This study evaluated the performance of a thermophilic two-phase anaerobic digestion (TTPAD) coupled with membrane process treating garbage leachate. The pilot-scale treatment system is consisted of thermophilic acidogenic reactor (TAR) and thermophilic methanogenic reactor (TMR) coupled with an ultrafiltration (UF) membrane unit. The hydraulic retention time of TAR and TMR were 4 and 20 days, respectively. Effluent TCOD and SCOD of the TTPAD were $25\;{\pm}\;6\;and\;12\;{\pm}\;3$ g/L, respectively, and the corresponding TCOD and SCOD removal efficiencies were 77% and 81%, respectively. Propionate was major acids as 75% in the effluent. Scum formation was not observed in TTPAD, which might be resulted from complete lipid degradation. However, TTPAD was appeared to be sensitive to free ammonia toxicity. The UF membrane was operated with constant pressure filtration at average TMP 1.3 atm. Permeate flux had a range of 15-30 $L/m^2/hr$. With UF membrane, TCOD removal increased from 77% to 93%, and this SS free effluent would be beneficial to subsequent processes such as ammonia stripping.
The anaerobic digester supernatant (ADS) with high $NH_4-N$ concentration often results in a $NH_4-N$ overloading to the mainstream process of municipal wastewater treatment plant (MWTP). The nitrogen removal from the ADS is therefore important in order to achieve a stable mainstream process performance as well as to prevent $NH_4-N$ overloading due to ADS. Recently because of several advantages compared to the full nitrification, many works have shown interests in controlling the build-up of $NO_2-N$ in nitritation processes. The application of nitritation could save the aeration power compared to the full nitrification processes. In addition, the denitrification of $NO_2-N$ could reduce organic carbon requirements compared to the $NO_3-N$ denitrification. The purpose of this research was to find out the characteristics of the ADS nitritation and $NO_2-N$ accumulating factors from the laboratory reactor study. As a result based on the long-term laboratory experiment, it can be concluded that the degree of nitritation was closely related with the availability of alkalinity, free ammonia (FA), solid retention time (SRT) and solid concentration in the nitritation reactor.
The purpose of this thesis is to investigate whether taurine supplementation in combination with fructose improves both energy metabolism and exercise capacity. Eight collegiate female subjects were recruited for the study. Each subject went through threecross-over designs: control(fluid), fructose, and taurine plus fructose supplementation trials. Subjects received taurine supplementation 100 mg/kg a day for two weeks. After the supplementation, all subjects take 10% fructose at 15 min prior to exercise, immediately before exercise, and every 15 min during exercise. Subjects received 150 ml fluid as placebo during the same procedure. The subjects performed submaximal exercise at the exercise intensity of 60% for 45 min and then 80% of maximal oxygen uptake (VO2max) until exhaustion time. A 10ml blood sample was taken for measuring the level of glucose, ammonia, lactate, free fatty acids, and insulin every 15 min during exercise at 60% of VO2max. The blood glucose levels was significantly higher at 45 min and 50 min exercise after supplementation of fructose, and immediately before exercise and 50 min exercise after taurine plus fructose compared to the placebo trial. However, the values tended to be lower in taurine plus fructose supplementation compared to the fructose trial. The levels of both lactate and ammonia were significantly lower compared to the placebo, while the exhaustion time was significantly increased. The level of free-fatty acids was significantly lower at 30, 45, and 50 min after fructoseand fructose plus taurine supplementation compared to the placebo trial. The level of glucagon was significantly lower at 15, 30, 45, and 50 min after fructose and fructose plus taurine supplementation compared to the placebo trial. There was no differences in insulin concentration among three treatments. This thesis concludes that combined taurine and fructose supplementation prior to exercise may improve exercise tolerance time and energy metabolism, lowering the muscle fatigue factors such as lactate and ammonia.
Shucked oysters were soaked in an equal weight of salt water and stored at $3^{\circ}C$ for 7 days. Changes in the free amino acid content of the whole body and in the adductor muscle were evaluated by a practical distribution method. With the exception of aspartic acid and tyrosine, no significant changes in free amino acids or ammonia were observed in whole-body shucked oysters during the storage period. In contrast, the majority of free amino acids in the adductor muscle decreased significantly. Most of these free amino acids were detected in considerable amounts in the surrounding salt water after 7 days of storage. Both the weight of the whole body and the salinity of the surrounding salt water decreased significantly during the storage period. These results suggest that free amino acids were eluted from the cutting surface of the adductor muscle and indicate that the free amino acid content per shucked oyster and in the adductor muscle, decreases during cold storage.
Uniform, hollow nanosilica spheres were prepared by the chemical coating of cationic polystyrene (cPS) with tetraethylorthosilicate (TEOS), followed by calcination at 600 $^{\circ}C$ under air. cPS was synthesized by surfactant-free emulsion polymerization using 2,2'-azobis (2-methyl propionamidine) dihydrochloride as the cationic initiator, and poly(vinyl pyrrolidone) as a stabilizer. The resulting cPS spheres were 280 nm in diameter, and showed monodispersion. After coating, the hollow silica product was spherically shaped, and 330 nm in diameter, with a narrow distribution of sizes. Dispersion was uniform. Wall thickness was 25 nm, and surface area was 96.4 $m^2/g$, as determined by BET. The uniformity of the wall thickness was strongly dependent upon the cPS surface charge. The effects of TEOS and ammonia concentrations on shape, size, wall thickness, and surface roughness of hollow $SiO_2$ spheres were investigated. We observed that the wall thicknesses of hollow $SiO_2$ spheres increased and that silica size was simultaneously enhanced with increases in TEOS concentrations. When ammonia concentrations were increased, the irregularity of rough surfaces and aggregation of spherical particles were more severe because higher concentrations of ammonia result in faster hydrolysis and condensation of TEOS. These changes caused the silica to grow faster, resulting in hollow $SiO_2$ spheres with irregular, rough surfaces.
Ammonia, $NH_3$, is a key chemical widely used in chemical industries and a toxic pollutant that impacts human health. Thus, there is a need for the development of effective adsorbents with high uptake capacities to adsorb $NH_3$. An adsorbent with a high surface area and a small pore size is generally preferred in order to have a high capacity for the removal of $NH_3$. The use inorganic nanoporous materials as gas adsorbents has increased substantially and emerged as an alternative to zeolite and activated carbon. Herein, mesoporous alumina (MA) was prepared and used as an $NH_3$ adsorbent. MA showed good pore properties such as a uniform pore size and interlinked pore system, when compared to commercial adsorbents (activated carbon, zeolite, and silica powder). MA has free hydroxyl groups, serving as useful adsorption sites for $NH_3$. In an adsorption isotherm test, MA exhibited 4.7-6.5 times higher uptake capacities for $NH_3$ than commercial adsorbents. Although the larger surface areas of adsorbents are important features of ideal adsorbents, a regular and interlinked adsorbent pore system was found to be a more crucial factor to adsorb $NH_3$.
In order to understand the regulation of glutamate dehydrogenase(GDH) synthesis in Brevibacterium flavum, we have isolated a mutant lacking NADP-linked GDH activity by ethlmethane sulfonate treatment. The $gdh^-$ mutant was grown on the minimal plate with 1mM ammonium chloride and not that with 300mM ammonium chloride. The cell-free extracts from $gdh^-$ mutant and prototroph were also examined with glutamine synthetase(GS) and glutamate synthase (GOGAT) production by niteogen sources. The growth of $gdh^-$ mutant in presence of 20mM ammonium chloride means that GOGAT synthesis is sufficient to allow growth in this condition. GS production of $gdh^-$ mutant as well as parental strain was induced by 1mM urea and ammonium tartrate, but it was repressed by higher concentration of ammonia, and also induced by 20mM to 50mM glutamate as a substrate. It was special attention that GOGAT synthesis from $gdh^-$ strain was more repressed by higher concentration of ammonia than prototroph as described in E. coli system.
In this study, real-time and label-free methods for monitoring the enzymatic reaction of urease, which releases ammonia through the hydrolysis of urea in an aqueous solution, were developed using a liquid crystal (LC)-based pH sensor. Nematic liquid crystal 4-cyano-4'-pentylbiphenyl (5CB), doped with 4'-pentyl-biphenyl-4-carboxylic acid (PBA), exhibited a shift in optical appearance from bright to dark when it was in contact with ammonia generated from the enzymatic reaction between urease and urea. This optical change was attributed to the anchoring transitions of LCs caused by hydrophobic interactions between the tails of deprotonted PBA ($PBA^-$) molecules and the LCs at the aqueous/LC interface. This novel technique holds great promise for the sensitive detection of urease along with its substrates and inhibitors.
Lactate and ammonia are two major toxic waste products formed during mammalian cell culture. Accumulation of the side products have negative effects of on cell growth and specific production rate. In this study, K-562 cells were used as the host cell of a recombinant protein. Effects of carbon sources were invetigated focused on the cell culture span, the accumulation of lactate and ammonia in culture of recombinant K-562 cells.
Involvement of ethylene and Ca2+ on the induction of phenylalanine ammonia-lyase (PAL) was investigated in Daucus carota L. suspension culture system. Ethylene production started to increase about 3 h after glucan treatment. And the maximal induction of ethylene was preceded by PAL induction by 30 min. After the treatment of ethrel, PAL activity was increased. When cells were treated with glucan and Co2+, PAL activity was simultaneously reduced. Ethylene production was reduced dramatically in calcium-free medium, even though glucan was treated. PAL activity and ethylene producton was inhibited conspicuously when ethylene glycolbis($\beta$-aminoethyl ether) N,N,N',N'-tetraacetic acid (EGTA) was treated with glucan. Verapamil and trifluoperazine also inhibited PAL activity. When cells were treated with calcium ionophore A23187, PAL activity was increased in nontreated medium. We report here PAl activity is increased in related to ethylene production and involvement of Ca2+ in glucan-treated carrot suspension cells.
