생약의 ${\beta}-Glucuronidase$ 저해와 간장보호효과 (${\beta}-Glucuronidase-inhibitory\;Activity$ and Hepatoprotective Effect of Herbal Medicines)

  • Inhibitory effect of water and ethyl acetate extract of 60 kinds of herbal medicines was investigated on ${\beta}-glucuronidase$. Among water extract of them, Galla Rhois had the most potent ${\beta}-glucuronidase-inhibitory\;activity$. Termaliae Fructus, Amomi Tsa-ko Fructus and Arecae Semen were also potent inhibitors. Among ethyl acetate extract of them, Galla Rhois had the most potent ${\beta}-glucuronidase-inhibitory\;activity$. Nelumbinis Semen. Ephedrae Radix and Termaliae Fructus were also potent inhibitors. The extract of Galla Rhois had potent hepatoprotective effect on $CCl_4-induced$ hepatotoxicity of rats. These results suggest that the ${\beta}-glucuronidase$ seems to be closely related to the liver injury, which could be prevented by the inhibitor of ${\beta}-glucuronidase$.

  • Dalwoneum is the one of Chinese traditional prescription used for the treatment of liver disease. This prescription consists of Arecae Semen (12 g), Magnoliae Cortex (9 g), Amomi tsao-ko Fructus (3 g), Anemarrhenae Rhizoma (12 g), Paeoniae Radix (9 g), Scutellariae Radix (12 g), and Glycyrrhizae Radix (3 g). Water extract of Dalwoneum showed the significant hepatoprotective effect on tacrine-induced cytotoxicity in Hep G2 cells. (omitted)

  • In order to search for antioxidants from the plants, eighty-two kinds of herbal medicines were investigated. The MeOH extracts of Euryales Semen, Alpiniae Officinari Rhizoma. Drynariae Rhizoma, Sophorae Flos, Trachelospermi Caulis, Crassirhizomae Rhizoma, Euphorbiae lathyridis Semen, Lini Semen, Myristicae Semen, Epimedii Herba, Santali Lignum rubrum, Perillae Herba, Amomi Tsao-Ko Fructus and Garanii Herba showed potent antioxidative activities using the 1,1-diphenyl-2-picrylhydrazyl free radical generating system. 생약의 Protein Tyrosine Phosphatase 1B 저해활성 검색 (Screening of Medicinal Herbs for Inhibitory Activity against Protein Tyrosine Phosphatase 1B)

  • Protein tyrosine phosphatase 1B (PTP1B) is predicted to be therapeutic target in treatment of type 2 diabetes and obesity. Thus, in order to search for PTP1B inhibitors, we screened the inhibitory activity of PTP1B in the water extracts of 84 medicinal herbs. Among them, the extracts of Pini Folium, Magnoliae Cortex, Artemisiae asiaticae Herba, Schizonepetae Herba, Menthae Herba, Mume Fructus, Cimicifugae Rhizoma, and Amomi Cardamomi Fructus showed relatively significant (58-68%) inhibitory activity against PTP1B. Especially, the methylene chloride fraction of the methanol extract of Menthae Herba (81% inhibition at 30 ${\mu}g$/ml) showed more potent inhibitory activity against PTP1B than others.

  • In vitro sensitivity testing was performed for 21 kinds putative anticancer drugs selected from references and information. Cellular damage of P815 mastocytoma cells following exposure to water extracts of drugs was evaluated by colony formation assay. Highly effective drugs with more than 50% inhibition of colony formation were seven (Houttuyniae Herba, Sanguisorbae Radix, Nepetae Herba, Manitis Squama, Lonicerae Flos, Amomi Semen, Polyporus), though not more effective than BCNU. According to the results of $^3H-thymidine$ incorporation assay for determination of selective cytotoxicity, 3 of these drugs (Houttuyniae Herba, Polyporus, Manitis Squama) were found to be low cytotoxic to normal mouse lymphoid cells. These findings suggest that the above 3 drugs may be used for effective anticancer drugs in vivo.

비만(肥滿) 치료(治療)를 위한 경혈(經穴) 및 약물(藥物)에 관한 고찰(考察) (A literatural study on acupoints and herbs for treating obesity by herbal acupuncture)

  • Objectives : This study was designed to investigate possibilities of herbal acupuncture for obesity. Methods : We researched into 23 kinds of literature and considered acupoints and herbs of herbal acupuncture for obesity. Results : Prescriptions for obesity include Erchentang(二陳湯), Liujunzitang(六君子湯) for phlegm-retention syndrome(痰飮), Wulingsan(五?散), Qianghuoshengshitang(羌活勝濕湯), Shenshitang(渗濕湯), Lingguishugantang(?桂朮甘湯) for Dampness syndrome(濕證) and Pingweisan(平胃散) for activating spleen and stomach(運化脾胃). And Rhizoma atractylodis(蒼朮), Tuber pinelliae(半夏), Rhizoma atractylodis macrocephalae(白朮), Fructus ponciri(枳殼), Fructus immaturus ponciri(枳實), Poria(茯?), Semen coicis(薏苡仁), Rhizoma alismatis(澤瀉) Fructus amomi(砂仁), Polyporus umbellatus(猪?), Medulla tetrapanacis(通草) can be used for both herbal medication and herbal acupuncture. And also there are some important acupoints for treating obesity, which are Sanyinjiao(三陰交), Pishu(脾兪), Gongsun(公孫), Fenglong(豊隆), Yinlingquan(陰陵泉), Zusanli(足三里), Qihai(氣海), Sanjiaoshu(三焦兪), Zhongwan(中脘), Neiguan(內關), Weishu(胃兪), Shenshu(腎兪) and Shangjuxu(上巨虛). Conclusions : We recommend that herbal acupuncture can be used for obesity. This method is a kind of injection using herbal extract, which can apply pharmacognosy theory and meridian system at the same time.

태원음(胎元飮)의 효능(效能) 및 가감법(加減法)에 관한 문헌적(文獻的) 고찰(考察) (Literatural Study on the utility and additional methods of TaeWonEum)

  • According to the literatural study on the utility of TaeWonEum, the results were as follows 1. TaeWonEum is the prescription originally registered on Zang jie Bin's ${\ll}$Jing-Yue-Quan-Shu${\gg}$ and because of the dificiency of the renmai, the chongmai in women, it is made for the discomfort of the pregnancy 2. On the treatment of miccarriage, they were used to the methods of tonificating qi and blood, tonificating insufficiency, making flesh, eliminating wind, tonificating the spleen, clearing heat, eliminating stagnant blood, tonificating blood, growing zhang qi, etc but it is almostly used to the method of tonificating qi and blood 3. TaeWonEum is used on the purposes of tonificating the spleen and stomach, qi and blood, on the case of both the discomfort of the pregnancy by the dificiency of the renmai, the chongmai in women, and the insufficiency of fetal growth by the difficiency of the spleen and stomach, qi and blood 4. The symptoms can be used which are the fetal problem of incipient pregnancy, quickening, lumbago, distension, pallor, weakness of the mind, dizziness, cold, anorexia 5. The additional methods of TaeWonEum are as follows when much turbid enuresis is concerned, Disscoreae Rhizoma, Psoraleae Fructus, Schizandrae Fructus can be added when a patient is very difficient of qi, Atractylodis Macrocephalae Rhizoma is doubled and Astragali Radix can be added when a patient is often vomiting with asthenia-cold, Typhae Pollen can be added when a patient has fever and weak, Scutellariae Radix or Rehmanniae Radix can be added and E ucommiae Cortex can be subtracted when a patient has the colicky pain because of the difficiency of yin, Lycii Fructus can be added when a patient is at a great rage and qi is ascenting, Cyperi Rhizoma or Amomi Fructus can be added when a patient has hemorrage with trauma, Dipsaci Radix, Asini Gelatinum can be added when a patient's vomiting can not be reduced, Pinelliae Rhizoma, Zingiberis Rhizoma Recens can be added

Anti-thrombotic Effects of Modified Jeho-tang using a $FeCl_3$-induced Carotid Arterial Thrombosis Model

  • Objectives: The aim of this study was to examine the antithrombotic effects of the four herbal ingredients (Mume Fructus, MF; Santali Albi Lignum, SAL; Amomi Tsao-Ko Fructus, ATF; and Amomi Fructus, AF) of modified Jeho-tang (MJHT) in a ferric chloride ($FeCl_3$)-induced carotid arterial thrombosis model. Methods: Thirty minutes prior to a 35% $FeCl_3$ application, Sprague-Dawley (SD) rats were injected with saline, MF, SAL, ATF or AF (100 mg/kg, intraperitoneal injection), respectively. The effect of the MJHT ingredients was examined for time to occlusion (TTO) and thrombus weight (TW) in a $FeCl_3$-induced thrombosis model. Histological analysis was performed to examine the effect of the MJHT ingredients on collagen fiber damage using hematoxylin & eosin and Masson's trichrome staining. Results: Compared with vehicle treatment, MF, SAL and ATF treatment delayed TTO (vehicle, $8.11{\pm}0.60$ min; MF, $16.67{\pm}1.03$ min; SAL, $17.50{\pm}1.52$ min and ATF, $13.33{\pm}1.21$ min; P < 0.001) and inhibited thrombus formation (vehicle, $0.79{\pm}0.03$ mg/mm; MF, $0.61{\pm}0.07$ mg/mm; SAL, $0.57{\pm}0.03$ mg/mm and ATF, $0.72{\pm}0.02$ mg/mm; P < 0.001). In addition, each herbal ingredient of MJHT except for AF prevented the collagen fiber damage induced by a 35% $FeCl_3$ application. These results indicate that the MJHT ingredients MF ${\geq}$ SAL ${\geq}$ ATF ${\geq}$ AF possess antithrombotic activity in a $FeCl_3$-induced carotid arterial thrombosis. Conclusions: Altogether, these results are the first evidence that the MJHT ingredients MF, SAL and ATF have the ability to prevent vascular damage and thrombus formation in $FeCl_3$-induced carotid arterial thrombosis.

  • In this dissertation, I will focus on the channel entry, the effect and the treatment throughout books of oriental medicine from ancient to modern in order to classify the medicines of the Spleen as main or supplementary organ. The results are as follows: 1. The medicines which work on the other viscera(他臟) chiefly were 12, which were Talcum(滑石), Bupleuri Radix(柴胡), Semen Lepidii Seu Descurainiae, Mori Cotex Radicis(桑白皮), Aurantii lmmaturi Pericarpium(靑皮), Gardeniae Fructus(梔子), Forsythiae Frucus(連翹), Antelopis cornu(羚羊角), Alimatis Rhizoma(澤瀉), Epimedii Herba(仙靈脾), Cyperi Rhizoma(香附子), Rhizome Chuanxiong(川芎). 3. medicines, effected on the Spleen functioned through any other viscera were as follows: Talcum(滑石) works to treat renal heat Entering the Spleen(腎熱入脾) Bupleuri Radix(柴胡) works to treat Hepatic Asthenia Entering the Spleen(肝虛入脾) Semen Lepidii Seu Descurainiae and Mori Cotex Radicis(桑白皮) works to treat Pulmonary gi Entering the Spleen(肺氣入脾) Aurantii lmmaturi Pericarpium(靑皮) works to treat Hepatic gi Entering the Spleen(肝氣入脾) Gardeniae Fructus(梔子) and Forsythiae Frucus(連翹) works to treat Cardiac Heat Entering the Spleen(心熱入脾) Antelopis cornu(羚羊角) works to treat Hepatic wind Entering the Spleen(肝風人脾) Alimatis Rhizoma(澤瀉) works to treat Hepatic heat Entering the Spleen(肝熱入脾) Epimedii Herba(仙靈脾) works to treat Renal asthenia Entering the Spleen(腎虛入脾) Cyperi Rhizoma(香附子) 와 Rhizome Chuanxiong(川芎) works to treat Hepatic gi Entering the Spleen(肝氣入脾) In the study of concerning the medicines effected on the spleen, It is considered that it dedicated to development of the medicines related to the spleen and making efficient use of the medicines.

  • 1. Objectives: The purpose of this study is to understand the origin, changes and the constructive principles of Hyangsayangyitang (XiangShaYangWeiTang). 2. Methods: Hyangsayangyi-tang and other related prescriptions were analyzed in terms of pathology, based on "Donguibogam(東醫寶鑑)", "Donguisusebowon Chobongwon(東醫壽世保元 草本卷)", "Donguisusebowon Gabobon(東醫壽世保元 甲午本)", "Donguisusebowon Sinchukbon(東醫壽世保元 辛丑本)". 3. Results and Conclusions: (1) The origin of Hyangsayangyi-tang is Hyangsayukgunja-tang from "Gongshin Uigamseo(龔信 醫鑑書"). Hyangsayangyi-tang inherited some of the principles from"Euihangangmok(醫學綱目)" Jeonssiyigong-san's (全氏異功散) application. (2) "Gabobon(甲午本)"'s prescription emphasizes the effect of lossen up by using Pogostemonis Herba(藿香). "Sinchukbon(辛丑本)"'s prescription emphasizes the effect of warm up stomach by using Zingiberis Rhizoma(乾薑). (3) Ginseng Radix(人蔘), Atractlodis Rhizoma White(白朮), Glycyrrhizae Radix(甘草), Paeoniae Radix Alba(白芍藥) support Soeumin's spleen root (脾元), and Zingiberis Rhizoma(乾薑), Myristicae Semen(肉豆蔲) warm up stomach, and Pinelliae Rhizoma(半夏), Citri Pericarpium(陳皮) circulate Qi, Crataegi Fructus(山楂), Amomi Fructus(砂仁), Cyperi Rhizoma(香附子) encourage digestion to maintain descending Yin.
