The G23 and G25 Genes of Temperate Mycobacteriophage L1 Are Essential for The Transcription of Its Late Genes

  • Two lysis-defective but DNA synthesis non-defective temperature-sensitive (ts) mutants of mycobacteriophage L1, L1G23ts23 and L1G25ts889 were found to be defective also in phage-specific RNA synthesis in the late period of their growth at 42$^{\circ}C$each to the extent of 50% of that at 32$^{\circ}C$The double mutant, L1G23ts23G25ts889 showed the ts defect in phage RNA synthesis that was nearly additive of those shown individually by the two single-mutant parents. Both G23 and G25 were shown to start functioning sometimes between 30 and 45 min after infection but the former gene might be dispensable after 45 min, while the latter was not. Northern analysis also shows that at 42$^{\circ}C$>, L1G23ts23 affects RNA synthesis more strongly than L1G25ts889 from L1 DNA segments that serve as the template for late gene transcription. Among the 21 virion and 12 non-virion late proteins synthesized by L1, L1G23ts23 is defective in the synthesis of at least 9 virion and all of non-virion proteins at 42$^{\circ}C$>. In contrast, L1G25ts889 is completely defective in synthesis of all the 33 late proteins. Possible roles of G23 and G25 in the positive regulation of transcription of different sets of late genes of L1 have been discussed.

Correlation between Serum D-Dimer Level and Volume in Acute Ischemic Stroke

  • Objective : D-dimer is a breakdown product of fibrin mesh after factor XIII stabilization. Previously, many authors have demonstrated a relationship between D-dimer level and stroke progression or type. This study aimed to investigate the relationship between D-dimer level and stroke volume. Methods : Between January 2008 and December 2009, we analyzed the D-dimer levels of 59 acute ischemic stroke patients in our neurosurgical department both upon admission and after seven days of initial treatment. Each patient's National Institute of Health Stroke Scale score, modified Rankin Scales score, Glasgow outcome score, and infarction volume were also evaluated. Results : Mean D-dimer level at admission was 626.6 ${\mu}g/L$ (range, 77-4,752 ${\mu}g/L$) and the mean level measured after seven days of treatment was 238.3 ${\mu}g/L$ (range, 50-924 ${\mu}g/L$). Mean D-dimer level at admission was 215.3 ${\mu}g/L$ in patients with focal infarctions, 385.7 ${\mu}g/L$ in patients with multiple embolic infarctions, 566.2 ${\mu}g/L$ in those with 1-19 cc infarctions, 668.8 ${\mu}g/L$ in 20-49 cc infarctions, 702.5 ${\mu}g/L$ in 50-199 cc infarctions, and 844.0 ${\mu}g/L$ in >200 cc infarctions (p=0.044). On the 7th day of treatment, the D-dimer levels had fallen to 201.0 ${\mu}g/L$, 293.2 ${\mu}g/L$, 272.0 ${\mu}g/L$, 232.8 ${\mu}g/L$, 336.6 ${\mu}g/L$, and 180.0 ${\mu}g/L$, respectively (p=0.530). Conclusion : Our study shows that D-dimer level has the positive correlation with infarction volume and can be use to predict infarction-volume.

Seasonal Variations of Chemical Composition of the Estuary Water at the Tidal Flat in Nack Dong River from Nov. 1962 to Oct. 1963 (洛東江 河口 干潟地 水質의 年間變化 (1962年 11月~1963年 10月))

  • Seasonal variations of the contents of some chemical constituents of the estuary water at two definite stations of the laver bed in Nack Dong River have been determined over one tidal cycle in spring tide from Nov. 1962 to Oct. 1963. The ranges of annual variations of the contents at station 1 and station 2 are as follows: water temp. $2.2-30.8^{\circ}C$, $3.3-28.0^{\circ}C$; pH 7.8-8.5, 7.9-8.4; chlorosity 0.025-19.66 g/l, 4.31-19.56 g/l; magnesium 0.00355-1.565 g/l, -1.524 g/l; calcium 0.00557-0.482 g/l, - -0.590 g/l; saturation % of dissolved oxygen 71.8-123.2%, 88.2-113.8%; silicate-Si 8.00-125.5 ${\mu}$g-at./l, 6.70-100.5 ${\mu}$g-at./l; phosphate-P 0.12-1.47 ${\mu}$g-at./l, 0.11-1.09 ${\mu}$g-at./l; ammonia-N 4.88-25.45 ${\mu}$g-at./l, 4.12-17.58 ${\mu}$g-at./l; nitrite-N 0.07-0.75 ${\mu}$g-at./l, 0.08-0.58 ${\mu}$g-at./l; nitrate-N 2.11-6.89 ${\mu}$g-at./l, 1.85-7.43 ${\mu}$g-at./l each. The annual tidal variations of the constituents at station 1 are more remarkable than of station 2. The chlorosity, magnesium and calcium contents are decreased nearing the slack after ebb, and increased abruptly then one hour after the slack. The contents of the other constituents are varied according to the chlorosity variety. The values of pH, chlorosity, magnesium and calcium contents are lower in summer than winter, while the difference of seasonal variations of the % saturation of dissolved oxygen is not remarkable. The phosphate-P and total nitrogen contents have a tendency of increasing within a definite range, while the silicate-Si increase proportionally, to the increasing of mixing percentage of fresh water. The average values of Si/P and N/P are several times greater than of the normal in sea water. The chemical composition considered from the value of Mg/Cl or Ca/Cl of estuarine water varies according to the variety of chlorosity, even at the high chlorosity of 19 g/l.

Effects of Culture Conditions on Mycelial Growth and Polysaccharide Production of Tricholoma matsutake in Bioreactor

  • This experiment was carried out to obtain the optimal liquid culture conditions for the mycelial growth and the polysaccharide production of Tricholoma matsutake. For the mycelial growth and polysaccharide production, the synthetic medium was optimized with containing glucose 40 g/L, yeast extract 30 g/L, $KH_2PO_4$ 1.5 g/L and $MgSO_4.7H_2O$ 1 g/L. The effects of agitation and aeration were investigated for the cell growth and the polysaccharide production in batch culture. The biomass and polysaccharide concentrations were 21.87 g/L at 150 rpm and 8.86 g/L at 300 rpm, respectively. And the biomass concentration and the polysaccharide production were 20.85 g/L at 0.5 vvm and 8.83 g/L at 1.5 vvm, respectively.

Continuous Alcohol Fermentation by a Flocculating Yeast (응집성 효모를 이용한 연속 알코올 발효)

  • In this study continuous alcohol fermentation of molasses by the recirculation system has investigated. After cultivation of yeast cells in the YPD medium with increasing the medium concentration from 10 to 183.5 g/l stepwisely, the fermentation medium was replaced by molasses. The maximum cell mass was 25 g/l, and the mean cell mass during the operation was 23.5g/1, which was 3.4 times higher compared with a conventional batch system. The optimum fermentation conditions with feeding molasses of 180 g/l were obtained when the fermentation was carried out at 500 rpm and at the dilution rate of 0.037 $h^{-1}$. Under these conditions we could safely operate the fermentor for 645 h without any trouble. The maximum alcohol productivity was 4.9 g$l\cdot h$ with an alcohol concentration of 53.9 g/l at the dilution rate of 0.091$h^{-1}$.

The Study of Radon and Uranium Distribution in the Groundwater at Regional Difference of Daejeon (대전지역별 지하수중의 우라늄, 라돈 농도준위 분포조사 연구)

  • To know radon and uranium concentration variations in groundwater with regional difference, groundwaters were sampled at five different Daejeon area. Seventy-five samples were analyzed and forty samples were collected twice at drying and after raining season to know surface water effect. The average radon and uranium concentration of five areas are $270.9{\pm}152.3\;Bq/L,\;43.8{\pm}23.5\;{\mu}g/L$ at Yusung-gu, $112.9{\pm}65.8\;Bq/L,\;0.45{\pm}0.23\;{\mu}g/L$ at Seo-gu, $41.3{\pm}24.0\;Bq/L,\;4.9{\pm}11.3\;{\mu}g/L$ at Dong-gu, $131.8{\pm}99.5\;Bq/L,\;54.3{\pm}127.5\;{\mu}g/L$ at Daeduk-gu and $44.0{\pm}43.0\;Bq/L,\;8.1{\pm}11.6\;{\mu}g/L$ at Jung-gu. The mean concentrations of analyzed samples to know surface water effect were ranged from 0.5 to 640 ${\mu}g/L$ for uranium and from 0.4 to 729 Bq/L for radon. The average concentration of radon and uranium after raining season were lower than those of drying season. The mean contents of radon End Uranium at drying season were $253{\pm}14\;Bq/L,\;63{\pm}12.2\;{\mu}g/L$, and $195{\pm}11\;Bq/L,\;45.4{\pm}11.7\;{\mu}g/L$ after raining season.

Monitoring the Manufacturing Characteristics of Aloe Gel-State Food (알로에 겔상 식품의 제조특성 모니터링)

  • Four-dimensional response surface methodology was used for monitoring the manufacturing characteristics of aloe gel-state food. The optimum conditions predicted for each corresponding sensory properties of aloe gel-state food were 87.38 mL (content of aloe juice), 0.16 g (content of konjac) and 0.19 g (content of carrageenan) in coloror of gel-state food, 83.84 mL, 0.17 g and 0.20 g in aroma of aloe gel-state food, 83.20 mL.0.15 g and 0.27 g taste of aloe gel-state food and 98.95 mL, 0.10 g and 0.23 g in texture of aloe gel-state food. Maximum chewiness of aloe gel-state food was in 113.05 mL aloe juice, 0.27 g konjac and 0.21 g carrageenan. The optimum conditions, which satisfied all sensory properties of gel-state food, were 88.23 mL, 0.15 g and 0.49 g in content of aloe juice, content of konjac and content of carrageenan, respectively.

The State of Marine Pollution in the Waters adjacent to Shipyards in Korea - 2. Assessment of the Pollution of Heavy Metals in Seawater around Major Shipyards in Summer 2010 (국내 조선소 주변해역의 해양오염 현황 - 2. 2010년 하계 대형조선소 주변 해수의 중금속오염 평가)

  • In order to elucidate the current state of marine pollution of heavy metals around major shipyards in Korea, seawater samples were collected at eleven sampling stations and four control stations around 4 major shipyards located in the southeastern coast of Korea in summer 2010, and 6 kinds of metals such as copper(Cu), zinc(Zn), iron(Fe), cadmium(Cd), lead(Pb) and mercury(Hg) in seawater samples were analyzed. The analyses of heavy metals in seawater showed that the mean Cu concentrations in seawater around 4 major shipyards were in the range of $0.817{\sim}1.638{\mu}g/L$ which were lower than Korean environmental standards of $20{\mu}g/L$ for the protection of human health(PHH) and of $3{\mu}g/L$ for short-term protection of marine ecosystem(SPME) but higher than Cu concentration at control station by a factor of up to 2.75. The mean Zn concentrations were in the range of $0.228{\sim}0.567{\mu}g/L$ which were lower than Korean environmental standards of $100{\mu}g/L$ for PHH and $34{\mu}g/L$ for SPME but higher than Zn concentration at control station by a factor of up to 5.91. The mean Fe concentrations were in the range of $3.332{\sim}7.410{\mu}g/L$ which were higher than Fe concentration at control station by a factor of up to 6.75. The mean Cd concentrations were in the range of $0.013{\sim}0.028{\mu}g/L$ which were lower than Korean environmental standards of $10{\mu}g/L$ for PHH and $19{\mu}g/L$ for SPME but higher than Cd concentration at control station by a factor of up to 2.33. The mean Pb concentrations were in the range of $0.007{\sim}0.126{\mu}g/L$ which were lower than Korean environmental standards of $50{\mu}g/L$ for PHH and $7.6{\mu}g/L$ for SPME. The mean Hg concentrations were in the range of $0.002{\sim}0.004{\mu}g/L$ which were lower than Korean environmental standards of $0.5{\mu}g/L$ for PHH and $1.8{\mu}g/L$ for SPME. Although the concentrations of metals such as Cu, Zn and Fe which were used in shipbuilding works were lower than Korean environmental standards for PHH and SPME, the fact that the concentrations of Cu, Zn and Fe at sampling stations around major shipyards were higher than those at control stations implies that the works in shipyards had some effects on marine water quality around shipyards. Therefore, marine environment management such as the prevention and control of the discharge of various pollutants from shipyards is required on national level.

Growth Performances of Container Seedlings of Deciduous Hardwood Species Grown at Three Different Fertilization Treatments (시비처리에 따른 활엽수 용기묘의 생장 특성 변화)

  • The objective of this study was to find optimal amount of fertilization of container seedling production for Zelkova serrata, Ulmus parvifolia, Betula costata and Tilia amurensis. To reach our goal, we measured root collar diameter (RCD), height, biomass and seedling quality index (SQI) of container seedlings of four deciduous hardwood species grown at three different fertilization treatments ($0.5g{\cdot}L^{-1}$, $1.0g{\cdot}L^{-1}$ and $2.0g{\cdot}L^{-1}$). Z. serrata seedlings grown at $2.0g{\cdot}L^{-1}$ fertilization and B. costata seedlings grown at $0.5g{\cdot}L^{-1}$ fertilization showed the highest RCD, height, biomass and SQI. The RCD and height of U. parvifolia and T. amurensis seedlings showed no significant differences by fertilization treatments. Seedlings of two species at $0.5g{\cdot}L^{-1}$ fertilization showed the lowest SQI, however, SQI at 1.0 and $2.0g{\cdot}L^{-1}$ fertilization treatments were not significantly different. Based on these results, it is appeared that container seedlings of Z. serrata at minimum $2.0g{\cdot}L^{-1}$ fertilization, U. parvifolia and T. amurensis at $1.0g{\cdot}L^{-1}$ fertilization and B. costata at maximum $0.5g{\cdot}L^{-1}$ fertilization were optimal nutrient conditions. Practice of optimal fertilization rate will make us get better quality seedlings and reduction of production costs in the container nursery system as well as good field performances with higher survival rate after planting.

Growth Characteristics of Azotobacter vinelandii UWD Using Glucose and Organic Acids as Substrates (포도당 및 유기산을 이용한 Azotobacter vinelandii UWD 의 생장 특성)

  • Azotobacter vinelandii UWD synthesizes poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV), one of the biodegradable polymers, when odd and even number carbon sources are simultaneously added to a medium. In this study, we investigated the specific growth rate of Azotobacter vinelandii UWD on propionic acid and valeric acid. The specific growth rates were $0.183 hr^{-1} and 0.137 hr^{-1}$ at 1.0∼1.5 g/L of propionic acid and 1.0 g/L of valeric acid, respectively. When a mixture of 0.75 g/L of propionic acid and 0.5 g/L of valeric acid was added to the medium, the specific growth rate was 0.196 hr(sup)-1, which was equal to or higher than those of the individual organic acids. Among 10∼50 g/L of glucose cell growth was best at 20 g/L.
