Antioxidant Activities of Perilla frutescens Britton Seed Extract and Its Inhibitory Effects against Major Characteristics of Cancer Cells (들깨 추출물의 항산화 활성과 암세포 기본 특성에 대한 억제 효과)

  • The aim of the study was to investigate the antioxidant activities of ethanol extract of perilla seed (PSE) and its inhibitory effects against major characteristics of cancer cells, such as unrestricted growth and activated metastasis in vitro. The total polyphenol and flavonoid levels of PSE were 222.6 mg gallic acid equivalent/100 g and 285.7 mg quercetin equivalent/100 g, respectively. The radical scavenging activity and ferric reducing antioxidant power of PSE at concentration of 87.5 to $350{\mu}g/mL$ were 24~45% and 28~62%, respectively. Treatment of HCT116 colorectal carcinoma cells and H1299 non-small cell lung carcinoma cells with PSE dose-dependently inhibited growth by 18~94% (at concentration range of 87.5 to $350{\mu}g/mL$) and completely abolished colony formation (at concentration of $175{\mu}g/mL$). PSE was also effective in inhibiting migration of H1299 cells (by 30~37% at concentration range of 87.5 to $350{\mu}g/mL$) and adhesion of both HCT116 and H1299 cells (by 14~16% at concentration of $350{\mu}g/mL$). These results indicate that PSE exerts antioxidant and anti-cancer activities in vitro. It needs to be determined whether or not similar effects can be reproduced in vivo.

Removal of Arsenic in Waste Water using Pachymeniopsis sp. and Its Application to Soil Washing Process (Pachymeniopsis sp.의 폐수 내 가용성 비소 제거 가능성 평가 및 토양세척공법에의 적용)

  • In this report, we provide experimental evidence that heavy metal ions could be removed using Pachymeniopsis sp., particularly soluble arsenic in leachate from soil contaminated by arsenic. We performed pilot scale of soil washing process based on our results. The adsorption of arsenic by Pachymeniopsis sp. indicated that it could be described with the Langmuir Model and the maximum adsorption capacity increased with decreasing pH (pH 3: 102.48 mg/g, pH 5: 98.32 mg/g, pH 7: 57.70 mg/g, pH 9: 43.34 mg/g) and increasing temperature (10$^{\circ}C$ : 60.38 mg/g, 20$^{\circ}C$ : 76.39 mg/g, 30$^{\circ}C$ : 112.12 mg/g). Our results revealed that soluble arsenic in leachate was removed from 24.03 mg/L to 0.6 ${\pm}$ 0.1 mg/L by Pachymeniopsis sp. for 48hours on pilot scale of soil washing process.

Fed-Batch Sorbose Fermentation Using Pulse and Multiple Feeding Strategies for Productivity Improvement

  • Microbial oxidation of D-sorbitol to L-sorbose by Acetobacter suboxydans is of commercial importance since it is the only biochemical process in vitamin C synthesis. The main bottleneck in the batch oxidation of sorbitol to sorbose is that the process is severely inhibited by sorbitol. Suitable fed-batch fermentation designs can eliminate the inherent substrate inhibition and improve sorbose productivity. Fed-batch sorbose fermentations were conducted by using two nutrient feeding strategies. For fed-batch fermentation with pulse feeding, highly concentrated sorbitor (600g/L) along with other nutrients were fed intermittently in four pulses of 0.5 liter in response to the increased DO signal. The fed-batch fermentation was over in 24h with a sorbose productivity of 13.40g/L/h and a final sorbose concentration of 320.48g/L. On the other hand, in fed-batch fermentation with multiple feeds, two pulse feeds of 0.5 liter nutrient medium containing 600g/L sorbitol was followed by the addition of 1.5 liter nutrient medium containing 600g/L sorbitol at a constant feed rate of 0.36L/h till the full working capacity of the reactor. The fermentation was completed in 24h with an enhanced sorbose productivity of 15.09g/L/h and a sorbose concentration of 332.60g/L. The sorbose concentration and productivity obtained by multiple feeding of nutrients was found to be higher than that obtained by pulse feeding and was therefore a better strategy for fed-batch sorbose fermentation.

Association of the Myeloperoxidase $^{-463}G{\to}A$ Polymorphism with Helicobacter pylori-induced Atrophic Gastritis

  • Although only a minority of the infected individuals develops atrophic gastritis and the malignancy, factors governing clinical outcomes subsequent to Helicobacter pylori (H. pylori) infection have not yet been defined. H. pylori infection is characterized by extensive infiltration of neutrophils. Myeloperoxidase (MPO) in neutrophils amplifies the oxidative potential of hydrogen peroxides that induce gastric mucosal damage, thus MPO is suspected to play a role in H. pylori-induced gastric injury. Therefore, we explored the association of host MPO genetic polymorphism with atrophic gastritis upon H. pylori infection. Biopsy specimens taken from the gastric mucosa were examined histologically in 87 patients. The PCR-RFLP assay was used to characterize MPO genotypes. The distributions of MPO genotypes were MPO (G/G) 82% and MPO (G/A) 18%. None of MPO (A/A) genotype was observed. A strong positive correlation between the levels of neutrophil infiltration and gastric atrophy found only in MPO (G/G) but not in MPO (G/A) genotype. These results suggest that MPO genotype is a critical determinant in the pathogenesis of atrophic gastritis subsequent to H. pylori infection. Further works need to clarify the functional relevance of MPO genetic polymorphisms on gastric cell injury.

  • It is necessary to perform a study on the interrace between a G7 pantograph and a KTX catenary system prior to G7 on-line tests in the Korea High Speed Test Track in order to predict how high current collection quality can be obtained during the on-line tests and check if safety problems shall be caused b)Y the tests or not. According to the simulation results, current collection quality of the G7 pantograph at 350km/h is lower than that of a GPU pantograph at 300km/h, but the contact wire uplifts and average contact forces are within the safe-zone. In addition, the ratio of running speed (350km/h) to safe running. Therefore, the G7 on-line tests at 350km/h in the Korea High Speed Test Track is expected not to cause the safety problem.

  • The determination of trace selenium in milk powder has been studied by octopole reaction cell(ORC)-ICP-MS. The interferences by polyatomic ions and other concomitant molecular species could be removed remarkably by using $H_2$ as reaction gas in ORC. Compared to the normal mode (no cell gas), the $H_2$ cell gas mode improved the accuracy and precision. The quantitative result was average 102.7% and it was slightly higher than certified standard value of milk powder and the RSD was 7.6%.

Dermal Penetration Rate and Pharmacokinetics of the Insecticide Methidathion in Sprague-Dawley Rats

  • The skin penetration rate of methidathion in vitro and pharmacokinetics of methidathion in vivo were studied with male Sprague-Dawley rats by dermal treatment. The in vitro skin penetration rates for Sprague-Dawley rats of methidathion technical (50 mg, 100 ${mu}ell$) and emulsifable concentrate (EC,40mg, 100${mu}ell$) were determined as 18.4 $\mu\textrm{g}$/c $m^2$/h (RSD : 6.5) and 18.5 $\mu\textrm{g}$/c $m^2$/h (RSD : 3.2), respectively. Dose-related systemic exposure (AUC) was observed in rats after dermal treatment. The corresponding AUC, $T_{max}$, $C_{max}$, and $T_{1}$2/ of methidathion in plasma were 1.5$\mu\textrm{g}$.hr/ml, 6 h, 0.10 $\mu\textrm{g}$/ml, and 16 h, for 116mg/kg doses, 3.2 $\mu\textrm{g}$. hr/ml, 8 h, 0.12 $\mu\textrm{g}$/ml, and 23 h, for 232 mg/kg doses and 10 $\mu\textrm{g}$. hr/ml, 12 h, 0.32 $\mu\textrm{g}$/ml, and 20 h, for 1,160 mg/kg doses respectively. The urinary excretion of methidathion, estimated wing an equation derived from the in vitro skin penetration study was 0.24~0.35% of the absorbed dose. The concentration of methidathion in kidney was higher than that in liver. Dose-dependent absorption and excretion of methidathion without saturation was observed under in vivo experimental condition.n.n.

Binding of Calcium and Iron by Job′s tears (Yulmoo: Coix lachryma-jobi L. var. Ma-yuen Stapf.) Total Dietary Fiber and Acid Detergent Fiber (율무 식이섬유의 칼슘, 철분과의 결합특성)

  • This study was performed to examine the mineral binding properties of total dietary fiber (TDF) and acid detergent fiber (ADF) from Job's tears (Yulmoo: Coix lachryma-jobi L. var. Ma-yuen Stapf.) bran. The endogenous Ca and Fe contents of dietary fiber extracted from Yulmoo bran were measured by atomic absorption spectrophotometer (AAS). Ca contents were 40, 118.6$\mu\textrm{g}$ Ca/g TDF and 131.6 $\mu\textrm{g}$ Ca/g ADF. Fe contents were 1,219.7 $\mu\textrm{g}$ Fe/g TDF and 277.5 $\mu\textrm{g}$ Fe/g ADF. The calcium binding capacities of dietary fiber at pH 5.0, 6.0, 7.0 were 50.77, 77.57, 86.64% for TDF, 40.27, 51.42, 40.90% for ADF, respectively. The iron binding capacity of dietary fiber at pH 5.0~7.0 was 95~97% for TDF and 85~95% for ADF.

Luminous efficiency of ZnS:Sm,F TFEL devices (ZnS:Sm,F 형광체 박막 EL 소자의 발광효율)

  • ZnS:Sm, F TFEL devices with double insulating layer are prepared by e-beam evaporation method. Electroluminescence and luminous efficiency of the device fabricated at various conditions are investigated. The main transitions on the emission spectra for ZnS:Sm, F TFEL device occur at$^4G_{5/2}\to^6H_{9/2}^4G_{5/2}\to^6H_{7/2}, \;^4G_{5/2}\to^6H_{5/2}\to$.Among them, the dominant spectral line and its corresponding transition occur at $^4G_{5/2}\to^6H_{9/2}$(650 nm) and results in an orange-red emission color. The optimum concentration and substrate temperature for the ZnS:Sm, F TFEL device are around 1 wt% and $200^{\circ}C$. Luminous efficiency for the device is the largest at optimum condition.
