Effects of Aralia elata extract on experimentally Alloxan Induced Diabetes in Rabbits (두릅나무추출물(抽出物)이 Alloxan으로 유발(誘發)된 가토(家兎)의 고혈당(高血糖)에 미치는 영향(影響))

  • Effect of Aralia elata extract on experimentally alloxan-induced diabetes ill rabbits and toxicity of mouse were studied; $LD_50$ of Aralia elata extract (A. E. E.) was 5.5g/kg intraperitoneally ill mice. A. E. E. 800mg/kg exhibited more excellent hypoglycemic effect after 6 days and 14 days in 200mg/kg, compared to the control group. Blood urea nitrogen level was significantly decreased after 6 days in 800mg/kg. and blood levels of total cholesterol and GPT activity were showed more significant decreasing effect ill 800mg/kg than 200mg/kg.

Effect of Prunus mume Extract on Gastric Secretion in Rats and Carbon Tetrachloride Induced Liver Damage of Rabbits (매실추출물이 흰쥐의 위액분비 및 사염화탄소로 유발시킨 가토의 간장장애에 미치는 영향)

  • The aim of this study is to investigate effects of Prunus mume extract on gastric secretion in rats and experimentally carbon tetrachloride-induced liver damage of rabbits. The groups of Prunus mume extract showed excellent increasing effect in total acidity and peptic activity and decreasing effect on gastric volume than the control group whereas clear differences were not found between sample adminstered group and control group in gastric pH. In liver function test Prunus mume extract exhibited rapid recuperation of liver function that is sGPT activity showed an apparant decreasing effect from the 6th day total cholesterol and alkaline phosphatase level from the 10th day respectively compared to the control group in carbon tetrachloride-intoxicated rabbits. It is, is suggested that Pruns mume extract can be used as the stomachics to promote the gastric digestion by increasing the peptic activity and health food to shorten the recovery time from liver diseases.

Effects of Ixeris Sonchifolia H. Fiet on Lipid Metabolism and Liver Function of Rats Administered with Ethanol (고들빼기 첨가 식이가 알콜투여 흰쥐의 지방대사와 간기능에 미치는 영향)

  • To investigate the effects of Ixeris sonchifolia Hance diets on serum and hepatic lipid levles and enzyme activities in rats administered with ethanol chronically, Sprague-Dawley male rats were AIN-76 diet(control), control diet plus ethanol, control plus Ixeris sonchifolia Hance diet, or control plus Ixeris sonchifolia Hance diet plus ethanol for 30 days. Ixeris sonchifolia Hance diets significantly decreased the serum total cholesterol, triglyceride, LDL-cholesterol, and GOT levels that were increased due to the chronic ethanol administration. In addition, Ixeris sonchifolia Hance diets significantly decreased the liver triglyceride and total lipid levels that were increased due to the ethanol administration. The present findings, combined with previous data showing differences in the effects of cabbage diets having a high or a low level of GABA on the lipid levels and the serum Y-GPT activity of rats(Cha and OH[2000] J. Korean Soc. Food Sci. Nutr. 29, 500-505) raise the possibility that GABA in plants could have a nytraceutical role in the recovery of chronic alcohol-related diseases.   • Ketoconazole is an imidazole antifungal agent which inhibits the biosynthesis of fungal cellmembrane ergosterol and has immunosuppressive properties in vitro. Biphenyl dimethyl dicarboxylate (PMC) has been utilized for antioxidative action and for liver-protective purposes. Studies were undertaken to investigate effects of biphenyl dimethyl dicarboxylate (PMC) on the immunosuppression of ketoconazole in ICR mice. In the combination of PMC and ketoconazole, as compared with the treatment of ketoconazole alone, there were significant increases in activities of natural killer (NK) cells and phagocytes along with circulation leukocytes. The elevation of serum glutamic-pyruvic transaminase (S-GPT) and total protein levels caused by ketoconazole were reduced by the combination of PMC and ketoconazole. In addition, lower serum albumin and albumin/globulin (A/G) ratio were also increased to normal level.

  • Based on equal number of cells, supplementation of 10$^{-6}$ M cadimium highly simulated the intracellular amylase. GCT, LDH as well as the glucose and erea content of E carotovora var, carotovaro cells. This was coupled with initiation of highly active GOT, CPK as well as accumulation of cholesterol in the cells. Lanthanum was less active and unable to initiate COT or CPK. Nickel was almost without effect though reduced LDH activity without initiating either enzyme or cholesterol production. Similar stiulations and/or initiations were observed, though to variable extents, when the same concentration of the three elements were supplied to E. carotovora var, citullis or E. toxica. The highest yield of amylase, GPT, GGT or glucose was obtained when E. carotovora var. carotovora was supplemented with Cd + Ni. The highest urea level was recorded in Erwinia carotovara var, cirullis, amended with Cd + La.

Antitumor Effects of Glycoportein Extracted from Sea Cucumber (Stichopus japonicus)

  • The anititumor and immunologic activities of the glycoproteins extracted from sea cucumber (Stichopus japonicus) on mice bearing sarcoma 180 cells were investigated . Maximum tumor suppression (64%) occurred at the dose of 100mg glycoprotein/kg. The highest prolongation ratio was achieved at the level of 100mg/kg an dincreased by 395 more than that of control. Glycoproteins from sea cucumber exhibited direct cytotoxic effect on the tumor cells. Dose dependent increase of leucocyte, peritoneal exudate cell and weights of immunoorgans revealed the improvement of immunity. When the glycoportein-administered group was compared with the control, a significant difference was not noted in the clinico-chemical values such as S-GOT, S-GPT , alkaline phoshatse activity, total protein, cholesterol, triglyceride, urea nitrogen and glucose levels in blood. These results suggests that the antitumor activity of sea cucumber glycoprotein is associated with activation of cells in the immune system.

  • Aminolysis, hydrazinolysis and alkylation of 4-methoxy and 4,9-dimethoxy-6-cyano-7-thione-5-methyl-7H furo [3,2-g] [1] benzopyridine (1 a-b) yielded 7N-substituted furobenzopyridine derivatives (2 a-e or the possible isomers 3 a-e and 4 a-b), (5 a,b and 6 a,b) and the ester (8 a,b). Hydrolysis of (la) with acetic acid gave the corresponding pyridone derivatives (7). Furobenzopyridinyl-7-thioacetyl hydrazide (9 a,b) have been prepared via alkylation of furobenzopyridine thione (1 a-b) with ethyl chloroacetate followed by condensation with hydrazine hydrate. Schiff base (11) was prepared by reacting (9a) with p. N,N-dimethyl aminobenzaldehyde in boiling ethanol. Treatment of (8a) with anthranilic acid gave the corresponding 7-substituted-4H-3,1-benzoxazine-4-one (10). We found that compound (11) increased bleeding, coagulating time, the total count of white blood cells, blood glucose level (cause hyperglycemia), enzymes (GOT, GPT) activities, concentration of urea and creatinine. On the other hand it decreased red blood cells number, haemoglobin content and haematocrite value.

  • Objectives : The purpose of this study is to observe the anti-oxidative effects of electroacupuncture at Taegye($KI_3$) in rats. Methods : The methods are measures of experimental items including change of body weight, relative liver weight, level of alblumin, total bilirubin, LDL-cholesterol, LDH, GOT and GPT in blood serum, level of SOD, GSH, catalase, NO and MDA concentration in liver, histological change of liver. Results: 1. The $KI_3$-EA group lost weight in the beginning, and gained it gradually. 2. The liver ratio of the $KI_3$-EA group increased compared to the holder group, without any statistical significance. 3. The SOD activity, Glutathione concentration, and Catalase concentration increased, and the NO concentration and MDA concentration decreased in the $KI_3$-EA group significantly, compared to the holder group. 4. $KI_3$-EA group had more similar density to the normal group, than the holder group. Conclusion : The electroacupuncture application on Kb has anti-oxidation effect, by improving the body defence against the oxidation stress- induced cytotoxicity, and by supressing the oxidative liver damage.

  • The Hypoglycemic effect of exo-polymers (EPs) produced from submerged mycelial cultures of five types of mushrooms on streptozotocin (STZ)-induced diabetic rats were investigated in this study. The five experimental groups were fed with EPs (50 mg/kg body weight) for 7 days. Significant reduction in plasma glucose, total cholesterol (TC), and triglyceride (TG) levels were observed in rats fed with Lentinus edodes and Cordyceps militaris EPs. Plasma glucose and TC were also reduced by administration of Phellinus linteus EPs, but the TG level was not changed significantly. The EPs of three mushroom species also demonstrated a marked reduction in the level of plasma glutamate-pyruvate transminase (GPT). The result proves the hypoglycemic activity of EPs of three fungal group in STZ-induced diabetic rats and indicates their potential in the control of diabetes mellitus.

  • The effects of fruiting body of Cordyceps militaris on the growth, the food intake, the food efficiency ratio, the lipid metabolism, the serum protein level ad enzyme activity in male rats were studied. Sprague-Dawley rats were fed four types of diets for five weeks, respectiely: a control diet, a control diet supplemented with 2%, 3% or 4% fruiting body of Cordyceps militaris(CF) powder. In rats fed 2% or 3% CF diets the body weight gain, the food intake, the concentratons of hepatic triglyceride and serum LDL-cholesterol, the atherogenic index, and the total lipid, total cholesterol, triglyceride and phospholipid in serum were similar to those in rats fed the control diet. Whereas, in 4% CF diet these were significantly decrased. But the all CF diets feeding could not decrease the food efficiency ratio, the weights of liver, pancreas, spleen, kidney and heart, and the concentration of serum HDL-cholesterol. Also, it was shown that the concentrations of glucose, total protein, albumin, urea and creatinine, and the GOT, GPT, LDH, ${\gamma}$-GTP and ALP activities were the same levels in serum of rats of fed all the experimental diets.
