Inhibitory Effect of Guanidine Hydrochloride on the Infectious Flacherie Virus of the Silkworm, Bombyx mori (누에 전염성 연화병의 발병 억제제에 의한 방제)

  • The prevalence of the infectious flacherie virus (FV) disease causes a severe damage to cocoon yield and various methods to control the disease have been studied. In this regard, guanidine hydrochloride (GH), one of the guanidine derivatives known as the most inhibitory agent against the replication of picorna virus, was applied to silkworms per os with mulberry leaves and the results were as follows. 1. The application of GH below 0.01% of the chemical concentration did not give any damage to silkworm larvae. 2. The transmission of the virus disease by introducing the FV infected larvae to the healthy larvae group was proportioned to the number of infected larvae. When l% of infected larvae was introduced to the rearing tray of healthy larvae, the pupation rate was 70.7%(79) and it was 38.4% (43) to 5% of infected larvae introduced, while the control of non-mixed with infected larvae gave 89.2% (100) of pupation rate. The cocoon yield from 10,000 larvae also showed the same tendency as the pupation rate. 3. The inhibitory effect of GH against the replication of FV showed ten times in treatment of 0,01% of the chemical agent compared to the non-treatment. 4. The successive application of GH after virus inoculation to silkworm larvae led to the most effective on the inhibition of the virus replication. 5. The immediate application of GH after the virus inoculation also gave the best effect on the inhibition of the virus replication in silkworm larvae. 6. The effect of GH on the inactivation of FV in vitro was not observed.

Growth Pattern of GH-Transgenic Coho Salmon, Oncorhynchus kisutch in Winter Season (GH-Transgenic Coho Salmon, Oncorhynchus kisutch의 월동기 성장 경향)

  • Transgenic coho salmon, Oncorhynchus kisutch containing a growth hormone gene construct have been examined for their hormone levels and ability to growth for 90 days in winter season. Food intake of the transgenic coho was approximately 4-fold higher than that of nontransgenic coho salmon of similar size, but feed efficiency of the transgenic coho was 1.1-fold lower than that of size-matched control. Specific growth rates of body weight of the transgenic coho were approximately 1.4-fold (length) or 3-fold(weight) higher than that of nontransgenic coho salmon. GH, total-T$_4$ and total-T$_3$ levels were Increased approximately 2-fold compared to size control salmon. The transgenic animals also displayed head, jaw and opercular abnormalities typical of the effects of this gene construct in coho salmon, indicating that some imbalance in growth processes were induced.

Stable Expression of hGH Transgene in the Milk of Transgenic Mice (유즙내 사람 성장 호르몬을 분비하는 형질전환생쥐의 형질 유전성에 관한 연구)

  • Rat $\beta$-casein 유전자와 사람 성장호르몬 (hGH) 유전자의 융합유전자를 생쥐 수정란의 웅성전핵에 미세주입하여 형질전환생쥐 (transgenic mouse) 6계통을 확립하였다. 이들로부터 사람성장호르몬(hGH) 유전자의 발현 여부를 조사한 결과, 6계통중 4계통의 생쥐 유즙에서 hGH가 2~900 ng/ml 수준으로 발현되고 있었으며, 혈중에서는 hGH가 검출되지 않았다. 따라서 이들 형질전환생쥐에서 사람성장호르몬이 우선특이적으로 발현, 분비되고 있음을 알 수 있었다. 한편, 사람성장호르몬의 발현이 확인된 두 계통 (ChGH2-2, CChGH2)의 형질전환생쥐를 대상으로 하여 세대별, 산차별로 유즙내 사람성장호르몬의 함량을 조사한 결과, 3세대에 걸쳐, 또한 제1세대에서의 세차례 반복된 비유기에도 유즙내 사람성장호르몬은 지속적으로 분비되고 있었다. 이상의 결과는 형질전환생쥐에서의 외래유전자의 발현성은 반복되는 비유기와 여러 세대에 걸쳐 안정적으로 유지됨을 보여 주고 있다.

The Effects of gl, gh and wx Gene on the Grain Yield and Yield Components of Rice Plant (수도의 gl, gh 및 wx유전자가 수도수량 및 수량구성요소에 미치는 영향)

  • Isogenic lines for glabrous leaf (gl), hull color(gh) and glutinous endosperm (wx) were used to clarify the effects of the gl, gh and wx gene on the grain yield and yield components in rice plant. Eight genotypes: glgl ghgh wxwx, glgl ghgh WxWx, glgl GH-wxwx, glgl GH-WxWx, GL-ghgh wxwx, GL-ghgh WxWx, GL-GH-wxwx and GL-GH-WxWx were evalutated in the backcrossed $F_2$ generation. Significant effects were measured for gl genotypes on heading date, gh genotypes on grain fertility, and wxwx genotypes on number of panicles per hill and 1, 000 grain weight. The grain yield of straw hull and non glutinous lines were higher than gold hull and glutinous lines. The interaction of three genes, gl, gh and wx was significant for heading date, but not significant in other characters. The average value of isogenic lines which were combined with two recessive genes and one dominant gene were lower and the C.V. value was higher than the isogenic lines which were combined with two dominant genes and one recessive gene in all characters except plant height and number of grains per plant. The average value of isogenic line combined with three recessive genes was lower and the C.V. was higher than the isogenic line combined three dominant genes in all characters measured.

  • Objective: Fibronectin 3 (FN3) and immunoglobulin like modules (Ig) are usually collocated beside modular cellulase catalytic domains. However, very few researches have investigated the role of these modules. In a previous study, we have sequenced and analyzed bacterial metagenomic DNA in Vietnamese goats' rumen and found that cellulase-producing bacteria and cellulase families were dominant. In this study, the properties of modular cellulases and the role of a FN3 in unique endoglucanase belonging to glycosyl hydorlase (GH) family 5 were determined. Methods: Based on Pfam analysis, the cellulases sequences containing FN3, Ig modules were extracted from 297 complete open reading frames (ORFs). The alkaline, thermostability, tertiary structure of deduced enzymes were predicted by AcalPred, TBI software, Phyre2 and Swiss models. Then, whole and truncated forms of a selected gene were expressed in Escherichia coli and purified by His-tag affinity column for assessment of FN3 ability to enhance enzyme activity, solubility and conformation. Results: From 297 complete ORFs coding for cellulases, 148 sequences containing FN3, Ig were identified. Mostly FN3 appeared in 90.9% beta-glucosidases belonging to glycosyl hydrolase family 3 (GH3) and situated downstream of catalytic domains. The Ig was found upstream of 100% endoglucanase GH9. Rarely FN3 was seen to be situated downstream of X domain and upstream of catalytic domain endoglucanase GH5. Whole enzyme (called XFN3GH5 based on modular structure) and truncate forms FN3, XFN3, FN3GH5, GH5 were cloned in pET22b (+) and pET22SUMO to be expressed in single and fusion forms with a small ubiquitin-related modifier partner (S). The FN3, SFN3 increased GH5 solubility in FN3GH5, SFN3GH5. The SFN3 partly served for GH5 conformation in SFN3GH5, increased modules interaction and enzyme-soluble substrate affinity to enhance SXFN3GH5, SFN3GH5 activities in mixtures. Both SFN3 and SXFN3 did not anchor enzyme on filter paper but exfoliate and separate cellulose chains on filter paper for enzyme hydrolysis. Conclusion: Based on these findings, the presence of FN3 module in certain cellulases was confirmed and it assisted for enzyme conformation and activity in both soluble and insoluble substrate.

  • Growth hormone (GH) treatment has been increasingly widely used for children with GH deficiencies as the survival rate of pediatric patients with malignancies has increased. Both GH and insulin-like growth factor-I have mitogenic and antiapoptotic activity, prompting concern that GH treatment may be associated with tumor development. In this review, the authors examined the relationship between GH treatment and cancer risk in terms of de novo malignancy, recurrence, and secondary neoplasm. Although the results from numerous studies were not entirely consistent, this review of various clinical and epidemiological studies demonstrated that there is no clear evidence of a causal relationship between GH treatment and tumor development. Nonetheless, a small number of studies reported that childhood cancer survivors who receive GH treatment have a small increased risk of developing de novo cancer and secondary malignant neoplasm. Therefore, regular follow-ups and careful examination for development of cancer should be required in children who receive GH treatment. Continued surveillance for an extended period is essential for monitoring long-term safety.

The Effect of 12 Weeks Rope Skipping and Band Exercise on GH, IGF-1 and Metabolic Syndrome Risk Factors in Female Obese Middle School Students (12주간 줄넘기, 밴드운동이 비만 여중생의 GH, IGF-1 및 대사증후군 위험 인자에 미치는 영향)

  • The purpose of this study was to investigate the effect of rope skipping and band exercise on GH, IGF-1 and metabolic syndrome risk factors in female obese middle school students. Twenty female obese middle school students(%BF > 30%) were randomly assigned to a exercise group(EX, n=10) or control group(CON, n=10). The exercise group has performed rope skipping and band exercise for 12 weeks, 3time per week. Exercise intensity was increased gradually, from 40% to 70% of Heart Rate Reserve(HRR) and Rating of Perceived Exertion(RPE) 11-15. GH, IGF-1 and metabolic syndrome rist factors were GH, IGF-1 and metabolic syndrome risk factors were measured pre- and post-exercise program including rope skipping and band exercise. Levels of GH and IGF-1 were significantly increased (p<.01 and p<0.001, respectively) in exercise group after 12 weeks of exercise training. WC and TG were significantly decreased(p<.01) in exercise group after 12 weeks of training. HDL-C was significantly increased(p<.05) in exercise group after 12 weeks of training. In conclusion, rope skipping and band exercise were effective exercise program in improving GH, IGF-1 and metabolic syndrome risk factors in female obese middle school students.

Efficacy and safety of growth hormone treatment for children born small for gestational age

  • Recombinant growth hormone (GH) is an effective treatment for short children who are born small for gestational age (SGA). Short children born SGA who fail to demonstrate catch-up growth by 2-4 years of age are candidates for GH treatment initiated to achieve catch-up growth to a normal height in early childhood, maintain a normal height gain throughout childhood, and achieve an adult height within the normal target range. GH treatment at a dose of $35-70{\mu}g/kg/day$ should be considered for those with very marked growth retardation, as these patients require rapid catch-up growth. Factors associated with response to GH treatment during the initial 2-3 years of therapy include age and height standard deviation scores at the start of therapy, midparental height, and GH dose. Adverse events due to GH treatment are no more common in the SGA population than in other conditions treated with GH. Early surveillance in growth clinics is strongly recommended for children born SGA who have not caught up. Although high dose of up to 0.067 mg/kg/day are relatively safe for short children with growth failure, clinicians need to remain aware of long-term mortality and morbidity after GH treatment.

4-Nonylphenol Increased NO Synthesis via a Non-genomic Action in GH3 Cells (뇌하수체 세포인 GH3세포에서 non-genomic action을 통한 Nonylphenol의 nitric oxide 증진효과)

  • 본 연구는 환경호르몬(endocrine disruptors)으로 분류되었으며, 에스트로젠 화합물의 특성을 지닌 4-Nonylphenol (NP)이 설치류 Pituitary 세포 중 성장호르몬을 분비하는 GH3 세포의 Nitric oxide(NO)을 증가시키는 작용기전을 규명코자 수행되었다 먼저 GH3세포에 NP처리 농도에 따른 NO의 생성을 측정한 결과 NP처리농도 의존적으로 증가시켰다. 이러한 NO의 증가가 genomic action인지를 확인하기 위해 GH3세포의 NO를 증가시키는 효소인 neuronal oxide synthase의 단백질량을 측정한 결과 GH3세포에서 NP에 의한 nNOS의 단백질의 변화는 없었다. 에스트로젠 화합물인 NP가 에스트로젠 리셉터 (ER)와의 관계를 조사하기 위해 ER억제제(ICI 168,780)클 처리한 경우 NP에 의해 증가한 NO가 감소하였다. 또한 유전자 전사억제제인 actinomycin D 및 단백질 발현 억제제인 cycloheximide을 처리한 경우는 NP에 의한 NO 증가억제효과가 없었다. 이러한 결과를 종합해 볼 때 GH3 세포에서 NP는 ER을 매개한 non-genomic action에 의해 NO를 증가키는 것으로 사료된다.

Somatotropic Axis and Nutrition in Young Ruminants around Weaning Time

  • The somatotropic (GH-IGF-I) axis consists of many hormonal and nutritional factors that control GH release from the somatotrophs in the anterior pituitary. The GH-releasing substances are GHRH and GHS (GHRP or ghrelin), while the GH release-inhibiting substances are somatostatin (SRIF), insulin-like growth factor-I (IGF-I), leptin and glucocorticoids. However, there is evidence showing that nutrition is involved in the control of the somatotropic axis. In addition, weaning is a drastic event for neonates because their alimentary and endocrine circumstances are changed due to the switch, even if gradual, from a liquid milk diet to one composed of such solids as hay and grains. The biological role of ghrelin is one of the hormonal factors that have been focused on ever since ghrelin was discovered at the end of the last century. A 27-amino acid peptide that is mainly synthesized and released from the abomasum epithelium, ghrelin has not been fully evaluated in relation to the somatotropic axis of the ruminant. It has also proven difficult even to investigate the cellular mechanisms of ghrelin action, because this hormone exerts animal-species-dependent actions via a complex set of intracellular signaling pathways. This is also the case for the action of leptin. Another substance, IGF-I, shows a partial inhibitory action on GH secretion in the ruminant. The effect of nutrition is also different among animal species. This is evident by the fact that undernutrition suppresses the circulating GH levels in rodents, but increases it in ruminants and humans. Recently, weaning has been shown to change the postprandial GH responses in ruminants; milk feeding increases, but hay and concentrate feeding suppress, the postprandial circulating GH levels. Even if the postprandial GH level is increased, the ghrelin level is decreased by milk feeding. Macronutrients also possess stimulatory and inhibitory actions on GH secretion in vivo and in vitro. These findings indicate the complexity of the control mechanisms of the somatotropic axis. In the present review, we summarize recent findings on the factors controlling the axis of the ruminant.
