한우 암송아지의 성성숙 전 유지 단백질 요구량 결정 (Determination of Crude Protein Requirements for Maintenance of Prepubertal Hanwoo Heifers)

  • 본 연구는 성성숙 전 한우 암송아지의 일일 유지 단백질(CPm) 요구량을 구하기 위하여 평균체중 143.1 kg (실험 1)과 257 kg (실험 2)의 각 6두씩을 3수준의 단백질을 급여하는 two 3 ${\times}$ 3 latin square design에 공시하였다. 실험 1에서는 기초사료로서 timothy 건초를 2.8 kg/d/head를 급여(LCP)하면서 분쇄옥수수와 corn gluten meal을 각각 250 g과 150 g (MCP) 그리고 500 g과 300 g (HCP) 보충 급여하였다. 실험 2에서는 기초사료로서 timothy 건초를 4.8kg/d/head를 급여(LCP)하면서 분쇄옥수수와 corn gluten meal을 각각 350g과 250g (MCP) 그리고 700g과 500g (HCP) 보충 급여하였다. 실험 1에서의 CP 섭취량은 LCP, MCP, HCP구에서 각각 236.6, 340.1, 459.8g/d 이었고, 대사체중당 조단백질 균형은 각각 0.51, 1.87, 3.20g 이었다. 실험 2에서의 CP 섭취량은 LCP, MCP, HCP구에서 각각 415.2, 606.9, 793.0g/d 이었고, 대사체중당 조단백질 균형은 각각 0.67, 1.03, 2.99g 이었다. 실험 1과 2에서 얻은 CPm 요구량은 각각 4.58과 5.02/$BW^{0.75}$ 이었고 한우 사양표준(농림부, 2007)에서 채택하고 있는 5.56g CP/$BW^{0.75}$ 보다 낮았다.

에르븀 첨가 광증폭기의 파장에 따른 이득 특성 측정 및 분석 (Spectral gain variation characteristics of the silica-based erbium doped fiber amplifier in the 1545-1557 nm wavelength region)

  • 실리카가 주 매질인 에르븀 첨가 광증폭기의 1545-1557 nm의 파장 영역에서 이득 스펙트럼 특성과 EDFA를 여러 번 통과하면서 누적되는 특성을 측정, 분석하였다. 파장에 따른 이득 차이가 최소인 이득은 에르븀 첨가 광섬유의 길이가 정해지면 단일하게 결정되고(G$_{0}$), 이득 불균일의 정도는 동작 이득이 G일 때 G$_{0}$-G에 거의 비례하며 비례상수는 0.1~0.2 dB/dB의 값을 갖는다. 에르븀 첨가 광증폭기를 여러 번 지날 때 파장에 따른 이득불균일 및 신호대 잡음비 불균일은 계속 누적되며, 그 양은 통과한 EDFA의 수에 비례한다. 입력 신호광 세기 -20 dBm/ch., 이득 21 dB에서 최적화된 EDFA를 제작하여 파장에 따른 이득 및 잡음지수 특성을 측정한 결과, EDFA 12회 통과 후 파장에 따른 이득차는 5 dB, 잡음지수는 3 dB의 값을 나타냈다.

Error-Prone and Error-Free Translesion DNA Synthesis over Site-Specifically Created DNA Adducts of Aryl Hydrocarbons (3-Nitrobenzanthrone and 4-Aminobiphenyl)

  • Aryl hydrocarbons such as 3-nitrobenzanthrone (NBA), 4-aminobiphenyl (ABP), acetylaminofluorene (AAF), benzo(a)pyrene (BaP), and 1-nitropyrene (NP) form bulky DNA adducts when absorbed by mammalian cells. These chemicals are metabolically activated to reactive forms in mammalian cells and preferentially get attached covalently to the $N^2$ or C8 positions of guanine or the $N^6$ position of adenine. The proportion of $N^2$ and C8 guanine adducts in DNA differs among chemicals. Although these adducts block DNA replication, cells have a mechanism allowing to continue replication by bypassing these adducts: translesion DNA synthesis (TLS). TLS is performed by translesion DNA polymerases-Pol ${\eta}$, ${\kappa}$, ${\iota}$, and ${\zeta}$ and Rev1-in an error-free or error-prone manner. Regarding the NBA adducts, namely, 2-(2'-deoxyguanosin-$N^2$-yl)-3-aminobenzanthrone (dG-$N^2$-ABA) and N-(2'-deoxyguanosin-8-yl)-3-aminobenzanthrone (dG-C8-ABA), dG-$N^2$-ABA is produced more often than dG-C8-ABA, whereas dG-C8-ABA blocks DNA replication more strongly than dG-$N^2$-ABA. dG-$N^2$-ABA allows for a less error-prone bypass than dG-C8-ABA does. Pol ${\eta}$ and ${\kappa}$ are stronger contributors to TLS over dG-C8-ABA, and Pol ${\kappa}$ bypasses dG-C8-ABA in an error-prone manner. TLS efficiency and error-proneness are affected by the sequences surrounding the adduct, as demonstrated in our previous study on an ABP adduct, N-(2'-deoxyguanosine-8-yl)-4-aminobiphenyl (dG-C8-ABP). Elucidation of the general mechanisms determining efficiency, error-proneness, and the polymerases involved in TLS over various adducts is the next step in the research on TLS. These TLS studies will clarify the mechanisms underlying aryl hydrocarbon mutagenesis and carcinogenesis in more detail.

The Performance Analysis of Cognitive-based Overlay D2D Communication in 5G Networks

  • In the near future, it is expected that there will be billions of connected devices using fifth generation (5G) network services. The recently available base stations (BSs) need to mitigate their loads without changing and at the least monetary cost. The available spectrum resources are limited and need to be exploited in an efficient way to meet the ever-increasing demand for services. Device to Device communication (D2D) technology will likely help satisfy the rapidly increasing capacity and also effectively offload traffic from the BS by distributing the transmission between D2D users from one side and the cellular users and the BS from the other side. In this paper, we propose to apply D2D overlay communication with cognitive radio capability in 5G networks to exploit unused spectrum resources taking into account the dynamic spectrum access. The performance metrics; throughput and delay are formulated and analyzed for CSMA-based medium access control (MAC) protocol that utilizes a common control channel for device users to negotiate the data channel and address the contention between those users. Device users can exploit the cognitive radio to access the data channels concurrently in the common interference area. Estimating the achievable throughput and delay in D2D communication in 5G networks is not exploited in previous studies using cognitive radio with CSMA-based MAC protocol to address the contention. From performance analysis, applying cognitive radio capability in D2D communication and allocating a common control channel for device users effectively improve the total aggregated network throughput by more than 60% compared to the individual D2D throughput without adding harmful interference to cellular network users. This approach can also reduce the delay.

연취급 근로자들의 혈중 ZPP 농도 선별기준에 따른 정확도의 변화 (The change of validity of blood zinc protoporphyrin test by different cut-off level in lead workers)

  • Measurement of blood lead (PbB) and blood zinc protoporphyrin (ZPP) are most common biological indices to identify the individual at risk for excess or the health sequences by lead exposure. Because PbB is known most important and reliable index of lead exposure, PbB is often regarded as a gold standard to detect lead exposure. But in Korea PbB is a secondary test item of detailed health check-up with positive finding of screening test in most occasion. Our lead standard requires all lead workers to take annual heath-check twice a year for investigation of their health effect due to lead exposure. Blood ZPP is one of most important index to detect high lead absorption in lead workers as a screening test. Measurement of blood ZPP is known ,well to correlate with PbB in steady state of exposure in most lead workers and is often used as a primary screening test to detect high lead absorption of lead workers with the advantage of simplicity, easiness, portability and low cost. The current cut-off criteria of blood ZPP for further detailed health check-up is $100{\mu}g/d\ell$ which is supposed to match the level of $40{\mu}g/d\ell$ of PbB according to our standard. Authors tried to investigate the validity of current criteria of cut-off level $(100{\mu}g/d\ell)$ of blood ZPP and possible another better cut-off level of it to detect the lead workers whose PbB level over $40{\mu}g/d\ell$. The subjects in our study were 212 male workers in three small scale storage battery industries. Blood ZPP, PbB and hemoglobin (Hb) were selected as the indices of lead exposure. The results were as follows. 1. The mean of blood ZPP, PbB and Hb in lead workers were $79.5{\pm}46.7{\mu}g/d\ell,\;38.7{\pm}15.1{\mu}g/d\ell,\;and\;14.8{\pm}1.2g/d\ell$, respectively. There were significant differences in blood ZPP, PbB and Hb by industry (P<0.01). 2. The percents of lead workers whose blood ZPP were above $100{\mu}g/d\ell$ in the group of work duration below 1, 1-4, 5-9 and above 10 years were 8.6%, 17.2%, 47.6%, and 50.0%, respectively. The percents of lead workers whose PbB were above $40{\mu}g/d\ell$ in those were 31.4%, 40.4%, 71.4%, and 86.4%, respectively. 3. The percents of lead workers whose PbB were below $40{\mu}g/d\ell$, $40-59{\mu}g/d\ell$ and above $60{\mu}g/d\ell$ were 54.7%, 34.9% and 10.4%, respectively. Those of lead workers whose blood ZPP were below $100{\mu}g/d\ell$, $100-149{\mu}g/d\ell$ and above $150{\mu}g/d\ell$ were 79.2%, 13.7% and 7.1%, respectively. 4. Simple linear regression of PbB on blood ZPP was statistically significant (P<0.01) and as PbB was $40{\mu}g/d\ell$, blood ZPP was $82.1{\mu}g/d\ell$. 5. While the highest sensitivity and specificity of blood ZPP test to detect lead workers with PbB eve. $40{\mu}g/d\ell$ were observed in the cut-off level of $50{\mu}g/d\ell$ and $100{\mu}g/d\ell$ of blood ZPP, respectively, the highest validity (sensitivity+specificity) of blood ZPP to detect lead workers with PbB over $40{\mu}g/d\ell$ was observed in the cut-off level of around $70{\mu}g/d\ell$ of blood ZPP. But even with optimal cut-off level of around $70{\mu}g/d\ell$ of blood ZPP, still 25.0% of false negative and 20.7% false positive lead workers were found. As the result of this study, it was suggested that reconsideration of current blood ZPP cut-off of our lead standard from $100{\mu}g/d\ell$ to somewhat lower level such as around $70{\mu}g/d\ell$ and the inclusion of PbB measurement as a primary screening test for lead workers was highly recommended for the effective prevention of lead workers.

낙동강 하구호에서의 인의 침강특성 (Settling Characteristics of Phosphorus in Nakdong Estuary Barrage)

  • 낙동강 하구호의 부영양화와 관련된 인의 순환과정을 구명하기 위해 낙동강 하구호에서 인의 침강과정에 대한 연구를 수행하였다. sediment trap에 포집된 침강성 물질의 침강 플럭스는 중층 및 저층에서 각각 $296g/m^2/d$, $955g/m^2/d$, 침강속도는 1.25m/hr, 3.31m/hr이었고, 인의 침강 플럭스는 중, 저층에서 각각 $0.64g/m^2/d$, $1.97g/m^2/d$, 침강속도는 0.85m/hr, 1.89m/hr이었다. 침강성 물질에 존재하는 POP의 총린에 대한 존재비율은 중층에서 $51.9\%$, 저층에서는 $48.3\%$로 나타났다. 이상의 결과에서 부영양화 모델을 낙동강 하구호에서 적용할 경우 기존의 인의 침강속도 가정치에 대한 보정이 반드시 행해져야 함을 알 수 있다.

한국인 모유의 수유기간별 비중, 충고형분 및 단백질 함량의 변화 (Changes in Specific Gravity, Total Solid and Protein Contents of Human Milk During the Course of Lactation in Korean Women)

  • 수유기간의 경과에 따른 모유중의 비중, 총고형분, 단백질 함량 변화를 살펴 보고자 수유부 27명을 대상으로 분만후 15일부터 150일까지 추적하여 채취한 총112개 모유시료로 부터 얻은 결과는 다음과 같다. 즉 모유 비중의 경시적인 변화는 15일경에 1.0298(S.D. 0.0044, n=25), 30일경에 1.0304(S.D. 0.0046, n=24), 60일경에 1.0270(S.D. 0.0035, n=20) 90일경에 1.0293(S.D. 0.0055, n=18), 120일경에 1.0254(S.D. 0.0038, n=16), 150일경에 1.0268(S.D. 0.0039, n=9)로서 나타났고 수유기간이 경과함에 따라 유의하게 감소하는 경향을 보였다. 총고형분은 모유 100$m\ell$중에 15일 경에 12.48g(S.D. 1.23, n=25), 30일경에 12.31g(S.D. 1.31, n=24) 60일경에 12.47g (S.D. 1.45, n=20), 90일경에 11.81g (S.D. 1.85, n=18), 120일경에 11.44g(S.D. 0.92, n=16) 150일경에 11.88 (S.D. 0.89, n=9)으로 나타났고 수유기간의 경과에 따른 유의한 차이는 보이지 않았다. 모유 100$m\ell$중 단백질함량은 1.07~1.46g의 범위였고, 수유기간별 변화는 15일경에 1.46g (S.D. 0.20, n=25), 30일 경에 1.32g (S.D. 0.19, n=24) 60일경에 1.15g (S.D. 0.10, n=20), 90일경에 1.09g (S.D. 0.17, n=18), 120일경에 1.11g (S.D. 0.17, n=16), 150일 경에 1.07g (S.D. 0.18, n=9)으로 수유기간이 경과 함에 따라 유의하게 감소하는 경향을 보였다. 또한 수유기간 15, 30, 50, 90, 120, 150일에 수집한 모유시료 전체의 총평균 단백질함량은 1.20g (S.D. 0.14, n=27, 총시료수 112)로 나타났다.

Evaluation of polyherbal formulation and synthetic choline chloride on choline deficiency model in broilers: implications on zootechnical parameters, serum biochemistry and liver histopathology

  • Objective: The study was designed to establish choline deficiency model (CDM) in broilers for evaluating efficacy of polyherbal formulation (PHF) in comparison with synthetic choline chloride (SCC). Methods: A total of 2,550 one-day-old Cobb 430 broiler chicks were randomly assigned to different groups in three experiments. In experiment 1, G1 and G2 served as normal controls and were fed a basal diet with 100% soybean meal (SBM) as a major protein source supplemented with and without SCC, respectively. In G3, G4, G5, and G6 groups, SBM was replaced at 25%, 50%, 75%, and 100% by soy protein isolate (SPI) to induce a graded level of choline deficiency. In experiment 2, PHF (500 and 1,000 g/ton) in comparison with SCC (1,000 g/ton) were evaluated. In experiment 3, dose-response of PHF (200, 400, and 500 g/ton) with SCC (400 g/ton) was determined. Results: Replacement of SBM by SPI produced a linear decrease in body weight gain (BWG) with a poor feed conversion ratio (FCR). 25% SBM replacement by SPI yielded an optimum negative impact on BWG and FCR; hence, it is considered for further studies. In experiment 2, PHF (500 and 1,000 g/ton) and SCC (1,000 g/ton) showed a similar performance in BWG, FCR and relative liver weight. In experiment 3, PHF produced an optimum efficacy at 400 g/ton and was comparable to SCC in the restoration of serum aspartate aminotransferase activity, abdominal fat, breast muscle lipid content and liver histopathological abnormalities. Conclusion: Replacement of SBM by SPI caused choline deficiency characterised by worsening of BWG, FCR, elevation in liver enzymes and histopathological changes indicating fatty liver. CDM was found valid for evaluating SCC and PHF. It is concluded that PHF has the potential to mimic biological activities of SCC through the restoration of negative effects caused by CDM.

Identification of the Marker-Genes for Dioxin(2, 3, 7, 8- tetradibenzo-p-dioxin)-Induced Immune Dysfunction by Using the High-Density Oligonucleotide Microarray

  • In a variety of animal species, the perinatal exposure of experimental animals to the 2,3,7,8-tetrachlorodibenzo­p-dioxin (TCDD) leads to the immune dysfunction, which is more severe and persistent than that caused by adult exposure. We report here the changes of gene expression and the identification of the marker-genes representing the dioxin exposure. The expressions of the transcripts were analyzed using the 11 K oligonucleotide­microarray from the bone marrow cells of male C57BL/6J mice after an intraperitoneal injection of $1{\mu}g$ TCDD/kg body weight at various time intervals: gestational 6.5 day(G6.5), 13.5 day(G13.5), 18.5 day(G18.5), and postnatal 3 (P3W)and 6 week (P6W). The type of self-organizing maps(SOM) representing the specific exposure dioxin could be identified as follows; G6.5D(C14), G13.5D(C0, C5, C10, C18), G18.5D(7): P3W(C2, C21), and P6W(C4, C15, C20). The candidate marker-genes were restricted to the transcripts, which could be consistently expressed greater than $\pm$2-fold in three experiments. The resulting candidates were 85 genes, the characteristics of that were involved in cell physiology and cell functions such as cell proliferation and immune function. We identified the biomarker-genes for dioxin exposure: smc -like 2 from SOM C14 for the dioxin exposure at G6.5D, focal adhesion kinase and 6 other genes from C0, and protein tyrosine phosphatase 4a2 and 3 other genes from C5 for G13.5D, platelet factor 4 from C7 for G18.5D, fos from C2 for P3W.

Delftia acidovorans EK2에 의한 고농도 Trichloroethylene (TCE)의 생물학적 분해 특성 (The Biological Degradation of High Concentration of Trichloroethylene (TCE) by Delftia acidovornas EK2)

  • 본 연구에서는 성장기질로써 다양한 방향족 화합물을 첨가하여 TCE 분해 균주를 분리하고 고효율 TCE 분해 균주에 의한 고농도의 TCE 분해 특징에 대해 연구하였다. TCE에 오염된 토양 및 폐수로부터 시료를 채취하였고 성장기질로써 벤젠(Benzene), 페놀(Phenol), 에틸벤젠(Ethylbenzene), 아닐린(Aniline), 큐멘(Cumene), 톨루엔(Toluene) 등을 사용하여 TCE에 내성을 가지는 179 균주를 순수 분리하였다. 순수 분리된 균주들을 TCE 농도 30 mg/L, 성장기질 농도 0.2 g/L, $30^{\circ}C$, pH 7, 균 농도 1.0 g/L (w/v)의 호기적 조건으로 21일 동안 분해효율을 측정한 결과, 아닐린을 성장기질로 이용한 EK2 균주가 74.4%의 가장 높은 효율을 보여주었다. EK2 균주는 형태학적 특징, 생화학적 특징 및 분자적 특징을 분석한 결과 Delftia acidovorans로 동정되었다. D. acidovorans EK2의 TCE 분해는 TCE 농도 10에서 200 mg/L까지 성장 및 분해할 수 있었으나 250 mg/L 이상의 농도에서는 성장과 분해가 보이지 않았다. 저농도(1.0 mg/L) 분해 실험을 위하여 D. acidovorans EK2를 각 0.01 g/L, 0.03 g/L, 0.05 g/L로 적용한 결과, 모든 조건에서 12일 동안 99.9%의 분해효율을 보였다. 고농도(200 mg/L)를 분해하기 위한 최적 배양조건은 균 농도 1.0 g/L, 아닐린 농도 0.5 g/L, pH 7, 온도 $30^{\circ}C$로 확인되었으며, 21일 동안 호기적으로 배양 시 71.0%의 가장 높은 TCE 분해효율을 보여주었고, 분해속도는 94.7 nmol/h이었다. 결과적으로 본 연구에서 개발된 D. acidovorans EK2를 이용하여 고농도의 TCE로 오염된 토양 및 지하수의 생물학적 처리에 효율적으로 이용될 수 있을 것으로 사료된다.
