Effect of PAH on CYP1B1 Gene Expression (PAH가 CYP1B1 유전자 발현에 미치는 영향)

  • Cytochrome P4501B1(CYP1B1) is known to be inducible by xenobiotic compounds such as policyclic aromatic hydrocarbon(PAH) and dioxins such as 2,3,7,8-tetrachloro-dibenzo-p-dioxin(TCDD). And these induction of CYP1B1 is also regulated by many categories of chemicals. In order to investigate the effects of several chemicals on CYP1B1 gene expression in Hepa-I and MCF-7 cells, 5' flanking DNA of human CYP1B1 was cloned into pGL3 basic vector containing luciferase gene, and then transfected into these cells. After treatment of chemicals, the luciferase activity was measured. CYP1B1 enzyme metabolize PAHs and estradiol. CYP1B1 metabolize estradiol to 4-hydrozyestradiol that is considered as carcinogenic metabolite. Luciferase activity was induced about 20 folds over that control by 1 nM TCDD (2,3,7,8-tetrachloto-p-dioxin). Recent industrialized society, human has been widely been exposed to widespread environmental contaminants such as PAHs(polycyclic aromatic hydrocarbon) that are originated from the imcomplete combustion of hydrocarbons. PAHs are known to be ligands of the AhR(aryl hydrocarbon receptor). Induction of cytochrome P4501B1(CYP1B1) in cell culture is widely used as a biomarker for PAHs. Therefore we have studied the effect of PAHs in the human breast cancer cells MCF-7 to evaluate bioactivity of PAHs. We have used the United State of America EPA selected 13 different PAHs, PAHs mixtures and extracts from environmental samples to evaluate the bioassay system. We examined effects of PAHs on the CYP1B1-luciferase reporter gene and CYP1B1 mRNA level. Benzo(k)fluoranthene and dibenzo(a, h)anthracene showed strong response to CYP1B1 promoter activity stimulation, and also CYP1B1 mRNAs increase in MCF-7 cells in a concentration-dependent manner. Acenaphthene, anthracene, benzo(b)fluoranthene, fluorene, fluoranthene, anphthanlene, pyrene, phenanthrene and carbazole were weak responders in MCF-7 cells. RT-PCR analysis indicated that PAHs significantly up-regulate the level of CYP1B1 mRNA.

  • A number of attempts to develop methods for measuring software effort have been focused on the area of software engineering and many models have also been suggested to estimate the effort of software projects. Almost all current models use algorithmic or statistical mechanisms, but the existing algorithmic effort estimation models have failed to produce accurate estimates. Furthermore, they are unable to reflect the rapidly changing technical environment of software development such as module reuse, 4GL, CASE tool, etc. In addition, these models do not consider the paradigm shift of software engineering and information systems(i.e., Object Oriented system, Client-Server architecture, Internet/Intranet based system etc.). Thus, a new approach to software effort estimation is needed. After reviewing and analyzing the problems of the current estimation models, we have developed a model and a system architecture that will improve estimation performance. In this paper, we have adopted a neural network model to overcome some drawbacks and to increase estimation performance. We will also address the efficient system architecture and estimation procedure by a similar case-based approach and finally suggest the heuristic search method to find the best estimate of target project through empirical experiments. According to our experiment with the optimally parsimonious neural network model the mean error rate was significantly reduced to 14.3%.

  • Recently, due to the standard of a new browser developed by the Google and Mozilla "Service Worker", future users is expected to be able to make use of favorite Web sites offline. Google's is, Web sites have developed a standard of a new browser so as to always respond to user requests. Service Worker, websites that provide space capable of offline work to the user's browser to store various document information, to provide the necessary resources. Then, in order to greatly reduce the data exchange operations between the browser and the server, the speed of the Web page increases. Not only cooks as native app that can use the Web application offline, in that us to also further enhance the characteristics of an existing Web application that is running without installing destructive high technology. Service worker specifications, use experience of Web application is very can be improved, is an innovative technology indicates the version of the web evolve as the future of the platform. Service Worker is not included in HTML5 standard final, is currently being continued standardization. Future Service Worker technology I expect what kind of thing unfolds when applied to the Web browser.

Using Digital Climate Modeling to Explore Potential Sites for Quality Apple Production (전자기후도를 이용한 고품질 사과생산 후보지역 탐색)

  • This study was carried out to establish a spatial decision support system for evaluating climatic aspects of a given geographic location in complex terrains with respect to the quality apple production. Monthly climate data from S6 synoptic stations across South Korea were collected for 1971-2000. A digital elevation model (DEM) with a 10-m cell spacing was used to spatially interpolate daily maximum and minimum temperatures based on relevant topoclimatological models applied to Jangsoo county in Korea. For daily minimum temperature, a spatial interpolation scheme accommodating the potential influences of cold air accumulation and the temperature inversion was used. For daily maximum temperature estimation, a spatial interpolation model loaded with the overheating index was used. Freezing risk in January was estimated under the recurrence intervals of 30 years. Frost risk at bud-burst and blossom was also estimated. Fruit quality was evaluated for soluble solids, anthocyanin content, Hunter L and A values, and LID ratio, which were expressed as empirical functions of temperature based on long-term field observations. AU themes were prepared as ArcGlS Grids with a 10-m cell spacing. Analysis showed that 11 percent of the whole land area of Jangsoo county might be suitable for quality 'Fuji' apple production. A computer program (MAPLE) was written to help utilize the results in decision-making for site-selection of new orchards in this region.

Cell Cycle Analysis of Bovine Cultured Somatic Cells by Flow Cytometry

  • The cell cycle phase in which donor nuclei exist prior to nuclear transfer is an important factor governing developmental rates of reconstituted embryos. It was suggested that quiescent G0 and cycling G1 cells could support normal development of reconstituted embryos. In a quest of optimized donor nuclei treatment prior to nuclear transfer, this study was undertaken to examine the cell cycle characteristics of bovine fetal and adult somatic cells when cultured under a variety of culture treatments and the cell cycle change with the lapse of time after trypsinization. This was archived by measuring the DNA content of cells using flow cytometry, Cultured fetal fibroblast cells, adult skin and muscle cells, and cumulus cells were divided by 3 culture treatments; 1) grown to 60-70% confluency (cycling), 2) serum starved culture, 3) culture to confluency. Trypsinized cells were fixed by 70% ethanol and stained with propidium iodide. For one experiment, trypsinized cells were resuspended in DMEM+10% FBS and incubated for 1.5, 3 and 6 h with occasional shaking before ethanol fixation. Cell cycle phases were determined by flow cytometry enabling calculation of percentages of G0+G1, S and G2+M. The majority of cells were in G0+Gl stage regardless of origin of cells. Cultures that were serum starved or cultured to confluency contained significantly (P<0.05) higher percentages of cells in G0+G1 (89.5-95.4%). For every cell lines and culture treatments, percentages of cells in existing in G0+G1 increased with decreasing of the cell size from large to small. In the serum starved and confluency groups, about 98% of small cells were in G0+G1 Serum starved culture contained higher percentages of small-sized cells (38.5-66.9%) than cycling and confluent cultures regardless of cell lines (P<0.05). After trypsinization of fetal fibroblast and adult skin cells that were serum starved and cultured to confluency, the percentages of cells in G0+G1 significantly increased by incubation for 1.5(95.7-99.5%) and 3.0 h (95.9-98.6%). The results suggest that the efficient synchronization of bovine somatic cells in G0+G1 for nuclear transfer can be established by incubation for a limited time period after trypsinization of serum starved or confluent cells.

  • Um die rationalen Zahlen auf die reellen Zahlen zu erweitern und dadurch die Stetigkeit der reellen Zahlen sicherzustellen, hat der deutsche Mathematiker R. Dedekind im Jahr 1872 in seinem Aufsatz "Stetigkeit und Irrationale Zahlen" einen neuen mathmatischen Begriff $eingef\ddot{u}hrt,\;n\ddot{a}mlich$ 'Schnitt'. Die Menge aller rationalen Zahlen Q wird durch eine rationale Zahl a zu zwei Untermengen $A_1=\{x|x{\leq}a,\;x{\in}Q\}$, $A_2=\{x|x>a,\;x{\in}Q\}$ $vollst\ddot{a}ndig$ geteilt. Wenn wir solche Teilung, d.i. solchen Schnitt mit "$(A_1,\;A_2)$" bezeichnen, ist ein $Identit\ddot{a}tssatz$ "a=$(A_1,\;A_2)$" absolut harmlos. Analog dazu glaubt Dedekind fest, $da{\beta}$ jede irrationale Zahl mit Hilfe von einem entsprechenden Schnitt $einzuf\ddot{u}hren$ ist. Zum Beispiel, falls die zwei Mengen $B_1=\{x|x^2<2,\;x{\in}Q\}$ und $B_2=\{x|x^2>2,\;x{\in}Q\}$ gegeben sind, dann $w\ddot{a}re$ die irrationale Zahl $^{\surd}2$ mit $(B_1,\;B_2)$ gleichzusetzen. Dennoch glaubt Dedekind an eine streng wissenschaftliche Fundierung der irrationalen Zahl fest, und $w\ddot{u}rde$ nach dem Grund seines Glaubens befragt, $k\ddot{o}nnte$ er nur seine Behauptung wiederholen, ein klarer Fall circulus vitiosus. Mit anderen Worten, die $L\ddot{u}cke$ zwischen $B_1$ und $B_2$ durch die $Einf\ddot{u}hrung$ einer [einzigen] wissenschaftlich fundierten irrationalen Zahl $\ddot{u}berbr\ddot{u}ckt$ und das Ganze zu einem Kontinium gemacht werden sollte, bleibt dieses Vorhaben von Dedekind erst als eine Hoffnung und dessen Resultat kann $h\ddot{o}chstens$ nur als ein Postulat, aber keineswegs als ein methodisch einwandfreier Beweis betrachtet werden. Die Probleme, die mit dem Versuch der $Einf\ddot{u}hrung$ der irrationalen Zahlen mit Hilfe von Schnitt verbunden sind, sind nicht spezifisch allein im Gebient der Mathmatik, sondern betreffen immer wieder die Rechtfertigungsfrage der $Einf\ddot{u}hrung$ der letzten Bestandteile im bezug auf eine Systemerstellung, egal ob dies System ein Wissenschaftliches oder unsere $allt\ddot{a}gliche$ Sprachhandlung ist. $F\ddot{u}r$ all diese Rechtfertigungsfragen gilt das in der klassischen Logik $g\ddot{a}ngige$ logische Prinzip tertium non datur nicht mehr, aber nicht nur wegen der von praktischen $Unm\ddot{o}glichkeit$, die unendlichen vielen $Gegenst\ddot{a}nde$ durchforschen zu $m\ddot{u}ssen$, das $hei{\beta}t$, wegen der erkenntnistheoretischen $Beschr\ddot{a}nktheit$ des jetzigen Erkenntnisniveau, sondern auch wegen des speziellen ontologischen modus der $eizuf\ddot{u}hrenden$ Objekten. Der Autor des Aufsatzes analysiert $\ddot{a}nliche$ $F\ddot{a}lle$ (das Urmeterbeispiel und die Chrakterisierungen der geometrischen Axiome von Wittgenstein), und versucht mit Hilfe von beiden Begriffen, 'interne' und 'externe' Relation, zu zeigen, $da\beta$ eine gemeinsame, invariante Struktur in den eben genannten $F\ddot{a}llen$ besteht. Am Ende des Aufsatzes setzt der Autor sich mit der logischen Argumentationsstruktur des Zitates tiber 'Grenze' aus Noten von Leonardo da Vinci auseinander, und weist auf einen $m\ddot{o}glichen$ Zusammenhang der Grundidee seienes Aufsatzes mit der Philosophie der indischen Denker $N\bar{a}g\bar{a}rjuna$ hin, obwohl die zitierten Versen aus dem Hauptwerk von $N\bar{a}g\bar{a}rjuna$, dem Mittleren Weg$(Madhyamakak\bar{a}rik\bar{a})$ nur andeutend sein $m\ddot{o}gen$.
