가시성을 표시한 사과 검출 데이터셋과 적응형 히트맵 회귀를 이용한 딥러닝 검출 (Apple detection dataset with visibility and deep learning detection using adaptive heatmap regression)

  • 과실 수확 분야에서 다양한 계절성과 수확 비용 상승 등으로 자동 로봇 수확에 대한 관심이 증가하고 있다. 빛의 변화, 바람에 의한 진동, 나뭇잎 및 가지 겹침 등 복잡한 과수원 환경에서 정확한 사과 검출은 어려운 문제이다. 본 논문에서는 로봇 자동 사과 수확에 유리한 데이터셋과 적응형 히트맵 회귀 모델을 소개한다. 사과 데이터셋은 사과 위치뿐만 아니라 가시성을 같이 레이블링하였다. 가시성에 따라 가우시안 모양을 조절하는 적응형 히트맵 회귀 모델을 사용하여 사과 중심점을 검출하는 방법을 제안한다. 실험 결과 MAP@K가 K=5와 K=10일 때 0.9809, 0.9801로 사과 수확 로봇에 응용 가능한 성능을 나타내었다.

Isolation and Development of Quantification Method for Cyanidin-3-Glucoside and Cyanidin-3-Rutinoside in Mulberry Fruit by High-Performance Countercurrent Chromatography and High-Performance Liquid Chromatography

  • Cyanidin-3-glucoside (C3G) and cyanidin-3-rutinoside (C3R) were isolated by high-performance countercurrent chromatography (HPCCC) using a two-phase solvent system composed of tert-butyl methyl ether/n-butanol/acetonitrile/water/trifluoroacetic acid (1 : 3 : 1 : 5 : 0.01, v/v) to give pure C3G (34.1 mg) and C3R (14.3 mg) from 1.5 g crude mulberry fruit extract. Using the pure C3G and C3R, a reliable high-performance liquid chromatography (HPLC) method was developed and validated to determine the C3G and C3R contents in mulberry fruit. C3G and C3R were separated simultaneously using an Eclipse XDB-C18 column ($4.6{\times}250mm$ I.D., $5{\mu}m$) coupled with a photodiode array detector (PDA). The gradient elution of the mobile phase consisting of acetonitrile (0.5% formic acid) and water (0.5% formic acid) was applied (1.0 mL/min), and the detection wavelength was 520 nm. The calibration curves of C3G and C3R showed good linearity (both with $r^2=0.9996$) in the concentration range $15.625-500{\mu}g/mL$, and the relative standard deviations (RSD%) of intra- and inter-day variability were in the ranges 2.1 - 8.2% and 4.1 - 17.1%, respectively. The accuracies were ranged 96.5 - 102.6% for C3G and C3R, respectively. The developed HPLC method was used to determine the contents of C3G and C3R in newly harvested mulberry from eight different provinces of Korea.

Improved RNA extraction for fruit tree viruses in RT-PCR assay

  • Tissues from woody plant contain higher amount of phenolic compounds and polysaccharides, which give inhibitory effects on reverse transcriptase and/or Taq ploymerase. The common multiple-step protocols using several additives to inhibit polyphenoic compounds during nucleic acid extraction are time consuming and laborious. Sodium sulfite (Na$_2$SO$_3$) was used as inhibitor of polyphenolic oxidases in extraction buffer and compare it's effect between commercial RNA extraction kit and small-scale double-stranded RNA (dsRNA) extraction by RT-PCR. During nucleic acid extraction procedure, addition of 0.5%-1.5% (w/v) sodium sulfite to Iysis buffer or STE buffer resulted in lighter color change than extracts without sodium sulfite and improve the RT-PCR detection. When commercial RNA extraction kit used, optimal concentration of sodium sulfite were variable according to the host plant. However, using dsRNA as RT-PCR template, 1.5% sodium sulfite in STE buffer improves the detection of both viruses and unspecific amplifications were reduced significantly, Furthermore, when viruses existed at low titers in host plant, small-scale dsRNA extractions were very reliable.

PCR Based Detection of Phellinus linteus using Specific Primers Generated from Universal Rice Primer(URP) Derived PCR Polymorphic Band

  • This study was carried out to develop specific primers for PCR detection of Phellinus linteus. Diverse genomes of 15 Phellinus spp. including five Phellinus linteus isolates were fingerprinted by Primer Universal rice primer(URP)1F. The URP-PCR pattern differentiated P. linteus isolates from other phellinus spp. A polymorphic band(2.8 kb), which is unique for P. linteus isolates, was isolated and sequenced. Twenty four-oligonucleotide primer pairs were designed based on information of DNA sequence. The primer set(PLSPF2/PLSPR1) amplified single band(2.2 kb) of expected size with genomic DNA from seven Phellinus linteus, but not with that of other Phellinus species tested. The primers could be used identically in both DNA samples from mycelium and fruit bodies. This specific primers could offer a useful tool for detecting and identifying P. linteus rapidly.

A Simple and Reliable Molecular Detection Method for Tomato yellow leaf curl virus in Solanum lycopersicum without DNA Extraction

  • In the present work, a pair of primers specific to Tomato yellow leaf curl virus (TYLCV) was designed to allow specific amplification of DNA fragments from any TYLCV isolates using an extensive alignment of the complete genome sequences of TYLCV isolates deposited in the GenBank database. A pair of primers which allows the specific amplification of tomato ${\beta}$-tubulin gene was also analyzed as an internal PCR control. A duplex PCR method with the developed primer sets showed that TYLCV could be directly detected from the leaf crude sap of infected tomato plants. In addition, our developed duplex PCR method could determine PCR errors for TYLCV diagnosis, suggesting that this duplex PCR method with the primer sets is a good tool for specific and sensitive TYLCV diagnosis. The developed duplex PCR method was further verified from tomato samples collected from some farms in Korea, suggesting that this developed PCR method is a simple and reliable tool for rapid and large-scale TYLCV detections in tomato plants.

Research on the Variable Rate Spraying System Based on Canopy Volume Measurement

  • Characteristics of fruit tree canopies are important target information for adjusting the pesticide application rate in variable rate spraying in orchards. Therefore, the target detection of the canopy characteristics is very important. In this study, a canopy volume measurement method for peach trees was presented and a variable rate spraying system based on canopy volume measurement was developed using the ultrasonic sensing, one of the most effective target detection method. Ten ultrasonic sensors and two flow control units were mounted on the orchard air-assisted sprayer. The ultrasonic sensors were used to detect the canopy diameters and the flow controls were used to modify the flow rate of the nozzles in real time. Two treatments were established: a constant application rate of $300Lha^{-1}$ was set as the control treatment for the comparison with the variable rate application at a $0.095Lm^{-3}$ canopy. The tracer deposition at different parts of peach trees and the tracer losses to the ground (between rows and within rows) were analyzed in detail under constant rate and variable rate application. The results showed that there were no significant differences between two treatments in the liquid distribution and the capability to reach the inner parts of the crop canopies.

휴면아 경정 배양법을 통한 포도 '거봉' 에서 Grapevine fleck virus의 제거 (Elimination of Grapevine fleck virus from infected grapevines 'Kyoho' through meristem-tip culture of dormant buds)

  • 본 연구진은 세계 최초로 GFkV에 단독 감염된 포도 품종 거봉의 휴면아로부터 직접 경정 분열조직을 절취하여 배양함으로써 별도의 열처리나 화학처리 없이 GFkV가 제거된 신초 재생에 성공하였다. GFkV는 포도 식물체의 체관에만 한정적으로 존재하고 접목으로 감염되는 포도 바이러스이다. 경정조직은 $4^{\circ}C$에서 일정기간 저장된 1년생 경화가지의 휴면아로부터 0.3 mm (73 절편체)와 0.8 mm (5절편체)를 절취하였는데, 절취 부위는 생장점(apical meristem)을 포함하여 2 ~ 5개의 엽원기(leaf primordia)와 미분화 원기(uncommitted primordia) 1 ~ 4개를 포함하였다. 절취된 경정조직을 BA 3.0 mg/L와 IBA 0.1 mg/L가 혼합된 배지에 16주간 배양한 결과, 0.3 mm와 0.8 mm의 경정조직에서 각각 4.1%와 40.0%의 신초 재생률이 관찰되었고, 재생된 신초에서의 바이러스 제거율(재생된 신초수에 대한 RT-PCR negative 신초수의 백분율)은 0.3 mm의 경정조직에서는 100%를, 0.8 mm에서는 50%를 나타내었다. 신초를 증식시킨 후 감염바이러스를 다시 진단한 결과, 신초가 처음 재생되었을 때의 진단결과와 동일하였다. 휴면아로부터 분열조직을 배양하여 바이러스가 제거된 신초를 확보한 예는 세계적으로 보고된 바가 없는 것으로서, 본 연구진의 새로운 바이러스 제거방법은 향후 포도 뿐만 아니라 과수를 포함한 낙엽성 수목의 무병묘 생산에 매우 유용하게 활용될 수 있을 것이다.

DNA Comet Assay를 이용한 과일의 방사선 조사 확인 (Detection of Irradiated Fruits Using the DNA Comet Assay)

  • 과일의 방사선 조사 여부를 DNA comet assay로 확인하였다. 포도, 자두, 딸기, 복숭아, 사과, 천도복숭아를 구입하여 1.0kGy이하의 저선량으로 조사하고 비조사 시료와 조사시료간의 DNA 손상청도를 육안 검사 및 측정된 comet tail length로 비교하였다. 모든 시료에서 비조사 시료보다 조사시료의 tail length가 더 길었으며 포도와 자두는 $0{\sim}0.5kGy$에서 뚜렷한 증가가 관찰되었다. 특히 자두의 비조사 시료는 포도, 딸기, 복숭아, 사과, 그리고 천도복숭아의 비조사 시료에 비해서 손상된 세포의 comet 모양의 핵이 많이 관찰되기는 하였으나 항상 비손상된 세포의 원형모양의 핵이 동반되었으며 조사된 시료에서는 모두 전반적으로 comet모양의 핵이 관찰되어 비조사 시료와 조사시료 간에 comet 양상을 비교할 수 있었다. Comet 분석을 이용한 과일의 방사선 조사 유무의 확인은 비조사 시료와 조사시료의 tail length의 측정 및 육안 검사 그리고 통계분석으로 확인이 가능하였다. 한편, 이미지 분석기와 같은 정밀한 시스템이 동반되면 저선량으로 조사된 시료들간의 comet 양상을 정확하게 확인하는 것이 가능하다고 생각된다. 따라서 식품의 'DNA comet assay'는 시료의 저장상태 및 화학적 물리적 충격에 대한 몇가지 제한점을 가지고 있지만 간단하고 신속하게 검지할 수 있으므로 다양한 식품의 방사선 조사 유무를 확인하는데 유용하게 활용될 수 있을 것으로 기대한다.

A Novel Sensor Based on Electropolymerization Poly(safranine) Film Electrode for Voltammetric Determination of 4-Nitrophenol

  • A novel poly(safranine)-modified electrode has been constructed for the determination of 4-nitrophenol (4-NP) in natural water sample. The electrochemical behavior of poly(safranine) film electrode and its electrocatalytic activity toward 4-NP were studied in detail by cyclic voltammetry (CV) and adsorptive linear stripping voltammetry (LSV). All experimental parameters were optimized and LSV was proposed for its determination. In optimal working conditions, the reduction current of 4-NP at this poly(safranine)-modified electrode exhibited a good linear relationship with 4-NP concentration in the range of $8.0{\times}10^{-8}$ to $4.0{\times}10^{-5}mol\;L^{-1}$. The detection limit was $3.0{\times}10^{-8}mol\;L^{-1}$. The high sensitivity and selectivity of the sensor were demonstrated by its practical application for the determination of trace amounts of 4-NP in natural water and fruit samples.

Quantitative Analysis of chiisanoside in Acanthopanax Species by HPLC

  • High performance liquid chromatography (HPLC) was used for the analysis of chiisanoside in Acanthopanax species. A reverse-phase system using a gradient of H$_2$O and acetonitrile as the mobile phase was developed and a wavelength of detection was at 210 nm. The analysis was successfully carried out for 30 min. Chiisanoside was measured in the fruit, stem and root of A. sessiliflorus, A. koreanus, A. divaricatus and A. senticosus.
