Some theoretical and experimental aspects of a new electrodynamic separator

  • A power operated (0.5 hp electric motor) grain flour separator was designed and developed for separation of grain (wheat, corn, chickpea and soybean) flour it no various fractions based on the size of the particles of the product. The separator is made of mild steel and consists of a hopper, power driven agitating mechanism, feed control , cylindrical separator unit and an eccentric mechanism. The machine was tested for wheat (variety : Subjata) flour separation into four fraction, viz : semolina ; Gr-I and II, flour (coarse) and white (fine) flour. Wheat samples (6.8% m.c., db) were first pearled by CIAE pearler for 15.8% bran removal . The product and machine characteristics were determined at different capacities varying from 24 kg/h to 143 kg/h. It was found that 76 kg/h capacity gave reasonably best results in terms of purity and recovery of semolina vis-a-vis the market product. The energy requirement of the machine at no-load was found to be 230 w and at load conditio s, it varied between 36.3-6.4kj per kg of feed separation. The machine could be used by small flour millers, small/medium size traders and retailers and other processors for making available various flour products of different particle size in the market for ready use fo the consumers.

Seed Storage Method and Seed Germination Characteristics of Synurus deltoides(Ait.) Nakai (수리취의 종자저장 방법과 발아특성)

  • This study was conducted to investigate the seed storage method and seed germination characteristics of Synurus deltoides (Ait.) Nakai, a kind of wild vegetables. Several parameters affecting seed germination, such as germination temperature, chilling duration, imbibition duration, and storage method were evaluated. The seed germination of Synurus deltoides showed the highest germination rate (GR), promptness index (PI), germination performance index (GPI), mean daily germination (MDG) at 25℃. This implies that the optimum germination temperature was 25℃. And also, Seeds stored at -4℃, after 150 days showed relatively high germination rate about 98%. Therefore, The optimum germination temperature of Synurus deltoides was 25℃ and to increase the germination rate over 98% of Synurus deltoides, cold-wet storage 20 days treatments at 4℃ were effective in the improvement of seed germination.

  • 1. Preparation of Titanium tetrachloride; The following precesses were strictly followed as the preliminary step to obtain pure $TiOCl_2$, titanyl chloride; First, pure Titanium Oxide mixed with carbon is rolled into pills. After drying up perfectly, these pills are heated at 900∼1000${\circ}C$. And then the pills are subjected to the flow of $Cl_2$ gas in a quartz tube heated to 900-1000${\circ}C$. Thus Titanium tetrachloride is obtained. 2. Preparation of $TiOCl_2$ ; Yellowish trobrown solution is made by pouring 80 g of conc. HCl (sp.gr. 1.19) to 45 gr of Titanium tetrachloride (approx. 2 times of theoretical amount). Then this solution is kept settled for 5-days in a desiccator filled with phosphorous pentoxide at room temperature. As the colorless amorphous solid thus obtained is washed with aceton, 36.5 g of the pure salt are obtained. 3. Determination of composition. The analysis of the sample taken from the deposit desiccated gives the following data; (A) Qualitative analysis; a) $Ti(OH)_4$ is precipitated by adding NaOH in water solution of the salt. b) Adding $AgNO_3$ solution, the water solution of the salt gives white precipitate of AgCl. c) When acid and $H_2O_2$ are added, the solution turns its color to redish brown (This proves that $TiO^{++}$ was converted into $TiO^{++}$ by oxidation of $H_2O_2$. (B) Quantitative analysis; a) $Ti(OH)_4$ precipitated by $10{\%}$ NaOH isalitatsubjected consecutively to the filtration and ignition in porcelain crucible at approx. 1000${\circ}C$. , then $TiO_2$ thus formed is weighed and calculated into Ti content. b) Chlorine involved in water solution of the salt is determined by Vorhardt method. Result: The values obtained from previous analysis, devied by their atomic weight gives the following composition: Ti : Cl = 1 : 2 Therefore $TiOCl_2$ should be given as its molecular formula. 4. Summary. When $TiCl_4$ is additated into conc. HCl, $TiO^{++}$ formed exists as a stable form, and forms $TiOCl_2$. However $TiOCl_2$ is unstable to heating. When the temperature is raised to $65{\circ}C$the decomposition of the solution is accelerated, and gives $TiO_2$ aq. $TiOCl_2$ in addition is highly hygroscopic.

Comprehensive investigations of key mitochondrial metabolic changes in senescent human fibroblasts

  • There is a paucity of detailed data related to the effect of senescence on the mitochondrial antioxidant capacity and redox state of senescent human cells. Activities of TCA cycle enzymes, respiratory chain complexes, hydrogen peroxide (H2O2), superoxide anions (SA), lipid peroxides (LPO), protein carbonyl content (PCC), thioredoxin reductase 2 (TrxR2), superoxide dismutase 2 (SOD2), glutathione peroxidase 1 (GPx1), glutathione reductase (GR), reduced glutathione (GSH), and oxidized glutathione (GSSG), along with levels of nicotinamide cofactors and ATP content were measured in young and senescent human foreskin fibroblasts. Primary and senescent cultures were biochemically identified by monitoring the augmented cellular activities of key glycolytic enzymes including phosphofructokinase, lactate dehydrogenase, and glycogen phosphorylase, and accumulation of H2O2, SA, LPO, PCC, and GSSG. Citrate synthase, aconitase, α-ketoglutarate dehydrogenase, succinate dehydrogenase, malate dehydrogenase, isocitrate dehydrogenase, and complex I-III, II-III, and IV activities were significantly diminished in P25 and P35 cells compared to P5 cells. This was accompanied by significant accumulation of mitochondrial H2O2, SA, LPO, and PCC, along with increased transcriptional and enzymatic activities of TrxR2, SOD2, GPx1, and GR. Notably, the GSH/GSSG ratio was significantly reduced whereas NAD+/NADH and NADP+/NADPH ratios were significantly elevated. Metabolic exhaustion was also evident in senescent cells underscored by the severely diminished ATP/ADP ratio. Profound oxidative stress may contribute, at least in part, to senescence pointing at a potential protective role of antioxidants in aging-associated disease.

Typen und Ursachen der $Ver\"{a}nderungen$ der Sprache und die historischen Entwicklungen des Deutschen (언어변화의 유형과 원인 및 독일어의 역사적인 발전)

  • Das Ziel dieses Aufsatzes liegt darin, Typen und Ursachen der $Ver\"{a}nderungen$ bei den morphologischen Kategorien und den Satz- strukturen zu untersuchen, und die historischen Entwicklungen des Deutschen zusammenfassend darzustellen. In der Sprachentwicklung $m\"{u}ssen$ wir die lebendige Wechselbeziehung zwischen Sprache und Leben beachten. Sprache und Leben sind nicht als zwei verschiedene $Gr\"{o}{\ss}en$ zu betrachten, sondern nur in ihrem gegenseitigen Bedingtsein zu verstehen. Sprache und Leben sind immer in ihren inneren $Zusammenh\"{a}ngen$ zu sehen, $ge\"{a}nderten\;Lebens-\;umst\"{a}nden$ werden sprachliche $Ver\"{a}nderungen$ folgen. Bei den $Ver\"{a}nderungstypen$ wurden einige Ursachen $erw\"{a}hnt$, z. B. die $Abschw\"{a}chung$ der Endungsvokale und die Tendenz der Entwicklung vom synthetischen zum analytischen Satzbau. Eine wirkliche Ursache unter den $Ver\"{a}nderungsfaktoren$ ist der Einfluss der fremden Sprachen auf die deutsche Syntax. Zu den $\"{a}u{\ss}eren$ Ursachen rechnen wir $s\"{a}mliche\;ungew\"{o}hnlich$ verschiedenartigen Impulse, die von der die Sprache umgebenden Umwelt ausgehen. Sprachliche Veranderungen $k\"{o}nnen$ auch innere Ursachen haben. Als innere Ursachen wurden das Prinzip der sprachlichen $\"{O}konomie$, die Tendenz zur emotionalen $Expressivit\"{a}t$, das Streben nach Anschaulichkeit sowie einige $\"{a}sthetisch$ und sozial bedingte $Gr\"{u}nde$ besprochen. In Bezug auf die historischen Entwicklungen des Deutschen wurden die Epochen der Sprachentwicklung in Anlehnung an Hogo Moser(1972) in 5 Epochen, d.h. Vorgeschichte der deutschen Sprache, Altdeutsche Zeit, Mittelalterliches Deutsch, Neudeutsche Zeit und Deutsch der Gegenwart untergliedert, und einige wichtige Entwicklungen des Deutschen in der jeweiligen Epoche zusammengefasst.

Adsorption of methylene blue from an aqueous dyeing solution by use of santa barbara amorphous-15 nanostructure: Kinetic and isotherm studies

  • Santa Barbara Amorphous-15(SBA-15) nanoparticles were utilized as the inexpensive and effective adsorbents to remove methylene blue dye from the aqueous solution.SBA-15 was created by Zhao et al method. Infrared spectroscopy, X-ray diffraction and scanning electron microscopy (SEM) were used for the evaluated physical properties of SBA-15. The results of diffraction X-ray indicated that was the crystalline structure for it. Also IR spectroscopy indicated was a silica the whole structure of the groups and SEM image verify the structure of relatively identical particles size of SBA-15. Factors affecting adsorption including the amounts of adsorbent, pH and contact time were investigated by a SBA-15 nanomaterial design. The extent of dye removal enhanced with increasing initial dye concentration and pH from 4 to 10. The higher percentage adsorption were obtained under optimum conditions of variables (sorbent dose of 200 mg/liter, initial MB concentration 10 mg/liter, initial pH of 10 and temperature of $25^{\circ}C$). Maximum adsorption happened after the 2 hour and the kinetic processes of the dyes adsorption were described by applying the pseudo-first-order and the pseudo-second-order and the relatively High correlation with the kinetic Ellovich models. It was found that the pseudo-second-order models kinetic equation described the data of dye adsorption with a good correlation (R2>0.999) which indicated chemisorption mechanism. Freundlich and Langmuir adsorption models were investigated in conditions of variables (adsorbent dose 0.01 gr/liter, MB concentration 10, 20, 30 mg/liter, pH of 4, 7, 10, contact time 90 min and temperature of $27^{\circ}C$). The adsorption data were represented by Langmuir isotherm model. These values are higher than the adsorption capacities of some other adsorbents that have recently been published in the literature.

An Investigation of the Control of Two-Spotted Spider Mites (Tetranychus urticae Koch) Resistant to Organo-Phosphates (유기인제저항성 점박이응애(Tetranychus urticae Koch)에 관한 연구)

  • The study involved determination of resistance levels of spider mites ta argano-phosphates using topical application and slide dip techniques; laboratory serening tests of alternative acaricides using an O/P resistant strain and a field trial of the screened materials. 1. Strains of Tetranychus were from Timaru(TR), Havelock Narth (HNR), Lincaln (LN). Germany (GR, GN). Comparisons of the resistant strains and normal strains at the LD50 and LC50 levels were as follows : (a) Using the topical application tochnique; with Parathian. resistant levels of the GR. TR and HNR strains of T. urticae were respeativuly, 1035. 484 and 452 times as resistant' as the LN strain. (b) Using the slide dip technique; with Phosdrin, resistant of GR, TR and HNR strains of T. urticae were 635, 274 and 266 times greater respeativuly, than the GN strain. 2. The laboratory sereaning tests were carried out far their contact plus stomach and residual effect to assess the toxicities of eleven alternative materials which would be used far control of O/P resistant strain of T. urticae. The acaricide groups represented were 3 organo-chlorines (Spidex, Kelthane and C 8514), 2 nitrophenyls (UC 19786 and Morocide), 2 cyclic carbonates(Eradex and Morestan). I carbamate (UC2004 7A), 1 mixture of carbamate and orano-chlorine and 2 other chemicals (C 8677 and M2527). From all acaricide tested. Kelthane and Morocide were the most effective, folowed by Spidex and M2527. Morestan, C8514. C8677 and RS 143 were intermediate, but Eradex, UC 19786 and UC 20046A were poor. 3, The number of sapmles required for estimation of the population in the field evaluation of acaricidal effects was one giving the highest practical precision. It was decided, after preliminary sampling trials. to use samples of 30 leaves per replicate which gave a $5.7\%$ standard error. 4. In the field trials, Morocide applied at the $0.05\%\;and\; 0.04\%$ a. i. conc. to black currant trees gave excellent control of O/P resistant population of T. urticae for about 12 days, but Morocide 0.025 and Kel thane $0.02\%$ a. i. cone. gave efficient control for about 6 days. In other words. first applications of Kel thane ane Moroeide gave very high degrees of control of O/P resistant population of the two-spotted spider mite. However, the results indicate that secondary application would sometimes be necessary. There was no foliage damage of black Currants and strawberries by either acaricides at the concentrations used. Use of CGF and CCW as Medium Components for Commercial Production of Plug Seedlings of Seogun Tomato (토마토 공정묘의 상업적 생산을 위한 배지구성물질로서 발포유리와 탄화 밤나무 칩의 이용)

  • This study was conducted in a commercial plug glasshouse in Sacheon to examine the possibility of producing tomato plug seedlings using various growing media containing cellular glass foam (CGF)and carbonized chestnut woodchips (CCW) as medium components. Plug seedlings of 'Seogun' tomato were grown in media containing 50% CCW+50% peatmoss, 33% CGF+67% peatmoss, and 50% peatmoss+50% granular rockwool. A commercial plug medium (Tosilee) was used as the control. All seeds were sown in 200 cell plug trays on November 28, 2001. Seedling growth was measured at 31 days after sowing. Each treatment showed a similar growth result as compared to the control. Plant height, root grade, fresh weight, and air space and bulk density of the medium were significantly greater in the 33% CGF+67% peatmoss treatment than those in the other media. However, growth was slightly suppressed in the 50% CCW+50% peatmoss. pH and EC of the media were the highest in the control treatment, although no toxicity symptoms had been observed. The results suggest that perlite can be replaced with a new material such as CGF in the commercial scale production of plug seedlings of 'Seogun' tomato.

The Effects of Dictamni Radicis Cortex Extracts on DNCB Induced Atopic Dermatitis-like Skin Lesions of NC/Nga Mouse (백선피(白鮮皮) 추출물이 DNCB로 유발된 NC/Nga 생쥐의 아토피 피부염에 미치는 영향)

  • Objectives Dictamni Radicis Cortex extracts (DRC) has been known to suppress allergic reaction, however the cellular target of DRC and its mode of action remain unclear. The purpose of this study is to investigate the effects of Dictamni Radicis Cortex extracts on DNCB induced atopic dermatitis-like skin lesions of NC/Nga mouse. Methods This study was designed to investigate the effects of DRC extract in the DNP-IgE-induced activation of MC/9 murine mast cell lines in vitro and in the DNCB-induced activation of NC/Nga mouse in vivo. For this investigation, We examined IL-4, IL-5, IL-6, IL-13, TNF-${\alpha}$ and GM-CSF mRNA expression by Real-time PCR, IL-13, MIP-$1{\alpha}$ production by ELISA analysis and manifestations of NFAT1, NFAT2, AP-1 and NF-${\kappa}B$ p65 transcription factors by western blotting in vitro. Then, we examined WBC, eosinophil and neutrophil in NC/Nga mouse, IL-5, IL-13 in serum, IFN-${\gamma}$, IL-4 in the spleenocyte culture supernatant, the absolute cell numbers of $CD4^+$, $CD8^+$, $^+Gr-1^+CD11b$, $B220^+CD23^+$ in the ALN, PBMCs and dorsal skin, IL-5, IL-13 in the dorsal skin by Real-time PCR and the distribution of mast cells by H&E and toluidine blue. Results In vitro the mRNA expression of IL-4, IL-5, IL-6, IL-13, TNF-${\alpha}$, GM-CSF and IL-13, MIP-$1{\alpha}$ production by ELISA analysis were completely abolished by DRC and the western blot analysis decreased the expression of mast cell-specific transcription factors including NFAT-1, NF-${\kappa}B$ p65. In vivo DRC oral adminstration also decreased the counts of WBC, eosinophils and inflammatory cytokines such as IL-13 and IgE in the serum. DRC oral adminstration elevated IL-4 level in the spleenocyte culture supernatant. DRC oral adminstration decreased total ALN cells, total skin cells, cell numbers of $CD4^+$, $B220^+CD23^+$ in the ALN, $^+Gr-1^+CD11b$ in the PBMCs and $CD4^+$, $CD8^+$ in the dorsal skin. The mRNA expression of IL-5, IL-13, thickness of epidermis, inflammation immune cells and mast cells were abolished by DRC in the dorsal skin. Conclusions Histological examination showed that infiltration levels of immune cells in the skin of AD-induced NC/Nga mouse were much improved by DRC oral adminstration. These results, therefore, suggest that DRC can regulate molecular mediators and immune cells that are functionally associated with atopic dermatitis induced in NC/Nga mouse, and may play an important role in recovering AD symptoms.

Therapeutic Effects of Yijungtang on Atopic Dermatitis-like Skin Lesions of NC/Nga Mouse Induced by Mite Antigen (이중탕(理中湯)이 Mite Antigen으로 유발된 NC/Nga 생쥐의 아토피 피부염에 미치는 영향)

  • Objectives: The purpose of this study is to investigate the effects of Yijungtang(YJT) on atopic dermatitis in an in-vitro and in-vivo experiment using a RBL-2H3 mast cells and a NC/Nga atopic dermatitis mouse. Methods: In-vitro experiment, IL-4, IL-13 mRNA expression were evaluated by a real-time PCR, IL-4, IL-13 production by ELISA and transcription factor as GATA-1, GATA-2, NF-AT1, NF-AT2, AP-1 and NF-kB by western blotting. In-vivo experiment, clinical skin score we evaluated by, hematology and Serum total IgE and IgG1 of NC/Nga atopic dermatitis mouse, cytokine level, total number of cell, Immunohistochemical staining and Histological features of auxiliary lymph node(ALN), draining lymph node(DLN), peripheral blood mononuclear cells(PBMCs) and dorsal skin tissue in NC/Nga mouse. Results: YJT decreased IL-4, IL-13 mRNA expression, IL-4, IL-13 production and prominently decreased the expression of mast cell specific transcription factors including GATA-2, NF-AT2, c-Fos and NF-kB. YJT oral administration reduced the levels of skin severity scores. It also decreased the level of inflammatory cytokines such as IL-5, IL-13, histamine and IgE in the serum. It elevated IFN-gamma level in the spleenocyte culture supernatant but decreased. $CD3e^+$, $CD19^+$, $CD4^+$, $CD8^+$, $CD3e^+CD69^+$, $CD11b^+Gr-1^+$, $CCR3^+$ in the PBMCs, $CD4^+$, $CD8^+$, $CD3e^+CD69^+$, $B220^+CD23^+$ in the ALN, $CD4^+$, $CD3e^+CD69^+$ in the ALN and $CD4^+$, $CD11b^+Gr-1^+$ in the dorsal skin. Histological examination showed that infiltration levels of immune cells in the skin of AD-induced NC/Nga mice were much improved by YJT oral administration. Conclusions: The anti-allergic activities of YJT may be mediated by down-regulation of Th2 cytokines, such as IL-4 and IL-13, through the regulation GATA-2, NF-AT2 and NF-kB transcription factors in mast cells. YJT would be regulate molecular mediators and immune cells which are functionally associated with atopic dermatitis induced in NC/Nga mice, and may play an important role in recovering AD symptoms.
