CHANGES IN SOME PHYSIOLOGICAL PARAMETERS OF ALBINO RATS AT DIFFERENT AMBIENT TEMPERATURES

  • Five experimental groups with five adult male rats in each, were exposed to 20, 35, 40 and $45^{\circ}C$ air temperature for 50-70 minutes, and to $50^{\circ}C$ for 30-50 minutes, respectively. Food and drinking water were not permitted during the exposure. Blood samples were obtained by heart puncture immediately after the thermal treatment. All the rats were hyperthermic (p<0.01) as compared to the controls ($20^{\circ}C$). Hyperthermia was associated with hypoglycemia which was significant (p<0.01) at 45 and $50^{\circ}C$ exposures. Plasma levels of GOT and GPT declined at 35 and $40^{\circ}C$ reaching the lowest (p<0.05) level at $45^{\circ}C$, while at $50^{\circ}C$ GOT level was elevated by 45% but GPT was normal as compared to the controls. Differences between groups were significant (p<0.01) for GOT and insignificant for GPT. Hematocrit value increased significantly (p<0.01) at 45 and $50^{\circ}C$, indicating hemoconcentration. It could be concluded that severe heat stress (45 and $50^{\circ}C$) resulted in critical hyperthermia, hypoglycemia, disturbed liver function, body dehydration, and hemoconcentration leading to death.

The study of the change in the blood when acupuncture points are stimulated or when doing aerobic exercises (유산소운동과 경혈점 자극후 혈청내 변화에 대한 비교 연구)

  • The purpose of this study is to find out what effects aerobic exercises and acupuncture point stimuli have on the blood. For this purpose, we reviewed 10 papers on aerobic exercises and 10 papers on the stimuli to acupuncture points and compared the changes of HDL-C, seroenzyme GOT and GPT. The results showed that HDL-C increased significantly after aerobic exercises; more so than before as stated in the papers on aerobic exercises. On the other hand, HDL-C decreased significantly after acupuncture point stimuli; more so than before as stated in the papers on the stimuli to acupuncture points. Seroenzyme GOT increased more significantly after aerobic exercises than shown before in the papers on aerobic exercises. However, there was a more significant decrease after the stimulus than shown before in the papers on acupuncture points stimuli. Seroenzyme GPT increased more significantly after aerobic exercises than shown before in the papers on aerobic exercises. There were no significant differences before and after on, the acupuncture points stimuli, according to those papers. Therefore, as HDL-C increased significantly when doing aerobic exercises, aerobic exercises prove to be a more effective treatment method than stimuli to the acupuncture points for patients with hyperlipidemia. However, from the facts that seroenzyme GOT and GPT increased significantly after aerobic exercises, we find that aerobic exercises may be more of a burden on the tissues of the liver, skeletal muscles, stomach, etc. than the stimuli to acupuncture points.

Effect of Ganoderma lucidum Extract on Experimentally Induced Hepatic Damage and Hyperlipemic Rats. (영지(靈芝)엑기스가 백서(白鼠)의 실험적(實驗的) 간장중독(肝臟中毒) 및 고지혈증(高脂血症)에 미치는 영향(影響))

  • This study was attempted to investigate the effect of Ganoderma lucidum (Young-Jii) extract on the activities of GPT GOT Al. P LDH and the level of total bilirubin and total cholesterol in serum of $CCl_4$-intoxicated rats, and on the level of total lipids triglyceride phospholipids and total cholesterol in the serum of experimentally induced hyperlipemic rats, and on the effect of body and liver weight in rats. The results were shown as follows; In $CCl_4$-intoxicated rats, the extract showed a significant decrease in the activities of GPT and Al.P, a slight decrease in the activity of GOT and LDH; The level of total bilirubin was slightly affected, but significantly decreased at a dose as high as 500 mg/kg; the level of total cholesterol was increased dose dependently. In hyperlipemic rats, the extract caused a significant decrease in the level of total lipids and triglyceride and the rate of decrease was more pronounced with repeated treatments for 10 days; the level of phospholipids and total cholesterol were slightly decreased with repeated treatment of the extract at a dose of 300 mg/kg for 10 days; A significant body weight gain was shown with the treatment of the extract.

  • The effects of ethanol extracts from Asparagus cochinchinensis, Polggonatum officinale, Poria cocos, Panax ginseng and Acanthopanax sessiliflorum on the toxicity of polychlorinated biphenyls(PCBs) were examined and following results were obtained. By the administration of these extracts, PCBs intoxicated rats showed the normalizing tendency in body weights and various hematological parameters-counts of red blood cell and white blood cell, hemoglobin content and hematocrit value. These extracts ameliorated the elevated levels of serum cholesterol, total lipids, $_{8}-GOT-,\;_{8}-GPT$ and s-alkaline phosphatase activities of PCBs intoxicated rats.

Protective Effects of the Phenolic Compounds from the Leaves of Hedera rhombea on Hepatic Injury (송악의 페놀성 물질의 간 보호효과)

  • Hedera rhombea (HR) has been used for treatments of hemorrage, chronic catarrh, jaundice, lithisis and convulsion. This study was done to isolate active compounds that have protective effect on liver damage. BuOH and EtOAc fractions of HR recovered serum glutamic pyruvic transaminase (GPT), glutamic oxaloacetic transaminase (GOT) and ${\gamma}-glutamyltranspeptidase$ (${\gamma}-GTP$) activities in CCl4 treated rats. We isolated 7 phenolic compounds from BuOH and EtOAc fractions, which were identified as 3-caffeoyl quinic acid, 3,4-di-O-caffeoyl quinic acid, 3,5-di-O-caffeoyl quinic acid, 4,5-di-O-caffeoyl quinic acid, caffeic acid, methyl 3,4-di-O-caffeoyl quinic acid and methyl 3,5-di-O-caffeoyl quinic acid by chemical and spectral analysis. These compounds reduced significantly serum GOT and GPT elevated by CCl4 treatment in rats, and 3-caffeoyl quinic acid, 3,5-di-O-caffeoyl quinic acid and caffeic acid also showed mild inhibitory activity against human immunodeficiency virus.

Effects of Raw Soy Flour(yellow and black) on Serum Protein Concentrations and Enzyme Activity in Streptozotocin-Diabetic Rats (대두급여가 당뇨쥐의 혈청과 간의 단백질농도 및 효소활성에 미치는 영향)

  • Effects of raw soy flour(RSY) and black(RSB) feeding on protein concentration of liver and serum, and GOT, GPT and alkaline phosphatase activities of serum in diabetic and nondiabetic rats were studied. Male rats(Sprague-Dawley), mean weight of (338.4$\pm$19.2g) were assigned to six dietary groups and fed with the assigned diet for 28 days. For each experimental, some rats were injected with streptozotocin intraperitoneally(L.P.) to induce diabets, and other rats were injected with buffer L.P. as a control group. The liver, kidney and spleen weights relative to bo요 weigth were higher in raw yellow soy flour diet diabetes(D-RSY) and black soy flour diet diabetes(D-RSB) groups than control, but the body weights were lower than control. The protein and albumin concentrations of liver and serum were lower in D-RSY and D-RSB groups than control. The albumin concentration of serum in D-RSB group was lower than control. The GOT activities of serum in RSY and RSB groups were increased compared with control, but the GPT activities were lower in diabetic control, D-RSY and D-RSB groups than control. The alkaline phosphatase activities of serum in RSY and RSB groups were higher than control, but those in D-RSY and D-RSB groups were lower than diabetic control.

Protective Effects of Galangin on Carbon tetrachloride-Induced Hepatotoxicity (사염화탄소에 의해 유발된 간독성에 대한 galangin의 보호효과)

  • In order to evaluate the protective effects of galangin on $CCl_4$-induced hepatotoxicity, GOT, GPT and malondialdehyde(MDA) values were measured in ICR mice. Galangin,a flavonoid compound, was administered orally for six days and immediately $CCl_4$ was injected intraperitoneally after the last dose of galangin. Mice were sacrificed at 24h after the administration of $CCl_4$. In the multiple pre-treatments for 6 consecutive days, galangin showed more potent protective effects than silymarin as reference active compound in serum GOT, GPT and MDA values in the liver at all doses tested. Antioxidative activity was determined by measuring the amounts of MDA formed from ethyl linoleate by $H_2O_2$ in vitro. Galangin showed higher inhibition than silymarin. These results demonstrate a possible hepato-protective role of galangin against $CCl_4$-induced hepatotoxicity in vivo and $H_2O_2$-induced lipid peroxidation in vitro. Therefore, galangin may be capable of protecting hepatotoxicity.

The Effect of HP4060 on the Brain Neurotransmitter and Hot Flushes in the Female Sprague-Dawley Rat

  • In this study, the effects of HP4060, a pomegranate extract, on Sprague-Dawley (SD) female rats were investigated. SD rats used in the experiment were divided into 3 groups: a control group, 100 mg HP4060/kg rat powder fed group, and 25 mg HP4060/kg rat liquid fed group. After 20 days of administration, the changes of the brain neurotransmitters were measured. The data showed that the concentration of the serotonin and the norepinephrine were increased, whereas that of the epinephrine was decreased in HP4060 administered groups. In addition, the improving effect of HP4060 on depression symptom of menopause women were shown by increased immobility time of the SD rates in a separate experiment. The uterus weight of HP4060 fed groups were also shown to be increased. In order to monitor toxic effect of HP4060, glutamic-oxaloacetic transaminase (GOT) and glutamic-pyruvic transaminase (GPT) levels were measured, and the results showed that no significant difference in GOT and GPT levels among experimental groups implying no significant toxic side effects of HP4060. According to these results, it seems clear that HP4060 may improve symptoms of hot flush and depression caused by menopause without significant level of toxic effects.

Case of Oriental Obesity Treatment's Effect on Improvement of Nonalcoholic Steatohepatitis Patient's Liver Function (한방비만치료를 통한 비알코올성 지방간염 의증 환자의 간기능 개선 1례 보고)

  • Nonalcoholic steatohepatitis(NASH) may progress to advanced liver disease. The diagnosis is made on liver biopsy when investigating a patient with raised transaminases and an otherwise negative biochemical and serological work-up. The subject was a obese male patient who had unexplained raised GOT, GPT. He had no alcoholic consumption and drug ingested. On serological examination, HBsAg and Anti-HCV test are negative. The subject was diagnosed as NASH, and was treated with oriental treatment for obesity. After 2months treatment the raised GOT, GPT decreased to normal range.

Anti-daibetic Effect of Solani nigri Herba on Diabetic Rat Induced by Streptozotocin (용규(龍葵)가 streptozotocin으로 유발된 백서(白鼠)의 실험적 당뇨에 미치는 영향)

  • The objective of the study was to observe the anti-diabetic effect of Solani nigri Herba(SH). The following experimental results were derived from the measurement of the levels of glucose, GOT and GPT in the serum and weight from a group of experimental rats by way of the injection of streptozotocin as well as the oral administration of Solani nigri Herba water extracts. The glucose level in the serum significantly decreased in the Solani nigri Herba-dosed group(SH2,12.6 mg/100 g rat weight) on the 10th day. The GOT level of SHI(4.2 mg/100 g Solani nigri Herba) and SH2 in the serum slightly decreased, but no significance to compare with control group. The GPT level of SH1 and SH2 in the serum slightly decreased, but no significance to compare with control group. The weight of rat of SH1 and SH2 in the serum slightly increased, but no significance to compare with control group. In conclusion, Solani nigri Herba appeared to have significance inhibitory effects on the rise of blood sugar level in the experimentally induced diabetics.
