Cymbidium orchids with blight and rot symptoms were collected, and a total of 63 isolates of Fusarium app. was obtained from pseudobulbs, roots, and leaves of the diseased plants. The isolates were identified based on their morphological characteristics. Out of the 63 isolates of Fusatium sup., 51 isolates were identified as F. oxysporum, 10 isolates as F. solani, and the rest as F. proliferatum. F. oxysporum was isolated from all the Cymbidium spp., while F. solani and F. proliferatum were isolated only from Cymbidium ensifolium and C. ginatum, respectively. Isolates of the three Fusarium spp. were tested for pathogenicity to their hosts by artificial inoculation. The strongly pathogenic isolates of Fusarium spp. induced severe dry rot of pseudobulbs and roots of the host plants. The symptoms progressed up to the basal part of the leaves, which later caused blight of the entire plant. The dry root symptoms induced on the plants by artificial inoculation with the isolates of Fusarium app. were similar to those observed in the growers'greenhouses. This is the first report of dry rot of Cymbidium spp. caused by F. oxysporum, F. solani, and F. proliferatum in Korea.
These experiments were conducted to compare the growth characteristics, cold and drought tolerances, disease tolerance and color of the 23 varieties of four different species of cool-season turfgrass introduced from the United States of America and Europe at Yonam College in 1995. Newport and Glade of 8 Kentucky bluegrass varieties were recorded better marks than others. Newport variety was recorded the good marks in the disease tolerance of summer seasons, uniformity, coverage and texture, but the bad marks in color, drought tolerance and rust tolerance and stay green of automn seasons. Glade variety was recorded the good marks in the disease tolerance of summer seasons, uniformity and coverage, but the bad marks in drought tolerance, texture and Pythium blight tolerance. Revel and Revel Jr of 5 tall fescue varieties were recorded better marks than others in color, shoot density, uniformity, coverage, and tolerance of Pythium and Fusarium blight disease. All of red fescue varieties died of disease for rainy season however texture was the finest of all the species in spring season. Dandy of 4 ryegrass varieties was recorded better marks than others in color, shoot density, coverage, drought tolerance and tolerance of Pythium and Fusarium blight disease but texture.
This study was conducted in order to find out biological control of sesame wilt and blight caused by Fusarium of oxysporum f. sp. vasinfectum and Phytophthora nicotianae var. parasitica by using Streptomyces spp. Two sesame pathogens, Fusarium oxysporum f. sp. vasinfectum and Phytophthora nicotianae var. parasitica were purely isolated from diseased sesame plants of the field. Streptomyces species were isolated from 72 soil samples collected from red pepper and sesame uplands in Chungbuk and selected as antagonists according to the results of dual culture. The selected Streptomyces isolates such as St-11 and St-20 were confirmed their antagonistic effect through mycelial inhibition zone and inhibitory effects on the mycelial growth of the pathogens by culture filterate of the antagonists. Inhibitory effects on the conidial germination of Fusarium oxysporum vasinfectum and Phytophthora nicotianae parasitica by the antagonists were also tested in addition to mycelial Iysis. The antagonists St-11 and St-20 showed inhibitory effect on growth of sesame seedlings after seeds soaked in the suspension. Effect of soil inoculation with antagonist St-11 showed 40 to 78 percent of control effect for two diseases in comparison with control under greenhouse.
Antifungal substances were isolated from solid cultures of Fusarium equiseti FO-68 obtained from arrowhead and Fusarium sp. FO-510 obtained from egg plant, and then their antifungal activities were investigated against plant pathogenic fungi in vitro and in vivo. An antifungal substance was purifed from rice solid cultures of F. equiseti FO-68 and identified as equisetin. In addition, two antibiotic substances were isolated from solid cultures of Fusarium sp. FO-510 and their chemical structures were determined to be zearalenone and 8'-hydroxyzearalenone. in vitro, equisetin and zearalenone inhibited mycelial growth of most of the plant pathogenic fungi tested, whereas 8'-hydroxyzearalenone hardly inhibited fungal growth. In vitro, equisetin effectively controlled the development of tomato gray mold and tomato late blight. Zearalenone exhibited in vivo antifungal activity against rice blast, rice sheath blight, tomato gray mold, and tomato late blight. However, 8'-hydroxyzearale-none did not control the development of plant diseases except tomato gray mold. This is the first report on the antifungal activities of equisetin, zearalenone, and 8'-hydroxyzearalenone.
Job's tears (Coix lacryma-jobi) is native to East Asia, and grains of the plant are used as health food and medicinal material. Head blight symptoms of the plant were frequently observed during disease surveys in Korea from 2006 to 2008. The symptoms were characterized as discoloration of husks, and subsequently inside of mature grains were shriveled or emptied. One hundred fifty nine isolates of Fusarium species were obtained from the disease symptoms of the plant collected from several locations in the country. Out of the isolates, the most frequently isolated Fusarium species were F. graminearum (34%), F. proliferatum (14.5%), F. verticillioides (10.1%), F. equiseti (6.9%), and F. fujikuroi (6.3%). Other Fusarium species isolated were F. subglutinans, F. semitectum, F. poae, and F. sporodochioides. Elongation factor 1 alpha gene sequences of the isolates were used for phylogenetic analysis. Analyses of the sequences revealed that the isolates were confirmed to be identical with each reference species of NCBI GenBank. Pathogenicity tests showed that F. graminearum, F. proliferatum and F. verticillioides were strongly virulent to grains of Job's tears. The present study is the first report of head blight of Job's tears caused by Fusarium species in Korea.

The rice diseases occurring on the transgenic rice plant carrying protox gene (CPPO06) was assessed and compared with other varieties of rice such as Dongjin, Chucheong, Ilpeum, and Onnuri in the fields located in Cheongwon of Chungbuk, Suwon of Gyeunggi, and Gwangju of Chonnam. In the field of Cheongwon, the diseases such as blast, leaf spot, sheath blight and Fusarium blight were observed. False smut were observed only in the field of Suwon, where the ratio of diseased plants was 0.28% in CPPO06 and 0.37% in Onnuri, respectively. In the field of Gwangju, leaf spot caused by Cochlioborus miyabeanus was the most severely occurring disease among rice diseases mentioned above. Fusarium blight occurred in all the 3 fields, which were more severe in CPPO06 plants treated with or without oxadiazon as the herbicide. Except for Fusarium blight, there was no significant difference in the rice diseases as blast, leaf spot, sheath blight and false smut between CPPO06 and other rice varieties.
The ascomycete fungus Fusarium graminearum is a major causal agent for Fusarium head blight in cereals and produces mycotoxins such as trichothecenes and zearalenone. Isolation of the fungal strains from air or cereals can be hampered by various other airborne fungal pathogens and saprophytic fungi. In this study, we developed a selective medium specific to F. graminearum using toxoflavin produced by the bacterial pathogen Burkholderia glumae. F. graminearum was resistant to toxoflavin, while other fungi were sensitive to this toxin. Supplementing toxoflavin into medium enhanced the isolation of F. graminearum from rice grains by suppressing the growth of saprophytic fungal species. In addition, a medium with or without toxoflavin exposed to wheat fields for 1 h had 84% or 25%, respectively, of colonies identified as F. graminearum. This selection medium provides an efficient tool for isolating F. graminearum, and can be adopted by research groups working on genetics and disease forecasting.
Fusarium graminearum, the causal agent of Fusarium head blight (FHB) in cereal crops, employs the production of sexual fruiting bodies (perithecia) on plant debris as a strategy for overwintering and dissemination. In an artificial condition (e.g., carrot agar medium), the F. graminearum Z3643 strain was capable of producing perithecia predominantly in the central region of the fungal culture where aerial hyphae naturally collapsed. To unravel the intricate relationship between natural aerial hyphae collapse and sexual development in this fungus, we focused on 699 genes differentially expressed during aerial hyphae collapse, with 26 selected for further analysis. Targeted gene deletion and quantitative real-time PCR analyses elucidated the functions of specific genes during natural aerial hyphae collapse and perithecium formation. Furthermore, comparative gene expression analyses between natural collapse and artificial removal conditions reveal distinct temporal profiles, with the latter inducing a more rapid and pronounced response, particularly in MAT gene expression. Notably, FGSG_09210 and FGSG_09896 play crucial roles in sexual development and aerial hyphae growth, respectively. Taken together, it is plausible that if aerial hyphae collapse occurs on plant debris, it may serve as a physical cue for inducing perithecium formation in crop fields, representing a survival strategy for F. graminearum during winter. Insights into the molecular mechanisms underlying aerial hyphae collapse provides offer potential strategies for disease control against FHB caused by F. graminearum.
Effects of chitosan on growth of tomato plant, and suppression of Fusaruim wilt caused by Fusarium oxysporum f. sp. lycopersici and late blight casued by Phytophthora infestans, were examined. Both late blight and fusarium wilt were suppressed by spray and irrigation of chitosan, respectively. Inhibition of mycelial growth was not greatly affected by molecular size of chitosan but, concentration dependent effects was observed. Ninty percent of P. infestans and 80% of F. oxysporum f. sp. lycopersici of mycelial growth was inhibited by 1,000 ppm of chitosan (MW 30,000~50,000) when amended in plate media. Induction of defense-related gene expression in plant by chitosan treatments were observed when chitosan treated tobacco and tomato RNA samples were hybridized with several defense-related genes as probes. The results revealed that $\beta$-1,3-glucanase and chitinase genes were strongly induced, while pathogenesis-related protein-1, 3-hydroxy-3-methylglutaryl coenzyme A reductase, anionic peroxidase, phenylalanine ammonia lyase genes were weakly induced by chitosan treatment. These results suggest that chitosan have dual effects on these host-pathogen interactions. Possible roles of chitosan in suppression of tomato diseases by inhibition of mycelial growth and activation of plant defense responses are discussed.

Fusarium head blight (FHB) is a devastating wheat disease with a significant economic impact. Fhb1 is the most important large effect and stable QTL for FHB resistance. A pore-forming toxin-like (PFT) gene was recently identified as an underlying gene for Fhb1 resistance. In this study, we developed and validated a PFT-based Kompetitive allele specific PCR (KASP) marker for Fhb1. The KASP marker, PFT_KASP, was used to screen 298 diverse wheat breeding lines and cultivars. The KASP clustering results were compared with gelbased gene specific markers and the widely used linked STS marker, UMN10. Eight disagreements were found between PFT_KASP and UMN10 assays among the tested lines. Based on the genotyping and sequencing of genes in the Fhb1 region, these genotypes were found to be common with a previously characterized susceptible haplotype. Therefore, our results indicate that PFT_KASP is a perfect diagnostic marker for Fhb1 and would be a valuable tool for introgression and pyramiding of FHB resistance in wheat cultivars.
