Isolation of a Pestalotiopsis Species Degrading Mucilage from Fruit of Opuntia ficus-indica var. Saboten

  • The high molecular-weight mucilage extracted and purified from cactus fruit of Opuntia ficus-indica var. Saboten was degraded by the cell-free culture filtrate of a fungus isolated from soil. TLC analysis of the polymeric mucilage after incubation with the fungal culture filtrate confirmed its degradation. When the degradation products were tested for their qualitative reactions with ninhydrin and phenol-sulfuric acid, only phenol-sulfuric acid gave positive development, and ninhydrin did not show any observable color reaction. This coloring reaction suggested the presence of a carbohydrate without an amino group within the mucilage. Analyses by HPLC and liquid gel permeation chromatography on sephadex G-100 also provided additional information on degradation of the mucilage by the fungal culture filtrate. The sequences of ITS-5.8S rDNA from the fungal isolate that was cultivated for the preparation of mucilage-degrading enzyme showed 99% similarity to those of Pestalotiopsis aquatica.

  • The endophytic fungi were isolated from red pine trees (Pinus densiflora) that were distributed in three sites of Chungbuk, Korea. Twenty fungal isolates were isolated from 16 trees and divided into 8 groups by morphological characters. The fungal isolates were identified using the sequences of ITS region of rDNA; Lophodermium complex, Sydowia polyspora, Hymenula sp., Sistotrema brinkmannii, Septoria pini-thunbergii, Earliella sp. Lophodermium spp. were the most frequently found fungal species the across sites and firstly detected from Pinus species in Korea by molecular work.

First Report of Albifimbria verrucaria and Deconica coprophila (Syn: Psylocybe coprophila) from Field Soil in Korea

  • During a survey of fungal diversity in Korea, two fungal strains, KNU17-1 and KNU17-199, were isolated from paddy field soil in Yangpyeong and Sancheong, respectively, in Korea. These fungal isolates were analyzed based on their morphological characteristics and the molecular phylogenetic analysis of the internal transcribed spacer (ITS) rDNA sequences. On the basis of their morphology and phylogeny, KNU17-1 and KNU17-199 isolates were identified as Albifimbria verrucaria and Deconica coprophila, respectively. To the best of our knowledge, A. verrucaria and D. coprophila have not yet been reported in Korea. Thus, this is the first report of these species in Korea.

Six New Recorded Species of Macrofungi on Gayasan National Park in Korea

  • The fungi play important roles in maintaining the balance and homeostasis of natural ecosystems. We investigated fungal specimens indigenous to the Gayasan National Park located in the deep inlands of southeastern Korea from 2017-2020. Six fungal species in the Korean macromycota-Chiua olivaceoreticulata, Entoloma conchatum, Galerina sulciceps, Hebeloma radicosoides, Spongiporus gloeoporus, and Tricholoma sinoacerbum-were identified based on morphological characteristics and rDNA sequences. The six fungal speices were newly revealed on the Korean Peninsula, and it supports that the continuous investigaton is the best way to realize the mycosis.

Acrodontium burrowsianum and Pestalotiopsis humicola: Two Previously Unrecorded Fungal Species Isolated from Conifer Leaves in Korea

  • Endophytic fungal strains were isolated from the leaves of two conifer species (Juniperus rigida and Pinus densiflora) in Korea and identified on the basis of their morphological and molecular characteristics. Internal transcribed spacer and large subunit regions of rDNA were used for the phylogenetic analysis, and the translation elongation factor 1-alpha (TEF) and RNA polymerase II second largest subunit (RPB2) genes were analyzed depending on the species. Two fungal species that were previously unrecorded in Korea were identified: Acrodontium burrowsianum and Pestalotiopsis humicola. Their morphological and phylogenetic characteristics are described herein.

PCR 기법을 이용한 Phoma glomerate 의 특이검출 (Specific and Sensitive Detection of Phoma glomerata Using PCR Techniques)

  • Phoma glomerata는 식물 잎이나 열매에 병을 일으키는 식물병원균으로 알려져 있다. 국내에서는 아직 피해사례가 없기 때문에 P. glomerata는 국내의 식물검역균으로 관리되고 있다. 본 연구는 국내에 들어오는 목재나 과일에 P. glomerata를 검출할 수 있는 방법 개발코자 수행되었다. Phoma 균주들의 translation elongation factor 1 alpha 유전자 염기서열에 기초하여 P. glomerata 특이적 PCR 프라이머를 디자인 하였고 그 특이성을 검정하였다. PCR 수행 결과 P. glomerata에서만 170 bp 크기의 밴드가 증폭되었고, 다른 비교 균주에서는 밴드가 증폭되지 않았다. 검출 감도를 평가하기 위해 기존 PCR방법과 real time PCR 방법을 이용하여 실험한 결과 최소 10 pg과 1 pg까지 각각 검출할 수 있었다. 본 연구결과는 디자인된 PCR 프라이머가 P. glomerata를 특이적으로 검출하는데 유용할 것임을 보여준다.

Diversity of Fungi from Dokdo Island Soil, Korea and Their Antimicrobial and Hydrolytic Enzyme Activity

  • Dokdo island is located in the northeastern part of Ulleungdo, known as volcanic island. In total, 53 fungal isolates were isolated from Dokdo island soil sample, using dilution plate technique. The isolates were identified on the basis of morphological characteristics and rDNA ITS sequence analysis. Out of them, 41 isolates were identified at the level of species. The dominant fungal species and genera included Fusarium spp., Mucor sp., Clonostachys spp., and Trichoderma sp. The % sequence identity (the number of matches/the complete alignment length) values via NCBI BLAST searching of EML-IF9, EML-MF30-1 and EML-DDSF4 represented 97.19% (485/499) with Clonostachys cf. rosea (GenBank accession no. KC313107), 98.33% (472/480) with Metarhizium guizhouense (GenBank accession no. HM055445), and 100% (350/350) with Mortierella oligospora (GenBank accession no. JX976032), respectively. Three species of C. rosea, M. guizhouense and M. oligospora represented new records of fungi from Dokdo island, Korea. The antimicrobial activities of the fungal strains varied with tested. Two isolates (EML-MFS30-1 and EML-IF9) showed antifungal activity against several fungi including Fusarium oxysporum and Rhizotonia solani. Clonostachys rosea (EML-IF9) showed strong hydrolytic enzyme activity. Our results showed that the antagonistic fungi including Clonostachys rosea will be used as potential biocontrol agents for control of fungal diseases.

Differences among Endophytic Fungal Communities Isolated from the Roots of Cephalanthera longibracteata Collected from Different Sites in Korea

  • Orchidaceous plants have symbiotic relationships with endophytic fungi, including mycorrhizal fungi, which play important roles in the seed germination and growth of the host plants. In this study, endophytic fungal communities isolated from the roots of Cephalanthera longibracteata collected from three different sites in Korea were analyzed, and it was determined whether fungal communities were preferentially correlated with the sites. The fungal isolates were identified by sequence analysis of the internal transcribed spacer regions of rDNA. In total, 30 species of endophytic fungi, including two species of mycorrhizal fungi belonging to the genus Tulasnella, were identified. Leptodontidium orchidicola showed the highest frequency and was isolated from all root samples. Species diversity and richness were not significantly different among sites. However, the community structure of the endophytic fungi significantly differed among sites, suggesting that the site characteristics affected the community composition of the endophytic fungi colonizing the roots of C. longibracteata. Our findings will aid in developing methods involving the use of symbiotic fungi for orchid conservation and restoration in native habitats.

Evaluation of genetic affinities among Fusarium oxysporum f. sp. fragariae by RAPD and rDNA RFLP

  • Fusarium oxysporum f. sp. fragariae is a fungal pathogen causing wilt disease on strawberry. The RAPD and RFLP of IGS region of rDNA were used to identify genetic affinity among 22 isolates of F. oxysporum f. sp. fagariae obtained from various location of major strawberry cultivating areas in Korea. Approximately 2.6kb DNA fragment was amplified with primer CNS1 and CNL12, and polymorphisms were observed with Avail and HinfI. A dendrogram was constructed using the UPGMA for cluster analysis. Eight distinct groups were clustered based on the banding pattern obtained from RAPD and rDNA RFLP. There was high level genetic variation among Korean isolates of Fusarium of sporum f. sp. fragariae.

Phylogenetic Relationships of the Mutualistic Fungi Associated with Macrotermes subhyalinus in Oman

  • The symbiotic association between fungus-gardening termites Macrotermes and its fungal symbiont has a moderate degree of specificity-although the symbiotic fungi (Termitomyces) form a monophyletic clade, there is not a one-to-one association between termite species and their fungus-garden associates. Here, we aim to determine the origin and phylogenetic relationships of Termitomyces in Oman. We used sequences of the internal transcribed spacer region (ITS) and the nuclear large subunit ribosomal RNA (LSU rRNA, 25S) gene and analyzed these with sequences of Termitomyces from other geographic areas. We find no evidence for more than a single colonization of Oman by Termitomyces. Unexpectedly, we find Termitomyces in Oman is most closely related to the symbiont of M. subhyalinus in West Africa rather than to those of geographically closer populations in East Africa.
