Genetic effects of sterol regulatory element binding proteins and fatty acid-binding protein4 on the fatty acid composition of Korean cattle (Hanwoo)

  • Objective: This study identifies single-nucleotide polymorphisms (SNP) or gene combinations that affect the flavor and quality of Korean cattle (Hanwoo) by using the SNP Harvester method. Methods: Four economic traits (oleic acid [C18:1], saturated fatty acids), monounsaturated fatty acids, and marbling score) were adjusted for environmental factors in order to focus solely on genetic effects. The SNP Harvester method was used to investigate gene combinations (two-way gene interactions) associated with these economic traits. Further, a multifactor dimensionality reduction method was used to identify superior genotypes in gene combinations. Results: Table 3 to 4 show the analysis results for differences between superior genotypes and others for selected major gene combinations using the multifactor dimensionality reduction method. Environmental factors were adjusted for in order to evaluate only the genetic effect. Table 5 shows the adjustment effect by comparing the accuracy before and after correction in two-way gene interactions. Conclusion: The g.3977-325 T>C and (g.2988 A>G, g.3977-325 T>C) combinations of fatty acid-binding protein4 were the superior gene, and the superior genotype combinations across all economic traits were the CC genotype at g.3977-325 T>C and the AACC, GACC, GGCC genotypes of (g.2988 A>G, g.3977-325 T>C).

유효등방복사전력(EIRP)을 활용한 5G 주파수 대역 총복사전력(TRP) 측정시간의 단축방안 검증 (Verification on the Reduction Technique of Measurement Time of Total Radiated Power (TRP) by Using Effective Isotropic Radiated Power (EIRP) in 5G Frequency Band)

  • 본 논문은 수십시간이 소요되는 총복사전력(TRP) 측정 시간을 단축하는 방안을 컴퓨터 모의실험과 실제 실험을 통하여 검증한 결과를 제시하고 있다. 5G의 고주파수 대역에서는 무선통신기기의 정확한 성능평가를 위하여 TRP 측정이 기본적으로 수행되지만 촘촘한 샘플링 간격 때문으로 인하여, 긴 측정 시간이 소요되는 문제점을 가지고 있다. 더구나 다양한 빔 형성 시나리오를 갖는 경우 전체 측정 시간은 기하급수적으로 증가한다. 따라서, TRP 측정 시간 단축 방안이 필요하며, 그 일환으로 본 논문에서는 유효등방복사전력(EIRP)과 TRP 관계식을 이용하여 측정시간을 단축하는 방법을 다루었다. 먼저 TRP와 EIRP의 관계식을 관찰하고, 피시험용 안테나의 설계 및 제작하였으며 모의실험 결과와 측정결과를 통한 오차를 분석하였다. 그 결과 EIRP를 통하여 TRP를 산출한 결과는 매우 작은 오차를 가짐을 확인하였다. 본 방법은 향후 5G 무선통신기기의 TRP 도출에 적용할 수 있을 것으로 예상한다.

Solid Phase Microextraction 및 Purge & Trap을 이용한 생물시료 중 휘발성 유기화합물의 GC/MS 분석비교 (Comparison Solid Phase Microextraction with Purge & Trap on the GC/MS Analysis of Volatile Organic Compounds in Biota Samples)

  • 생물시료에 존재하는 휘발성 유기화합물 중 n-butylbenzene과 1,2-dibromo-3-chloropropane (DBCP)를 기체크로마토그래피/질량분석기-선택이온검색법에 의해 수행하였다. 시료 중 휘발성 유기화합물은 $100{\mu}m$ polydimethyl siloxane (PDMS) fiber를 사용하여 headspace solid phase microextractio (SPME) 및 purge & trap 방법에 의해 추출 및 비교하였다. SPME에 의한 회수율은 n-butylbenzene의 경우 85.8%, DBCP의 경우 92.4%로 나타났고 검출한계는 각각 $0.15{\mu}g/kg$, $0.05{\mu}g/kg$로 나타났다. 반면, purge & trap의 경우 회수율은 n-butylbenzene의 경우 115.2%, DBCP의 경우는 80.9%로 나타났고 검출한계는 각각 $0.04{\mu}g/kg$$0.70{\mu}g/kg$로 나타나 두 방법에 있어 국내에서 규제하는 검출한계측면에서 큰 차이는 없었다.

Method for Simultaneous Determination of Anatoxin-a and Microcystins in Korean Water Systems by Using LC/MS/MS

  • This study was purposed to develop an effective LC/MS/MS method for simultaneously determining five pre-treated cyanotoxins (anatoxin-a, microcystins-RR, -YR, -LR and -LA) of cyanobacteria blooms. Cyanobacterial bloom samples were collected from 11 major lakes and three downstream areas of river around Korea during 2005~2009. Cyanotoxins were identified in 38 samples from the lakes. The validity of the method was evaluated and the recovery rates were found ranging from 83~87%. The MDL turned out to be $0.046\;{\mu}g\;L^{-1}$ for anatoxin-a and $0.066\;{\mu}g\;L^{-1}$ for microcystins (RR, YR, LR and LA), which indicates that the method has high sensitivity and accuracy. The most dominant genus of the cyanobacterial blooms was Microcystis, which accounted for 71% of the analysed samples. Microcystis also contained the largest amount of microcystins ($398.5\;{\mu}g\;gDW^{-1}$) among the analyzed cyanobacteria. The analysis of the five cyanotoxins showed that anatoxin-a ranged between $0{\sim}41.833\;{\mu}g\;gDW^{-1}$ and microcystins ranged between $6.311{\sim}2,148.786\;{\mu}g\;gDW^{-1}$. Among the microcystins, micocystin-RR took up 58.3%, the largest portion. Anatoxin-a was found to account for 77.8% of the samples. This study has its significance in that it allowed the establishment of toxin criteria appropriate for the Korean water systems. Further studies may be necessary to conduct for improving water treatment methods.

비장 조직에서 에탄올 대사체(EtG, EtS)를 검출하는 방법과 유효성 확인 (Method validation of detecting ethanol metabolites (EtG, EtS) in post-mortem spleen)

  • 에탄올 대사체인 에틸글루쿠로나이드(ethyl glucuronide, EtG)와 에틸설페이트(ethyl sulfate, EtS)는 에탄올 섭취의 직접적인 지표물질로서 여러 생체 시료에서 법과학적으로 다양하게 연구되고 있다. 부검 변사자에게서 검출된 에탄올이 사망 전 음주에 의한 것인지 사후 부패에 의해 생성된 에탄올인지 판단하기 위해서는 에탄올 대사체를 분석할 필요가 있다. 일반적으로 혈액에서 에탄올과 함께 EtG, EtS를 검출하고 있지만 부패 상황에 따라 혈액을 확보하는 것이 어려우므로 조직 생체시료를 이용해 에탄올 대사체를 검출함으로써 이를 대체해야 한다. 본 연구에서는 조직 중에서 비장을 선택하여 LC-MS/MS를 이용해 EtG, EtS를 검출하였으며 시험법이 적합한 방법인지를 증명하기 위해 밸리데이션을 수행하여 법과학적 감정에 사용할 수 있는 분석법을 완성하고 실제 부검 사례에 적용하였다.

근외적 분광분석법을 이용한 담배 중 전휘발성염기 분석 (Determination of Total Volatile Bases of Tobacco Using Near Infrared Spectroscopy)

  • This study was carried out to develop calibration equation of total volatile bases of tobacco leaf using near infrared spectroscopy(NIRS). Burley, imported flue-cured and oriental leaf tobacco samples were collected in 2005 crop year. Calibration equation was developed by modified partial least square method. The standard error of calibration and $R^2$ between traditional analytical method and NIRS analytical method were $0.038\%$, 0.983 for burley and $0.027\%$, 0.986 for imported flue-cured and oriental leaf, respectively. The standard error of performance and $R^2$ between traditional analytical method and NIRS analytical method were $0.048\%$, 0.940 for burley and $0.024\%$, 0.986 for imported flue-cured and oriental leaf, respectively. From these results, the NIRS analytical method seems to be applicable in analyzing total volatile bases of tobacco.

추출방법에 따른 잎담배 종류별 휘발성 향기성분 특성비교 (Comparison of the volatile flavor compounds in different tobacco types by different extraction methods)

  • Traditional simultaneous distillation extraction(SDE) and solid-phase micro extraction(SPME) methods using GC/MS were compared for their effectiveness in the extraction of volatile flavor compounds from different tobacco leaves types(flue-cured, burley, oriental). The major volatile flavor compounds of flue-cured and burley tobacco were similar such as neophytadiene, solanone, megastigmatrienone isomers, ${\beta}$-damascenone and ${\beta}$-ionone. On the other hand, volatile flavor compounds such as norambreinolide, sclareolide were specifically identified in oriental tobacco. Each method was used to evaluate the responses of some analytes from real samples and standards in order to provide sensitivity comparisons between two techniques. Among three types of SPME fibers such as PDMS(Polydimethylsiloxane), PA(Polyacrylate) and PDMS/DVB (Polydimethylsiloxane/Divinylbenzene) which were investigated to determine the selectivity and adsorption efficiency, PDMS/DVB fiber was selected for the extractions of the volatile flavor compounds due to its effectiveness. The qualitative analysis showed that the total amount of volatile flavor compounds in SDE method(130 species) was much more than that in SPME method(85 species). SPME method was more efficient for all the highly volatile compounds than SDE method, but on the other hand, low-volatile compounds such as fatty acids or high-molecular hydrocabons were detected in SDE method. SPME method based on a short-time sampling can be adjusted to favor a selected group compounds in tobacco. Furthermore this results could be used to estimate the aroma characteristics of cigarette blending by using a different type of tobacco with more effectiveness and convenience.

An HPLC-UV-based quantitative analytical method for Chrysanthemum morifolium: development, validation, and application

  • A simple and reliable analytical method based on high-performance liquid chromatography-ultraviolet detection was established for the analysis of the flowers of Chrysanthemum morifolium (CM). Luteolin-7-O-glucoside (LU7G) was chosen as a target analyte considering its content, availability, and ease of analysis. Chromatographic separation of LU7G was achieved using a Phenomenex Gemini $C_{18}$ column ($250{\times}4.6mm$, $5{\mu}m$) run with a mobile phase consisting of 0.5 % acetic acid in water and 0.5 % acetic acid in acetonitrile at a flow rate of $1.0mL\;min^{-1}$. The detection wavelength and column temperature were set at 350 nm and $40^{\circ}C$, respectively. Method validation was performed according to the AOAC guidelines and the method was specific, linear ($R^2=0.9991$ for $50-300{\mu}g\;mL^{-1}$), precise (${\leq}3.91%$RSD), and accurate (100.1-105.7 %). The limits of detection and quantification were 3.62 and $10.96{\mu}g\;mL^{-1}$, respectively. The established method was successfully applied to determine the contents of LU7G in various batches of bulk CM extracts and labscale CM extract. The developed method is a readily applicable method for the quality assessment of CM and its related products.

어류에서의 과불화합물 분석을 위한 전처리법 확립 및 모니터링 (Monitoring and Development of Pretreatment Method for Perfluorinated Compounds in Fish Samples)

  • 본 연구에서는 어류에서 16종의 과불화합물을 분석하기 위한 최적 전처리법을 확립하고자 이온쌍 추출법(IPE)과, 액-액 추출법(LLE), 고체상 추출법(SPE), 이온쌍 + 고체상 추출법(IPE + SPE) 각각에 대하여 내부표준물질의 주입 시기를 달리하여 회수율을 비교하였다. 내부표준물질을 추출 전에 주입하는 IPE-before법이 가장 효율적인 전처리법으로 평가되었다. 추가로 RPM 및 pH 테스트를 통해 IPE-before법을 적용한 분석 전처리 효율을 더 개선할 수 있었다. 확립된 전처리법을 이용해 국내 마트에서 구입한 33개의 어류 및 어류의 간과 내장시료에서 과불화합물을 분석하였다. 16종 과불화합물의 검출범위는 ND~1.67 ng/g이었으며, 검출빈도는 100%로 나타났다. 어류의 근육부와 간, 내장을 비교 모니터링 한 결과, 간(17.8 ng/g) > 내장(13.3 ng/g) > 근육부(1.67 ng/g) 순으로 과불화합물이 검출됨을 확인하였다. 국외 선행연구 결과와 비교하였을 때, 본 연구에서의 어류의 농도는 비슷하거나 낮게 검출되었다.

잎담배 중 neutral volatile flavor 화합물 분석 (The Analysis of Neutral Volatile Flavor Compounds in Tobacco)

  • This work has been conducted to develop a method for the analysis of neutral volatile flavors and their precursors in tobacco. The neutral volatile compounds and precursors in tobaccos have been investigated by Neutral Volatile scan method(NV scan) using Soxhlet extractor. The method has been used to analyze a range of different tobaccos and tobacco products. Neutral flavor compounds were classified as three sections(1st Volatile Fraction, Breakdown Flavor Products and Cembranoid Precursors). The major components of the First Volatile Fraction were 2-cyclohexene-1-one, 6-methyl-5-hepten-2-one, limonene and phenyl ethanol. The major components of Breakdown Flavor Products were isophorone, solanone, damascenone, 3-hydroxy-$\beta$-damascone, geranyl acetone, $\beta$-ionone, dihydroactinidiolide, norsolanadione, neophytadiene, hexahydrofarnesylacetone, farnesyl acetone and megastigmatrienone. The major cembranoid precursor compounds were dibutyl phthalate, duvatrenediols, 8,12-epoxy-14-labden-13-ol, 11-hydroperoxy-2,7,12(20)-cembratriene-4,6-diol, 12,15-epoxy-12,14-labadien-8-ol, 2,7,11-cembratrien-4,6-diol and 8,13-epoxy-14-labdien-12-ol. The NV scna results of tobacco types(flue-cured, burley and oriental) showed that each tobacco type has a characteristic flavor component profile.
