참게(Eriocheir sinensis)와 꽃게(Portunus trituberculatus)의 2종으로부터 genomic DNA를 분리 추출하였다. 선택된 7개의 OPA-05, OPA-13, OPA-16, OPB-06, OPB-15, OPB-17 and OPD-10의 RAPD primer를 이용하여 identical, polymorphic 그리고 specific fragment를 얻어냈다. 본 연구에서 부안산 참게 집단에서는 505개의 fragment가 나타났고, 꽃게 집단에서는 513개의 fragment가 확인되었다. 참게 집단에서는 165개의 identical fragment가 나타났으며, 이는 primer당 평균적으로 23.6개의 fragment로 확인되었다. 또한 꽃게 집단에서는 66개로서 평균해서 primer당 9.4개의 identical fragment가 나타났다. 참게 집단과 꽃게 집단의 polymorphic fragment는 각각 50개와 14개로 나타났고, 참게 집단과 꽃게 집단의 경우 OPB-17에서 identical fragment가 300 bp의 크기에서 확인되었다. 각각을 비교해 보았을 때 유전적 차이는 참게 집단에서보다 꽃게 집단에서 더 높은 수치를 나타내었고, 2종 사이에서 $0.726{\pm}0.004$의 수치를 나타내었다. 7개의 primer를 사용하여 얻어진 dendrogram은 cluster 1(FRESHWATER 01), cluster 2(FRESHWATER 02, 03, 04, 05 및 06), cluster 3(FRESHWATER 07, 08, 09, 10 및 11) 및 cluster 4(SWIMMING 12, 13, 14, 15, 16, 17, 18, 19, 20, 21 and 22)와 같이 4개의 유전적 클러스터로 나뉘어졌다. 꽃게 집단에서 18번째 개체(SWIMMING no. 18)와 17번째 개체 (SWIMMING no. 17) 사이가 가장 가까운 유전적 관계(genetic distance 0.096)를 나타내었다. 궁극적으로 볼 때 참게 집단의 2번째(FRESHWATER no. 02)와 참게 집단의 3번째(FRESHWATER no. 03) 개체 사이가 가장 먼 유전적 거리 (genetic distance=0.770)를 나타내었다. 위에서 언급했던 것처럼 RAPD-PCR 방법은 참게 및 꽃게 2종의 종 판별을 하기 위한 진단적 표지 (diagnostic marker)로 이용할 수 있는 잠재력을 가지고 있는 것으로 확인되었다.
The effect of freshwater crab meat additive on protein quality of tofu was studied. Tofu containing freshwater crab meat(TCM) prepared by the formulation for the best sensory qualities had about 20% more lipid and ash content than those of commercial tofu. TCM showed a higher content of lysine, methionine, cysteine and tyrosine compared with those in commercial tofu. TCM was comparable to commercial tofu(CT) regarding the in vitro protein digestibility(95%). A considerable difference in computed protein efficiency ratio(C-PER) resulted between CT(1.76-1.81) and TCM(2.61). Therefore, crab meat has potential as an ingredient for enriching essential amino acid and improving protein quality of CT.
Genomic DNA samples isolated from four geographical freshwater crab (Eriocheir sinensis) populations collected in the inland of the Korean Peninsula (Gunsan, Paju, and Nampo) and a Chinese site, were used for PCR amplification. Seven decamer primers generated 19 specific loci (19/243 loci, 7.81%) in the Gunsan population, 32 (32/215 loci, 14.88%) in the Paju population, 19 (19/231 loci, 8.23%) in the Nampo population and 62 (62/340 loci, 18.24%) in a Chinese population. The average 8.9 specific loci exhibited inter-individual-specific characteristics, thus revealing DNA polymorphisms in the Chinese population. The number of unique shared loci to each population and number of shared loci by the four populations were generated by molecular analysis using seven primers in four populations. 35 unique shared loci to each population, with an average of 5.0 per primer, were observed in the Gunsan population, and 50 loci, with an average of 7.1 per primer, were observed in the Chinese population. The hierarchical dendrogram indicates three main branches: cluster 1 (GUNSAN 01$\sim$GUNSAN 05, PAJU 06$\sim$PAJU 10 and NAMPO 11$\sim$NAMPO 15) and cluster 2 (CHINESE 16, 17, 18, 19 and 20). Conclusively individual no. 20 of the PAJU 10 freshwater crab was most distantly related to CHINESE no. 20 (genetic distance = 0.667). Taken together, these results demonstrate the potential of RAPD analysis to identify diagnostic markers for the identification of four freshwater crab populations.
To optimize the mixing ratio of ingredients for optimal sensory qualities of seasoned tofu, a response surface methodology with a central composite design was performed on tofu containing freshwater crab meat (TCM). Using the desirability function technique, the optimal formulation was determined to be 3.67 g of freeze dried meat, 5.54 g of garlic powder, and 2,120 mL of soymilk. In the texture profile analysis, tofu prepared using the optimal ingredient ratio had a higher hardness, cohesiveness and gumminess than commercial tofu but TCM value for adhesiveness, springiness and chewiness were lower. However, the texture properties of TCM were not significantly different than those of commercial tofu.
Phenotypic plasticity is a rapid response mechanism that enables organisms to acclimate and survive in changing environments. The Chinese mitten crab (Eriocheir sinensis) survives and thrives in different and even introduced habitats, thereby indicating its high phenotypic plasticity. However, the underpinnings of the high plasticity of E. sinensis have not been comprehensively investigated. In this study, we conducted an integrated gut microbiome and muscle metabolome analysis on E. sinensis collected from three different environments, namely, an artificial pond, Yangcheng Lake, and Yangtze River, to uncover the mechanism of its high phenotypic plasticity. Our study presents three divergent gut microbiotas and muscle metabolic profiles that corresponded to the three environments. The composition and diversity of the core gut microbiota (Proteobacteria, Bacteroidetes, Tenericutes, and Firmicutes) varied among the different environments while the metabolites associated with amino acids, fatty acids, and terpene compounds displayed significantly different concentration levels. The results revealed that the gut microbiome community and muscle metabolome were significantly affected by the habitat environments. Our findings indicate the high phenotypic plasticity in terms of gut microbiome and muscle metabolome of E. sinensis when it faces environmental changes, which would also facilitate its acclimation and adaptation to diverse and even introduced environments.
To determine that Paragonimus sp. is actively transmitted in a tropical area of the Pacific region of Ecuador where human cases of pulmonary paragonimiasis have recently been documented, a total of 75 freshwater crabs were collected from 2 different streams in the Pedernales area of $Manab\acute{i}$ Province, Ecuador. All collected crabs were identified as Hypolobocera guayaquilensis based on morphological characteristics of the male gonopods. The hepatopancreas of each crab was examined by compressing it between 2 glass plates followed by observation under a stereomicroscope. Excysted Paragonimus metacercariae were detected in 39 (52.0%) crabs and their densities varied from 1 to 32 per infected crab. There was a positive relationship between crab size and metacercarial density. Sequences of the second internal transcribed spacer region of the ribosomal RNA gene of the Paragonimus metacercariae obtained in this study were identical to those of Paragonimus mexicanus deposited in the DDBJ/EMBL/GenBank database. Thus, the present study is the first to confirm that the crab species H. guayaquilensis is the second intermediate host of P. mexicanus in $Manab\acute{i}$ Province, Ecuador. Because this crab might be the possible source of human infections in this area, residents should pay attention to improper crab-eating habits related with a neglected parasitic disease, i.e., paragonimiasis.
폐흡충증은 우리나라에서 아직도 중요한 기생충질환으로서 서울의 한 대학병원에서 지난 10년간 105례가 발견되는 등 감염이 계속 발생하고 있으며 이 환자중 시장에서 민물게를 구입하여 게장을 담궈 먹은 과거력이 54% 에서 증명되었다. 이 관습은 일부 가정에 남아 있으므로 폐홉충증은 가족단위로 발생하는 경향을 보이고 있다. 민물게장이 폐흡충증의 중요 원인임에도 불구하고 민물게의 퍼낭유충 감염상태에 대한 조사결과는 대단히 적다. 따라서 1990년 9월부터 12월까지 서울의 시장에서 동남참게 85마리를 구입하고 폐홉충 피낭유충 감염상태를 조사하였다. 그 결과는 다음과 같다. 1. 구입한 민물게는 분류상 모두 동남참게 (nriocheir iaponicus)이었고 참게(5. sinensis)는 없었다. 종별 분 포에 대한 현지조사가 필요하다고 생각한다 2. 참게를 파쇄(파수), 3회 세척 여과하고, 침전물을 해부현미경으로 관찰한 결과 감염율은 11.8%이었고 평균 감염 충체수는 2.1개이었다. 3. 참게 무게별 감염율은 509 이하 43마리중 11.6%, 51∼1009 31마리중 12.7%, 1019이상 11마리중 9. l%이었다. 전국 각지에서 서울로 모아 판매하는 참게는 아직도 사람의 폐흡충 감염원이라고 판단하였다.
동남참게, Eriocheir japonicus 유생의 수온 및 염분 농도에 따른 이온 농도, 삼투질 농도 및 산소 소비에 대한 실험 결과는 다음과 같다. 체액의 $Na+$ 및 $Cl^-$ 이온 농도는 저염분 농도 ($30\%$ 해수) 이하일 때는 저온 $(16^{\circ}C)$에서 더 높았고, 고염분 농도 ($90\%$ 해수)일 떼는 고온 $(30^{\circ}C)$에서 더 높았다. 유생 령기가 어릴수록 매질의 염분 농도와 수온에 따른 체액의 이온 농도 변화폭이 컸다. 체액의 $K^+$ 이온 농도는 매질의 염분 농도의 영향을 많이 받았으나 수온의 영향은 거의 없었다. 체액의 삼투질 농도는 모든 유생 령기들에서 매질이 $70\%\;(24.5\%o)$ 해수 농도일 때 거의 등장액을 나타내었다. 개체당 산소 소비는 모든 실험구에서 zoea 1, 2령기에서 특히 낮았고, $70\%$ 해수 농도에서 모든 령기들의 산소 소비가 가장 높았다.

The present study investigated the infection status of Paragonimus westermani metacercariae in freshwater crabs (n = 363) and crayfish (n = 31) from October 2007 to October 2008 using the crush method. All of the freshwater crabs, Eriocheir japonicus, were negative for P. westermani metacercariae while 10 (32.3%) of the 31 examined crayfish were positive. The 10 positive crayfish were caught in Haenam, Jeollanam-do, and there were 8-59 (mean 28.4) metacrcariae per infected crayfish. These results suggest that P. westermani metacerariae are still transmitted by crayfish enzootically in southern Korea, and that freshwater crabs may transmit metacercariae only on rare occasions.
Among Paragonimus species, P. paishuihoensis is one of the most mysterious and poorly understood species. Metacercariae are characterized by having a unique dendritically branched excretory bladder. However, the morphology of the adult worm remains unknown. To date, metacercariae of this species have been reported only in China and Thailand. In this study, we first found P. paishuihoensis metacercariae in freshwater crabs, Potamon lipkei, in Hinheub District, Vientiane, Lao PDR, with a prevalence of 77.7% and the average intensity of 10.3 (range 1-28) metacercariae per crab. The molecular data based on ITS2 and CO1 markers indicated that P. paishuihoensis from Laos and Thailand were almost completely identical and were close to members of the Paragonimus bangkokensis/Paragonimus harinasutai complex. Attempts to infect experimental animals (cats, dogs, and rats) with P. paishuihoensis were unsuccessful, suggesting that these animals might be unsuitable definitive hosts for the species. Further studies are necessary to elucidate the taxonomic status and life cycle of P. paishuihoensis.
