Contraction of smooth muscle is initiated by an increase in cytosolic $Ca^{2+}$ leading to activation of $Ca^{2+}$/ calmodulin-dependnet myosin light chain (MLC) kinase and phosphorylation of MLC. The types of contraction and signaling mechanisms mediating contraction differ depending on the region. The involvement of these different mechanisms varies depending on the source of $Ca^{2+}$ and the kinetic of $Ca^{2+}$ mobilization. $Ca^{2+}$ mobilizing agonists stimulate different phospholipases $(PLC-{\beta},\;PLD\;and\;PLA_2)$ to generate one or more $Ca^{2+}$ mobilizing messengers $(IP_3\;and\;AA),$ and diacylglycerol (DAG), an activator of protein kinase C (PKC). The relative contributions of $PLC-{\beta},\;PLA_2$ and PLD to generate second messengers vary greatly between cells and types of contraction. In smooth muscle cell derived form the circular muscle layer of the intestine, preferential hydrolysis of $PIP_2$ and generation of $IP_3$ and $IP_3-dependent\;Ca^{2+}$ release initiate the contraction. In smooth muscle cells derived from longitudinal muscle layer of the intestine, preferential hydrolysis of PC by PLA2, generation of AA and AA-mediated $Ca^{2+}$ influx, cADP ribose formation and $Ca^{2+}-induced\;Ca^{2+}$ release initiate the contraction. Sustained contraction, however, in both cell types is mediated by $Ca^{2+}-independent$ mechanism involving activation of $PKC-{\varepsilon}$ by DAG derived form PLD. A functional linkage between $G_{13},$ RhoA, ROCK, $PKC-{\varepsilon},$ CPI-17 and MLC phosphorylation in sustained contraction has been implicated. Contraction of normal esophageal circular muscle (ESO) in response to acetylcholine (ACh) is linked to $M_2$ muscarinic receptors activating at least three intracellular phospholipases, i.e. phosphatidylcholine-specific phospholipase C (PC-PLC), phospholipase D (PLD) and the high molecular weight (85 kDa) cytosolic phospholipase $A_2\;(cPLA_2)$ to induce phosphatidylcholine (PC) metabolism, production of diacylglycerol (DAG) and arachidonic acid (AA), resulting in activation of a protein kinase C (PKC)-dependent pathway. In contrast, lower esophageal sphincter (LES) contraction induced by maximally effective doses of ACh is mediated by muscarinic $M_3$ receptors, linked to pertussis toxin-insensitive GTP-binding proteins of the $G_{q/11}$ type. They activate phospholipase C, which hydrolyzes phosphatidylinositol bisphosphate $(PIP_2),$ producing inositol 1, 4, 5-trisphosphate $(IP_3)$ and DAG. $IP_3$ causes release of intracellular $Ca^{2+}$ and formation of a $Ca^{2+}$-calmodulin complex, resulting in activation of myosin light chain kinase and contraction through a calmodulin-dependent pathway.
Background: This study was aimed to differentiate two forms of CTLA-4 (CD152) in activated peripheral blood lymphocyte and clarify the mechanism how cytoplasmic form of this molecule is targeted to cell surface. Methods: For this purpose we generated 2 different anti-human CD152 peptide antibodies and 5 different N'-terminal deletion mutant CTLA4Ig fusion proteins and carried out a series of Western blot and ELISA analyses. Antipeptide antibodies made in this study were anti-CTLA4pB and anti-CTLA4pN. The former recognized a region on extracellular single V-like domain and the latter recognized N'-terminal sequence of leader domain of human CD152. Results: In Western blot, the former antibody recognized recombinant human CTLA4Ig fusion protein as an antigen. And this recognition was completely blocked by preincubating antipeptide antibody with the peptide used for the antibody generation at the peptide concentration of 200 ug/ml. These antibodies were recognized human CD152 as a cytoplasmic sequestered- and a membrane bound- forms in phytohemagglutinin (PHA)-stimulated peripheral blood lymphocyte (PBL). These two forms of CD152 were further differentiated by using anti-CTLA4pN and anti-CTLA4pB antibodies such that former recognized cytosolic form only while latter recognized both cytoplasmic- and membraneforms of this molecule. Furthermore, in a transfection expression study of 5 different N'-terminal deletion mutant CTLA4Ig, mutated proteins were secreted out from transfected cell surface only when more than 6 amino acids from N'-terminal were deleted. Conclusion: Our results implies that cytosolic form of CTLA-4 has leader sequence while membrane form of this molecule does not. And also suggested is that at least N'-terminal 6 amino acid residues of human CTLA-4 are required for regulation of targeting this molecule from cytosolic- to membrane- area of activated human peripheral blood T lymphocyte.

The development of technology has achieved digital innovation throughout the media industry, including production techniques and editing technologies, and has brought diversity in the form of consumer viewing through the OTT service and streaming era. The convergence of big data and deep learning networks automatically generated text in format such as news articles, novels, and scripts, but there were insufficient studies that reflected the author's intention and generated story with contextually smooth. In this paper, we describe the flow of pictures in the storyboard with image caption generation techniques, and the automatic generation of story-tailored scenarios through language models. Image caption using CNN and Attention Mechanism, we generate sentences describing pictures on the storyboard, and input the generated sentences into the artificial intelligence natural language processing model KoGPT-2 in order to automatically generate scenarios that meet the planning intention. Through this paper, the author's intention and story customized scenarios are created in large quantities to alleviate the pain of content creation, and artificial intelligence participates in the overall process of digital content production to activate media intelligence.
A neurotoxin, 6-hydroxydopamine (6-OHDA) has long been used to form a Parkinson's disease (PD) model by inducing the lesion in catecholaminergic pathways, particularly the nigrostriatal dopamine (DA) pathway. Whereas l-deprenyl, a selective inhibitor of monoamine oxidase (MAO) type B, is now widely used in the treatment of PD, the precise action mechanism of the drug remains elusive. In this study, we investigated whether l-deprenyl shows protective effect against the DA depletion induced by 6-OHDA in rat brain, and against 6-OHDA-induced neurotoxicity and oxidative stress in catecholaminergic neuroblastoma SH-SY5Y cells that are known to lack MAO-B activity. Pretreatment of l-deprenyl significantly enhanced the striatal DA, 3,4-dihydroxyphenylacetic acid, homovanilic acid, and 3-methoxytyramine levels compared to the untreated 6-OHDA-lesioned rat, indicating that l-deprenyl pretreatment prevents 6-OHDA-induced depletion of not only striatal dopamine but also its metabolites. Treatment of 6-OHDA for 24hrs decreased the cell viability and increase the generation of ROS in dose-dependent manners. We further investigated whether caspase activity is involved in the action of l-deprenyl. Treatment of l-deprenyl $(0.1\~100{\mu}M)$ did not produce any changes in 6-OHDA-induced cleavage of poly (ADP-ridose) polymerase in SH-SY5Y cells. Our results suggest that the neuroprotective effect of l-deprenyl against 6-OHDA is due to its increased scavenger activity, but independent of inhibition of MAO-B or caspase-3 activation.

Start Ongoing next generation networks are expected to be deployed over current existing networks, in the form of overlayed heterogeneous networks, in particular, in hot spot areas. Therefore, it will be necessary to develop an interworking technique such as load balancing, to achieve increased overall resource utilization in the various heterogeneous networks. In this paper, we present a new load balancing mechanism termed 'soft' load balancing where the IP(Internet Protocol) traffic of a user is divided into sub-traffic, each of which flows into a different access network. The terminology of soft load balancing involves the use of both load sharing and handover techniques. Through a numerical analysis, we obtain an optimal LBR (Load Balancing Ratio) for determining the volume of traffic delivered to each network over an overlayed multi-cell environment. Using the optimal LBR, a more reliable channel transmission can be achieved by reducing the outage probability efficiently for a given user traffic.

This paper presents experimental evaluation of an insect-mimicking flapping-wing device actuated by a unimorph piezoceramic actuator. Length of each rod and hinge point in the linkage/amplification system are carefully chosen such that the resulting wing motion can mimic clapping of wings in a real insect at the end of upstroke. In addition to this, a pair of corrugated wings are fabricated mimicking zig-zag cross section of a real insect wing. Thanks to the two additional implementation, the improved flapping wing device can generate a larger lift force than the previous model even though area of the new wing is about 50% less than that of the previous wing. In this work, effects of the wing clapping, the wing corrugation, and the input wave form on the lift force generation have been also experimentally investigated. Finally, the vortex generated by the flapping device has been captured by a high speed camera, showing that vortices are produced during up- and down-strokes.
Many studies have investigated the smart grid architecture and communication models in the past few years. However, the communication model and architecture for a smart grid still remain unclear. Today's electric power distribution is very complex and maladapted because of the lack of efficient and cost-effective energy generation, distribution, and consumption management systems. A wireless smart grid communication system can play an important role in achieving these goals. In this paper, we describe a smart grid communication architecture in which we merge customers and distributors into a single domain. In the proposed architecture, all the home area networks, neighborhood area networks, and local electrical equipment form a local wireless mesh network (LWMN). Each device or meter can act as a source, router, or relay. The data generated in any node (device/meter) reaches the data collector via other nodes. The data collector transmits this data via the access point of a wide area network (WAN). Finally, data is transferred to the service provider or to the control center of the smart grid. We propose a wireless cooperative communication model for the LWMN.We deploy a limited number of smart relays to improve the performance of the network. A novel relay selection mechanism is also proposed to reduce the relay selection overhead. Simulation results show that our cooperative smart grid (coopSG) communication model improves the end-to-end packet delivery latency, throughput, and energy efficiency over both the Wang et al. and Niyato et al. models.

The implosion phenomena of pressure vessels operating in deep water under extremely high external pressure have been well known. The drastic energy release to ambient field in the form of pressure pulse is accompanied with catastrophic collapse of shell structure. Such a proximity shock wave could be a serious threat to the structural integrity of adjacent submerged body and several suspected accidents have been reported. In this study, basic research for the occurrence and development of shock wave due to implosion was carried out. The mechanism of pressure pulse generation and energy dissipation were investigated, and a simplified kinematic model to approximate the collapse modes of circular tubes which can be generated by external pressure and implosion was examined. Using the simplified kinematic model, the process of energy dissipation was formulated, and the magnitude of released pressure shock wave was estimated quantitatively. To investigate the validity of developed kinematic model and shock wave estimation process, the results from a nonlinear FE analysis code and collapse test carried out using pressure chamber were compared with the results from the developed kinematic model.

The current theories on the interaction between surrounding rock and support in deep-buried tunnels do not consider the form of pre-reinforcement support or the flexibility of primary support, leading to a discrepancy between theoretical solutions and practical applications. To address this gap, a comprehensive mechanical model of the tunnel with pre-reinforced rock was established in this study. The equations for internal stress, displacement, and the radius of the plastic zone in the surrounding rock were derived. By understanding the interaction mechanism between flexible support and surrounding rock, the three-dimensional construction analysis solution of the tunnel could be corrected. The validity of the proposed model was verified through numerical simulations. The results indicate that the reduction of pre-deformation significantly influences the final support pressure. The pre-reinforcement support zone primarily inhibits pre-deformation, thereby reducing the support pressure. The support pressure mainly affects the accelerated and uniform movement stage of the surrounding rock. The generation of support pressure is linked to the deformation of the surrounding rock during the accelerated movement stage. Furthermore, the strength of the pre-reinforcement zone of the surrounding rock and the strength of the shotcrete have opposite effects on the support pressure. The parameters of the pre-reinforcement zones and support materials can be optimized to achieve a balance between surrounding rock deformation, support pressure, cost, and safety. Overall, this study provides valuable insights for predicting the deformation of surrounding rock and support pressure during the dynamic construction of deep-buried weak rock tunnels. These findings can guide engineers in improving the construction process, ensuring better safety and cost-effectiveness.

To investigate the characteristics of rip current occurring at Haeundae beach, observations obtained from a buoy and a CCTV were analyzed and numerical experiments were conducted. During observed rip-current events, the CCTV images showed that a couple of wave-trains, which are close to regular waves with slightly different directions, propagated to the beach, and wavelet analyses of data from the buoy showed very narrow-banded spectra with a peak frequency. From the evidences, it was inferred that a known mechanism of generating rip current due to the nodal line area of honeycomb-patterned wave crest was one of the significant factors of rip current occurrences of Haeundae beach. The mechanism has been explained by the following: When two wave-trains with slightly different directions propagate to a beach, wave crests of the incident wave-trains form honeycomb pattern due to nonlinear interaction. The nodal lines of honeycomb pattern are developed in the cross-shore direction. And longshore currents flow toward the nodal line area which has very low wave energy. Consequently their mass flux is expelled through the area toward the sea direction. To confirm the generation, numerical experiments were performed using a nonlinear Boussinesq equation model. In the cases with two incident wave-trains with slightly different directions and with a monochromatic wave propagating over a submerged shoal, it was seen that the honeycomb pattern of wave crests was well developed, and thus rip currents were evolved along the nodal lines.
