Plant regeneration via direct and indirect adventitious shoot formation and chromosome-doubled somaclonal variation in Titanotrichum oldhamii (Hemsl.) Solereder

  • The gesneriaceous perennial plant Titanotrichum oldhamii has beautiful foliage and attractive bright yellow flowers. However, breeding of T. oldhamii by conventional sexual hybridization may be difficult because sexual reproduction of this species is very rare. In the present study, plant regeneration systems via both direct and indirect formation of adventitious shoots from leaf explants were established as the first step toward breeding T. oldhamii by using biotechnological techniques. Adventitious shoots were formed efficiently on medium containing $0.1mg\;l^{-1}$ benzyladenine. Histological observation showed that shoot formation on this medium occurred directly from leaf epidermal cells without callus formation. On the other hand, leaf explants formed calluses on medium containing $0.1mg\;l^{-1}$ 2,4-dichlorophenoxyacetic acid. The calluses could be maintained by monthly subculturing to fresh medium of the same composition. When the calluses were transferred to plant growth regulator-free medium, they formed adventitious shoots. Directly and indirectly formed shoots rooted well on medium containing $0.1mg\;l^{-1}$ indole-3-butyric acid. Plantlets thus obtained were successfully acclimatized and grew vigorously in the greenhouse. Flow cytometry analysis indicated that no variation in the ploidy level was observed in plants regenerated via direct shoot formation, whereas chromosome doubling occurred in several plants regenerated via indirect shoot formation. Regenerated plants with the same ploidy level as the mother plants showed almost the same phenotype as the mother plants, whereas chromosome-doubled plants showed apparent morphological alterations: they had small and crispate flowers, and round and deep green leaves.

  • Peptides are formed in the rumen as the result of microbial proteinase activity. The predominant type of activity is cysteine ptoteinase, but others, such as serine proteinases, are also present. Many species of protozoa, bacteria and fungi are involved in ptoteolysis; large animal-to-animal variability is found when proteinase activities in different animals are compared. The peptides formed from proteolysis are broken down to amino acids by peptidases. Different peptides are broken down at different rates, depending on their chemical composition and particularly their N-terminal structure. Indeed, chemical addition to the N-terminus of small peptides, such as by acetylation, causes the peptides to become stable to breakdown by the rumen microbial population; the microorganisms do not appear to adapt to hydrolyse acetylated peptides even after several weeks exposure to dietary acetylated peptides, and the amino acids present in acetylated peptides are absorbed from the small intestine. The amino acids present in some acetylated peptides remain available in nutritional trials with rats, but the nutritive value of the whole amino acid mixture is decreased by acetylation. The genus Prevotella is responsible for most of the catabolic peptidase activity in the rumen, via its dipeptidyl peptidase activities, which release dipeptides rather than free amino acids from the N-terminus of oligopeptides. Studies with dipeptidyl peptidase mutants of Prevotella suggest that it may be possible to slow the rate of peptide hydrolysis by the mixed rumen microbial population by inhibiting dipeptidyl peptidase activity of Prevotella or the rate of peptide uptake by this genus. Peptides and amino acids also stimulate the growth of rumen microorganisms, and are necessary for optimal growth rates of many species growing on tapidly fermented substrates; in rich medium, most bacteria use pre-formed amino acids for more than 90% of their amino acid requirements. Cellulolytic species are exceptional in this respect, but they still incorporate about half of their cell N from pre-formed amino acids in rich medium. However, the extent to which bacteria use ammonia vs. peptides and amino acids for protein synthesis also depends on the concentrations of each, such that preformed amino acids and peptides are probably used to a much lesser extent in vivo than many in vitro experiments might suggest.

Behaviour of cold-formed steel concrete infilled RHS connections and frames

  • This paper presents the results of a series of tests carried out on cold-formed steel rectangular hollow and concrete infilled beam to column connections and frames. A stub column was chosen such that overall buckling does not influence the connection behaviour. The beam chosen was a short-span cantilever with a concentrated load applied at the free end. The beam was connected to the columns along the strong and weak axes of columns and these connections were tested to failure. Twelve experiments were conducted on cold-formed steel direct welded tubular beam to column connections and twelve experiments on connections with concrete infilled column subjected to monotonic loading. In all the experiments conducted, the stiffness of the connection, the ductility characteristics and the moment rotation behaviour were studied. The dominant mode of failure in hollow section connections was chord face yielding and not weld failure. Provision of concrete infill increases the stiffness and the ultimate moment carrying capacity substantially, irrespective of the axis of loading of the column. Weld failure and bearing failure due to transverse compression occurred in connections with concrete infilled columns. Six single-bay two storied frames both with and without concrete infill, and columns loaded along the major and minor axes were tested to failure. Concentrated load was applied at the midspan of first floor beam. The change in behaviour of the frame due to provision of infill in the column and in the entire frame was compared with hollow frames. Failure of the weld at the junction of the beam occurred for frames with infilled columns. Design expressions are suggested for the yielding of the column face in hollow sections and bearing failure in infilled columns which closely predicted the experimental failure loads.

Characteristic of Oxidized Components Formed in Heated Soybean Oil and Lard (가열산화 대두유 및 돈지에서 생성된 산화물의 특성)

  • Total polar lipid produced from the soybean oil and lard by heating with different surface area at $185^{\circ}C$ were measured by silica gel column chromatography. Further, the polar lipid was fractionated by high performance size exclusion chromatography-evaporative light scattering detector (HPSEC-ELSD). The HPSEC system was composed of two GPC columns $(100\;{\AA}\;and\;500\;{\AA})$ and a THF mobile phase. With this system it was possible to fractionate into the free fatty acid, diglyceride, triglyceride monomer, triglyceride dimer and triglyceride polymers. The triglyceride monomer, triglyceride dimer and triglyceride polymers significantly increased as the heating time and surface area increased. But diglyceride and free fatty acid did not increased as the heating time and surface area increased. Triglyceride polymer (r>0.93), triglyceride dimer (r>0.97), triglyceride monomer (r>0.95) showed a high correlation with polar lipid content. On the other hand, diglyceride (r<0.68) and free fatty acid (r<0.76) were not significantly correlated with the polar lipid content. These results indicated that a major oxidative components formed during thermal oxidation were triglyceride polymers and triglyceride dimer and triglyceride monomer.

  • To investigate the inhibitory effects of Peonia japonica water extract(PJWE) on skin wrinkle formation, skin wrinkles were induced by both the irradiation of UVB and the application of squalene monohydroperoxide to the backs of hairless mice for 4 weeks. And at the same time experimental materials were applied topically. Wrinkles for the control (C) group were formed as a pattern of deep furrows and thick crests. Whereas wrinkles for the positive control (PC, 0.01% retinoic acid) and experimental(E, PJWE) groups were formed as a pattern of shallow furrows and thin crests, which were similar to that of the normal(N) group. Collagen and elastic fibers in dermis of the PC and E groups were almost intact with a regular arrangement, which were similar to those of the N group. The activity of xanthine oxidase, the free radical generating enzyme, was significantly lower in the E group than the C and PC groups. The activities of superoxide dismutase and catalase, the free radical scavenging enzymes, were much higher in the E group than the C and PC groups and similar to the N group. As for the amount of matrix metalloproteinase-3(MMP-3) expression, PC and E groups were significantly lower than the C group. Therefore, PJWE could be very effective natural herbal material for the inhibition or improvement of wrinkle formation in hairless mice skin.

The Occurrence of Laminarionema elsbetiae (Phaeophyceae) on Rhodymenia pseudopalmata (Rhodophyta) from the Patagonian Coasts of Argentina: Characteristics of the Relationship in Natural and Experimental Infections, and Morphology of the Epi-endophyte in Unialgal Free Cultures

  • The occurrence of Laminarionema elsbetiae (Ectocarpaceae, Phaeophyceae), as epi-endophyte of Rhodymenia pseudopalmata (Rhodymeniales, Rhodophyta), described from Santa Isabel, Rawson, Argentina. L. elsbetiae grows in the host tissues forming epi-endophytic relationship in the epidermal, cortical and medullar layers. Epiphytic thalli of L. elsbetiae were unbranched filaments emerging from hostis surface. Reproductive structures of L. elsbetiae on the host were absent. On the contrary, free cultured individuals formed different reproductive structures. Macrozoosporangia containing a single large motile zoospore originated from vegetative cells, they were conical to cylindrical in shape, 30-50 ${\mu}m$ in length and 18-20 ${\mu}m$ in wide. Uniseriate plurilocular zoosporangia were cylindrical shape, 40 ${\mu}m$ in length and 10-13 ${\mu}m$ in wide. Sexual fusion was not seen. In mixed cultures of L. elsbetiae with R. pseudopalmata fronds, L. elsbetiae infected the host, grew as in natural host and, formed macrosporangia between host subcortical cells. Gametophytes of L. elsbetiae were filaments with diffuse growth, branched with a branch pattern alternate or opposite. Gametangia were plurilocular, uni or biseriate and lateral. When mature they contained 2 to 6 isogametes. The presence L. elsbetiae on R. pseudopalmata could be defined as an epi-endophytic relationship. The percentage of infection of R. pseudopalmata thalli by L. elsbetiae was 34%.A25% of the infected thalli presented a low, non-symptomatic level infection, whereas a 62% and a 13% of them exhibited respectively moderate and high indexes of infection.

Properties Analysis of Environment Friendly Coating Films Formed by Using Electrodeposition Principle on Seawater (해수환경중 전착원리에 의해 형성시킨 환경친화적인 코팅막의 특성 분석)

  • Cathodic protection is one of the successful ways to prevent corrosion of steel structures in marine environments. The unique feature of cathodic protection in seawater is the formation of calcareous deposits on cathodic metal surface. The formation principles of calcareous deposit seawater had been known for a long time. That is, cathodic reduction reactions associated with cathodic protection in seawater generate $OH^-$ at the metal surface in accordance with the formular ; 1/2 $O_2$ + $H_2O$ + $2e^-$ $2OH^-$ and $2H_2O$ + $2e^-$ ${\rightarrow}$ $H_2$ + $2OH^-$. These reactions increase the pH at the metal / seawater interface. The high pH causes precipitation of $Mg(OH)_2$ and $CaCO_3$ in accordance with the formular ; $Mg^{2+}$ + $2(OH)^-$ ${\rightarrow}$ $Mg(OH)_2$ and $Ca^{2+}$ + $HCO_3^-$ + $OH^-$ ${\rightarrow}$ $H_2O$ + $CaCO_3$. These are typically the main compounds in calcareous deposits. It obviously has several advantages compared to the conventional coatings, since the environment-friendly calcareous deposit coating is formed by the elements($Mg^{2+}$, $Ca^{2+}$) naturally present in seawater. In this study, environmental friendly calcareous deposit films were prepared on steel plates by electro plating technic in natural seawater. The influence of current density on composition ratio, structure and morphology of the coated films were investigated by scanning electron microscopy formation process of calcareous deposits films in natural seawater. And we confirmed the properties of all the films can be improved greatly by controlling the material structure and morphology with effective use of the electroplating method in natural seawater.

A Study of Kirkendall Void Formation and Impact Reliability at the Electroplated Cu/Sn-3.5Ag Solder Joint (전해도금 Cu와 Sn-3.5Ag 솔더 접합부의 Kirkendall void 형성과 충격 신뢰성에 관한 연구)

  • A noticeable amount of Kirkendall voids formed at the Sn-3.5Ag solder joint with electroplated Cu, and that became even more significant when an additive was added to Cu electroplating bath. With SPS, a large amount of voids formed at the $Cu/Cu_3Sn$ interface of the solder joint during thermal aging at $150^{\circ}C$. The in-situ AES analysis of fractured joints revealed S segregation on the void surface. Only Cu, Sn, and S peaks were detected at the fractured $Cu/Cu_3Sn$ interfaces, and the S peak decreased rapidly with AES depth profiling. The segregation of S at the $Cu/Cu_3Sn$ interface lowered interface energy and thereby reduced the free energy barrier for the Kirkendall void nucleation. The drop impact test revealed that the electrodeposited Cu film with SPS degraded drastically with aging time. Fracture occurred at the $Cu/Cu_3Sn$ interface where a lot of voids existed. Therefore, voids occupied at the $Cu/Cu_3Sn$ interface are shown to seriously degrade drop reliability of solder joints.

Studies on Sexing of Bisected Mouse Embryos by Rat H-Y Antibody (Rat H-Y 항체에 의한 생쥐 분할란의 성 조절에 관한 연구)

  • This experiment was carried out to develop a new technique of identifying XX of XY-bearing bisected embryos prior to implantation by immunological method. H-Y antiserum prepared in inbred Wastar female rats by repeated immunization with spleen cells from males of the same strain. The reactivity of H-Y antibody was confirmed by culturing mouse embryos in the medium containing H-Y antiserum and complement obtained from the guinea pig. The optimal condition for the activity of H-Y antibody was also investigated by culturing embryos under the concentraton or affected H-Y antibody was also investigated by culturing embryos under the concentration or affected H-Y antibody and culture rate. However, production of live young or sex rates of male and female from embryos transferred with psudopregnant. The biological test with the morula stage embryos showed that H-Y antibody was formed in all female rats immunized with spleen cell, but it was formed only in 80% female rats immunized with the antigen. When the bisected mouse embryos were cultured in vitro for 5~6 hours in morula stage, of 457 bisected embryos 81.4% of then were developed to the blastocyst stage. When the concentration rate of complement to H-Y antiserum varied from 1.0~5.0${mu}ell$, the lysis-rate of embryo was 19.5 to 67.3%. The concentration rate of complement did not influence the lysis-rate of embryos(P<0.05). The morphology embryos of bisected, zona-free and intact embryos showed the embryos lysis rate of 58.6, 42.7 and 48.5% respectively(P<0.05). Pregnancy rate were 50.0, 45.5 and 57.1% in psudopregnant recipient transferred with bisected, zona-free and intact blastocyst embryos. However, production of live youngs, sexual rate of male or female was 24(50.0:50.0), 22(45.5:55.5) and 36(58.3:41.7)mice, but affected and non affected half embryos with H-Y antiserum treatment was 23.1 and 26.7%. Also production of live youngs and sexual rate was 14(92.9:7.1) and 17(17.6:82.4)mice in affected and non affected half embryos in H-Y antiserum treatment(P<0.05).

Effect of Dewpoints on Annealing Behavior and Coating Characteristics in IF High Strength Steels Containing Si and Mn (Si, Mn함유 IF 고강도강의 소둔거동 및 도금특성에 미치는 이슬점 온도의 영향)

  • The effect of dewpoints on annealing behavior and coating characteristics such as wettability and galvannealing kinetics was studied by annealing 0.3wt%Si - 0.1~0.4wt% Mn added interstitial-free high strength steels(IF-HSS). The 0.3wt%Si-0.1wt%Mn steel exhibited good wettability with molten zinc and galvannealing kinetics after annealing when the dewpoint of $H_2-N_2$ mixed gas was above $-20^{\circ}C$. It is shown that the wettability and galvannealing kinetics are directly related to the coverage of the external(surface) oxide formed by selective oxidation during annealing. At $N_2-15%H_2$ annealing atmosphere, the increase of dewpoint results in a gradual transition from external to internal selective oxidation. The decrease of external oxidation of alloying elements with a concurrent increase of their subsurface enrichment in the substrate, showing a larger surface area that was free of oxide particles, contributed to the improved wettability and galvannealing kinetics. On the other hand, the corresponding wettability and galvannealing kinetics were deteriorated with the dewpoints below $-20^{\circ}C$. The continuous oxide layer of network and/or film type was formed on the steel surface, leading to the poor wettability and galvannealing kinetics. It causes a high contact angle between annealed surface and molten zinc and plays an interrupting role in interdiffusion of Zn and Fe during galvannealing process.
