Damping-off is a critical disease on ginseng seedling, which caused by the fungal pathogen Rhizoctonia solani. The disease has been prevented by tolclofos-methyl for the last 20 years. However, the tolclofos-methyl usually detected on the harvested roots of 6-year-old ginseng. Herein, we tried to select an alternative pesticide which not only must be safe but also efficiently inhibits the fungal pathogen. Four fungicides (fludioxonil, flutolanil, pencycuron, and thifluzamide) were applied to their inhibition efficacy against the pathogen. In in vitro test, fludioxonil treatment showed 80% inhibition activity for 25 days. Thifluzamide and flutolanil showed the activity for 10 days. Pencycuron showed for 1 days. In addition, the fludioxonil was more effective to control the pathogen comparing to other three fungicides in field. The incidence of damping-off was reduced to 71% by fludioxonil treatment. The level of the fungicide residue in seedling was 0.44 mg/kg, which value will be a negligible level in final products after 5 years. Consequently, the fludioxonil is a conceivable alternative for tolclofos-methyl to cope with R. solani.

dsRNA was found in malformed cultures of Lentinula edodes strain FMRI0339, one of the three most popular sawdust cultivated commercial strains of shiitake, and was also found in healthy-looking fruiting bodies and actively growing mycelia. Cloning of the partial genome of the dsRNA revealed the presence of the RdRp sequence of a novel L. edodes mycovirus (LeV), and sequence comparison of the cloned amplicon showed an identical sequence to known RdRp genes of LeV found in strain HKA. The meiotic stability of dsRNA was examined by measuring the ratio of the presence of dsRNA among sexual monokaryotic progeny. More than 40% of the monokaryotic progeny still contained the dsRNA, indicating the persistence of dsRNA during sexual reproduction. Comparing the mycelia growth of monokaryotic progeny suggested that, although variations in the growth rate existed among progeny and virus infection was observed in highly actively growing progeny, there appeared to be a tendency toward a lower frequency of virus incidence in actively growing progeny. This study attempted to cure the edible mushroom L. edodes strain FMRI0339 of the L. edodes mycovirus (LeV) in order to obtain an isogenic virus-free fungal strain as well as a virus-infected strain for comparison. Mycelial fragmentation, followed by being spread on a plate with serial dilutions resulted in a virus-free colony. Viral absence was confirmed with gel electrophoresis after dsRNA-specific virus purification, Northern blot analysis, and PCR using reverse transcriptase (RT-PCR). Once cured, all of fungal cultures remained virus-free over the next two years. Interestingly, the viral titer of LeV varied depending on the culture condition. The titer from the plate culture showed at least a 20-fold higher concentration than that grown in the liquid culture. However, the reduced virus titer in the liquid culture was recovered by transferring the mycelia to a plate containing the same medium. In addition, oxygen-depleted culture conditions resulted in a significant decrease of viral concentration, but not to the extent seen in the submerged liquid culture. Although no $discernable phenotypic changes in colony morphology were observed, virus-cured strains showed significantly higher growth rates and mycelial mass than virus-infected strains. We were also explored effects of LeV on fruiting body formation and mushroom yield. The fruiting body formation yield of virus-free L. edodes was larger than virus-infected L. edodes. These results indicate that LeV infection has a deleterious effect on mycelial growth and fruiting body formation. In addition, we have been investigated host-parasite interaction between L. edodes and its mycovirus interaction to study viral mechanism by establishment of proteomics.

To select bacterial strains with antifungal activity against an anthracnose fungal disease causing damage severely on hot pepper, previous isolates obtained from plant root samples were screened. Among 457 isolates, IA70-5 isolate was finally selected and identified as Streptomyces padanus based on 16S rDNA sequence analysis. Strain IA70-5 is non-pigmenteous, non-mobile, and filamentous. S. padanus IA70-5 inhibited effectively the mycelium growth, spore germination, and appressorium formation of Colletotrichum acutatum in vitro. The results of this study demonstrated that IA70-5 strain, especially applied on fruit of hot pepper, decreased disease incidence 90% for pre-inoculation before pathogen treatment. Taken together, S. padanus IA70-5 strain is a promising biological control agent to control of a major fungal pepper disease, anthracnose.
In order to develop a environmentally friendly control protocol for managing tomato leaf mold disease in the field, we employed bacteria- and fungi-based commercially available microbial preparations. The field experiment was conducted from April to July in 2010. Average incidence rates tomato leaf mold caused by Fulvia fulva were 13.1% at the two plastic houses located in Jangsung, Jeonnam area. Initially 11 microbial preparations were tested for antifungal activity against F. fulva in vitro. Among them, 7 selected preparations showed to be inhibited the mycelial growth of the fungal pathogen over 50%. Four microbes suppressed disease incidence as much 50% under greenhouse condition. Eventually in the field two microbial products including Bacillus subtilis GB-0365 and B. subtilis KB-401 respectively were showed control value up to 71.8% for four times sprays from 20 days to 70 days after transplanting. Furthermore, the control value of three times spray program demonstrated 79.3%. Efficacy of the three and four spray programs was more effective than that of non-spray control treatment. Our results indicated that adjustment of application method of commercially available microbial preparation could be used to control a target plant disease as an effective and efficient crop protection system for organic farming.
In 2004, average temperature was higher, and rainfall was less than those of previous year, The diseases of rice, barley, pepper, chinese melon, apple and oriental pear were surveyed. Seedling diseases, leaf blast, sheath blight and bacterial blight of rice, phytophthova blight, virus diseases and anthracnose of pepper, and sudden wilt syndrome and powdery mildew of chinese melon grown in plastic greenhouse were severe. Especially, sheath blight and bacterial blight of rice occurred two times higher than those of previous year, Panicle blight of rice decreased about 4 times, compared with the previous year, presumed that the lower rainy days, rainfall and RH suppressed spread of the fungal pathogens to panicles of rice. Lower rainfall during mid- and late Aug caused three-times less occurrence of phytophtora blight of red-pepper than that of the previous year, Another diseases surveyed occurred similar or less than those of the previous year.
Ginseng damping-off, caused by the fungal pathogens Rhizoctonia solani and Pythium sp., is a critical disease in ginseng seedling. In a continuing effort to find microorganisms with the potential of acting as a biocontrol agent against Rhizoctonia damping-off, we found that a Streptomyces sp. A501 showed significant antifungal activity against Rhizoctonia solani. In field experiment to test the efficacy of Streptomyces sp. A501 in controlling ginseng damping-off, the incidence of damping-off disease was meaningfully reduced when ginseng seeds were soaked in the culture broth of Streptomyces sp. A501 before sowing. To perform characterization of the antifungal compound, we isolated it from the culture broth of strain A501 through Diaion HP-20 and silica gel column chromatographies and preparative high-performance liquid chromatography. The structure of the antifungal compound was assigned as fungichromin by spectroscopic methods, mainly nuclear magnetic resonance and electrospray ionization-mass analysis.

The filamentous Ascomycota Colletotrichum gloeosporioides sensu lato is a fungus that has been reported worldwide as a causal agent of anthracnose disease in avocado and other crops. In Mexico, this species affects fruits from an early stage of development in the orchard until the post-harvest stage. Although fungicides are continuously applied to control Colletotrichum species, pericarp cankers and soft rot mesocarp in fruits are still frequently observed. Considering the lack of a precise description of the causative agent, the aim of the current study was to determine the pathogens involved in this symptomatology. Twenty-four isolates were consistently obtained from the pericarp of avocado fruits cv. "Hass" collected in the central avocado-producing area of Mexico. Morphological features such as colony growth, conidia size, and mycelial appressorium were assessed. Bayesian multilocus phylogenetic analyses were performed using amplified sequences of the internal transcribed spacer region of the nuclear ribosomal DNA; actin, chitin synthase, glyceraldehyde-3-phosphate dehydrogenase partial genes; and APn2-Mat1-2 intergenic spacer and mating type Mat1-2 partial gene from the nine selected isolates. In addition, fruits were inoculated with a conidial suspension and reproducible symptoms confirmed the presence of Colletotrichum fructicola in this area. This pathogenic species can now be added to those previously reported in the country, such as C. acutatum, C. boninense, C. godetiae, C. gloeosporioides, and C. karstii. Disease management programs to reduce the incidence of anthracnose should include C. fructicola to determine its response to fungicides that are routinely applied, considering that the appearance of new species is affecting the commercial quality of the fruits and shifting the original population structure.

Pulmonary cavities are caused by bacterial pneumonia, fungal diseases, lung cancer, and tuberculosis (TB). However, in Korea, patients with cavitary lung lesions are generally considered to have pulmonary TB, where the incidence of TB is approximately 70/100,000 per year. We report a case of chronic necrotizing pulmonary aspergillosis that was obscured by multidrug-resistant pulmonary TB.

We tested for the presence of double-stranded RNA (dsRNA) mycovirus in 827 Fusarium graminearum isolated from diseased barley and maize. dsRNA mycoviruses with various sizes were isolated. Of them, it was previously reported that dsRNA from DK2l isolate had pronounced morphological changes, including reduction in mycelial growth, increased to red pigmentation, reduced virulence and sporulation. (Chu et al., Appl. Environ. Microbiol. 2002). For better understanding of this hypovirulence associated with DK2l dsRNA virus, we determined the complete nucleotide sequence of dsRNA genome and named Fusarium hypovirus DK2l strain (Fhv-DK2l ). Genomic RNA of Fhv-DK2l was determined to be 6625 nucleotides in length excluding the poly (A) tail and contained three putative open reading frame. RNA-dependent RNA polymerase (RdRp) and helicase domain were expected in ORF A, 54 to 4709 nucleotide position. ORE B, 4752 to 5216 nucleotide position, and ORF C, 5475 to 6578 nucleotide position, were predicted to encode 16.7kDa and 41.3kDa protein respectively each. We could not detect any conserved domains from these two proteins. Phylogenetic analysis showed Fhv-DK2l was related to Cryphonectria hypovirus 3. Ten additional isolates were found that were infected with dsRNA mycoviruses. These mycoviruses contain 2 to 4 different segments of dsRNAs with the size range of approximately 1.7 to 10-kbp in length. The presence of dsRNAs isolates did not affect colony morphology and were transmissible through conidia and ascospore with incidence of 30-100%. These results indicate that there is genomic diversity of dsRNA mycoviruses that infect F. graminearum isolates and that impact of virus infection on host's morphology and virulence is determined by the interaction between dsRNAs and the fungal host, not by the mere presence of the dsRNAs

Maize is a socioeconomically important crop in many countries. Recently, a high incidence of stalk rot disease has been reported in several maize fields in Gangwon province. In this report, we show that maize stalk rot is associated with the fungal pathogens Fusarium subglutinans and F. temperatum. Since no fungicides are available to control these pathogens on maize plants, we selected six fungicides (tebuconazole, difenoconazole, fluquinconazole, azoxystrobin, prochloraz and kresoxim-methyl) and examined their effectiveness against the two pathogens. The in vitro antifungal effects of the six fungicides on mycelial growth and colony formation were investigated. Based on the inhibition of mycelial growth, the most toxic fungicide was tebuconazole with 50% effective concentrations ($EC_{50}$) of < $0.1{\mu}g/ml$ and $EC_{90}$ values of $0.9{\mu}g/ml$ for both pathogens, while the least toxic fungicide was azoxystrobin with $EC_{50}$ values of 0.7 and $0.5{\mu}g/ml$ for F. subglutinans and F. temperatum, respectively, and $EC_{90}$ values of > $3,000{\mu}g/ml$ for both pathogens. Based on the inhibition of colony formation by the two pathogens, kresoxim-methyl was the most toxic fungicide with complete inhibition of colony formation at concentrations of 0.1 and $0.01{\mu}g/ml$ for F. subglutinans and F. temperatum, respectively, whereas azoxystrobin was the least toxic fungicide with complete inhibition of colony formation at concentrations > $3,000{\mu}g/ml$ for both pathogens.
