Diversity, distribution, and antagonistic activities of rhizobacteria of Panax notoginseng

  • Background: Rhizobacteria play an important role in plant defense and could be promising sources of biocontrol agents. This study aimed to screen antagonistic bacteria and develop a biocontrol system for root rot complex of Panax notoginseng. Methods: Pure-culture methods were used to isolate bacteria from the rhizosphere soil of notoginseng plants. The identification of isolates was based on the analysis of 16S ribosomal RNA (rRNA) sequences. Results: A total of 279 bacteria were obtained from rhizosphere soils of healthy and root-rot notoginseng plants, and uncultivated soil. Among all the isolates, 88 showed antagonistic activity to at least one of three phytopathogenic fungi, Fusarium oxysporum, Fusarium solani, and Phoma herbarum mainly causing root rot disease of P. notoginseng. Based on the 16S rRNA sequencing, the antagonistic bacteria were characterized into four clusters, Firmicutes, Proteobacteria, Actinobacteria, and Bacteroidetesi. The genus Bacillus was the most frequently isolated, and Bacillus siamensis (Hs02), Bacillus atrophaeus (Hs09) showed strong antagonistic activity to the three pathogens. The distribution pattern differed in soil types, genera Achromobacter, Acidovorax, Brevibacterium, Brevundimonas, Flavimonas, and Streptomyces were only found in rhizosphere of healthy plants, while Delftia, Leclercia, Brevibacillus, Microbacterium, Pantoea, Rhizobium, and Stenotrophomonas only exist in soil of diseased plant, and Acinetobacter only exist in uncultivated soil. Conclusion: The results suggest that diverse bacteria exist in the P. notoginseng rhizosphere soil, with differences in community in the same field, and antagonistic isolates may be good potential biological control agent for the notoginseng root-rot diseases caused by F. oxysporum, Fusarium solani, and Panax herbarum.

생물방제균 Bacillus sp. YBL-7의 형질전환조건 (Genetic Transformation of Biocontrol Agent Bacillus sp, YBL-7 by Plasmid pE194)

  • 식물근부균 Fusarium solani의 생육을 강력히 길항하는 Bacillus sp. YBL-7을 토양으로부터 분리하였으며 그 길항기작이 항생물질임을 이미 보고하였다. 분리된 생물방제균 Bacillus sp. YBL-7 균주들 siderophore, 가수분해능 등의 타 방제기능을 도입할 수 있는 다기능적인 방제균으로 육종하기 위해 plasmid vector pE194를 이용하여 효율적인 형질전환 system을 확립하고자 하였으며, 이때 필요한 여러가지 조건을 조사하였다. 생물방제균 Bacillus sp. YBL-7 원형질체 형성의 최적조건은 5시간 배양된 균체를 사용함으로써, 최종농도가 200${\mu}g$/ml인 lysozyme을 $37^{\circ}C$에서 90분간 처리함으로써 원형질체 생성율이 가장 높았다. 삼투압안정 완충제인 SMM bufferso의 sucrose 농도는 0.5M에서 안정하였고, mannitol 재생 배지에서 원형질체를 재생시켰을 경우 첨가된 agar의 농도가 1.2%에서, mannitol은 0.7M의 농도에서 가장 재생율이 높았다. 형질전환율은 polyethylene glycol 농도가 40%(w/v)일 때 가장 높았으며, plasmid DNA와의 접촉시간과 발현배양시간은 각각 10분, 120분에서 가장 효과가 좋았다. 또한 plasmid DNA의 농도는 5$\mu$g/ml 첨가에서 가장 높았으며, 형질전환된 숙주균주 Bacillus sp. YBL-7내에서의 pE194는 매우 안정하게 유지되었으며, 외붕 전자가 도입된 전환체와 숙주균주가 동일한 억제력을 갖는 것으로 확인되었으며, agarose gel 상에서도 고유의 plasmid pE194의 band를 확인 할 수 있었다.

감자 맹아억제제 Chlorpropham의 마른썩음병 억제 효과 (Inhibitory Effect of Potato Sprouting Inhibitor Chlorpropham on Dry Rot)

  • 감자 마른썩음병은 Fusarium 종에 의해 발병하는 감자 저장병 중 하나이며 저장중 수확후 손실을 일으키는 대표적인 병리장해이다. 맹아억제를 위해 허용된 chlorpropham 처리는 주로 가공용 감자의 상온저장 시 이용되고 있다. 본 연구에서는 감자 저장 중 맹아의 출현을 억제하기 위해 사용되는 chlorpropham 처리가 Fusarium에 의한 마른썩음병(Fusarium solani Appel & Wollenw) 발생의 억제 효과가 있는지 확인하기 위해 감자 '단오' 품종을 이용하여 수행되었다. 마른썩음병균의 균사 생장률을 조사하기 위해균사 생장은 F. oxysporum 균사를 치상한 chlorpropham(5.0, 50.4, 503.8, 5,038ppm)과 prochloraz(0.1, 1.0, 10.0, 100.0ppm) 약제배지를 이용하여 in vitro에서 조사하였다. 20℃에서 14일간 치상한 균사는 chlorpropham과 prochloraz 처리 시 처리 농도가 높을수록 생장이 더욱 억제되었으며, 억제율은 chlorpropham 503.8ppm과 prochloraz 10ppm 처리 시 각각98.2%와 100%를 나타냈다. 20℃ 상온과 4℃ 저온저장 중 마른썩음병 방제 효과는 F. oxysporum을 접종한 감자 괴경을 chlorpropham(5.0, 50.4, 503.8ppm)과 prochloraz(100ppm)에 침지처리하여 조사하였다. 상온 저장한 감자(20℃)의 병반 직경은 저장 70일 때에 무처리구 괴경의 13.7mm에 비해 chlorpropham 50.4ppm 처리구에서만10.7mm로 유의성이 있게 억제되었다. 저온에서는 모든 처리구에서 병반 진행은 통계적 유의차가 없이 유사하였다. 본 연구 결과에서 저장 감자의 마른썩음병을 억제하기 위해서는 수확 후 chlorpropham 50.4ppm 처리가 유용할 것으로 판단된다.

버섯중 항균활성물질의 개발 -버섯중의 식물병원성 곰팡이에 대한 항균활성 물질 검색- (Development of Antibiotics in Mushroom -The Screening of Antifungal Activities in Basidiomycetes-)

  • 한국산 흰주름버섯외 50종 버섯을 채집하여 석유에테르, 80%에탄올 및 멸균증류수로 각각 추출하여 153종의 추출물을 얻어 식물병원성 곰팡이에 대한 항균활성을 검색하였다. A. alternata에 항균활성은 독우산광대버섯의 물 추출물, MPC-3 버섯의 에탄올 추출물 그리고 큰낙엽버섯과 Xanthoconium affine의 석유에테르 추출물이 활성을 보였다. 잔디탄저병균, C. graminicola에 대하여는 애광대버섯, 노란길민그물버섯 그리고 흰둘레무당버섯의 석유에테르 추출물이, 해면버섯, MPC-3 버섯 그리고 주걱송편버섯의 에탄올 추출물이, 조개껍질 버섯의 물 추출물이 각각 항균활성을 보였다. 인삼뿌리썩음병균, C. destructans에 대하여는 애광대버섯, 수원그물버섯, 해면버섯, MPC-3 버섯, 주걱 송편버섯, 띠미로버섯 그리고 외대덧버섯의 에탄올 추출물이, 애광대버섯, 흰둘레무당버섯, 향버섯, 큰낙엽버섯의 석유에테르 추출물이 각각 항균활성을 보였다. 오이덩굴쪼김병균, F. o. cucumerinum에 대하여는 MPC-3 버섯의 에탄올 추출물이 항균활성을 보였다. 감자시들음병균, F. solani에 대하여는 진갈색주름버섯의 물 추출물이 그리고 무변색무당버섯의 에탄올 추출물이 각각 항균활성을 보였다. 잔디탄저병균, I. bolleyte에 대하여는 진갈색주름버섯의 물 추출물이 그리고 MPC-3버섯의 에탄올 추출물이 각각 항균활성을 보였다.

Isolation and Characterization of Fungal Diversity from Crop Field Soils of Nigeria

  • In order to find indigenous beneficial fungal species from crop field soils of Nigeria, 23 soil samples were collected from various places of Nigeria in June, 2013 and fungi were isolated through serial dilution technique. Isolated fungi were purified and differentiated according to their morphological and microscopic characteristics. In total, 38 different representative isolates were recovered and the genomic DNA of each isolates was extracted using QIAGEN$^{(R)}$ Plasmid Mini Kit (QIAGEN Sciences, USA) and the identification of fungi was carried out by sequence analysis of internal transcribed spacer (ITS) region of the 18S ribosomal DNA (18S rDNA). Recovered isolates belonged to 9 fungal genera comprising Fusarium, Aspergillus, Chaetomium, Coniothyrium, Dipodascaceae, Myrothecium, Neosartorya, Penicillium and Trichoderma. Aspergillus spp., Penicillium spp. and Trichoderma spp. were the most dominant taxa in this study. The antagonistic potentiality of species belonged to Trichoderma against 10 phytopathogenic fungi (F. oxysporum, C. gloesporoides, P. cytrophthora, A. alternata, A. solani, S. rolfsii, F. solani, R. solani, S. sclerotiorum and P. nicotiana) was assessed in vitro using dual culture assay. The dual culture assay results showed varied degree of antagonism against the tested phytopathogens. The potential Trichoderma spp. will be further evaluated for their antagonistic and plant growth promotion potentiality under in vivo conditions.

BIOLOGICAL CONTROL OF GINSENG ROOT ROTS WITH SOIL AMENDMENTS

  • The phenomenon of 'soil sickness' is one of the most important limiting factors for ginseng(Panax ginseng) production in Korea. The principal cause is known to be due to the root rots caused by Cylindrocarpon destructans and Fusarium solani. Attempts were made to control the root rots with non-polluting cultural methods or soil amendments. Among the nine soil amendments tested, crab shell, cow bone and pig feces were selected for further testing. Each of the three amendments increased the populations or various actinomycetes in the range of 10-25 times over that of non-amended soil, whereas the population of C. destructans was reduced to about $50-70\%$ as compared with the control. Five isolates of Streptomyces with clear zones on chitin-agar medium were selected and then tested for their antagonistic effects on C. destructans. When anyone of the five isolates of Streptomyces and C. destructans was grown together in a modified peptone broth, growth of the latter was highly inhibited. When three levels of crab shell, cow bone, or pig feces were used to amend potted soil infested with C. destruetans, the root rot ratings of ginseng seedlings were reduced to less than one half in all the treatments as compared to the control. In another similar experiment, crab shell and cow bone amendments resulted in almost complete control of the seedling root rots in soil infested with C. destructans or F. solani. In conclusion, biological control with soil amendments of ginseng root rots caused by C. destructans and F. solani was successful. Further basic studies should be pursued using soil amendments for better control. In addition, field experiments are needed to complement the soil amendment control measures in an integrated pest control program.

One Pot Synthesis of Novel Cyanopyridones as an Intermediate of Bioactive Pyrido[2,3-d]Pyrimidines

  • Synthesis, structural characterization, and biological activity studies of novel pyrido[2,3-d]pyrimidines (10a-h, 11a-h) are described. Cyclization of cynoacetamides (4, 5) with malonitrile (7) and aldehyde (6a-h) via Hantzsch pyridine synthesis afforded cyanopyridones (8a-h, 9a-h), which on cyclization with formic acid under microwave conditions led to the final product. All the reactions are significantly faster and the isolated yields are remarkably higher in microwave conditions compared to the conventionally heated reactions. The compounds were tested in vitro for their antibacterial activity against Escherichia coli, Pseudomonas aeruginosa, Bacillus subtillus, Staphylococcus aureus, and Micrococcus luteus and antifungal activity against Trichphyton longifusus, Candida albicans, Microsporum canis, Fusarium solani. Compounds 10b, 10e, 11b and 11e exhibited good antibacterial and antifungal activities compared with standards.

식물병원성 곰팡이에 대한 몇 가지 식물정유 및 주성분의 성장억제 효과 (Antifungal Activity of Some Essential Oils and Their Major Constituents on 3 Plant Pathogenic Fungi)

  • 11 plant essential oils are screened in vitro for their antifungal activities against Botrytis cinerea, Fusarium oxysporum and Rhizoctonia solani, which are causative agents of serious plant diseases. The radial growth of the test fungi were reduced in response to the oils. Among them, the essential oil from the bark of Cinnamomum zeylanicum inhibited 3 tested fungi growth, strongly, followed by those of oregano and thyme. The major constituents of the three essential oils, cinnaldehyde, carvacrol and thymol were tested for their effects on the fungi. From the results obtained, cinnamaldehyde, the major constituents of C. zeylanicum bark esential oil, has potential to be developed as a biopesticide for controlling phytopathogenic fungi causing serious damages on the important crops cultivated in Korea.

Damping-off Disease in Mulberry Seedlings and Its Management

  • During the routine survey, the mortality of mulberry seedlings was noticed due to damping-off disease. The disease recognized by rotting of emerged seedlings near the soil line (just below the soil level) resulting in collapse of the seedlings. Two fungi were isolated from affected samples and identified as Alternaria alternata (Fr.) Keissler and Fusarium solani (Mart.) Sacc. Both the fungi were found to be responsible in causing pre and post emergence damping-off of seedlings in mulberry. For management of the disease, an experiment was conducted using fungicides. These fungicides were applied as seed treatment; soil drenching and foliar spray alone and in combination. Among the different treatments, integration of seed treatment and soil application of Dithane M-45 (Mancozeb 75% WP) + Bavistin (Carbendazim 50% WP) followed by foliar spray of these fungicides (after 35 days of sowing) resulted in better survivability of seedlings (93.3 %) on $90^th$ day and controlled the pre and post emergence damping off by 100 and 89.5%, respectively over the check.

Screening for In Vitro Antifungal Activity of Soil Bacteria Against Plant Pathogens

  • Antifungal bacteria for biological control of plant diseases or production of novel antibiotics to plant pathogens were isolated in 1997 from various soils of Ansung, Chunan, Koyang, and Paju in Korea. Sixty-four bacterial strains pre-screened from approximately 1,400 strains were tested on V-8 juice agar against eight plant pathogenic fungi using in vitro bioassay technique for inhibition of mycelial growth. Test pathogens were Alternaria mali, Colletotrichum gloeosporioides, C. orbiculare, Fusarium oxysporum f. sp. cucumerinum, F. oxysporum f. sp. lycopersici, Magnaporthe grisea, Phytophthora capsici, and Rhizoctonia solani. A wide range of antifungal activity of bacterial strains was found against the pathogenic fungi, and strain RC-B77 showed the best antifungal activity. Correlation analysis between inhibition of each fungus and mean inhibition of all eight fungi by 64 bacterial strains revealed that C. gloeosporioides would be best appropriate for detecting bacterial strains producing antibiotics with potential as biocontrol agents for plant pathogens.
