Hematological Aspects in A Endotoxemic Young Rabbit Model

  • Gram-negative septicemia, which continues to be a serious clinical problem, is one of the major causes of morbidity and mortality in hospitalized patients. Endotoxin of gram-negative bacteria is a pivotal pathogen of sepsis. To understand the effect of endotoxin on hematological aspect and the time course in early childhood, this study was designed with experimental septic model of young rabbits (8 week-old). Rabbits were divided into control (n=7) and endotoxin group (0.50 mg/kg of endotoxin). The endotoxin group was subdivided into six groups by the sampling times: 3, 6, 12, 24, 48 and 72 hr-group (E-G$_{3}$, E-G$_{6}$, E-G$_{12}$, E-G$_{24}$, E-G$_{48}$ and E-G$_{72hrs}$, each n=7). The evaluation of CBC, activated partial thromboplastin time (APTT), prothrombin time (PT), fibrinogen concentration, coagulation factors and D-dimer were taken from the bloods. The number of leukocytes was lower in E-G$_{3}$ and E-G$_{6hrs}$ (due to pantocytopenia), whereas it was higher in E-G$_{24}$ and E-G$_{48}$ (due to neutrophilia and/or lymphophilia) than in control group (P<0.05). Platelet counts in E-G$_{3}$, E-G$_{6}$, E-G$_{12}$, E-G$_{24}$ and E-G$_{48hrs}$ were lower than those of control group (P<0.05). Normoblast counts in E-G$_{3}$, E-G$_{6}$, E-G$_{12}$, E-G$_{24}$ and E-G$_{48hrs}$ were higher than those of control group (p<0.01). APTT in E-G$_{3}$, E-G$_{6}$, E-G$_{12}$, E-G$_{24}$ and E-G$_{72hrs}$ were longer while PT in E-G$_{3}$, E-G$_{6}$, E-G$_{48}$ and E-G$_{72hrs}$ were higher than those of control group (p<0.05). Fibrinogen concentrations were lower in E-G$_{3}$, E-G$_{6}$ and E-G$_{12}$ but higher in E-G$_{48}$ and E-G$_{72hrs}$ than those of control (p<0.05). Intrinsic coagulation factors (XII, XI, IX, VIII) in all endotoxin groups were significantly lower than those of control group (p<0.05). Extrinsic coagulation factor (X, VII, V, II) were lower in E-G$_{3}$, E-G$_{6}$, E-G$_{12}$ and E-G$_{24hrs}$ whereas they were higher in E-G$_{48}$ and E-G$_{72hrs}$ than in control group (p<0.05). D-dimer concentrations in E-G$_{48}$ and E-G$_{72hrs}$ were higher than those of control group (P<0.001). We concluded that endotoxin led to extensive hematological disturbances including disseminated intravascular coagulation in the young rabbits and that this pathologic condition in the infant and childhood groups will cause the grave results.

인간 전과립 세포로부터 항미생물 인자의 생산에 관한 연구 (Studies on Producing Anti-microbial Factor from Human Promyelocytic Cells)

  • 본 실험에서는 인간 전과립 세포를 회분배양 조건 에서 10%의 혈청을 포함하는 배지로 배양하였다. 이 와같은 조건에서 세포의 비성장율은 0.374C1/day) 이고 세포내 존재하는 살균활성을 가진 목적 단백질 의 생산성은 0.435(mg/10' viable cells)으로 나타났 다. 세포내 존재하는 목척 단백질의 순수 분리를 위 해 sephadex G-75, sephadex G-100, Bio→Rex70 을 이용해 분리하였다. 정제된 목적 단백질은 감수성 균주, E. coli IFO 13168과 St. aureus IFO 3060 을 표준 균주로 살균 측정을 하였을 때, 기존의 항생 물질은 균주에 대해 40~701땅1m!에서 활성을 나타 낸 반면, 정제된 목적 단백질은 G(+)균(St. aureus IFO 3060)에서는 $0.5\mu\textrm{g}$/ml이고, G(-)균(E. coli IFO 13168)에서는 $0.4\mu\textrm{g}$/ml에서 살균 활성을 보였다. 따라서 AMF가 기존 항생물질보다 살균 활성이 강력한 단백질임을 알 수 있다. 해당 분자량을 측정 하기 위해 SDS-PAGE로 측정한 결과 약 15,000 Dalton의 분자량을 갖고 있음이 확인됐다.

단백질합성인자 eIF5B의 저 발현 효모벡터의 제조 및 특성 (Construction and Characterization of Vector Expressing Low Level of Translation Factor eIF5B)

  • eIF5B는 단백질합성의 개시 인자로서 Met-$tRNA^{Met}$을 AUG 개시코돈에 전달하고, 리보솜의 두 소단위체 결합을 유도한다. 이 인자의 기능을 연구할 목적으로 eIF5B를 코딩하는 FUN12 유전자의 5'말단 부위를 삭제하는 연구를 수행하였다. 프로모터의 대부분을 삭제한 FUN12를 함유한 pRS효모벡터를 FUN12가 삭제되어 천천히 자라는 돌연변이주 ($fun12{\Delta}g$)에 전달하였을 때 그 표현형을 부분적으로 상보하였다. 위와 같이 제조된 벡터에서 N-말단이 상실된 eIF5B 단백질이 발현되었고, 그 양이 정상 균주에서 발현되는 eIF5B 양의 약 5%에 불과하였다. 이와 같이 부분적으로 성장을 상보한 균주에서 발현된 적은 양의 단백질합성개시 인자 eIF5B는 직접적으로 그 성장을 제한하는 요소로 작용한다. 이러한 균주에서 성장의 제한인자인 eIF5B는 in vitro 에서도 역시 전체 단백질 합성의 활성을 조절하였다.

CJ-50001 (rG-CSF)의 일반약리작용 (Giniral pharmacology of CJ-50001 (rG-CSF))

  • CJ-50001 is a recombinant granulocyte-colony stimulating factor (rG-CSF) synthesized by recombi-nant DNA technology using E. coli as an expression system. The general pharmacological properties of CJ-50001 were evaluated in mice, rats, dogs and isolated guinea pig ileum. The doses are 100, 300 and 1, 0007g/kg, i.v. for mice and rats, 1, 10 and 100$\mu$g/kg, 1.v. for dogs and 1 and 10$\mu$g/ml for isolated guinea pig ileum. Intravenous administration of CJ-50001 at this dose range did not affect general behavior, central nervous system, smooth muscles, gastrointestinal system, cardiovascular and respiratory system and water and electro-lytes excretion. In summary, CJ-50001 had no harmful pharmacological erect in these studies even up to the 200-fold expected clinical dose, 2507g/man.

수학·과학 성취도의 요인 분석으로 본 과학고등학교 학생들의 화학 교과에 대한 인식 연구 (A Study on the Students' Cognition of Chemistry in Science High School by Factor Analysis of Mathematics and Science Achievement)

  • 과학 고등학교에서 다양한 재능을 지닌 학생들의 효과적인 교수- 학습 활동을 위해서, 교수자는 학생들의 다양한 재능과 교과목의 특성에 따라 자연세계의 정보를 인식하고 처리과정에서의 개인차를 이해하는 것이 중요하다. 본 연구의 목적은 수학/과학 성취도의 교과 간 상관 및 요인 분석으로 과학고 학생들의 화학 교과에 대한 인식을 규명하는데 있다. 아울러 R&E 학급에 따른 화학 교과의 인식을 알아보고자 하였다. 연구 대상은 입학 전형과 교육과정의 개편 시기에 따른 G과학고 3개년 입학생(296명)이 주 연구 대상자이고, 경남·울산 지역 2개 과학고를 포함하였다. 성취도의 상관 및 요인 분석은 SPSS 25를 이용하여 탐색적 요인분석법으로 수행하였다. 본 연구의 수행 결과, 다음과 같은 결론을 얻었다. 첫째, 수학·과학 성취도 간의 상관분석에서 화학의 Pearson 상관계수는 다른 교과목들에 비해 높은 정적 상관을 보이는 것이 확인되었다. 둘째, 수학·과학 성취도의 요인 분석에서 요인 지표는 수리-논리(수학, 물리)와 자연 이해(생명과학, 지구과학)의 2개 요인으로 구분되는 것이 확인되었다. 셋째, 요인 분석에서 화학 교과는 수리-논리와 자연 이해 능력이 모두 요구되는 교과로 인식하고 있음을 확인할 수 있었다. 마지막으로, R&E 학급에 따라서 화학 교과에 대한 학생들의 인식이 다르다는 것을 확인하였다. 즉, R&E 화학반 학생들은 다른 학생들과 달리 화학 교과를 수리-논리가 요구되는 과목으로 인식하고 있음이 확인되었다.

Tissue Factor Inhibitory Flavonoids from the Fruits of Chaenomeles sinensis

  • Tissue factor (TF, tissue thromboplastin or coagulation factor III) accelerates the blood clotting, activating both the intrinsic and the extrinsic pathways to serve as a cofactor. In order to isolate TF inhibitors from the fruits of Chaenomeles sinensis, an activity-guided purification utilizing a bio-assay method of prothrombin time prolongation, was carried out to yield five active flavoniods such as hovetrichoside C (1) ($IC_{50}$ = 14.0 $\mu$g), luteolin-7-Ο-$\beta$-D-glucuronide (3) ($IC_{50}$ = 31.9$\mu$g), hyperin (4) ($IC_{50}$ = 20.8 $\mu$g), avicularin (6) ($IC_{50}$ = 54.8 $\mu$g) and quercitrin (10) ($IC_{50}$ = 135.7 $\mu$g), along with other inactive compounds such as ($\pm$)-(2E,4E)-Ο-$\beta$-D-glucopyranosyl-4'-hydroxy-$\beta$-ionylideneacetic acid ester (2), genistein-7-Ο-$\beta$-D-glucopyranoside (5), luteolin-3'-methoxy-4'-Ο-$\beta$-D-glucopyranoside (7), luteolin-7-Ο-$\beta$-D-glucuronide methyl ester (8), tricetin-3'-methoxy-4'-Ο-$\beta$-D-glucopyranoside (selagin-4'-Ο-$\beta$-D-glucopyranoside) (9), (-)-epicatechin (11), luteolin-4'-Ο-$\beta$-D-glucopyranoside (12) and apigenin-7-Ο-$\beta$-D-glucuronide methyl ester (13). The structures of the isolated compounds were elucidated through spectral analysis. Among them, compounds 1 to 9, 12 and 13 were isolated for the first time from the fruits of this plant and the compound 9 is a new flavonoid.

  • Let N be a $II_1$ factor and G be a finite group acting outerly on N. Then the crossed product algebra $M = N \rtimes G$ is also a $II_1$ factor and $N' \cap M = CI$, i.e. N is irreducible in M. Moreover, N is regular in M, in other words, M is generated by the normalizer $N_M (N)$.

  • This paper considers backing structure of non-contact steel plate using magnectically levitation and aspect-quality factor is defined by estimation of electromagnet-aspect attraction steel plate. Characteristics of attraction power is studied in cylindrical electromagnet, U-type electromagnet, E-type electromagnet by aspect-quality factor. Designing and manufacturing electromagnet attraction steel plate, propriety of experimental results is confirmed.

No Association between Tumor Necrosis Factor-alpha Gene Polymorphisms and Lung Cancer Risk

  • Objectives The role of genetic polymorphisms of tumor necrosis factor-alpha (TNF-${\alpha}$) for lung cancer development was evaluated. Methods Genotypes of the TNF-${\alpha}$ polymorphisms, -1210C>T, -487A>G, -417A>G, IVS1+123G>A, and IVS3+51A>G, were determined in 616 lung cancer cases and 616 lung cancer-free controls. Results After adjusting for body mass index and smoking, each TNF-${\alpha}$ genotype or haplotype composed of five TNF-${\alpha}$ single nucleotide polymorphisms did not show an association with lung cancer risk (p>0.05). The statistical power was found to be 88.4%, 89.3%, 93.3%, 69.7%, and 93.9% for 1210C>T, -487A>G, -417A>G, IVS1+123G>A, and IVS3+51A>G, respectively. Furthermore, the effects of each SNP or haplotype on lung cancer risk were not found to be different according to the cell type of lung cancer (p>0.05). In the repeated analysis with only subjects without other diseases related to inflammation, there was also no association between polymorphisms or haplotypes of the TNF-${\alpha}$ gene and lung cancer risk (p>0.05). Conclusions This study found no association between common variants of the TNF-${\alpha}$ gene and lung cancer risk.

생쥐난의 체외 배양에서 인간난포액과 표피 성장 인자가 난성숙에 미치는 영향 (The Effect of E.G.F. and Human Follicular fluid on the Maturation of Mouse Oocytes in in vitro Culture)

  • The human follicular f1uids(F.F.) may be considered to contribute the maturation of the oocytes on the in vitro culture. To investigate the effects of epidermal growth factor (E.G.F.), which is present in mature and immature follicular fluids, we had experiments of mouse oocytes maturation in vitro. The endpoints assayed were rated as percentage of oocytes undergoing germinal vesicle breakdown(G.V.B.D.) and polar body(P.B.) formation at 12 hours after in vitro culture. The rates of G.B.B.D. were 87% in mature F.F. 68% in immature F.F. and 78% in Ham's F-10 medium respectively. And overall the mature F.F. seem to stimulate on in vitro oocyte maturation compared with either immature F.F. or Ham's F-10 medium. As the concentration of addition of E.G.F. in immature F.F., the rates of G.V.B.D. and P.B. formation were 82 %, 23% in addition with 2 ng/ml while 84%, 32% in addition with 4 ng/ml respectivly. And at the concentration of addition of E.G.F. in Ham's F-10 media as well, the rates of G.V.B.D. and P.B. formation were 84%, 40% and 82%, 44% in addition with each 2ng, 4ng. AccordinglY there was no influence on the oocytes maturation at the addition of E.G.F. to each immature F.F. and Ham's F-10 medium. In conclusion, the E.G.F. is not able to induce oocyte maturation independent of it's effects in immature F.F. and Ham's F-10 media.
