Recombinant plasmid, pBKH4, containing NPT II and P35S-BcHSP17.6 was constructed by ligation of Bum H I -digested pBKSl-l and BcHSP 17.6 (thermotolerance gene) 6om pBLH4. The tobacco leaf disc was cocultivated with transformed Agmbacterium tumefaciens bearing pBKH4 for 24 hours and transformed shoots were selected on MS-n/B medium containing $100\;{\mu\textrm{g}}/ml$ of kanamycin. Heat-killing temperature of Nicotima tabacum was $50^{\circ}$ for >15min, and transformed tobacco plants with BcHSP17.6 cDNA exhibited thermotolerance at the heat-killing temperature. The transgenic plants were analyzed by Southern blot hybridization with the probe of ${\alpha}^{_32}P$ labelled BcHSP17.6 cDNA. Transcription and expression level of BcHSP17.6 cDNA were also continued by Northern blot analysis and Ouchterlony double immunodiffusion assay. In this study, we suggest that the BcHSP17.6 cDNA introduced to tobacco plant is related to thenuoto-lerance and 17.6-kD LMW HSP acts as a protector from heat damage in plants.

This experiment was carried out to compare chemical composition of corn-cowpea inter cropping and corn monocropping plants at different harvestion time and obtained the following results. 1. In both cropping systems, the contents of crude fat and nitrogen free extract (NFE) were significantly increased (P<0.01), while that of crude protein crude fiber and crude ash were decreased (P<0.01) with each harvesting time. 2. The acid detergent fiber (ADF) and crude fiber contents were decreased same patterns in growing period, however, according to maturing of corn ears the difference between ADF and crude fiber contents reduced. 3. At mature stage, crude protein yields per 10a in corn monocropping and corn-cowpea intercropping system were 127.6kg and 152.1kg, respectively. The difference of crude protein content between corn-cowpea intercropping and corn monocropping system was 19.2%. 4. TDN yields of each cropping system, at mature stage, obtained similar results and TDN yields per 10a of corn monocropping and corn-cowpea intercropping system were 1006.1kg and 990.1, respectively. 5. Conseqently, corn-cowpea intercropping system could be increased protein yield without decreasing of dry matter yield in comparison with corn monocropping system.

Spring sown oat crops were grown during early March to late June in 1993~1994. The crops were harvested at different stage of maturity to determine the yield performance, chemical components and net energy value of the plants for silage utilization. The pattern of dry matter accumulation, absolute growth rate(Abs. GR), leaf weight ratio(LWR), stalk diameter(SD) and other growth parameters were analysed to evaluate the suitability of the plant materials for ensiling. The effect5 of wilting treatment and formic acid addition on the quality of silage was also disscused during the experiments. Spring sown oat crops was evaluated as a suitable materials for whole plant silage making. The best qualitative oat silage was obtained at the stage of soti dough to hard dough, when the plant reached 32.2~35.7% in dry matter concentration. Spring sown oat produced 9.43 ton in DM basis, 5.79 ton in TDN and 57.550 MJ-NEL/ha in net energy lactation. Early harvesting of the oat corps needed wilting treatment of the material before ensiling. Under pre-witing treatment (1/2 day) and formic acid addition(0.3%) silage quality point of oat crops harvested at the bloom stage was improved from 58 to 84 point.

In order to investigate effects of starch addition on the fermentative quality and dry matter digestibility (DMD) of Kudzu, Puerarie thunbergii Bentham plants were ensiled by the conventional method in small experimental plastic silo of 7.5 liter with different levels of starch addition. The fermentative quality, neutral detergent fiber (NDF), acid detergent fiber (ADF), and acid detergent lignin (ADL) of silage were determined by chemical analysis, and in vitro dry matter digestibility (DMD) by pepsin-cellulase technique. Total digestibile nutrients (TDN), digestible energy (DE) and metabolizable energy (ME) were calculated with DMD. The results obtained were as follows; 1. Acetate, butyrate, PH, NDF, ADF, and ADL of Kudzu silage were reduced with increasing of starch addition but the contents of lactate and total acid were increased. 2. The DMD was marked 47.5, 49.5, 51.2, 57.9 and 62.0 % under starch addition with a rate of 0, 2, 4, 6 and 8 %, respectively. 3. TDN, DF and ME were increased with high rate of starch addition. 4. The regression equation and correlation of Kudzu silage between level of starch (X) and DMD (Y) were Y= 41.6 + 1.86 X (I= 0.96, P < 0.01), of silage between Flieg's score (X) and DMD (Y) were Y= 41.6 + 0.23 X (I= 0.96, P < 0.01) and of silage between level of starch (X) and Flieg's score (Y) were Y= 21.3 + 7.97 X (I= 0.97, P < 0.01), respectively.

Drought is one of the detrimental factors that impair plant growth and productivity. In this study, we applied annealing control primer (ACP)-based reverse transcriptase PCR (polymerase chain reaction) technique to identify differentially expressed genes (DEGs) in maize leaves in response to drought stress. Two-week-old maize seedlings were exposed to drought (DT) by suspending water supply. DEGs were screened after 3 days of DT-treated samples using the ACP-based technique. Several DEGs encoding 16.9 protein, antimicrobial protein, hypothetical protein NCLIV_068840, thioredoxin M-type were identified in maize leaves under drought stress. These genes have putative functions in plant defense response, growth and development. These identified genes would be useful for predictive markers of plant defense, and growth responses under drought stress in plants.

To understand the role of small heat shock protein (sHSPs) in rice plant response to various stresses such as the heat and oxidative stresses, a cDNA encoding a 24.1 kDa mitochondrial small HSP (Oshsp24.1) was isolated from rice by rapid amplification of cDNA ends (RACE) PCR. The deduced amino acid sequence shows very high similarity with other plant small HSPs. DNA gel blot analysis suggests that the rice genome contains more than one copy of Oshsp24.1. High level of expression of Oshsp24.1 transcript was observed in rice seedlings in response to heat, methyl viologen, hydrogen peroxide, ozone, salt and heavy metal stresses. Recombinant OsHSP24.1 protein was produced in E. coli cells for biochemical assay. The protein formed oligomeric complex when incubated with Sulfo-EGS (ethylene glycol bis (succinimidyl succinate)). Our results shows that Oshsp24.1 has an important role in abiotic stress response and have potential for developing stress-tolerant plants.

Drought stress is a condition that occurs frequently in the field, it reduces of the agricultural yield of field crops. The aim of the study was to screen drought-adapted genotype of Italian rye grass. The experiments were conducted between the two Italian ryegrass (Lolium multiflorum L.) cultivars viz. Hwasan (H) and Kowinearly (KE). The plants were exposed to drought for 14 days. The results suggest that the morphological traits and biomass yield of KE significantly affected by drought stress-induced oxidative stress as the hydrogen peroxide (H2O2) level was induced, while these parameters were unchanged or less affected in H. Furthermore, the cultivar H showed better adaptation by maintaining several physiological parameter including photosystem-II (Fv/Fm), water use efficiency (WUE) and relative water content (RWC%) level in response to drought stress. These results indicate that the cultivar H shows improved drought tolerance by generic variation, improving photosynthetic efficiency and reducing oxidative stress damages under drought stress. These findings can be useful to the breeder and farmer for improving drought tolerance in Italian rye grass through breeding programs.

A vegetation survey of the native pasture was conducted in Cheju from July to September, 1980. The purpose of this investigation were, to determine the botanical composition of the native pasture and to estimate how they can be improved to the feed resources of livestock industry. The results obtained are summarized as follows: 1. Grass species existed in native pasture were 64 species with Pwceae 13, Fabaceae 8, Carduacea 12 and other 31. Most of them were identified as perennial grasses. 2. Botanical composition of major grass species were Imperata cylindrim 32.7 %, Pteridium aquilinum 18.9 70, Miscanthus sinensis 8.4 %, and Arternisia japonicn 2.8 % respectively. 3. The average dry matter yield of forage taken on the native pasture was 383 kg per 10a and these yields were increased as the growing stages progressed. 4. Seasonal distribution of edible plants were increased as the growing stages progressed.

A field experiment was conducted at the forage experimental plots, Seoul National University, Suweon h m 1995 to 1996 to determine the effect of legume residues as a N source and N fertilizer on corn (Zea mays L.) silage yield, N uptake, and availability of inorganic N in the soil. Corn was grown following (i) red clover (Trifolium pratense L.), (ii) crimson clover (Trifolium incarnatum L.), (iii) alfalfa (Medicago sativa L.) and (iv) winter fallow. The plots were split into two rates of fertilizer N (0 and 90kg Nlha) in a split-plot experimental plan. Compared with fallow treatment, legumes depleted soil water in the surface 15cm at corn planting by 17 to 26%. As a result, corn emergence was markedly delayed with legume residues by 8 to 11 days. Corn silage DM yield was significantly reduced in the presence of legume residues by 2.0 to 3.4 and 1.5 to 2.5 tonlha compared with winter fallow treatment at 0 and 90kg fertilizer Nlha, respectively, but no significant difference in the corn DM yield was found between legume residue treatments. There was an overall tendency for increased corn yields with 90kg fertilizer Nlha compared to Okg fertilizer Nlha, although not all yield increases were significantly greater.
