Objective: Panax ginseng is one of the most medicinally used herbal medicines in the world. Wild ginseng is widely accepted to be more active than cultivated ginseng in chemoprevention. However, little has actually been reported on the differences between wild ginseng and cultivated ginseng. Method: To identify wild ginseng-specific genes, we used suppressive subtraction hybridization. Results: We report that one of the clones isolated in this screen was the GAPDH (glyceraldehyde 3-phosphate dehydrogenase) gene (designated pGAPDH-w). DNA BLAST sequence analysis revealed that this pGAPDH-w gene contained novel sequences of 94 bp. RT-PCR results showed that the expression of the pGAPDH-w gene was significantly up-regulated in the wild ginseng as compared with the cultivated ginseng. Conclusion: The pGAPDH-w gene may be one of the important markers of wild ginseng.
이 연구는 유산소운동이 1형 당뇨쥐의 체장세포질 GAPDH 및 미토콘드리아 MnSOD 활성에 미치는 영향을 알아보기 위한 것이다. 실험동물로는 SD계열 흰쥐를 대상으로 strepzotosine을 주입하여 당뇨를 유발시킨 후 수영운동을 8주간 적용시켰다. 실험 종료 후 췌장조직을 적출하여 시료를 분리한 다음 Western Blotting을 실시하여 GAPDH와 MnSOD를 분석하였다. 본 실험결과 췌장세포질 GAPDH의 그룹 간 활성정도는 정상 대조군(CON)과 당뇨군(D.M) 및 당뇨 운동군(D.M-Ex)의 MnSOD 활성 정도는 각 그룹 간 유의한 활성차이를 나타냈다[F(3, 27) = 57.9, P = .000]. 대조군과 비교했을 때 당뇨그룹의 활성은 나타났으며, 당뇨운동그룹 또한 대조군에 비해 낮은 활성을 나타냈다. 반면 당뇨운동그룹은 당뇨그룹과 비교했을 때 상대적으로 높은 활성을 보였다. 췌장의 미토콘드리아 MnSOD 활성 또한 유사한 결과를 보여 이들 또한 각 그룹 간 유의한 활성차이를 나타냈다[F(3, 27) = 572.472, P = .000]. 따라서 본 연구결과를 종합해 볼 때 유산소운동에 의한 GAPDH와 MnSOD 활성증가는 당뇨병 병소와 관련된 다양한 경로의 활성을 억제하고 미토콘드리아 기능회복에 긍정적인 영양을 미쳐 당뇨병 발병의 진행을 막는데 효과적인 것으로 생각된다.
In order to study the biological function of gapdh gene in yak, and prove whether the gapdh gene was a useful intra-reference gene that can be given an important role in molecular biology research of yak, the cDNA sequence encoding glyceraldehyde-3-phosphate dehydrogenase from yak was cloned by the RT-PCR method using gene specific PCR primers. The sequence results indicated that the cloned cDNA fragment (1,008 bp) contained a 1,002 bp open reading frame, encoding 333 amino acids (AAs) with a molecular mass of 35.753 kDa. The deduced amino acids sequence showed a high level of sequence identity to Bos Taurus (99.70%), Xenopus laevis (94.29%), Homo sapiens (97.01%), Mus musculus (97.90%) and Sus scrofa (98.20%). The expression of yak's gapdh gene in heart, spleen, kidney and brain tissues was also detected; the results showed that the gapdh gene was expressed in all these tissues. Further analysis of yak GAPDH amino acid sequence implied that it contained a complete glyceraldehyde-3-phosphate dehydrogenase active site (ASCTTNCL) which ranged from 148 to 155 amino acid residues. It also contained two conserved domains, a NAD binding domain in its N-terminal and a complete catalytic domain of sugar transport in its C-terminal. The phylogenetic analysis showed that yak and Bos taurus were the closest species. The prediction of secondary structures indicated that GAPDH of yak had a similar secondary structure to other isolated GAPDH. The results of this study suggested that the gapdh gene of yak was similar to other species and could be used as the intra-reference to analyze the expression of other genes in yak.
Aberrant GAPDH expression following S-nitrosoglutathione (GSNO) treatment was compared in HepG2 cells, which express functional p53, and Hep3B cells, which lack functional p53. The results of Western blotting and fluorescent immunocytochemistry revealed that nuclear translocation and accumulation of GAPDH occur in both HepG2 and Hep3B cells. This finding suggests that p53 may not be necessary for the GSNO-induced translocation of GAPDH to the nucleus during apoptotic cell death in hepatoma cells.
Transcriptional response of the second isoform of glyceraldehyde (GAPDH-2) to infectious challenges using various bacterial species and the rockbream iridovirus (RBIV) was examined in the spleen of the barred knifejaw (Oplegnathus fasciatus). Bacterial challenges of the juvenile barred knifejaws with Escherichia coli, Edwardsiella tarda, Vibrio anguillarum and Streptococcus iniae resulted in the significant elevation of the GAPDH-2 transcripts in the spleen up to 12-fold relative to that in the non-challenged controls (PBS-injected). In addition, the barred knifejaw fingerlings injected with RBIV stock also represented the highly upregulated mRNA expression of the GAPDH-2 up to more than 20-fold when compared to that of control fingerings. Results obtained from this study strongly suggest that the GAPDH-2 is no longer a housekeeping glycolytic protein and rather than that it might be associated with immune-relevant cellular activities. From this finding, the traditional dogma for the use of GAPDH as an invariant standard for gene expression assays should be carefully revised depending on the kinds of biological stimulations applied in this species.
Streptococcus equi ssp. zooepidemicus (Streptococcus zooepidemicus, SEZ) is an important pathogen associated with opportunistic infections of a wide range of species, including pigs and humans. The absence of a suitable vaccine makes it difficult to control SEZ infection. Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) has been previously identified as an immunogenic protein using immunoproteomic techniques. In the present study, we confirmed that the sequence of GAPDH was highly conserved with other Streptococcus spp. The purified recombinant GAPDH could elicit a significant humoral antibody response in mice and confer significant protection against challenge with a lethal dose of SEZ. GAPDH could adhere to the Hep-2 cells, confirmed by flow cytometry, and inhibit adherence of SEZ to Hep-2 cells in an adherence inhibition assay. In addition, real-time PCR demonstrated that GAPDH was induced in vivo following infection of mice with SEZ. These suggest that GAPDH could play an important role in the pathogenesis of SEZ infection and could be a target for vaccination against SEZ.
S. cerevisiae의 대표적인 구성적 promoter로 GAPDH, ADH1 및 ENO1를 사용하고 이들 promoter 하류에 INUl의 ORF를 in frame으로 연결한 각각의 plasmid pYIGP, pADHl-lNU 및 pENO-INU를 구축하였다. 이들 plasmid 를 함유한 재조합 S. cerevisiae SEY2102 균주들을 sucrose 함유 평판배지에서 선별한 후, 초기 포도당 농도가 2$\%$ 또는 4$\%$인 배지에서 배양했을 때, 모든 균주들은 12시간 이후부터 정지기에 들어갔으며, 정지기에서도 느리지만 균체증식과 inulinase 발현은 계속되었다. 4% 포도당 배지에서 inulinase 총발현량은 ADH1 promoter 계를 제외하고 GAPDH와 ENO1 promoter의 경우 2$\%$ 포도당 배지 때 보다 약 2배 증가한 2.0 unit/mL과 1.4 unit/mL를 각각 보였다. 단위균체농도당 inulinase 활성 즉, 비활성은 GAPDH와 ENOl promoter계의 경우 포도당 농도가 4$\%$일 때 2$\%$때보다 약 63$\%$ 정도의 비활성 증가를 나타내었다. 하지만 ADH1 promoter의 경우는 오히려 비활성이 약 40$\%$ 감소하였다. 그러나, plasmid 안정성 측면에서는 ADH1과 ENO1 promoter 발현계가 GAPDH promoter 경우의 34$\%$보다 훨씬 뛰어난 80$\%$이상을 보였다. 결론적으로 높은 포도당 농도에서 구성적 promoter의 활성 (발현능)은 GAPDH, ENO1, ADH1 promoter 순으로 나타났지만, 초기 포도당 농도가 높을 때나 에탄을 생산이 심각한 유가식 배양에서는 ENO1 promoter가 inulinase의 구성적 발현ㆍ생산에 더 적합할 것으로 사료된다.
본 연구는 5-L 발효기를 이용하여 재조합 고스트 박테리아(E.coli $DH5{\alpha}$/ pHCE-InaN-(enolase, GAPDH, sagA or piaA)-ghost 37 SDM) 백신의 산업화를 위해 탄소원 공급조건, 교반속도, 산소공급 조건등의 최적 배양조건과 고스트 박테리아 발현 유도를 위한 온도조절 시점과 그에 따른 발현효율 최적화를 조사하기 위해 수행하였다. 각각 다른 4종의 항원 유전자를 보유한 고스트 박테리아를 LB 배지를 이용하여 배양한 결과 모두 1 g / 1 glucose, 300 rpm, 1vvm에서 최대 균주 성장을 나타내었다. 고스트 박테리아 생성 효율의 경우 초기 대수증식기(OD$_{600}$=1.0)에서 고스트 발현을 유도했을 때 각각 최대효율인 99.99%를 나타내었으나 증기 대수증식기(OD$_{600}$=2.0)와 말기 대수증식기 (OD$_{600}$=3.0)에서는 고스트 박테리아 생성이 낮은 효율을 나타내었다. 또한 SDS-PAGE 와 western blot를 이용하여 각각 다른 4종의 항원 단백질 발현 여주를 확인한 결과 enolase (78kda), GAPDH (67kda),sagA(26kDa), piaA(26kDa)에서 항원 단백질 band를 확인할 수 있었다. 따라서 본 연구결과 확립된 배양 조건과 발현효율 최적화 조건은 연쇄구균증 질병에 대해 E.coli를 이용한 고스트 박테리아 백신이 양식 산업에 있어 상업적으로 유용한 백신의 최적생산을 위해 사용 될 수 있을 것으로 사료된다.
Dirofilaria immitis (D. immitis)는 개의 심폐사상충증을 일으키는 선형사상충이다. 이 기생충에 감염된 개는 감염 후기 단계에서 하나 이상의 증상과 혈관 주위의 염증을 동반한 심화된 심장 질환을 보인다. 감염 초기단계에 특이적이고 효율적으로 D. immitis를 검출하기 위해서, 선행연구에서 밝혀낸 D. immitis의 cytochrome c oxidase subunit I (COI)와 개의 glyceraldehyde-3-phosphate dehydrogenase (GAPDH)를 검출하는 특이적인 프라이머와 프로브를 이용하여 이중 TaqMan qPCR 방법을 개발했다. 양성 대조군인 플라스미드 유전자는 TA-cloning vector와 D. immitis의 COI나 개의 GAPDH로 구성되었다. 단일과 이중 TaqMan qPCR 방법은 특이적인 프라이머와 프로브, 그리고 게놈 유전자나 플라스미드 유전자로 수행했다. 프라이머의 농도를 최적한 후, 본 연구에서 개발한 이중 반응은 D. immitis의 COI와 개의 GAPDH를 동일 시료에서 동시에 검출했다. 검출 한계는 단일과 이중 방법 모두 25 copies였고, 두 방법 모두 좋은 선형성과 높은 민감도, 그리고 우수한 PCR 효율을 보여주었다. 병원체를 검출하기 위한 이중 방법은 단일 방법에 비해 비용과 노동력, 시간이 적게 든다. 따라서 이중 TaqMan qPCR 방법의 개발은 많은 수의 시료로부터 동시에 효율적으로 D. immitis 검출과 정량이 가능하게 할 것이다.
Background: Quantitative real time reverse transcription PCR (qRT-PCR) is one of the most important techniques for gene-expression analysis in molecular based studies. Selecting a proper internal control gene for normalizing data is a crucial step in gene expression analysis via this method. The expression levels of reference genes should be remained constant among cells in different tissues. However, it seems that the location of cells in different tissues might influence their expression. The purpose of this study was to determine whether the source of mesenchymal stem cells (MSCs) has any effect on expression level of three common reference genes (GAPDH, ${\beta}$-actin and ${\beta}2$-microglobulin) in equine marrow- and adipose-derived undifferentiated MSCs and consequently their reliability for comparative qRT-PCR. Materials and methods: Adipose tissue (AT) and bone marrow (BM) samples were harvested from 3 mares. MSCs were isolated and cultured until passage 3 (P3). Total RNA of P3 cells was extracted for cDNA synthesis. The generated cDNAs were analyzed by quantitative real-time PCR. The PCR reactions were ended with a melting curve analysis to verify the specificity of amplicon. Results: The expression levels of GAPDH were significantly different between AT- and BM-derived MSCs (p < 0.05). Differences in expression level of ${\beta}$-actin (P < 0.001) and B2M (P < 0.006.) between MSCs derived from AT and BM were substantially higher than GAPDH. In addition, the fold change in expression levels of GAPDH, ${\beta}$-actin and B2M in AT-derived MSCs compared to BM-derived MSCs were 2.38, 6.76 and 7.76, respectively. Conclusion: This study demonstrated that GAPDH and especially ${\beta}$-actin and B2M express in different levels in equine AT- and BM-derived MSCs. Thus they cannot be considered as reliable reference genes for comparative quantitative gene expression analysis in MSCs derived from equine bone marrow and adipose tissue.
