Total polysaccharide contents in Panax ginseng roots were evaluated by a spectrophotometry, utilizing the complex formation of ginseng polysaccharide with alcian blue dye in 50 mM ammonium biphosphate, pH 4.2. The total polysaccharide content in red ginseng was about three times higher than that in fresh ginseng when both were extracted with water, and was increased about two times when red ginseng was extracted with an alkaline solution. The determination of total polysaccharide in various parts of ginseng revealed that main roots contained the component more than fine roots. Fresh ginseng sections stained by the dye showed polysaccharide mainly found in cortex and cambium but not in epidermis. Pattern-analysis on total and acidic polysaccharides from fresh and red ginsengs exhibited that the chemical compositions of the polysaccharides extracted from both ginsengs quite differed from each other.
Volatile flavor components of fresh ginseng (Panax ginseng C.A. Meyer) were studied by a combination of SE-54 fused silica capillary gas chromatography and mass spectrometry. Steam distillate of fresh ginseng roots was extracted with oxygen-free diethylether and concentrated. This aroma concentrate was separated into neutral, acidic, phenolic, and basic fractions. The neutral fraction, containing over two hundred compounds, was aromatically the closest to fresh ginseng with the key-flavor components predominantly being monoterpenes and sesquiterpenes. The sesquiterpene compounds were identified as being azulenic, menthenic, and napthalenic with a mass of 204.
Seventy percent ethanol extracts were prepared from 6-year-old fresh, white and red ginseng cultivated in Kangwha, and administered orally into rats for the comparison of their effects. And their effects on the production of lipid peroxide in the livers of ethanol-administered rats were measured. Red ginseng administered group showed the largest body weight increase. However, fresh, white, red ginseng extract administered group showed no significant change in concentration of total lipid, triglyceride and free cholesterol in serum. White and red ginseng extract administeration decreased blood-sugar levels significantly. Lipid peroxide content in livers of white and red ginseng administered groups was decreased significantly. Red ginseng administered group showed greater decrcase in lipid peroxide content than that of white ginseng administred group. The increase of lipid peroxide content in the livers of ethanol-administered group was inhibited by administration of fresh, white, and red ginseng extract. And the strongest-inhibitory action was observed in red ginseng administered group. Therefore, it is supposed that red ginseng has the most powerful antiaging effect.
In the browning reaction of Korean ginseng, it appears that enzymatic and non-enzymatic browning reaction occurred in the initial stage of heating fresh ginseng at low temperature, and then non-enzymatic browning reaction followed in the drying period after heating. Activation energy of the browning reaction for red ginseng was about 9.0 kcal/mol. Browning reaction of red ginseng was accede- rated with an increase in steaming time, and a great extent of browning reaction occurred between 60-90 min of steaming at 10$0^{\circ}C$. Browning pigments of red ginseng were mostly water soluble subset.
This study was undertaken to examine the effects of fresh ginseng on the physicochemical and microbiological changes in dongchimi juice fermented under various conditions. The pH was somewhat lower in dongchimi juice added with 2.0% and 4.0% of fresh ginseng than that without ginseng, whereas the titratable acidity was higher in dongchimi juice with 2.0% and 4.0% of ginseng addition than the control. The addition of fresh ginseng to dongchimi preparation increased the numbers of total viable bacteria, lactic acid bacteria including Leuconostac mesenterotdes in dongchimi juice during fermentation. The changes in the counts of lactic acid bacteria were similar to those of total viable cells throughout the experiment except the initial stage of fermentation. Polyamine constituents of fresh Panax ginseng root and dried white ginseng were extracted with 5% trichloroacetic acid, respectively. The isolation of polyamine constituents was conducted by the cation exchange chromatography using $Dowex-50W{\times}8$ resin and the detection was performed with TLC. Identification of the polyamine was carried out by the methods of IR, NMR, MS spectroscopy and GLC. Polyamine constituents of white and fresh ginseng root were identical and composed of five different polyamines. The major polyamine in white and fresh ginseng root was determined as putrescine.
Background: Dried and red ginseng are well-known types of processed ginseng and are widely used as healthy food. The dried and red ginseng quality may vary with the storage period of raw ginseng. Therefore, herein, the effect of the storage period of fresh ginseng on processed ginseng quality was evaluated through multicomponent quantification with statistical analysis. Methods: A method based on ultrahigh performance liquid chromatography coupled to triple quadrupole mass spectrometry in multiple-reaction monitoring mode (UPLC-MRM-MS) was developed for quantitation of ginsenosides and oligosaccharides in dried and red ginseng. Principal component analysis and partial least squares discriminant analysis were conducted to evaluate the dynamic distributions of ginsenosides and oligosaccharides after different storage periods. Results: Eighteen PPD, PPT and OLE ginsenosides and nine reducing and nonreducing oligosaccharides were identified and quantified. With storage period extension, the ginsenoside content in the processed ginseng increased slightly in the first 2 weeks and decreased gradually in the following 9 weeks. The content of reducing oligosaccharides decreased continuously as storage time extending, while that of the nonreducing oligosaccharides increased. Chemical conversions occurred during storage, based on which potential chemical markers for the storage period evaluation of fresh ginseng were screened. Conclusion: According to ginsenoside and oligosaccharide distributions, it was found that the optimal storage period was 2 weeks and that the storage period of fresh ginseng should not exceed 4 weeks at 0 ℃. This study provides deep insights into the quality control of processed ginseng and comprehensive factors for storage of raw ginseng.
To investigate optimum temperature for storage of fresh ginseng (Panax ginseng C. A. Meyer), the quality of the ginseng was compared during its storage at $-3^{\circ}C$, $-1.5^{\circ}C$ and $0^{\circ}C$. The deterioration rate of fresh ginseng stored at $-3^{\circ}C$ was the lowest for 8 weeks after storage. The rate was rapidly increased after that time and the rate at $-3^{\circ}C$ was higher than that of fresh ginseng stored at $-1.5^{\circ}C$ or $0^{\circ}C$ after the 12th week of storage. The deterioration severity of the fresh ginseng stored at $0^{\circ}C$ was much higher than that of the ginseng stored at $-1.5^{\circ}C$ and $-3^{\circ}C$. The weight loss of fresh ginseng ranged from 0.7---- to 1.6---- after 16th week; it was the lowest in the ginseng stored at $-1.5^{\circ}C$ and similar in fresh ginseng stored at $0^{\circ}C$ and $-3^{\circ}C$. The number of viable cells and molds in the fresh ginseng stored at $-3^{\circ}C$ was smaller than the fresh ginseng that was stored at other temperatures for 12 weeks, but did not differ with different storage temperatures after the 14th week of storage. The surface color of the fresh ginseng at $0^{\circ}C$ or $-1.5^{\circ}C$ was changed little while the discoloration of fresh ginseng at $-3^{\circ}C$ was relatively great. The electrolytic leakage from the rhizome of the fresh ginseng stored at $-3^{\circ}C$ was higher than that of the rhizome stored at $-1.5^{\circ}C$ and $0^{\circ}C$. The overall sensory quality of the fresh ginseng dropped below 3.0 in the S-point scale after the 10th week of storage at $-3^{\circ}C$ and after the 14th week of storage at $-1.5^{\circ}C$ and $0^{\circ}C$ (p<0.05).
Objectives : The purpose of this study was to investigate anti-cancer effects of various ginseng herbal acupuncture in mice and expression of cytokine mRNA. Methods: Anti-cancer effects of various ginseng herbal acupuncture were tested by measuring the increase of life span of mice suffering from peritoneal cancer induced by Sarcoma-180, and expression of mRNA manifestation using RT-PCR. The results are as follows: Results: 1. Increase of life span of mice suffering from peritoneal cancer induced by Sarcoma-180 was measured for anti-cancer effects. As a result, 115% increase was shown in the cultivated wild ginseng group, 11.1% increase in the red ginseng group, and no increase was detected in either white ginseng and fresh ginseng groups. 2. Measuring the expression of cytokine mRNA manifestation, expression of $interferon-{\gamma}$ was slightly increased in the cultivated wild ginseng group compared to the control group, but manifestation of interleukin-10 was slightly decreased. 3. For the red ginseng, white ginseng, and fresh ginseng experiment groups II, IL-2, IL-4, $INF-{\gamma}$, and IL -10 all showed increase suggesting possible error occurring during the test process. Conclusion: From the results obtained in this study, we can reason that the ginseng we use may not match the ginseng cited in the texts of the past. Anti-cancer effects of cultivated wild ginseng can be more potent than those of white and fresh ginseng.

Background : This study was conducted to determine out the effect of storage temperature on the quality of fresh ginseng (Panax ginseng C. A. Meyer) during distribution. Methods and Results : Fresh ginseng was washed, packed in $30{\mu}m$ low density polyethylene (LDPE) film, then stored at 0, -2 and $-4^{\circ}C$. After 4 weeks of storage, ginseng was then stored at $5^{\circ}C$, as a simulation of the distribution process. Ginseng stored at $-4^{\circ}C$ showed higher respiration rate, ethylene production and electrolyte conductivity during the distribution phase than those stored at 0 and $-2^{\circ}C$. Decay and browning rate rapidly increased following 3 weeks of distribution in samples stored $-4^{\circ}C$. However ginseng stored $-2^{\circ}C$, which is below freezing point, for 4 weeks did not show the physiological change or quality deterioration. Ginsenoside contents decreased during storage for all plant, but did not differ significantly between storage temperatures. Conclusions : Storage at temperatures below $-2^{\circ}C$ can negatively affect respiratory characteristics and electrolyte leakage and increase quality deterioration and decay rates during distribution.
