꾸지뽕잎(Cudrania tricuspidata) 추출물의 3TS-L1 세포 분화 억제 (Inhibition of Adipogenesis in 3T3-Ll Adipocytes with Water and Ethanol Extracts of Cudrania tricuspidata Leaves)

  • 꾸지뽕잎의 물 및 70% 에탄올 추출물에 대한 세포의 독성 및 지방생성에 미치는 영향과 DNJ 함량과 rutin 함량을 측정하였다. Cell viability test에서 꾸지뽕잎 추출물 0.5-5 mg/mL의 농도일 때 물 및 에탄올 추출물 모두 세포의 생장에 영향을 미치지 않는 것으로 나타나 세포에 대한 독성이 없다고 판단되었다. 동일한 농도로 3T3-L1 지방세포의 지방축적에 미치는 영향은 물 및 에탄올 추출물 모두 농도 의존적으로 지방축적을 저해하는 것으로 나타났으며 추출물 농도가 5mg/mL 일 때, 물 추출물은 86%, 에탄올 추출물 50%의 지방축적을 보였다. DNJ 함량은 꾸지뽕잎의 에탄올 추출물에서 $3921.3{\mu}g/g$로 높았고 rutin의 함량은 꾸지뽕잎의 물 추출물에서 $4592.9{\mu}g/g$로 높게 나타냈다. 따라서 꾸지뽕잎 추출물에는 기능성 성분인 DNJ 함량과 rutin 함량이 다량 함유되어 있고 지방의 축적을 저해하는 효과가 있어 비만과 대사관련 질환개선을 위한 기능성 소재로 활용될 수 있다고 생각한다.

Rutin, Catechin Derivatives, and Chemical Components of Tartary Buckwheat (Fagopyrum tataricum Gaertn.) Sprouts

  • The aim of this study was to develop the tartary buckwheat (Fagopyrum tataricum Gaertn.) sprouts and to clarify the biological and chemical characteristics of the sprouts. At 7 days after seeding, hypocotyls length and thickness, and root length of tartary buckwheat sprouts were 137 cm, 1.4 mm, and 12.6 cm, respectively. Fresh weight, dry weight, and moisture contents of an individual sprout at 7 days after seeding were 202 mg, 5.4 mg, and 95.3%, respectively. Protein content in tartary buckwheat sprouts was 23.0% which relatively higher than that of seeds, while lipid and ash contents were 3.5% and 5.3%. Among 7 minerals, the content of phosphorus showed the highest level (1,383.5 mg/100 g), while the contents of sodium and potassium were 1,197.5 mg/100 g and 1,106 mg/100 g, respectively. The contents of other minerals were Mg (795.5 mg/100 g), Ca (149 mg/100 g), Zn (16.4 mg/ 100 g), and Fe (14.7 mg/100 g). The rutin content of tatary buckwheat sprouts including root parts was the highest (5644.9 mg/100 g) at 7 days after seeding. The concentration of catechin derivatives in tartary buckwheat sprouts was high in order of catechin (59 mg/100 g), epicatechin gallate (47 mg/100 g), and epicatechin (14 mg/100 g).

Effect of SiO2 on yield and phenolic profile of Tartary buckwheat

  • The aim of this study was to enhance the Tartary buckwheat growth and phenolic profile by the application of $SiO_2$ on potsin the glasshouse. The liquid of $SiO_2$ were applied at three times after three weeks of sowing at ten days interval. The doses of the $SiO_2$ was low (2.5 ml/5 L water), high (10 ml in 5 L water) and control experiment was done without $SiO_2$. In this study, it is clearly shown that high dose of $SiO_2$ increased the buckwheat plant growth including, plant height, leaf length and width, stem diameter, fresh weight and number of seed per plant compared to control treatment. In the same way, the total phenolic compound (1421 mg/100 g), total flavonoid (35.1 mg/100g), rutin (3,130 mg/100g) and DPPH (82%) in plant were also increased high dose of $SiO_2$ compared to control (1,175 mg/100g, 31.9mg/100g, 860 mg/100g, 59%, respectively). Moreover, higher phenolic compound (1,421 mg/100g) and DPPH (82%) was observed in plant compared to seed (196 mg/100mg, 72 %, respectively). In the contrary, total flavonoid (36.2 mg/100g) and rutin (1,400 mg/100g) was higher in buckwheat seed compared to plant (35.1 mg/100g, 3,130 mg/100g, respectively). Finally, it is concluded that higher dose of $SiO_2$ enhance buckwheat growth and phenolic profile. Further investigation is needed to evaluate the optimum dose of $SiO_2$ according to soil conditions in the field.

루틴의 피부 흡수 증진을 위한 탄성 리포좀 제형 연구 (Elastic Liposome Formulation for Transdermal Delivery of Rutin)

  • 본 연구에서는 항산화 활성이 우수한 루틴을 담지한 탄성 리포좀을 제조하여, 이 제형의 물리적 특성과 증진된 피부 투과 효과를 평가하였다. 탄성 리포좀은 인지질과 계면활성제의 비율을 달리하여 제조하였다. 루틴을 담지한 탄성리포좀의 평균 입자 크기는 205.7 ~ 298.0 nm, 가변형성은 20.9 ~ 42.5, 포집효율은 52.0 ~ 71.0 %로 측정되었다. 0.1 % 루틴을 담지한 탄성 리포좀 중에서 인지질과 계면활성제 비율이 85 : 15 인 경우가 가장 높은 포집효율(71.0 %)과 가변형성 지수(42.5)를 나타내었다. 이 제형을 대상으로 피부 투과 실험을 진행하였다. 그 결과 대조군으로 사용된 일반 리포좀(98.0 ${\mu}g/cm^2$)과 1,3-butylene glycol (76.3 ${\mu} g/cm^2$) 용액보다 탄성 리포좀의 피부 투과능(129.9 ${\mu}g/cm^2$)이 훨씬 더 크게 나타났다. 이러한 결과들로 미루어 보아 $Tego^{(R)}$ care 450을 이용한 탄성 리포좀이 피부를 통한 유효성분 전달에 유용하게 이용될 수 있음을 확인하였다.

Quantitative Analysis of Bioactive Compounds in the Fruits of Crataegus pinnatifida by High-Performance Liquid Chromatography

  • In order to facilitate the quality control of the fruits of Crataegus pinnatifida, a simple, accurate and reliable HPLC method was developed for the simultaneous determination of the three bioactive compounds: chlorogenic acid (1), rutin (2), and hyperin (3), which were selected as the chemical markers of C. pinnatifida. Separation was achieved on an Agilent Eclipse XDB-C18 column with a gradient solvent system of 0.1% trifluoroacetic acid aqueous-acetonitrile at a flow-rate of 1.0 mL/min and detected at 254nm. All three calibration curves showed good linearity ($R^2$ > 0.998). The recoveries of three marker compounds were in the range of 94.87~111.52 %. The contents of chlorogenic acid (1), rutin (2), and hyperin (3) of the fruits of C. pinnatifida collected from 23 district markets in Korea, Japan, and China were 0.16~0.65 mg/g, 0.07~1.24 mg/g, and 0.03~0.62 mg/g, respectively. The results demonstrated that this method is simple and reliable for the quality control of the fruits of C. pinnatifida.

발아중 메밀의 영양성분의 변화 (Nutritional Changes of Buckwheat During Germination)

  • To produce buckwheat sprouts, buckwheats were germinated at $23{\pm}2^{\circ}C$ up to 8 days in total darkness. In proximate analysis, moisture content increased from 13.87% of buckwheat groats to 93.75% of buckwheat sprouts on the 8th day of germination. On dry weight basis, lipid and ash contents increased like as protein content increased from 13.45% to 21.82% while carbohydrate content decreased due to enzyme hydrolysis. Amino acids were rich in glutamic acid, aspartic acids, and lysine, and also the ratio of the essential amino acids to total amino acids increased from 26.84% to 36.84%. Vitamin C did not exist in buckwheat groats but its content has continued to increase as far as 99.56 mg/100 g buckwheat sprouts. Rutin continued to increase by HPLC analysis from 4.71 mg/100 g buckwheat groats to 1,524 mg/100 g buckwheat sprouts. In analysis of organic acids, oxalic, maleic, and citric acids were commonly found in buckwheat sprouts. Fagopyrin was found almost none in buckwheat sprouts. In conclusion, the conversion of buckwheat seeds into sprouts through germination in darkness results in physically different final product with nutritional changes such as higher content of rutin, generation of vitamin C, abundance of the essential amino acids, and the existence of fagopyrin on very little.

메밀 싹 추출물 에멀젼의 제조 및 에멀젼의 열 안정 특성 (Production and Heat-Stable Characteristics of Emulsion Made from Buckwheat Sprout Extracts)

  • 메밀 성분 중 루틴의 추출 효율을 높이기 위해 메밀을 발아한 후 루틴 및 polyphenol 추출 효율이 높은 추출방법을 선택하였다. 메밀의 발아는 흙 재배가 효율적이었으며, 8일 후에는 polyphenol 함량이 10.66 mg/g으로 메밀 종자에 함유되어 있던 polyphenol 함량의 10배 이상 높은 양을 얻을 수 있었다. 발아 메밀에서 추출된 루틴을 포함한 polyphenol 성분은 에멀젼을 제조하여 열에 대한 안정성을 높이는 데 효율이 있는지를 확인하였다. 에멀젼은 homo mixer만을 통과시키는 것보다 유압식 균질기를 통과시켰을 때 포접 효율이 높을 뿐만 아니라 열처리 온도에 대하여 열 안정성이 우수한 것을 알 수 있었다.

Heterologous Expression and Characterization of a Thermostable α-L-Rhamnosidase from Thermoclostridium stercorarium subsp. thermolacticum DSM 2910 and Its Application in the Biotransformation of Rutin

  • An α-L-rhamnosidase gene from Thermoclostridium. stercorarium subsp. thermolacticum DSM 2910 (TstRhaA) was cloned and expressed. The maximum TstRhaA activity of the protein reached 25.2 U/ml, and the molecular mass was approximately 106.6 kDa. The protein was purified 8.0-fold by Ni-TED affinity with an overall recovery of 16.6% and a specific activity of 187.9 U/mg. TstRhaA activity was the highest at 65℃ and pH 6.5. In addition, it exhibited excellent thermal stability, better pH stability, good tolerance to low concentrations of organic reagents, and high catalytic activity for p-nitrophenyl-α-L-rhamnopyranoside (pNPR). Substrate specificity studies showed that TstRhaA exhibited a high specific activity for rutin. At 60℃, pH 6.5, and 0.3 U/ml enzyme dosage, 60 g/l rutin was converted to 45.55 g/l isoquercitrin within 150 min. The molar conversion rate of rutin and the yield of isoquercitrin were 99.8% and 12.22 g/l/h, respectively. The results suggested that TstRhaA could be used for mass production of isoquercitrin.

가공 및 추출 방법에 따른 일반메밀차와 쓴메밀차의 루틴 함량 및 항산화 활성 비교 (Comparison of rutin content and quality characteristics of tea products from common buckwheat (Fagopyrum esculentum) and tartary buckwheat (Fagopyrum tataricum) by different processing and brewing methods)

  • 메밀의 식품가공기술 개발을 통해 이용성을 확대하고 새로운 소비수요를 창출하고자 일반메밀과 쓴메밀을 대상으로 가공단계 및 추출방법에 따른 메밀 볶음차의 품질 특성을 비교 분석하였다. 일반메밀의 원료, 탈피, 찌기 및 볶음 단계의 루틴 함량은 21.6, 17.8, 9.8 및 6.8 mg/100 g으로 차 가공공정이 진행됨에 따라 유의하게 감소하였다. 쓴메밀의 원료, 찌기, 탈피 및 볶음 과정의 루틴 함량은 1,679.9, 1,044.2, 703.1 및 834.5 mg/100 g으로 감소하였으나, 퀘세틴 함량은 0, 12.3, 36.0 및 32.3 mg/100 g으로 가공공정이 진행됨에 따라 증가하였다. 메밀차는 메밀의 종류, 제형 및 추출방법에 따라 루틴 함량에서 큰 차이를 보였다. 루틴 함량은 일반메밀차보다 쓴메밀차에서 평균 61배 높았다. 관능평가 결과를 종합하면, 쓴메밀차의 제형별 비교에서는 알곡보다 분말이 우림색, 풍미, 구수한 맛, 단맛 및 선호도에서 높은 값을 보였다. 특히, 루틴은 퀘세틴이나 쓴맛과 높은 상관관계를 나타냈다. 본 연구에서는 쓴메밀 품종인 '황금미소'를 대상으로 쓴맛을 최대한 줄이면서 루틴 함량이 높은 메밀차 제조를 위해 볶음 과정의 열처리 조건을 70-80℃에서 2-3분으로 확립하였다. 루틴 함량이 많은 메밀차를 섭취하고자 할 경우에는 핸드드립으로 추출한 방법이 적합하며, 쓴맛이 적고 달콤한 맛과 풍미가 좋은 메밀차를 원할 경우 침출식 콜드브루 방식을 선택하는 것이 가장 유리할 것으로 판단된다.

루틴 첨가가 고지방 식이 랫드의 혈청 지질 농도의 변화에 미치는 영향 (Effect of Rutin on Major Lipid-Related Parameters in Sera of Rats Fed High Fat Diet)

  • This study was performed to investigate the changes of the serological lipid-related parameters of the rats when they were fed with the high fat diets supplemented with or without rutin for five weeks. Twenty-four Sprague-Dawley male rats ($272.2{\pm}7.2 g$ of body weight) were randomly divided into three groups: control (C) group and two treatment groups. Rats in the C group were fed with the high-fat diet containing 20% lard, 1% cholesterol and 0.5% sodium cholate (w/w) which was modified from the formula of the American Institute of Nutrition-76 (AIN-76) diet. Rats in treatment groups were fed with above diet supplemented with 0.75% rutin (R-0.75) or 1.5% rutin (R-1.5) on the weight to weight basis, respectively. The supplementation of rutin did not induce any significant difference on the final body weight, gain of body weight, the amount of feed intake and the feed efficiency of rats in both control and treatment groups. In addition the values of glucose concentration, total protein, albumin, globulin and albumin/globulin (A/G) ratio showed no significant differences among groups. The values of total cholesterol (TC) and low density lipoprotein-cholesterol (LDL-C) in sera of rats in both R-0.75 and R-1.5 groups were lower than those in C group but the significances were showed in only between R-0.75 and C group (p < 0.05 and p < 0.01, respectively). The values of high density lipoprotein-cholesterol (HDL-C) in sera of rats in both R-0.75 and R-1.5 groups were higher than those in C group but the significances were showed in only between R-1.5 and C group (p < 0.01). The values of atherogenic index(AI) of rats in both R-0.75 and R-1.5 groups were the lower than those in C group (p < 0.01 and p < 0.05, respectively). The values of triglyceride in sera of rats showed no significant differences among groups. The values of AST and ALT in sera of rats showed no significant differences among groups. Therefore the supplementation of rutin to high fat diet in rats reduced effectively the serum lipid levels such as TC and LDL-C which were regarded as to cause the cardiovascular diseases, and moreover it elevated effectively HDL-C value which was regarded to protect cardiovascular diseases.
