron and malondialdehyde(MDA) contents, glutathione peroxidase(GSHPx), superoxided dismutase(SOD) and catalase(CAT) activity. Iron and MDA contents and GSHPx activities were increased with aging. Vitamin C and Vitamin E supplementation increased iron contents of the plasma. Vitamin C raised iron contents, but vitamin E decreased iron contents of the liver. In the brain vitamin C and vitamin E did not affect the iron level. MDA levels were decreased with vitamin C and vitamin E supplementation in the erythrocyte and liver, and vitamin C supplementation elevated MDA levels in the brain. GSHPx activity was increased with vitamin C and vitamin E supplementation. SOD activities of erythroucyte and brain were not affected with age, but in the liver, SOD activity was raised with age and vitamin C supplementation. Vitamin C and vitamin E supplementation promoted CAT activity of erythroucyte and liver, and CAT activity of brain was eleveated with vitamin addition but was decreaed with vitamin E addition. Vitamin C and vitamin E decreased iron contents of blood plasma, MDA contents of plasma and liver, and CAT activity of erythrocyte. Above results indicated that iron contents and biomarkers of oxidative stress were more affected by age than antioxidant action of vitamin C and vitamin E.

Enhancement of performance and anti-oxidant variables in broiler chicken fed diets containing sub-optimal methionine level with graded concentrations of sulphur and folic acid

  • Objective: An experiment was conducted to determine the effects of supplementing graded concentrations of inorganic sulphur (S) without and with folic acid (FA) in maize-soybean meal diets on performance, slaughter and anti-oxidant variables, immune responses and serum protein fractions in broiler chicken. Methods: Inorganic S was supplemented at 0.05%, 0.10%, 0.15%, and 0.20% alone or in combination with FA (4 mg/kg) in basal diet (BD) containing no supplemental methionine (Met) and FA. A control group was fed with the recommended concentration of Met. Each diet was offered to 10 pens of 5 male broiler chicks (Cobb 400) and fed ad libitum from day 1 to 42. Results: The broilers fed the BD had lower body weight gain (BWG), feed efficiency (FE), higher lipid peroxidation (LP), lower activity of glutathione peroxidase (GSHPx), lower lymphocyte proliferation ratio (LPR), and reduced concentrations of total protein, albumin, and globulin in serum. Supplementation of FA and S to the BD improved the BWG (all concentrations of S) and FE (0.20% S) similar to the control group. Similarly, the combination of S and FA significantly improved the concentrations of total protein, albumin, and globulin in serum, reduced the LP and increased the activity of GSHPx and LPR. However, responses in the above parameters were related to the concentration of S in the diet. The slaughter variables and antibody titres against the Newcastle disease were not affected with the treatments. Conclusion: Based on the results, it is concluded that the combination of S (0.2%) and FA (4 mg/kg) improved the BWG and FE, similarly supplementation of these nutrients improved the concentration of protein fractions and reduced the stress (reduced LP and improved GSHPx) variables in serum and improved the cell mediated immune response (LPR) in broilers fed sub-optimal concentrations of Met in diet.

  • 해양오염의 진단을 위한 생화학적 오염지표 설정의 기초연구의 일환으로서, 오염이 심각한 서해 (또는 황해)산 넙치(Paralichthys olivaceus)에 이어 서해산 도다리 (Pleuronichthys cornutus)의 혈액 및 뇌중의 활성산소종 및 그들의 제거효소의 활성을 동해안의 포항의 자연산 도다리를 대조군으로 하여 분석 평가하였다. 이들 넙치의 혈청중의 단백질의 함량은 해역이나 서식환경에 따라 뚜렷한 차이를 발견할 수 없었지만, 서해안 자연산 도다리의 뇌 및 근육중의 단백질의 함량은 동해안의 포항산 도다리 대비 뇌는 $15\~45\%$, 근육은 $35\~45\%$나 현저히 감소되고 있었다. 서해안의 자연산 도다리의 혈청중의 과산화지질 (MDA)의 함량은 포항의 자연산 도다리의 대조군 대비 $30\~70\%$나 현저히 높았다. 서해안의 자연산 도다리의 혈청중의 히드록시 라디칼의 생성이 동해안 포항의 자연산 도다리의 혈청중의 히드록시 라디칼의 생성 대비 $15\~90\%$ 정도나 유의적으로 높았다. 서해안의 자연산 도다리의 혈청중의 SOD의 활성은 동해안 포항의 자연산 도다리의 혈청중의 SOD의 활성 대비 $20\~40\%$나 SOD의 활성이 유의적으로 낮았을뿐만 아니라 서해안의 자연산 도다리의 뇌중의 GSHPx의 활성도 동해안 포항의 자연산 도다리의 뇌중의 GSHPx의 활성 $10\~60\%$정도나 유의적으로 낮았다. 이러한 사실도 도다리의 SOD의 활성과 마찬가지로 해양오염에 의해 활성산소의 제거효소로서 생체의 방어효소가 그 만큼 활성이 저하되고 있다는 사실을 입증하고 있다. 이상의 실험결과에서 볼 때 뇌나 근육중의 단백질 함량의 감소, 혈청중의 SOD 및 뇌중의 GSHPx 활성의 저하, 그리고 혈청중의 과산화지질의 함량이 서해안의 도다리가 동해안의 대조군 대비 유의적인 변화를 나타낸다는 사실은 생화학적 오염지표로서의 가능성을 검토할 가치가 있다고 판단된다.
