Anti-inflammatory Modulating Effect of Rengyolone in Rat

  • Hepatitis refers to inflammation of hepatocytes and liver tissue, and is mainly caused by viruses, alcohol, and drugs. Forsythiae Fructus has traditionally been used as a diuretic, anti-inflammatory and antipyretic. Research on rengyolone, a bioactive substance extracted from Forsythiae Fructus, is rarely found in Korea and abroad. First, an acute animal toxicity test for rengyolone was conducted for the animal experiment. 4 week-old SD rats were injected intraperitoneally with acetaminophen for 2 weeks to induce chronic liver inflammation. Rengyolone was orally administered into two groups during 4 weeks: pre-inflammatory group and post-inflammatory group. Oral doses were also divided into 1 mg/kg and 5 mg/kg. Liver function tests (ALT, AST, ALP), western blot analysis of liver tissue, and level of inflammatory cytokine were performed to evaluate the improvement of hepatitis. Experimental results showed that rengyolone inhibited the development of acute inflammation and thus could reduce hepatitis symptoms.

Screening of Natural Products that have Activities against Skin-Aging (피부노화 억제활성을 갖는 천연 추출물의 탐색)

  • To screen antioxidative and whitening effects of some medicinal plants, 70% ethanol extracts were tested for their electron donating abilities to DPPH ( ${\alpha}$, ${\alpha}$-diphenyl- ${\beta}$-picrylhydrazyl) and inhibitory activities against lipid peroxidation, and tyrosinase inhibition in vitro. The electron donating activities of 70% ethanol extracts at the concentration of 100 $\mu\textrm{g}$/ml were in the order of Forsythiae fructus > Polyporaceae > Glycyrrhizae radix > Schizandrae fructus > Ramulus mori > Carthami semen. The order was Forsythiae fructus > Carthami semen > Schizandrae fructus > Polyporaceae > Glycyrrhizae radix when tested at 10 $\mu\textrm{g}$/ml, indicating that the activities depend on the concentration. The electron donating activities of Forsythiae fructus and Carthami semen at the concentration of 1 $\mu\textrm{g}$/ml were 37% and 31% respectively. Lipid peroxidation inhibitory activities were in the order of Artemisia messerschimidtiana > Glycyrrhizae uadix > Rehmaniae radix > Ramulus mori. Tyrosinase inhibitory activities were in the order of Ramulus mori > Artemisia messerschimidtiana > Oyster mushroom. The results suggest that Forsythiae fructus, Artemisia messerschimidtiana and Ramulus mori could be used as materials for anti-skin aging functional cosmetics.

  • The purpose of this thesis is to dedicate to get the objectivity of herbal medicine treatments by choosing herbs likely to work as antibacterial agents, Moutan Cortex Radicis, Caesalpiniae Lignum, Houttuyniae Herba, Forsythiae Fructus, Prunellae Herba, Scrophulariae Radix against Gardnerella vaginalis which is associated with Bacterial vaginosis, making experiments on them and getting the significant results. Each herb's efficacy on control the number of Bacterial vaginosis is noticed by using Disk Susceptibility test with six herbs medicine and Broth dilution assay of the culture. Disk Suseptibility Test : The efficiency strength is as follows in a row : Caesaipiniae Lignum, Moutan Cortex Radicis, Prunellae Herba, Forsythiae Fructus, Houttuyniae Herba, Scrophulariae Radix. MIC Prunellae Herba is 1.5 mg/ml, Caesalpiniae Lignum is 5 mg/ml, Forsythiae Fructus and Moutan Corex Radicis is 10mg/ml for G. vaginalis. As a results, herbmed which tested in this study have an antibacterial effects against G. vaginalis and that may be used for treament of Bacterial vaginosis. Assistant use of external medicine to p.o medicine is expected to bring the good effect of treatment which is used for the origin of this disease.

Ethanol Extract of Forsythiae Fructus Inhibits the Production of Inflammatory Mediators in LPS-stimulated BV-2 Microglial Cells (연교 추출물의 Microglia에서 LPS에 의해 유도되는 염증매개물질 생성 억제 효과)

  • Objectives : Forsythiae Fructus (Forsythia koreana Nakai) has been used anti-inflammatory, diuretics, antidote, and antibacterials in traditional herbal medicine. The present study is focused on the inhibitory effect of Forsythiae Fructus ethanol extract (FF-E) on the production of inflammatory mediators such as NO, iNOS and proinflammatory cytokines ($TNF-{\alpha}$, $IL-1{\beta}$ and IL-6) in LPS-stimulated BV-2 cells, a mouse microglial cell line, and investigated the scavenging activity of FF-E. Methods : BV-2 cells were pre-incubated with FF-E for 30 min and then stimulated with LPS (1 ${\mu}g/m{\ell}$) at indicated times. Cell toxicity of GCF was determined by MTT assay. The levels of NO, PGE2 and cytokines were measured by Griess assay and ELISA. The mRNA and protein expressions of iNOS and cytokines were determined by RT-PCR and Western blotting. Free radical scavenging activity of GCF was determined by DPPH assay in tube test. Results : FF-E significantly inhibited the excessive production of NO, $PGE_2$, $TNF-{\alpha}$, and $IL-1{\beta}$ in LPS-stimulated BV-2 cells. In addition, FF-E attenuated the mRNA and protein expressions of iNOS, and proinflammatory cytokines. FF-E also significantly scavenged the DPPH free radicals in a dose-dependent manner. Conclusions : These results indicate that FF-E exhibits anti-inflammatory property by suppressing the transcription of inflammatory mediator genes, suggesting the anti-inflammatory property of FF-E may make it useful as a therapeutic agent for the treatment of human neurodegenerative diseases.

Forsythiae Fructus Extract Inhibits RANKL-Induced Osteoclast Differentiation and Prevent Bone Loss in OVX-Induced Osteoporosis Rat (연교의 파골세포 분화 및 골 흡수 억제 기전 연구)

  • Objectives : Osteoporosis is a condition characterized by low bone mass and increased bone fragility. It has become a major problem of senior citizens. The purpose of this study is to experiment the effect of water extract of Forsythiae Fructus (wFF) on osteoclast differentiation; and the other purpose is to examine the effect of wFF on osteoporosis in ovariectomized rat. Methods : To investigate the effect of wFF on osteoclast differentiation and activity, RAW 264.7 cells were used. The number of TRAP positive cell, TRAP activity, pit area, mRNA expression of makers (RANK, TRAP, CA II, CTK, MMP-9, NFATc1, c-Fos), protein expression of makers (NFATc1, c-Fos) were investigated. For in vivo study, 40 female Sprague-Dawley (SD) rats were induced osteoporosis by ovariectomy (OVX) and then tested for anti-osteoporosis effect by administration of wFF. Results : wFF suppressed osteoclatogenesis, TRAP activity and pit area formation. Moreover, wFF decreased the expression of master differentiation factors (NFATc1, c-Fos) and also reduced the osteoclastogenesis-related markers (TRAP, CA II, CTK, MMP-9). These suggest that wFF inhibit osteoclasts differentiation and bone resorption. In the OVX rat model, wFF inhibited decreasing of BMD and trabecular area. Conclusions : Forsythiae Fructus should be effective for osteoporosis prevention and treatment.

Cytotoxic Constituents from the Forsythiae Fructus against L1210 and HL60 cells (L1210 및 HL60 Cell에 대한 연교의 세포독성 성분)

  • Forsythiae Fructus was studied on cytotoxic activities for the purpose of finding out active consituents against L1210 and HL60 cells. To isolate the active ones, the methanolic extract was partitioned into water insoluble and water soluble fractions. Furthermore, the water soluble fraction was fractionated into four parts, n-hexane, benzene, ethylacetate and water fractions. Among these, the water insoluble fraction showed the most potent cytotoxic activities on L1210 and HL60 cells in vitro. The water insoluble fraction was applied to silica gel column chromatography and divided into 5 fractions(fr. 1-5). The active constituents I and II were isolated from fr.2 and 3, respectively, by repeated silica gel column chromatography and recrystallization. The constituents were identified as 3${\beta}$-acetylbetulinic acid and betulinic acid by means of physicochemical data. The $ED_{50}$ values of 3${\beta}$-acetylbetulinic acid and betulinic acid were 9.10 and 16.43${\mu}g$/ml against L1210 cells and 2.72 and 2.41${\mu}g$/ml against HL60 cells, respectively.

  • Among 14 medicinal plants selected for the study ethanol (70%) extract of Coptis japonica Makino showed the highest anti-microbial activity against Helicobacter pylori followed by Perilla frutescens var. acuta KUDO, Caesalpinia sappan L. and Schizonepeta tenuifolia Briq. However, anti-urease activity of methanol (80%) extracts was best for Forsythiae Fructus followed by Caesaipinia sappan L. and Schizonepeta tenuifolia Briq. In the second fractionation using water, ethyl acetate and butanol more than 90% of the anti-urease activity was detected in the ethyl acetate fraction.

Effects of Forsythiae Fructus, Ulmi Cortex and JwaGwiEum in Allergic Rhinitis Induced by Ovalbumin in Mice (난알부민으로 유발된 생쥐의 알레르기 비염에 연교, 유근피, 좌귀음이 미치는 영향)

  • Objectives In the past, studies have been shown that Forsythiae Fructus (FF), Ulmi Cortex (UC) and JwaGwiEum (JGE) are effective in treating allergic reactions. However, no report shows the difference in effects of FF, UC and JGE in allergic rhinitis (AR). Therefore, the purpose of this study was to differentiate the effects of FF, UC and JGE in AR induced by ovalbumin (OVA) in mice. Methods From this experiment, the effects of FF, UC and JGE were several - changes in body weight, hematologic changes such as WBC, RBC, Hb and PLT counts, immunological changes such as levels of histamine, IgE, IL-4 and expressions of iNOS & COX-2 mRNA. Moreover, histological change of nasal mucosa was also investigated. Results FF administration group and JGE administration group significantly inhibited level of IgE compared to the control group. Also, FF administration group, UC administration group and JGE administration group inhibited IL-4 and expressions of iNOS & COX-2 mRNA. In histological assessment, FF administration group, UC administration group and JGE administration group showed reduced amount of subepithelial edema, desquamated mucosa and hyperplasia of basement membrane from OVA.

The Combined Anti-apoptotic Effect from Tamiflu and Pinoresinol of Forsythia fructus Extract Against Influenza Virus Infection (연교 추출물 Pinoresinol와 Tamiflu의 병용효과로부터 Influenza Virus 감염에 의한 세포사멸 억제효과)

  • The fruit body of Forsythiae Fructus (Oleaceae), a common Korean medical herb, is widely used in the treatment of cold and inflammation. In order to elucidate the action mechanism and the active principles from the plant against anti-influenza virus, the influenza virus hemagglutinin (HA) and neuraminidase (NA) gene RT-PCR and Viral Screening & Identification (VSI) assay were conducted, and the activity against viral replication was also investigated. Consequently, one active constituent, namely pinoresinol showed the in vitro antiviral principle using a cytopathic effect (CPE) reduction method, indicating pinoresinol possessed anti-influenza viral activity. Furthermore, combination of pinoresinol and Tamiflu exhibited higher activities than Tamiflu alone against influenza virus (H3N2) infection. The results suggested that combination of pinoresinol with Tamiflu could be a better candidate for an ant-H3N2 viral agent in the treatment of the influenza.

Ameliorative Potential of Rengyolone Against CCI-induced Neuropathic Pain in Rats

  • The sciatic nerve is the largest nerve among the peripheral nerves, and the damage to the sciatic nerve is caused by mechanical and physical pressure. This is an important disease that consumes a lot of time and money in the treatment process. Among them, research on relieving nerve pain caused by damage to the peripheral sciatic nerve has been made efforts to prevent and treat this disease through various methods such as drugs, natural products, electrical stimulation, exercise therapy, and massage. Existing treatments are not very effective in neurological pain, and countermeasures are needed. Forsythia Fructus, used in this study, has been used as a therapeutic agent for infectious diseases and a pain reliever for cancer from the past, and in past studies, it has been known to properly control the inflammatory response. In this study, rengyolone, a physiologically active substance of Forsythiae Fructus, was administered to rats that caused chronic left nerve pain to verify the pain relief effect. As a result of the experiment, it was found that mechanical pain and cold stimulation pain were significantly reduced in the rengyolone-treated group compared to the non-administered group. In addition, it was found that nerve growth factor (NGF) mRNA expression was significantly reduced and Cyclin-dependent kinase 2 (Cdc2) expression was increased in the rengyolone administration group. This increase in NGF expression is thought to be related to rengyolone's anti-inflammatory regulatory mechanism. It is expected that the reduced NGF was directly involved in pain relief.
