One exotherm was detected in the intact ginseng seeds containing more than 35% water, but in seeds with 20% there was no exotherm. The shapes of exotherm were remarkably uniform without relation to water content above 35%. The temperature at the initiation of freezing varied from -3.5$^{\circ}C$ to -9.6$^{\circ}C$ with the different water content in the seeds, and the Initial temperature of freezing delayed with the decrease of water content. The resistance damage at low temperature appeared in order of maln body, rhizome, lateral root of 3-year-old yearling rhizome, and fine root of 3-year-old. Ginseng roots didn't receive any damage at -5$^{\circ}C$ for 24 hours. Otherwise they received serious damage below -1$0^{\circ}C$ even for 5 hours'exposure. Hence, alternative low temperature gave more severe damage compared to constant low temperature. This result suggests that the Possibility of receiving injury at low temperature was higher during the thawing season of the early spring than in the winter.

In order to predict the risk of freeze injury for 'Changhowon Hwangdo' peach trees, we used the dormancy depth (i.e., the daily chill unit accumulation during the overwintering period) as a proxy for the short-term, physiological tolerance to freezing temperatures. A Chill-days model was employed and its parameters such as base temperature and chilling requirement were optimized for peach trees based on the 12 observational experiments during the 2008-2009 winter. The model predicted the flowering dates much closer to the observations than other models without considering dormancy depth, showing the strength of employing dormancy depth into consideration. To derive empirical equations for calculating the probabilistic freeze risk, the dormancy depth was then combined with the browning ratio and the budburst ratio of frozen peach fruit branches. Given the exact date and the predicted minimum temperature, the equations calculate the probability of freeze damages such as a failure in budburst or tissue browning. This method of employing dormancy depth in addition to freezing temperature would be useful in locating in advance the risky areas of freezing injury for peach trees production under the projected climate change.

The study was performed to evaluate thermal insulation covering materials (TICMs) to protect peach trunks against freezing temperatures in winter season by investigating thermo-physical properties and practical thermal insulation effect of the TICMs which was made of white non-woven fabrics, yellow paper sheets, and waterproof fabric pads. Among the three TICMs, Waterproof fabric pad (double layer) possessed the best performance about thermal insulation rate and thermal resistance among three kinds of TICMs. Day thermal insulation effects of waterproof fabric pad, which prevent from temperature rise on the bark tissues of trunks during the day time, were $14.09^{\circ}C$. Night thermal insulation effects of them, which prevent from temperature decline on the bark tissues of trunks at night time, were $7.23^{\circ}C$. Waterproof fabric pad showed the highest day and night thermal insulation effects. Thus our results suggest that development of TICMs using waterproof fabric pad might be helpful to protect the bark tissues of trunks from freezing injury.

We investigated the patterns of freeze injury in dormant 'Changhowon Hwangdo' peach fruit by observing the extent of browning and germination of the branches that were treated with freezing temperature sets simulating the process of natural freezing incidences in orchards. Under the treatment of freezing temperature of $-15^{\circ}C$, the browning ratios were 15% for flower bud and less than 3% for both leaf bud and cambium. Under the $-20^{\circ}C$ treatment, the browning ratios were 40% for both flower and leaf buds and 1% for cambium. The browning ratios were 86%, 68% and 40% respectively for flower bud, leaf bud, and cambium under the $-25^{\circ}C$ treatment. All the samples showed 100% browning ratio under the $-30^{\circ}C$ treatment. The budburst ratios of leaf buds were 85%, 66%, 32%, and 0% under the -15, -20, -25 and $-30^{\circ}C$ treatments, respectively. The branches of peach fruit treated with the same freezing temperature showed different responses depending on the sampling date. In January the browning ratio was low and the budburst ratio was high whereas in February the opposite was the case, showing vulnerability of peach trees to low temperature after endo-dormancy release.
In order to examine the viability depending on rehydration process and membrane injury of Nocardia mediterranei upon lyophilization, We labeled $3^H$-thymidine in deoxyribonucleic acid of N. mediterrranei to obtain information on the mechanisms of injury caused by lyophilization. Suspensions of rehydrated cells were incubated with added DNase in a buffer solution. Extracellular radioactivity levels appeared to be high in the rehydrated solutions after lyophilization than freezing-thawing. Thus, the membrane systems were injured by lyophilization, but not ovenvhelmed. These considerations were confirmed by electron microscopy. In effects of rehydration, the cell membrane was seriously damaged by strong atmospheric pressure as soon as the inner ampule was opened, but this was not the case without admitting air under vacuum. N. rnediterranei cells, with no additives, were lyophilized and reconstituted without admitting air, virtually about 84% of the cells were viable.

Extremely low values of the daily minimum temperature occurred in January 2010, ranging from -18 to $-29^{\circ}C$ at various locations growing peach trees in the Gangwon province. Due to the extreme cold temperature during the winter dormancy period of peach trees, the growth of the peach trees was damaged and the damaged areas increased as the extent of 17 to 144 ha. In order to provide information on mitigation measure of the cold temperature on the peach trees in the Gangwon province, we assessed the distribution of the damaged areas of growing the peach trees in 2010 and compared it with freezing risk estimated from the dormancy depth of the peach trees and the daily minimum temperature. The dormancy depth of 'Changhowon Hwando (Prunus persica (L.) Batsch)' ranged from -62 to -90 and the freezing risk was greater than 51%. The relationship between the freezing risk and the actual damaged area ratio showed reasonable agreement ($r^2$ of 0.5 with p < 0.01). The results imply that the estimates of the freezing risk based on the dormancy depth can be used as a mitigation measure to identify susceptible peach growing areas to freezing damage injury.
This study was undertaken to find out the effect of methyl-beta-cyclodextrin (MBCD) on cold shock and membrane cholesterol quantity of sperm during the freezing process in miniature pigs. For this study, semen ejaculated from PWG M-type miniature pig was diluted that freezing solution (with egg yolk group) and m-Modena B (without egg yolk group) treated with 0, 1, 5, 10 and 20 mM MBCD before freezing process. The diluted semen was monitored sperm ability at room temperature, after cooled until $5^{\circ}C$ and after forzen-thawed for cold shock test of spermatozoa. Also, membrane cholesterol of sperm was extracted by folch solution at the same time sperm ability was assessed for viability and acrosomal status. The membrane cholesterol quantity was measured by thin-layer chromatography (TLC) method. The result, viability and acrosome integrity in semen diluted without egg yolk groups were decreased at all temperature range by increasing of MBCD concentration. In particular, sperm of egg yolk group was showed that significantly higher viability and lower acrosome damage when treated with 5 mM MBCD (p<0.05). The results of TLC experiment, cholesterol amounts were increased with MBCD cocentration in egg yolk, and decreased with MBCD concentration in m-Modena B. In cryopreservation efficiency, there was no significant difference at viability, and acrosomal state of sperm in 5 mM MBCD concentration was significantly lower in acrosome damage than other groups (p<0.05). Therefore, the addition MBCD in egg yolk was protected spermatozoa from cold shock injury. This protective effect of MBCD may be due to addition of sperm membrane cholesterol.
This study was carried out to investigate the effect of vitrification and slow freezing methods on the post-thaw developmental rate of rabbit zygotes. After exposing rabbit zygotes in EFS solution for 0.5, 1, 2, 3 and S min at room temperature, they were washed with 0.5 M sucrose solution, D-PBS and TCM-199 and then cultured in TCM-199 plus 10% FBS with bovine oviduct epithelial cells(BOEC) to examine whether the cryoprotectant induced injury during the various exposure periods. The embryo development rates to hatched blastocyst after exposing in EFS solution for 3 and 5 min(40.0 and 16.7%) were significantly lower than in 0.5, 1 and 2 min(63.0, 72.0 and 54.5%), respectively. The post-thaw development rates to hatched blastocyst were significantly(P<0.05) higher in in vivo morula with intact mucin coat(85.2%) and mucin seperated morula(77.8%) than those of in vitro morula(58.5%) and zygote(5.9%), hut no difference was shown between in vitro morulae and mucin separated morula. The cryoprotectant dilution procedures showed no effects on the post-thaw development rates to hatched blastocyst under the present culture conditions. The post-thaw development to hatched blastocyst in the rabbit zygotes was not significantly different between the slow freezing(12.8%) and vitrification(5.9%). These results indicated that the rabbit frozen zygotes could he successfully developed in vitro to hatched blastocysts, though their developmental rate was very low, compared with morula stage embryos, in either vitrification or slow freezing procedure under the present conditions.

Background: To analyze the effects of the factors associated with different types of injury (superficial wounds, dislocations and sprains, bone fractures, concussion and internal injuries, burns scalding and freezing) caused by occupational accidents in automotive repair workshops. Methods: Study of a sample consisting of 89,954 industry accidents reported from 2003 to 2008. Odds ratios were calculated with a 95% confidence interval. Results: Belonging to a small company is a risk factor for suffering three of the five types of injury studied. Women are less likely to suffer burns and superficial wounds, and more likely to suffer dislocations or sprains. Foreign workers are more likely to suffer concussion and internal injuries. Conclusion: Health and safety strategies and accident prevention measures should be individualized and adapted to the type of worker most likely to be injured in each type of accident. Occupational health and safety training courses designed according to worker profile, and improving the participation of the workers in small firms creating regional or roving safety representatives would improve working conditions.
This study aimed to know difference in freezing resistance among different clonal seedlings or different seed source seedlings of Cryptomeria japonica which has been selected where extreme cold prevails in Korea and Japan. The freezing resistance of three 12-50 year old trees was also measured in the experiment. The freezing resistance was measured in different tissue parts: mainly leaf, cambiam and xylem, at three different collection dates in two different collection places during the winter of 1977-1978. The following results and discussions were made: 1. The clonal difference in freezing resistance of Cryptomeria japonica was $9^{\circ}$ to $15^{\circ}C$ in maximum according to the collection place. However, the clonal difference in freezing resistance was not related to the difference in climatic conditions where the parent tree have been growing. This impiled that the natural selection of cold resistant genes in Cryptomeria japonica has not reached its evolutional equilibrium yet since most of the Cryptomeria forest has been established by artificial regeneration. 2. The difference in freezing resistance among leaf, cambium and xylem was not apparent except that leaf of several clones showed higher freezing resistance than cambium or xylem when they collected at mid-winter. The least freezing resistant tissue part, thought its freezing resistance was not measure in all clones and all temperatures were appeared in the apical buds. The new shoot growth was observed in the next spring with being replaced by its dormant or adventitious bud growth when the apical bud was injured dy cold during winter. 3. The freezing resistance of leaf, cambium and xylem was shown high enough so that freezing resistance Cryptomeria clones in this experiment were supposed to be able to survive in cold winter conditions at the middle part of Korea. However, it was reported that the most susceptible tissue part to winter injury was the basal stem, but of which freezing resistance was not-measured in this experiment. Several silvicultural methods for prevention of Cryptomeria seedlings from cold damage were discussed in literature.
