Successful Development of Cordyceps bassiana Stromata from Beauveria bassiana

  • A specimen of Beauveria bassiana was collected from Yang-yang of Gangwon province, Korea in October 2006. Conidial isolates were prepared from the specimen by the dilution method and inoculated in brown rice medium for fruiting body production. After nearly two months incubation for perithecial stromata developed from single isolates as well as from their combinations. They were determined as Cordyceps bassiana by observing the stromatal characters and their conidial structures. This is the first report of the development of C. bassiana from B. bassiana cultures.

  • The basidomycetes fungus, Agaricus blazei Murill has been well known as the biologically active polysaccharides. They do not attack cancer cells directly, but produce their antitumor effects by activating various immune responses in the host. This present study was designed to investigate the immune activity of polysaccharides extracted from fruiting body, culture broth, and mycelia. In order to examine the optimum conditions of polysaccharide production, medium and C/N ratio were examined in flask culture. The fine polysaccharides were isolated and purified by DEAE cellulose and Sephadex CL-6B, respectively. Each fraction was spplied to recognize NO synthesis and TNF-${\alpha}$ for immune activity test.

Characteristics of newly bred spore-less cultivar Lentinula edodes 'Daedam' for sawdust cultivation (표고 톱밥재배용 무포자 신품종 '대담' 육성 및 특성)

  • A new spore-less cultivar Lentinula edodes 'Daedam' was bred from monokaryotic strains of 'LE15401-24' and 'LE192118-10'. The optimum temperature for mycelial growth of 'Daedam' on potato dextrose agar was 22~25℃. Total cultivation period of the new cultivar, from inoculation to its first harvest, was 134 days, similar to that of the control cultivar 'Hwadam'. Total yield of 'Daedam' was 222g per 3kg substrate, and was lower than that of control cultivar(266.0g). The fruiting body of 'Daedam' had a thick and small pileus and a longer stem compare to control cultivar. As a result of a analyzing the productivity of 'Daedam' on the different substrate types, the biological efficiency was 26.7% in the 1.2kg cylindrical substrate(CS), which was higher than that of the 3kg rod-type substrate(RS). 'Daedam' had a similar yield compared to 'Hanacham' in first fruiting body production, but the cultivation period was 40 days shorter. Therefore, 'Daedam' can only harvest fruiting bodies once, it is thought that it can be used as spore-less oak mushroom cultivar for short-term cultivation instead of 'Hanacham' in mushroom farms.

Isolation of New Strain of Cordyceps militaris HB8 and Optimal Condition for Production of Adenosine and Cordycepin in Fruit Body

  • Cordyceps has been used in traditional Chinese medicine more than 2000 year ago. In this study, the new Cordyceps militaris was founded and isolated from O-dae mountain in Korea, and was identified its genetic characteristics. The newly isolation strain HB8 was most closet to Cordyceps militaris W141449 (99.82%), Cordyceps militaris JLCY-LI819 (99.82%) and Cordyceps militaris 4642 (99.81%), respectively. the genotypic result was show that train HB8 was belonging to the Cordyceps militaris genus, therefore, Cordycep militaris HB8 proposed with accession number MT835161. This study we find the optimal condition for production of cordycepin from Cordyceps militaris HB8 was 8 mg/g (200 g of pupa, 1 g of KH2PO4, 0.5 g of K2HPO4, 20 g of glucose, 1 g of MgSO4, 0.05 g of vitamin B1, and 1 mg of NAA per liter; light condition 300-700 Lux and day/night was 14 h/10 h) and the optimum condition for the production of adenosine was 2.6 mg/g (15 g of skim milk powder, 1 g of KH2PO4, 0.5 g of K2HPO4, 20 g of glucose, 1 g of MgSO4, 0.05 g of vitamin B1, and 1 mg of NAA per liter; light condition 300-700 Lux and day/night was 14 h/10 h).

Varietal characteristics of cross-bred Cordyceps militaris 'Dowonhongcho' (교배육종에 의한 밀리타리스 동충하초 '도원홍초'의 품종특성)

  • Cordyceps militaris is being studied and cultivated as a medicinal mushroom having many valuable biological and pharmaceutical activities. In the breeding of new C. militaris mushroom, single ascospores were isolated and examined their mycelial growth, mycelial density, and production of stroma and perithecia. Among them selected isolates were crossed and hybrids were produced showing high quality fruiting bodies on artificial media. Mycelial growth rate of new strain 'Dowonhongcho' was higher than that of better on 'Yedang 3' on SDAY at $10-25^{\circ}C$. The stromata of new strain were club-shaped and bright orange-red. Its height was 6.1 cm and the cordycepin content was 0.34% on average. The new strain showed 9% higher yield than 'Yedang 3' with producing firmer fruit bodies. The optimum temperature for mycelial growth was $22{\sim}25^{\circ}C$ and the optimum temperature for stroma development was $18{\sim}22^{\circ}C$. Fruiting bodies were began to produce 45 days later after inoculation. This new cultivar may serve as a valuable one for artificial cultivation and industrial-scale production of C. militaris.

Varietal characteristics of new Cordyceps militaris 'Dowonhongcho 2ho' improved by mating type molecular markers (교배형 분자마커를 이용한 신품종 밀리타리스 동충하초 '도원홍초 2호'의 품종 특성)

  • The mushroom species Cordyceps militaris has been studied and cultivated as a medicinal mushroom due to its multiple valuable biological and pharmaceutical activities. For breeding new strains of C. militaris, multiplex PCR assays were performed using primers specific for its mating type genes, MAT1-1 and MAT1-2. Mating types and mating status were confirmed, as evidenced by DNA bands at 233-bp and 191-bp for MAT1-1 and MAT1-2 respectively. The novel 'Dowonhongcho 2ho' was developed through mating; they were found to possess high-quality fruiting bodies when grown in artificial media. The stromata of the new strain were club-shaped, with a bright orange-red color, and measured 7.1 cm in length. They had an average cordycepin content of 0.33%. Compared to 'Dowonhongcho,' the new strain had a 7% higher yield, as well as firm fruiting bodies. The optimum temperature for mycelial growth was $20{\sim}25^{\circ}C$, and the optimum temperature for stroma development was $18{\sim}22^{\circ}C$. The fruiting bodies developed after 49.1 days from inoculation. The use of mating type molecular markers improved the breeding efficiency of the new strain 'Dowonhongcho 2ho.' Thus, they may be valuable for artificial cultivation and industrial-scale production of C. militaris with excellent characteristics.

Various physiological effects from fruiting body extracts of Phellinus baumii (장수진흙버섯 자실체 추출물의 다양한 생리활성 효과)

  • Phellinus baumii, a white-rot fungus, has been used for centuries as folk medicine in China, Japan, and Korea. This study aimed to evaluate the in vitro anti-diabetic, and anti-cholinesterase, and in vivo anti-inflammatory effects of the fruiting bodies of P. baumii. The methanol (ME) and hot water (HE) extracts (2.0 mg/mL) of P. baumii fruiting bodies suppressed α-amylase activity, exactly 61.33%, and 65.00%, respectively; of note, acarbose, the positive control, inhibited 93.33% of the α-amylase activity. Moreover, the ME and HE (2.0 mg/mL) inhibited 89.67% and 91.00%, respectively, of the activity of α-glucosidase activity, whereas the same concentration of acarbose suppressed 84.67% of the α-glucosidase activity. The ME and HE (1.0 mg/mL) also inhibited 96.05% and 94.58%, respectively, of the acetylcholinesterase (AChE) activity; galanthamine, the positive control, led to an inhibition of 81.12%. The butyrylcholinesterase (BChE) activity was also inhibited by ME and HE (1.0 mg/mL; 91.05% and 82.27%, respectively); of note, the same concentration of galanthamine suppressed 81.12% of the BChE activity. The production of NO in LPS-induced RAW 264.7 macrophages was significantly suppressed by both ME and HE treatments. Importantly, the carrageenan-activated rat hind-paw edema was significantly reduced 2-6 h after ME administration (50 mg/mL). Taken together, the results suggest that the fruiting bodies of P. baumii have α-amylase, α-glucosidase, α-cholinesterase, and anti-inflammatory activities, and, therefore, may be good natural sources for the promotion of human health.

Ethanol Production from Various Sugars and Cellulosic Biomass by White Rot Fungus Lenzites betulinus

  • Lenzites betulinus, known as gilled polypore belongs to Basidiomycota was isolated from fruiting body on broadleaf dead trees. It was found that the mycelia of white rot fungus Lenzites betulinus IUM 5468 produced ethanol from various sugars, including glucose, mannose, galactose, and cellobiose with a yield of 0.38, 0.26, 0.07, and 0.26 g of ethanol per gram of sugar consumed, respectively. This fungus relatively exhibited a good ethanol production from xylose at 0.26 g of ethanol per gram of sugar consumed. However, the ethanol conversion rate of arabinose was relatively low (at 0.07 g of ethanol per gram sugar). L. betulinus was capable of producing ethanol directly from rice straw and corn stalks at 0.22 g and 0.16 g of ethanol per gram of substrates, respectively, when this fungus was cultured in a basal medium containing 20 g/L rice straw or corn stalks. These results indicate that L. betulinus can produce ethanol efficiently from glucose, mannose, and cellobiose and produce ethanol very poorly from galactose and arabinose. Therefore, it is suggested that this fungus can ferment ethanol from various sugars and hydrolyze cellulosic materials to sugars and convert them to ethanol simultaneously.

The Medium Development for Entomopathogenic Fungi by Using Silkworm Powder (누에분말을 이용한 곤충기생균의 최적생장배지 개발)

  • This study was carried out to investigate the optimal medium and optimal dextrose concentration of 12 entomopathogenic fungi. The colonies of Cordyceps gracilioides, C. sp.(J23), Paecilomyces farinosus(J5), P tenuipes, Paecilomyces sp.(J4), Beauveria bassiana and B. brongniartii on SLA(silkworm larva agar) medium are similar to those on PDA(Potato dextrose agar) medium. On the contrary, the colonies of 5 fungi are very various according to its medium. The optimal medium for 1 1 fungi was SLA-A medium, which was better than PDA medium with regard to the mycelial growth. The optimal concentration on mycelial growth was superior to 6% for 6 fungi. When silkworm larvae was injected with P. tenuipes on SLA medium for fruiting body production. Infection rate, fruiting body formation rate, synnemata number and synnemata length were 91.3 $\pm$ 1.5%, 88.6 $\pm$ 2.3%, 56.6 $\pm$ 3.2 and 33.4 $\pm$ 2.88 ㎜ respectively. In economical analysis, the prototype medium can reduce 61.9% of production cost per 1 liter compared with PDA medium.

Optimization of Cultivation and Extraction Conditions of Pupae-Cordyceps for Cordycepin Production

  • Cordycepin is a characteristic bioactive compound of Cordyceps militaris with various beneficial effects. Cordyceps grows on both grains and insects, and the content of cordycepin varies depending on the cultivation conditions. In this study, the effect of culture conditions on the cordycepin content was analyzed and the extraction conditions were optimized. Analysis of cordycepin content in Pupae-Cordyceps found that it was highly affected by temperature in culture conditions. In the case of mycelium, it grows well at 20 and 25 ℃, but not at 30 ℃. However, the content of cordycepin was highest at 30℃ and less at 20 ℃. The fruiting body also showed a similar tendency: growth was 20 ℃ > 25 ℃ > 30 ℃, but the cordycepin content was 30 ℃ > 25 ℃ > 20 ℃. The content of cordycepin decreased after the fruiting bodies were produced. Next, extraction conditions such as solvent and time were optimized for maximum cordycepin content using response surface methodology (RSM). There was a large difference in the content of cordycepin according to the content of ethanol and the extraction temperature. Through RSM, it was confirmed that the optimum condition for extraction of cordycepin was 48.9 ℃ using 49.0% ethanol, and 160.9 mg/g extract could be obtained under this condition. In conclusion, this study suggested the optimized conditions for the cultivation and extraction of Pupae-Cordyceps for maximizing the content of cordycepin, and this may be applied to the discovery of materials using cordycepin.
