  • 새조개(Fulvia mutica)는 우리 나라, 동중국해, 홍콩 일본 등에 분포하며, 수심 10∼50m의 사니질에 서식한다. 패각이 얇고, 패각내면은 분홍색을 띄고 있으며, 육질부의 발은 길고 검은 색이며, 맛이 매우 좋아 식용 패류로서의 가치가 매우 높다. 본 종은 일본의 경우 성장(Tain and Makoto, 1997; 1998), 자원량 평가(Tain and Makoto, 1997), 어획량과 개체군 구조(Tain and Makoto, 1997) 등 여러 분야에서 많은 연구가 이루어지고 있다. (중략)

Effects of Water Temperatures on the Larval Development and Spat Growth of the Cockle Shell, Fulvia mutica (새조개 (Fulvia mutica) 난 발생 및 유생 성장에 미치는 수온의 영향)

  • The effect of water temperature on spawning induction, larval development, spat settling and its growth of the cockle shell, Fulvia mutica, were investigated to obtain the basic data for effective seed production. The eggs, which were randomly divided into 6 groups of water temperatures of 14, 17, 20, 23, 26 and $29^{\circ}C$, were transferred into 1 L beaker, respectively. The relationships between the water temperature and the required time (1/h, hour) by each egg developmental stage were calculated. Biological minimum water temperature and the cumulative water temperature until egg development of the veliger stage were calculated to be $0.1^{\circ}C$ and $397.3^{\circ}C$, respectively. The optimal water temperature for developmental bioassay of F. mutica was clarified to be $23^{\circ}C$. The required time for the embryo to become veliger larvae was 20 hours at $23^{\circ}C$.

Geographic Variations of Three Fulvia mutica Populations

  • In the present study, the seven primers BION-33, BION-34, BION-37, BION-41, BION-44, BION-45 and BION-42 generated the total number of loci, average number of loci per lane and specific loci in Hongseong, Yeosu and Goheung population of F. mutica, respectively. 7 primers generated 19 specific loci in the Hongseong population, 29.3 in the Yeosu population and 23.1 in the Goheung population, respectively. Especially, the decamer primer BION-37 generated 7 unique loci to each population, which were identifying each population, approximately 700 bp in Hongseong population. In this study, the dendrogram obtained by the seven primers indicates three genetic clusters: cluster 1 (HONGSEONG 01-HONGSEONG 07), cluster 2 (YEOSU 08-YEOSU 14) and cluster 3 (GOHEUNG 15-GOHEUNG 21). Among the twenty one cockles, the shortest genetic distance that displayed significant molecular differences was between individuals 17 and 19 from the Goheung population (genetic distance = 0.051), while the longest genetic distance among the twenty-one cockle individuals that displayed significant molecular differences was between individuals HONGSEONG no. 03 and YEOSU no. 12 (genetic distance = 0.616). Relatively, individuals of YEOSU population were fairly closely related to that of GOHEUNG population. Ultimately, PCR fragments revealed of in this study may be useful as a DNA marker the three geographic populations to distinguish.

Processing and Pigment Stability of Cooked and Frozen Cockle, Fulvia mutica

  • Processing condition and pigment maintenance of cockle, Fulvia mutica were studied. Proximate composition of whole meat was $82.3\%$ moisture, $10.8\%$ crude protein, $0.8\%$ crude lipid, $2.5\%$ carbohydrate and $3.1\%$ crude ash, and that of foot muscle was $80.6\%,\;12.3\%,\;0.3\%,\;2.9\%\;and\;3.3\%$ respectively. When the living cockle was soaked in $2\%$ NaCl solutions, about $90\%$ of silt and mud was removed after 10 hours soaking, and over $92\%$ was removed when the pH was adjusted to 7.5. When the pigment destruction was tested by 40 seconds at $75^{\circ}C,\;80^{\circ}C,\;85^{\circ}C,\;90^{\circ}C\;and\;95^{\circ}C$, retention ratios of pigment in cockle were above $95\%$ at all temperature. Soaking in ethanol for 5 minutes resulted in strong adhesion of pigment to meat. Soaking in seasoning liquid containing $10\%$ soy sauce, $5\%$ wasabi, $5\%$ sugar, $2\%$ vinegar, $2\%$ powdered garlic for 3 minutes was effective for instant processing of cooked and frozen cockle after thawing. After 60 days storage at $-20^{\circ}C$, the contents of moisture, crude lipid, carbohydrate, ash and salinity were not changed so much, and pH and TBA values increased to 6.6 and 0.3 compared with 6.2 and 0.2, respectively, while pigment absorbance at 226 nm was decreased from 2.7 to 2.3. However, in case of 60 days storage at $-45^{\circ}C$, there was no change in these compositions.

  • The structure of gonads, gametogenesis and reproductive cycle of the cockle, Fulvia mutice, were studied mainly by histological observation. The materials were monthly sampled in the southern area of Yeosu from October 1980 to September 1981. F. mutica was monoecious. The gonads were situated between the liver tissues and the outer fibronuscular layers compacted by the connective tissue fibers and muscle fibers beneath the outermost layer of simple cuboidal epithelium. The gonad was composed of a number of the ovarian sacs and the testicular tubules which form the tubular structure. Testicular tubules in the mature stage sometimes contained 'testis-ova' The undifferentiated mesenchymal tissues and the eosinophilic cells were abundantly distributed on the germinal epithelium in the early development stage. With the further development of the ovary and testis, these tissues and cells gradually disapprared. The undifferentiated mesenchymal tissues and the eosinophilic cells are related to the growing of the oocytes and spermatocytes . Early multiplicating oogonium was about $10{\mu}m$ in diameter. As the oocytes grow to $27-34\times50-58{\mu}m$ by increasing cytoplasm, the oocytes connected to the basement membrane by their egg-stalks. The ripe eggs were about $60{\mu}m$ in diameter and they were surrounded by gelatinous membrane. Most male germ cells in mature stage were transformed into the spermatozoa and they formed the sperm bundles. After spawning, undischarged ripe eggs and spermatozoa remained in the ovarian sac and the testicular tubule respectively for some time, then they finally degenerated. Especially the early spent ovarian sacs in May did not contract significantly and then they took part in the secondary maturation within two or three months during the summer season. The monthly changes of the fatness well agreed with the reproductive cycle. The reproductive cycle of F. mutica could be classified into six successive stages : multiplicative, growing, mature, spent, degenerative and recovery stage. It seems that the spawning season is closely rotated to the water temperature, and the spawning occurs from May to October at about $20^{\circ}C$ in water temperature. The peak spawning seasons appeared twice a year between June and July and in September. Nuclear DNA content determinations in 15 seawater shellfish species in Korea (한국 해산 패류 15종의 DNA 함량)

  • The object of this study was to obtain nuclear DNA content data for representatives of the 15 shellfish species that inhabit the coast of Korea. In the gastropoda group, the DNA content (pg DNA nucleus-1) was 3.3±0.08 in Haliotis discus hannai and 2.4±0.18 in Batillus cornutus. In the bivalvia group, the DNA content(pg DNA nucleus-1) was 2.0±0.15 in Scapharca broughtonii, 3.0±0.12 in Mytilus galloprovincialis, 2.9±0.05 in Meretrix lusoria, 2.2±0.03 in Meretrix lamarkii, 2.6±0.05 in Fulvia mutica, 1.8±0.18 in Tegillarca granosa, 3.3±0.01 in Solen corneus, 2.2±0.04 in Barnea manilensis, in 2.5±0.32 in Crassostrea gigas, 3.9±0.24 in Atrina pectinate, 3.5±0.15 in Patinopecten yessoensis, 1.9±0.16 in Amygdala philippinarum, and 2.3±0.14 in Pseudocardium sachalinensis. The results of this study provide new information for a better understanding of the genomic evolution process of the shellfish species investigated in this experiment.

Environmental Characteristics of Natural Conditions of the Flat Oyster, Ostrea denselamellosa in Haechang Bay, Korea (해창만 벗굴, Ostrea denselamellosa 서식지의 환경특성)

  • This study was measured environmental factors of flat oyster habitats and biomass of flat oyster to improve the productivity of flat oyster. Water temperature and salinity of the flat oyster habitat ranged from 5.5 to 27.4$^{\circ}C$ and from 31.2 to 33.4 , respectively. Average concentrations of DO, COD, DIN and PO$\_$4/$\^$3/ -P were 7.11 mg/l, 4.55 $\mu\textrm{g}$-at./l and 0.36 $\mu\textrm{g}$-at./l respectively. Surface sediments at the sampling area were composed of coarse sand, sandy silt and silty sand. Average level of IL, COD and AVS in the surface sediments were 2.6%, 13.70 mg/g dry and 0.33 mg/g dry respectively. In each sampling station, total standing crops of phytoplankton showed peaks twice in February and August. Dominant species of phytoplankton occurred in Haechang Bay throughout the year were Skeletonema costatum, Paralia sulcata, Eucampia zodiacus, Chaetoceros curvicetus, C. affinis, C. debilis, C. decipiens, Asterionella glacialis, Pseudonitzschia longissima, Pseudonitzschia seriata, Ceratium furca and C. fusus. Ten species of the bivalves were collected at the flat oyster habitat. Most of bivalves were the eutrophic species Ostrea denselamellosa, Crassostrea gigas, Ruditapes philippinarum, Scaphraca subcrenata, Scapharca broughtonii, Atrina pectinata, Fulvia mutica, Mytilus edulis, Protothaca jedoensis and Megacardita ferruginosa. The mean density of them was 21 inds./㎡ (479.14 g/㎡), while that of the flat oyster was at 0.25 inds./㎡ (231.25 g/㎡).
