Background:Liquid nitrogen freezing techniques have already met with widespread success in biology and medicine as a means of long-term storage for cells and tissues. The use of cryoprotectants such as glycerol and dimethylsulphoxide to prevent ice crystal formation, with carefully controlled rates of freezing and thawing, allows both structure and viability to be retained almost indefinitely. Cryopreservation of various tissues has various con-trolled rates of freezing. Material and Method: To find the optimal freezing curve and the chamber temperature, we approached the thermodynamic calculation of tissues in two ways. One is the direct calculation method. We should know the thermophysical characteristics of all components, latent heat of fusion, area, density and volume, etc. This kind of calculation is so sophisticated and some variables may not be determined. The other is the indirect calculation method. We performed the tissue freezing with already used freezing curve and we observed the actual freezing curve of that tissue. And we modified the freezing curve with several steps of calculation, polynomial regression analysis, time constant calculation, thermal response calculation and inverse calculation of chamber temperature. Result: We applied that freezing program on mesenchymal stem cell, chondrocyte, and osteoblast. The tissue temperature decreased according to the ideal freezing curve without temperature rising. We did not find any differences in survival. The reason is postulated to be that freezing material is too small and contains cellular components. We expect the significant difference in cellular viability if the freezing curve is applied on a large scale of tissues. Conclusion: This program would be helpful in finding the chamber temperature for the ideal freezing curie easily.

In cold-climate regions, deicing agents is used for smooth traffic on the road due to freezing and snowdrift in winter. The use of de-icing salts has resulted in the accelerated scaling damage of concrete with salt damage under freezing and thawing condition. Scaling is the deterioration of concrete where in the paste-mortar structure delaminates in flakes from the surface of the concrete. Due to such damage, concrete pavement causes various problems such as early deterioration according to the decrease in the thickness of cover concrete and user's stability issues. Accordingly, various tests and evaluation methods have been suggested in order to evaluate these phenomena in other countries. However, there have been no regulations for the evaluation method in South Korea, and related studies are also very rare. Therefore, in this study, the evaluation methods proposed by each institution and country were investigated and the experiments were performed according to each regulation, followed by the comparison and analysis of the results. Furthermore, this study aims to suggest the optimized experimental method adopted to domestic field through the discussion of such experimental methods and results.
This study was undertaken to find out the effect of methyl-beta-cyclodextrin (MBCD) on cold shock and membrane cholesterol quantity of sperm during the freezing process in miniature pigs. For this study, semen ejaculated from PWG M-type miniature pig was diluted that freezing solution (with egg yolk group) and m-Modena B (without egg yolk group) treated with 0, 1, 5, 10 and 20 mM MBCD before freezing process. The diluted semen was monitored sperm ability at room temperature, after cooled until $5^{\circ}C$ and after forzen-thawed for cold shock test of spermatozoa. Also, membrane cholesterol of sperm was extracted by folch solution at the same time sperm ability was assessed for viability and acrosomal status. The membrane cholesterol quantity was measured by thin-layer chromatography (TLC) method. The result, viability and acrosome integrity in semen diluted without egg yolk groups were decreased at all temperature range by increasing of MBCD concentration. In particular, sperm of egg yolk group was showed that significantly higher viability and lower acrosome damage when treated with 5 mM MBCD (p<0.05). The results of TLC experiment, cholesterol amounts were increased with MBCD cocentration in egg yolk, and decreased with MBCD concentration in m-Modena B. In cryopreservation efficiency, there was no significant difference at viability, and acrosomal state of sperm in 5 mM MBCD concentration was significantly lower in acrosome damage than other groups (p<0.05). Therefore, the addition MBCD in egg yolk was protected spermatozoa from cold shock injury. This protective effect of MBCD may be due to addition of sperm membrane cholesterol.
Frost damage of a storage tank for chemical admixtures caused by a low temperature in winter and quality deterioration of chemical admixtures have been often reported. However, there have been few regulations and researches related to the frost damage of chemical admixtures and facilities. Therefore, this paper is intend to investigate not only, the freezing points of chemical admixtures such as AE admixtures and water-reducing AE admixtures, etc., but also the quality variation of concrete used with chemical admixtures before freezing and after freeze-thaw cycles. According to the results of experiments, most chemical admixtures except anti-freezing agent and accelerating water-reducing AE admixtures should be kept from being frozen in low winter temperatures. However, full agitation of frozen chemical admixtures dose not cause the problems of concrete related to the quality of chemical admixtures.
The quality of the surface layer in concrete structures plays an important role in the durability of the concrete. The concrete factory products are made as they improve the appearance of the surface and compressive strength in need. A common criterion to judge the quality of concrete products frequently seen in our daily life appears to be "beauty" in terms of consistent shaping. However, as for most concrete curb in such areas where a large amount of anti-freezing agents(NaCl) and ice and snow melting agents(CaCl2) are spread over roads to ensure road safety during the winter season, since deterioration advances from the surface, scaling is seen on the surface concrete due to deterioration which combined freezing damage and salt damage. Especially, In cold northern districts, the spreading amount of deicing salts increases by regulation of studded tire use, and the scaling of the concrete products, the various parts of concrete structures for roads is increasing in recent years. In this study, L-shape concrete curb were targeted, the permeable form method with the commercial permeable sheet was applied to it and the improvements of the quality were examined. By the permeable form method, surface layers got strengthened, which prevented permeation of the deterioration factor from the outside, and the scaling resistance of the upper surface where the permeable sheet was applied improved exceedingly. It will be expected by applying the permeable form method to various concrete products that frost resistance improves and scaling damage decreases.

Tunnels constructed in cold regions can cause serious defects such as cracks and leaks due to external temperature changes in the portals and vents. In order to prevent the freezing damage of the tunnel, appropriate measures should be applied to the section where the freeze damage is concerned. However, the specific criteria and contents for judging whether or not the anti-freeze measures are applied are not presented. In this study, the laboratory freezing tests on the temperature changes of the concrete specimens under freezing conditions were carried out. And the freeze-thaw repetition cycle (F), which can judge the possibility of freezing damage, were presented based on the heat transfer quantity (W) by experimental results of case studies. Also, we propose a classification of cold regions considering the climatic characteristics of Korea for using it to efficient design and maintenance.

This research delves into the evaluation of the suitability of ultrasonic pulse velocity as a diagnostic tool for early detection of frost damage in concrete. The investigation involves the measurement of compressive strength and ultrasonic pulse velocity concerning the depth of freezing for individual mortar specimens, followed by an analysis of their microstructure and their interrelation. The findings indicate a consistent decrease in both compressive strength and ultrasonic pulse velocity with increasing freezing depth. Furthermore, a correlation between compressive strength and ultrasonic pulse velocity concerning the depth of early frost damage is established. Consequently, the study asserts the potential of utilizing the ultrasonic pulse velocity method for early detection of frost damage in concrete, with prospects for quantifying the depth of damage through further research endeavors.
Human umbilical Mesenchymal Stem Cell(uMSC) has been known as one of major component to regenerate connective tissues such as bone, cartilage, fat and others. The effect of low(5%), normotensive(20%) oxygen and freezing-thawing damage on proliferation of uMSC were investigated. low oxygen concentration culture of uMSC resulted in enhanced proliferation significantly($p$ <0.05) than 20% of oxygen culture. After the freezing-thawing injury to uMSC, 5% oxygen culture showed marked proliferation of uMSC than that of 20% oxygen($p$ <0.05) in the 5th passage of uMSC. Expression of antioxidant enzymes such as superoxide anion 1 and glutathione peroxidase 1 appeared marked in 20% oxygen cultured uMSC, which suggest oxidative stress could induce less proliferation of uMSC. Above findings would suggest proliferation of uMSC in 5% of oxygen will give more yields.

When fresh concrete is exposed to sufficiently low temperature, the free water in the concrete is cooled below its freezing point and transforms into ice, which causes decrease in compressive strength of concrete. Of the many influencing factors on the loss of compressive strength, the age of concrete at the beginning of freezing, water-cement ratio, and cement-type are significantly important. The objective of this study is to examine how the these factors affect the compressive strength of concrete frozen at early ages. The results from the tests showed that as age at the beginning of freezing is delayed and water-cement ratio is low, the loss of compressive strength decreases. In addition, concrete made with high-early-strength cement is less susceptible to frost damage than concrete made with ordinary portland cement.

In a heat only boiler system of a steam power plant, outdoor air required for combustion is made to pass through indoor space for increasing the boiler efficiency. Due to heat generated by various equipments, temperature of the air that enters the boiler will increase resulting in combustion efficiency. If the outdoor air temperature is low, however, this will cause freezing and bursting of pipes which are filled with water. It is especially fatal to small diameter pipes and pipes connected to measuring instruments. The purpose of this study is find operation and outdoor conditions where this phenomena can happen and also establish preventive measures to avoid this problem.
