일산화탄소를 이용하여 자가영양적으로 성장한 Acinetobacter sp. 1 의 세포추출액은 혐 기성 실험조건하에서 thionin, methylene blue, 2,6-dichlorophenol-indophenol둥올 일산화탄소의 산회를 위한 전자수용체로 사용할 수 있었으나 NAD, NADP, FAD, 또는 FMN등은 천자수용체로 이용하지 못하였다. 이 세균에 존재하는 일산화탄소 산화효소는 유도효소로 밝혀졌고, pH 7.5와 $60^{\circ}C$에서 최대의 활성을 나타내었다. 이 효소의 활성화에너지는 6.1kcal/mol (25.5 kJ/mol)이며 일산화탄소에 대한 Km값은 $154{\mu}M$로 밝혀졌다. 그리고 잘 알려진 몇가지 금속 chelat tIng agent와 2가의 양이온들은 이 효소의 활성에 거의 영향을 미치지 않았는데 $Cu^{2+}$ 이온만은 이 효소의 활성을 완전히 억제시켰다. 또한 이 효소는 포도당과 숙신산에 의해 활성이 저해되었으며, hydrogenase의 활성도 나타내었다. 그리고 Acinetobacter sp. 1의 일산화탄소 산화효소는 Pseudomonas carboxydohydrogena의 일산화탄소 산화효소와 연역학적인 연관성이 없는 것으로 나타났다.
To determine 8 anti-impotence drug/drug-like compounds such as sildenafil, vardenafil, tadalafil, homosildenafil, hydroxyhomosildenafil, aminotadalafil, pseudovardenafil and hongdenafil in foods, simultaneously, high performance liquid chromatography(HPLC) and liquid chromatography-mass spectrometry (LC/MS) were used. The HPLC/UV analysis was performed on a column of capcellpak $C_{18}$ with 0.1% sodium-1-hexansulfonate in 0.2M ammonium formate/acetonitrile as a mobile phase. Mass spectra of the compounds by LC/MS were investigated with SCAN mode(Mass range and Fragment voltage) and SIM(Selected Ion Monitoring) mode (Ion target and Fragment voltage). The results follow as; 1. The HPLC/UV analysis was detected from 5 out of 63 samples. The content of sildenafil was in the range of 32.80 ppm ${\sim}$ 60.13 ppm from 4 out of 5 samples. The contents of sildenafil, vardenafil, homosildenafil were in the range 47.14 ppm from 1 out of 5 samples. 2. The conformed result of LC/MS was equal of detected from 5 out of 63 samples in HPLC/UV analysis. An easily available, simultaneous determination of 8 standards in adulterated health related foods was established by using a combination of LC/MS methods.
본 연구는 메탄산화균을 분리 배양하여 생물학적 질소 및 인 제거에 응용하는데 있다. 수도권 매립지의 상부 토양으로부터 NMS (nitrate mineral salt solution)배지로 분리 배양된 메탄산화균을 이용하여 영양염 제거 및 성장특성을 분석하였다. 분리 배양된 메탄산화균은 탈질의 탄소원으로서 이용될 수 있는 메탄올, 포름알데히드, 포름산으로 구성된 상당한 양의 유기물(COD 증가)을 생산하였다. 이때 메탄올의 생성속도는 $8\;mg/L{\cdot}hr$로 나타났다. 메탄산화균의 슬러지에 함유되어 있는 질소와 인의 함량을 볼 때 메탄산화균은 탈질에 필요한 탄소원 생성 뿐만 아니라 자체적으로 질소와 인을 성장기질로서 사용하는 것으로 나타났다.
The concentrations of ten inorganic (sodium, chloride, sulfate, ammonia, etc.) and three organic (acetate, formate, and MSA) ions associated with airborne particulate matter were measured from Cheju Island, Korea during the three field intensive campaigns conducted in (1) Sept./oct. 1997 (fall), (2) Dec. 1997 (winter), and (3) April 1998 (spring). The results of our measurements indicated that the concentration levels of most ionic species were decreasing significantly across the three experimental periods. The patterns of concentration reduction were clear as the sum of all cation and anion species changed dramatically across those periods such as 294> 144 > 65 and 193 >96>74 nequiv/m3, respectively. The changes were best explained in terms of the wind rose patterns of the study site. Since our sampling spot is located on the western-end point of Cheju Island, it is likely to reflect the effects of diverse sources such as natural, marine processes during NW and local non-maritime ones during SE winds. .Hence, the periodical changes in ionic concentrations may be accounted for by the comparable changes in wind direction. To further investigate environmental characteristics of these ionic components, correlation analysis was conducted not only between meteorological and ion data but between different ion-pairs. The results of these analyses confirm that the concentration levels of ionic species are strongly affected by wind speed and temperature and that there are certain patterns between ion species to which such effects apply. In light of the significance of the wind rose patterns in the area, we further extended these analyses into four data groups that were divided on the basis of wind direction. The results of these analyses showed that the strength of correlations between important pairs (e.g.:. between windspeed and most of major inorganic species including sodium and chloride) can be ranked on the distribution of major ions are very diverse, depending on data grouping scheme for such analysis. The results of this study thus suggest that environmental behavior of chemical components be analyzed in various respects, rather than simple standard, especially if measurements are made in complex environmental condition under which both natural and anthropogenic effects are competing each other.

This study was performed to determine the effect of field pea silages which were the organic acid (OA) alternative of oregano and cinnamon essential oils on fermentation quality and aerobic stability. Whole crop pea was harvested at full pod stage and wilted in the laboratory at the 48 h. The chopped pea was mixed and divided into equal portions allocated to five groups: CON (non-treated), distilled water, denoted as control group; OA group, a mixture of 60% formic acid, 20% sodium formate and 20% water applied at a rate of 5 g/kg fresh forage (Silofarm Liquid, Farmavet); origanum (ORE) group, Origanum onites essential oil at 400 mg/kg fresh forage; cinnamon (CIN) group, cinnamon essential oil at 400 mg/kg fresh forage; origanum+cinnamon (ORECIN) group, a mixture of ORE and CIN applied at an equal rate of 400 mg/kg fresh forage. Cinnamon decreased acetic acid (AA), ammonia nitrogen ($NH_3-N$) and weight loss (WL) at the end of 60 days silage. Crude protein (CP) and dry matter (DM) increased by cinnamon essential oil. Yeasts were not detected in any treatments, including the control, after 7 days of air exposure. The $CO_2$ amount decreased and the formation mold was inhibited in the aerobic period by the addition of cinnamon oil. Oregano did not show a similar effect, but when it was used with cinnamon, it showed synergic effect on AA and during aerobic period, it showed antagonistic effect on mold formation and DM losses. It was found in this study that cinnamon can be an alternative to organic acids.

A rapid and specific high performance liquid chromatography-tandem mass (LC/MS/MS) method for the analysis of montelukast in human plasma has been developed and validated. After cold acetonitrile-induced precipitation of the plasma samples, montelukast and glipizide (internal standard, IS) were eluted on a reverse-phase $C_{18}$ column by isocratic mobile phase consisted of 10 mM ammonium formate buffer (adjusted to pH 3.5 with formic acid) and acetonitrile (3:97, v/v). Acquisition was performed with multiple reaction monitoring (MRM) mode by monitoring the transitions: m/z 587.2${\rightarrow}$ 423.2 for montelukast and m/z 446.0${\rightarrow}$321.2 for IS. Ranges of concentration for calibration curves (10-1000 ng/mL) showed correlation coefficients ($r^2$) were better than 0.9948. Precision of intra- and inter-day ranged from 3.70 to 11.68% and from 3.04 to 12.95%, accuracy of intra-day and inter-day ranged from 93.34 to 102.75% and from 100.79 to 107.63%, respectively. The described method provides a fast and sensitive analytical tool for determining montelukast levels in plasma, and was successfully applied to a pharmacokinetic study in 16 healthy human subjects after oral administration of 10mg tablet formulation of montelukast sodium under fasting conditions.
The aerosol concentrations of ionic components were measured on a daily basis from a coastal monitoring site located at Kosan, Cheju Island from 26 September to 5 October 1997 as a field-intensive for a LRTAP project The chemical species we investigated include most of important inorganic species (i.e., Cl-, NO3-, F-, SO42-, Na+, NH4+, and K+) and some organic species (i.e. formats, acetate, and methanesulfonate (MSA) ions). The concentration data of those important inorganic and organic species obtained during this study were evaluated to properly address their chemical and physical characteristics. Most of major inorganic components including sulfate, sodium, chloride, and potassium ions exhibited very conservative relationships with each other such that the concentration ratios of any pair are quite analogous to that of seawater ratio. Since the oceans serve as the major sources of ionic constituents, their concentration changes appear to be senstively reflected by the factors affecting air-sea processes such as an increase in wind speed or changes in wind direction. A comparative analysis of sulfur-containing species such as seasalt (SS) and nonseasalt (NSS) sulfate and MSA were also made to assess the factors influencing the S cycling. An evaluation of NSS/SS ratios suggests that most of sulfate be associated with NSS fraction rather than 55 one. The finding of lower MSA/NSS-SO42- ratio along with a line of physical evidence such as intrusion of anthropogenically affected air mass suggests that the oxidation of S species have been promoted under the conditions encountered during the study period. Finally, the concentration data of carboxylic species (such as formats and acetate ions) were also analyzed. Although the existence of temporal trends were difficult to assess, these data indicate that their contribution to the precipitation acidity may not be significant enough.
A rapid, sensitive and selective hydrophilic interaction liquid chromatography-tandem mass spectrometric(HILIC-MS/MS) method for the determination of tiapride in human plasma was developed. Tiapride and internal standard, metoclopramide were extracted from human plasma with dichloromethane at basic pH and analyzed on an Atlantis HILIC silica column with the mobile phase of acetonitrile-ammonium formate (190 mM, pH 3.0) (94:6, v/v). The ana-Iytes were detected using an electrospray ionization tandem mass spectrometry in the multi-ple-reaction-monitoring mode. The standard curve was linear (r=0.999) over the concentration range of 1.00-200 ng/mL. The coefficient of variation and relative error for intra- and inter-assay at three QC levels were 6.4∼8.8% and -2.0∼3.6%, respectively. The recoveries of tiapride ranged from 96.3 to 97.4%, with that of metoclopramide (internal standard) being 94.2%. The lower limit of quantification for tiapride was 1.00 ng/mL using 1 00 $\mu$L of plasma sample.
A methane-oxidizing bacterium was isolated from rice paddy field soil around Jeollanam-do province, Korea, and characterized. The isolate was gram-negative, orange pigmented and short rod ($1.1-1.2{\times}1.6-1.9{\mu}m$). It was catalase and urease-negative but oxidase-positive. The strain utilized methane and methanol as sole carbon and energy sources. It had an ability to grow with an optimum pH 7.0 and an optimum growth temperature $30^{\circ}C$. The strain was resistant to antibiotic polymyxin B but sensitive to streptomycin, kanamycin, ampicillin, chloramphenicol and rifampicin. The isolate required copper for their growth with concentration range of $2-25{\mu}M$, with an optimum of $10{\mu}M$. Under optimal culture condition, specific cell growth rate and generation time were found to be $0.046hr^{-1}$ and 15.13 hr, respectively. Phylogenetic analysis based on 16S rDNA sequences indicated that the strain formed a tight phylogenetic lineage with Methylomonas koyamae with a value of 99.4% gene sequence homology. So, we named the isolate as Methylomonas sp. SM4. 8.6 mM methanol was accumulated in the reaction mixture containing 70 mM sodium formate and 40 mM $MgCl_2$ (MDH inhibitor) under atmosphere of methane:air (40:60) mixture for 24 hr at $30^{\circ}C$.

Natural photosynthesis utilizes solar energy to convert carbon dioxide and water to energy-rich carbohydrates. Substantial use of sunlight to meet world energy demands requires energy storage in useful fuels via chemical bonds because sunlight is intermittent. Artificial photosynthesis research focuses the fundamental natural process to design solar energy conversion systems. Nicotinamide adenine dinucleotide ($NAD^+$) and $NADP^+$ are ubiquitous as electron transporters in biological systems. Enzymatic, chemical, and electrochemical methods have been reported for NADH regeneration. As photochemical systems, visible light-driven catalytic activity of NADH regeneration was carried out using platinum nanoparticles, molecular rhodium and cobalt complexes in the presence of triethanolamine as a sacrificial electron donor. Pt nanoparticles showed photochemical NADH regeneration activity without additional visible light collector molecules, demonstrating that both photoactivating and catalytic activities exist together in Pt nanoparticles. The NADH regeneration of the Pt nanoparticle system was not interfered with the reduction of $O_2$. Molecular cobalt complexes containing dimethylglyoxime ligands also transfer their hydrides to $NAD^+$ with photoactivation of eosin Y in the presence of TEOA. In this photocatalytic reaction, the $NAD^+$ reduction process competed with a proton reduction.
