Antitumor Activities of Red Ginseng Acidic Polysaccharide(RGAP) as an Immunomodulator

  • A red ginseng acidic polysaccharide(RGAP) with immunomodulating antitumor activities was isolated from Korean red ginseng, The molecular weight of RGAP was estimated to be 12-450 kDa by gel filtration chromatography, RGAP was found to increase survival rate and to inhibit of tumor growth significantly in a dose dependent manner in mice transplanted with tumor cells. RGAP significantly promoted nitric oxide(NO) production from peritoneal macrophages bothin vivo and in vitro. Western blot analysis exhibited a newly synthesized inducible nitric oxide synthase(iNOS) protein band in the RGAP treated group. It seems likely that immunomodulating antitumor activities of RGAP are mainly mediated by NO production of macrophage. RGAP was further purified by ultrafiltration and anion exchange chromatography on DEAE-sepharose, followed by gel filtration on Sephacryl S-300 to give an active fraction(GFP) with stronger NO production in murine macrophages. GFP increased survival rate ten times compared to RGAP in male ICR mice transplanted with sarcoma 180 and also showed more potent tumoricidal activities of natural killer cells than RGAP. Sugar $composition(mol\%)$ of GFP was found to be arabinose:rhamnose:xylose:galacturonic acid:mannose:galactose:glucose(10:9:1:25:8:20:27) by GC/MS. The results suggest that clinical trials of RGAP in immunotherapy against cancer are highly feasible.

Quantification and Varietal Variation of Fatty Acids in Mulberry Fruits (뽕나무 계통별 오디종자의 지방산 조성 및 함량)

  • Fatty acids analysis was carried out with mulberry seed. As a result, palmitic acid, stearic acid, oleic cid and linoleic acid were detected as a major fatty acid. Their rates of composition were 8.8${\pm}$3.3, 4.2${\pm}$1.6, 5.8${\pm}$2.3 and 81.2${\pm}$6.9(%), respectively. The unsaturated fatty acids such as oleic acid and linoleic acid were abundant in mulberry seed. Paticularly, five accessions including ‘Gukkwang’, ‘Jaelaenosang’, ‘Sangbansibmunja (Jeonnam) ’, ‘Neunggeum’ and ‘Suwonnosang’ were observed only with linoleic acid. Unsaturated fatty acids are known to have physiological effects decreasing the serum cholesterol. From the above result, we confirm that mulberry fruits have abundant unsaturated fatty acids and anthocyanin pigments, and they are of great value as functional food resources.

  • Upon chemical agent release, it is of importance to study the characteristic persistence and evaporation of chemical agents from surfaces for the prediction of dispersion hazard. We have recently developed a fast and near real-time wind tunnel system proving the controlled environment(air flow, temperature, and humidity), continuously collects agent vapor and analyzes it. A thermal sorber/desorber is unnecessary to collect the vapor in the system we have developed. Instead, a tandem thermal sorber collects the vapor, which is then directly transferred to a fast gas chromatography(GC) for analysis. As a proof of concept, the evaporation of sulfur mustard agent(HD) was studied from glass, sand and concrete. The results were in an excellent agreement with those obtained from the conventional wind tunnel system.

  • In this study GC and PAS were used to calculate $N_2O$ concentration of exhaust gas from Wood Chip combustion system. Fuel supplied to the incinerator was collected and analyzed and then the analysis result was used to calculate $N_2O$ emissions. Tier 3 and Tier 4 Method were used to calculate the $N_2O$ emissions. Plant's Specific emission factor of $N_2O$ by Tier 3 Method was 0.35 kg/TJ, while default emission factor of Wood?Wood Waste proposed by 2006 IPCC G/L was 4 kg/TJ. So the $N_2O$ emission factor of this study was 3.65 kg/TJ lower compared to the IPCC G/L. The total emissions calculated by Plant's specific emission factor was 4.22 kg during the measuring period, but by Tier 4 Method it was 7.88 kg. This difference in emissions was caused by the difference of continuous measuring and intermittent sampling. It would be necessary to apply continuous measuring to calculate emissions of $Non-CO_2$ gas whose the density distribution is relatively high. However currently, according to the target management guideline of greenhouse gas and energy, the continuous measuring method to calculate greenhouse gas emission is applied only to $CO_2$. Therefore for reliable greenhouse gas emission calculation it would be necessary to apply continuous measuring to calculate $Non-CO_2$ gas emission.

Microtensile bond strength of CAD/CAM-fabricated polymer-ceramics to different adhesive resin cements

  • Objectives: This study evaluated the microtensile bond strength (${\mu}TBS$) of polymer-ceramic and indirect composite resin with 3 classes of resin cements. Materials and Methods: Two computer-aided design/computer-aided manufacturing (CAD/CAM)-fabricated polymer-ceramics (Enamic [ENA; Vita] and Lava Ultimate [LAV; 3M ESPE]) and a laboratory indirect composite resin (Gradia [GRA; GC Corp.]) were equally divided into 6 groups (n = 18) with 3 classes of resin cements: Variolink N (VAR; Vivadent), RelyX U200 (RXU; 3M ESPE), and Panavia F2 (PAN; Kuraray). The ${\mu}TBS$ values were compared between groups by 2-way analysis of variance and the post hoc Tamhane test (${\alpha}=0.05$). Results: Restorative materials and resin cements significantly influenced ${\mu}TBS$ (p < 0.05). In the GRA group, the highest ${\mu}TBS$ was found with RXU ($27.40{\pm}5.39N$) and the lowest with VAR ($13.54{\pm}6.04N$) (p < 0.05). Similar trends were observed in the ENA group. In the LAV group, the highest ${\mu}TBS$ was observed with VAR ($27.45{\pm}5.84N$) and the lowest with PAN ($10.67{\pm}4.37N$) (p < 0.05). PAN had comparable results to those of ENA and GRA, whereas the ${\mu}TBS$ values were significantly lower with LAV (p = 0.001). The highest bond strength of RXU was found with GRA ($27.40{\pm}5.39N$, p = 0.001). PAN showed the lowest ${\mu}TBS$ with LAV ($10.67{\pm}4.37N$; p < 0.001). Conclusions: When applied according to the manufacturers' recommendations, the ${\mu}TBS$ of polymer-ceramic CAD/CAM materials and indirect composites is influenced by the luting cements.

Synergistic Anticancer Effect of the Cinnamomi Cortex Ethanol Extract (CcEE) and Hyperthermia in AGS Human Gastric Cancer Cells (AGS 인체 위암세포에서 육계 에탄올 추출물(CcEE)과 온열치료의 항암 시너지 효과)

  • Objectives : In this study, we investigated the combination effects of Cinnamomi cortex Ethanol Extract (CcEE) and hyperthermia in the human AGS gastric cancer cell line. Methods : AGS cells were treated with the indicated concentrations of CcEE (0, 50 or $60{\mu}g/mL$) for 1h prior to hyperthermia. And then incubated for a further 30 min at the indicated temperatures (37, 42 or $43^{\circ}C$) in a humidified incubator containing 5% $CO_2$ or a thermostatically controlled water bath for hyperthermia. The cell viability was measured by MTT assay, Morphology assay and Trypan blue assay. To investigate the possible molecular signaling pathways, the activation of mitogen-activated protein kinase (MAPK) proteins (ERK, p38 and JNK) and expression of various anti-apoptotic proteins such as Caspase-3, Caspase-9, p53, Cyclin D1 and MMP-2 were assessed by Western blot analysis. In addition, Annexin V and 7-amino-actinomycin D (7-AAD) staining was performed to examine the apoptotic mechanism. Results : Combination of CcEE with hyperthermia effectively suppressed the cell viability and changed cellmorphology compared with CcEE or hyperthermia treatment alone. Combined treatment also abated the expression of Caspase-3, Caspase-9, Cyclin D1 and MMP-2. Whereas, the expression level of p53 was up-regulated by co-treatment. Moreover, combination treatment enhanced phosphorylation of ERK, p38 and JNK. In addition, this combination increased anti-cancer effect by inducing cell death through the apoptosis. Conclusions : Taken together, all these findings suggest that the combination treatment with CcEE and hyperthermia may have therapeutic potential as a promising approach to patients with stomach cancer.

In vivo wear determination of novel CAD/CAM ceramic crowns by using 3D alignment

  • PURPOSE. To determine wear amount of single molar crowns, made from four different restoratives, and opposing natural teeth through computerized fabrication techniques using 3D image alignment. MATERIALS AND METHODS. A total of 24 single crowns (N = 24 patients, age range: 18 - 50) were made from lithium disilicate (IPS E-max CAD), lithium silicate and zirconia based (Vita Suprinity CAD), resin matrix ceramic material (Cerasmart, GC), and dual matrix (Vita Enamic CAD) blocks. After digital impressions (Cerec 3D Bluecam, DentsplySirona), the crowns were designed and manufactured (Cerec 3, DentsplySirona). A dualcuring resin cement was used for cementation (Variolink Esthetic DC, Ivoclar). Then, measurement and recording of crowns and the opposing enamel surfaces with the intraoral scanner were made as well as at the third and sixth month follow-ups. All measurements were superimposed with a software (David-Laserscanner, V3.10.4). Volume loss due to wear was calculated from baseline to follow-up periods with Siemens Unigraphics NX 10 software. Statistical analysis was accomplished by Repeated Measures for ANOVA (SPSS 21) at = .05 significance level. RESULTS. After 6 months, insignificant differences of the glass matrix and resin matrix materials for restoration/enamel wear were observed (P>.05). While there were no significant differences between the glass matrix groups (P>.05), significant differences between the resin matrix group materials (P<.05) were obtained. Although Cerasmart and Enamic were both resin matrix based, they exhibited different wear characteristics. CONCLUSION. Glass matrix materials showed less wear both on their own and opposing enamel surfaces than resin matrix ceramic materials.

Monitoring and Risk Assessment of Pesticide Residues in Pre-harvest Fresh Ginseng (Panax ginseng C. A. Meyer) in Chungbuk Province in 2019 (충북지역 생산단계 수삼 중 잔류농약 모니터링 및 위해성 평가(2019))

  • BACKGROUND: It is very important to monitor the residual characteristics of pesticides in pre-harvest fresh ginseng to ensure consumer safety. METHODS AND RESULTS: Forty-eight fresh ginseng samples were collected from 8 ginseng-growing fields 10 days before harvest and pesticide residues in fresh ginseng with and without rhizome (head of ginseng) and rhizome were analyzed for 320 pesticides by using GC-MS/MS and LC-MS/MS. As a result of the pesticide residue analysis, the number of pesticides detected from the fresh ginseng with rhizome, that without rhizome and rhizome were 26, 25, and 40, respectively, indicating that high number of pesticides found in rhizome, compared with the other parts. Pyraclostrobin was detected with the highest frequency in all samples, reaching to 21.2% in fresh ginseng with rhizome, 16.8% in that without rhizome, and 14.8% in rhizome. CONCLUSION: The residue levels of pesticides detected did not exceed their maximum residue limits, in spite of residual data in fresh ginseng before harvest. The amounts of the estimated daily intakes of all the detected pesticides were found to be from 0.018 to 1.818% of their acceptable daily intakes, indicating that concentrations of pesticides detected from fresh ginseng with and without rhizome collected before harvest do not pose the immediate health risks.

Effect of tack cure time on polymerization shrinkage of dual-cure resin cement

  • When luting indirect restorations with dual-cure resin cement (DCRC), excess cement can be easily removed by performing tack cure of DCRC for a few seconds. The purpose of this study was to evaluate whether different tack cure times affect polymerization shrinkage (PS) of the selected DCRC. One dual-cure resin cement (G-CEM LinkAce, GC) was used for measuring PS in light-cure (LC group), self-cure (SC group), and two tack-cure modes. In the first tack-cure subgroup, tack cure was performed for 1, 2, 3, and 5 seconds, followed by light cure after 2 minutes of remnant removal time in each case (TC-LC groups). In the other tack-cure subgroup, tack cure was performed for the same lengths of time, but followed by self-cure in each case (TC-SC groups). PS was measured by a modified bonded disc method for 1,800 seconds. One-way analysis of variance followed by Duncan's post hoc test was used to determine any statistically significant differences among the test groups (α = 0.05). When the DCRC was self-cured after tack cure, PS was significantly lower than when it was only self-cured (p < 0.05); however, tack cure time did not affect PS (p > 0.05). When the DCRC was light-cured, PS was not affected by tack cure or tack cure time (p > 0.05). Therefore, tack cure within 5 seconds did not negatively affect the final PS when the DCRC was light-cured after cement remnant removal.

Variovorax terrae sp. nov. Isolated from Soil with Potential Antioxidant Activity

  • A white-pigmented, non-motile, gram-negative, and rod-shaped bacterium, designated CYS-02T, was isolated from soil sampled at Suwon, Gyeonggi-do, Republic of Korea. Cells were strictly aerobic, grew optimally at 20-28℃ and hydrolyzed Tween 40. Phylogenetic analysis based on 16S rRNA gene sequence indicated that strain CYS-02T formed a lineage within the family Comamonadaceae and clustered as members of the genus Variovorax. The closest members were Variovorax guangxiensis DSM 27352T (98.6% sequence similarity), Variovorax paradoxus NBRC 15149T (98.5%), and Variovorax gossypii JM-310T (98.3%). The principal respiratory quinone was Q-8 and the major polar lipids contain phosphatidylethanolamine (PE), phosphatidylethanolamine (PG), and diphosphatidylglycerol (DPG). The predominant cellular fatty acids were C16:0, summed feature 3 (C16:1ω7c and/or C16:1ω6c) and summed feature 8 (C18:1ω7c and/or C18:1ω6c). The DNA GC content was 67.7 mol%. The ANI and dDDH values between strain CYS-02T and the closest members in the genus Variovorax were ≤ 79.0 and 22.4%, respectively, and the AAI and POCP values between CYS-02T and the other related species in the family Comamonadaceae were > 70% and > 50%, respectively. The genome of strain CYS-02T showed a putative terpene biosynthetic cluster responsible for antioxidant activity which was supported by DPPH radical scavenging activity test. Based on genomic, phenotypic and chemotaxonomic analyses, strain CYS-02T was classified into a novel species in the genus Variovorax, for which the name Variovorax terrae sp. nov., has been proposed. The type strain is CYS-02T (= KACC 22656T = NBRC 00115645T).
