여포성숙의 대사에 관한 생화학적인 연구 II. 돼지 난소내 중여포액에 존재하는 단백질의 조성 및 동일여포액내 스테로이드 호르몬의 농도 (Biochemical studies on the metabolism of follicular maturation II. Protein composition and stenoid concentration in individually isolated medium-sized follicular fluid of pig ovary)

  • 포유류의 난소내 여포의 폐쇄기작을 알아보기 위하여 본 실험을 행하였다. 정상과 폐쇄를 형태적으로 판정한 돼지의 난소내 개개 중여포의 여표액내 스테로이드 호르몬과 단백질을 방사면역측정법과 SDS-전기영동으로 각각 분석하여 다음과 같은 결과를 얻었다. Progesterone은 여포성숙기의 여포액내 폐쇄여포에서 높은 농도를 나타내었고testosterone의 경우 황체기의 정상여포 및 여포성숙기의 폐쇄여포에서 높은 농도를 나타내었다. estradiol은 여포성숙기의 폐쇄여포에서 정상여포에 비해 현저히 낮았다. 여포액내 존재하는 단백질의 SDS-PAGE 결과, 혈청에 존재하지 않는 4종의 단백질을 확인하였다. 난소주기에 따라 112K, 141K의 단백질이 확인되었고, 폐쇄여포액내 특이하게 존재하는 23K, 24K의 단백질도 확인할 수 있었다. 이상의 결과로 보아 난소주기에 따라 여포액재에 존재하는 스테로이드 호르몬의 농도와 단백질의 조성이 서로 같지 않음을 알 수 있었으며 여포의 폐쇄기작에 있어서도 난소주기에 따라 분명한 차이가 있음을 알 수 있었다. 그러나 이들 폐쇄여포에 존재하는 단백질들의 생리적인 역할을 이해하기 위하여는 많은 연구가 수행되어야 한다고 사료된다.

  • 면역학적인 방법을 사용하여 가돈 여포액내의 특이단백질의 존재여부와 이들을 추출하여 난자성숙에 미치는 영향을 조사한 결과 다음과 같았다. 1. 가돈 혈청 및 혈장에는 존재하지 않는 면역학적으로 특이한 두 단백질이 가돈 여포액내에서 검출되었다. 2. 이들 두 단백질은 전기영동적으로 각각 fast alpha-I globulin 과 beta-globulin 의 이동성을 나타냈다. 3. 이들 두 단백질은 생쥐 여포난자의 성숙을 억제시켰다. 본 실험의 결과로 미루어 가돈 여포액내에는 난자성숙을 억제시키는 특이단백질이 존재하는 것으로 사료된다.

폐쇄에 따른 돼지 여포액내 단백질의 전기영동 양상에 관한 연구 (A Study on the Pattern of Proteins on Electrophoresis in the Porcine Follicular Fluid During Atresia)

  • This experiment has been done to evaluate the relationship between the follicular atresia and the protein patterns on electrophoreais of the follicular fluids in porcine ovary. The protein concentration of the follicular fluids was lower than that of serum, and gradually decreased as the follicle siae became larger. The number of protein bands of follicular fluid on electrophoresis was less than that of serum, and gradually increased as follicle size became larger. Three specific bands were detected on disc PAGE and one band(M W. 75,000) on SDS PAGE in the follicular fluids, while not in serum. One band (A) at ${\beta}$-globulin region on disc PAGE became heavier, as follicles became atretic. Two bands less than(M. W. 20,000) were detected only in the large follicular fluid. Another band(M. W. 43,000) was not detected in necrotic group, whereas all other groups showed it. It could be concluded that the component and composition of the proteins follicular fluids changes according to the follicular size during atresia. Therefore detection of the changing pattern of proteins in the follicular fluid can be used as a basic criterion for the identification of follicular atretic stage.

소 정자의 운동성에 영향을 미치는 난포액 성분에 관한 연구 (Study on Components of Bovine Follicular Fluid Affecting on Sperm Movement)

  • Follicular fluid influxed into the oviduct during ovulation may affect movement of sperm for fertilization Thus, in this study, the effect of follicular fluid, obtained from follicles of l0mm in diameter, on number and quality of sperm recovered by swim-up separation was investigated and sperm-movement stimulating components extracted from follicular fluid with methanol and isooctane were separated by gel filtration with Sepadex G-1O, G-25 and G-1OO gels, and were isolated by electrophoresis with SDS-PAGE mini gel. The results obtained were as follows; 1. Diluted follicular fluid stimulated sperm movement. 2. Sperm-movement stimulating factors were in methanol extract. 3. Sperm-movement stimulating effect of methanol extract appeared in fraction I among fractions recovered after gel filtration. And the fraction I contained proteins indicating 4 major bands as about 47, 43, 25 and 14 kilodaldons and 5 minor bands as about 67, 58, 23, 22 and 21 kilodaldons. 4. The fraction I recovered from G-100 gel showed significantly low percentage of motile sperm and had no protein indicating the band of 67 kilodaldons among the minor bands.

Effect of Follicular Fluid Proteins and Gonadotropins on Progesterone Secretion by Buffalo Granulosa Cells In vitro

  • In the mammalian ovary the follicular fluid contains proteins and peptides which play an important role in growth, development and maturation of oocytes. The gonadotropins and some other factors work synergistically and regulate ovarian functions. In the present study the effect of follicular fluid proteins (FFP) and gonadotropins on progesterone secretion by granulosa cells (GC) from buffalo ovary, was investigated during culture. The follicular fluid was collected from small (<5 mm), and medium (5-8 mm) follicles obtained from buffalo ovaries. The follicular fluid from medium follicles was fractionated with ammonium sulphate at 80% saturation. The precipitated protein fraction was further resolved in to minor (peaks I, III) and major (peak II) proteins using gel filtration (Sephadex G-200). The FFP from small follicles and major FFP (peak II) at a dose of 200 $\mu$g/well, significantly stimulated progesterone secretion by pooled GC (3${\times}10^{5}$ cells/2 ml medium/well). The minor FFP did not show any stimulatory effect. There was a significant increase in progesterone secretion by pooled GC in presence of FFP and LH (10 ng/well), however, FSH (20 ng/well) with FFP exhibited an inhibitory effect. The major FFP and gonadotropins were also studied for their effect on progesterone production by GC isolated from medium and large size follicles. The GC from medium follicles were more responsive to FSH and FFP whereas GC from large follicles exhibited enhanced progesterone secretion with LH and FFP. These results indicated that FFP have their own stimulatory effect and also act synergistically with gonadotropins. The significantly different response shown by GC, for steroid hormone secretion, is based on their stage of growth and differentiation. The purification and characterization of such steroidogenic proteins may help in elucidating their role in growth and differentiation of granulosa cells.

Identification of Proteins in Human Follicular Fluid by Proteomic Profiling

  • Human follicular fluid (HFF) is the in vivo microenvironment for oocyte maturation and includes a variety of proteins that could be involved in oocyte development and fertilization. We therefore used a proteomic approach to identify new HFF proteins. HFF from mature human follicles was obtained from five women following oocyte collection for in vitro fertilization (IVF). Ethanol-precipitated HFF run on two-dimensional gel electrophoresis (2DE) produced approximately 250 Coomassie brilliant blue-stained spots, 64 of which were identified using matrix-assisted laser desorption/ionization-mass spectrometry (MALDIMS). In this study, several proteins including complement factor H, inter-${\alpha}$ (globulin) inhibitor H4, inter-${\alpha}$-trypsin inhibitor heavy chain H4 precursor, human zinc-${\alpha}$-2-glycoprotein chain B, PRO2619, PRO02044, and complex-forming glycoprotein HC were new proteins that have not been previously reported in HFF using proteomic methods. Additionally, we identified alloalbumin venezia for the first time from trichloroacetic acid (TCA)-precipitated HFF. These HFF proteins could serve as new biomarkers for important human reproductive processes.

난포액내 정자유인물질의 분석 (Analysis of Sperm Chemoattractant in Follicular Fluid)

  • Among proteins separated from methanol extract of follicular fluid with superose column, the components inducing sperm swim-up separation through sucrose layer were analysed with superose column in Smart system and SDS-PAGE. And the results obtained were as follows; The fractions of retention volume (RV) 0.83ml and RV 1.36ml separated with superose column should stimulate sperm migration and movement. However, RV 0.83 fraction was consisted of complex materials containing RV 1.36 component. RV 1.36 fraction contained a BSA analogue of 67 kilodaltons (Kd) and showed identical peak pattern with BSA fraction V. In conclusion, the protein of 67 Kd in follicular fluid should stimulate sperm migration and movement.

Comparative Proteomic Analysis of Yak Follicular Fluid during Estrus

  • The breeding of yaks is highly seasonal, there are many crucial proteins involved in the reproduction control program, especially in follicular development. In order to isolate differential proteins between mature and immature follicular fluid (FF) of yak, the FF from yak follicles with different sizes were sampled respectively, and two-dimensional gel electrophoresis (2-DE) of the proteins was carried out. After silver staining, the Image Master 2D platinum software was used for protein analysis and matrix-assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS) was performed for differential protein identification. The expression level of transferrin and enolase superfamily member 1 (ENOSF1) was determined by Western blotting for verification analysis. The results showed that 2-DE obtained an electrophoresis map of proteins from mature and immature yak FF with high resolution and repeatability. A comparison of protein profiles identified 12 differently expressed proteins, out of which 10 of them were upregulated while 2 were downregulated. Western blotting showed that the expression of transferrin and ENOSF1 was enhanced with follicular development. Both the obtained protein profiles and the differently expressed proteins identified in this study provided experimental data related to follicular development during yak breeding seasons. This study also laid the foundation for understanding the microenvironment during oocyte development.

돼지의 난포액 내 단백질인자의 탐색과 변화에 관한 연구 (Studies on Investigation and Change of Protein Factors in Porcine Follicular Fluids)

  • When fully grown oocytes are removed from their follicles, they can resume meiosis and mature spontaneously under in vitro conditions. However, nuclear maturation under in vitro condition is not accompanied by complete cytoplasmic maturation, which is essential for successful fertilization and the initiation of zygotic development. This study analyzed change of proteins in follicular fluids during the porcine follicular development. Follicular fluids were collected from follicles of diameter 1~2 mm, 2~6 mm and 6~10 mm in ovary of slaughtered pigs. Total proteins were extracted from follicular fluids by M-PER Mammalian Protein Extraction Reagent. We confirmed totally 27 same spots, 1 spot from follicle fluid of 2~6 mm follicle and 5 spots from follicle fluid of 6~10 mm in diameter were analyzed by MALDI mass spectrometry and searched on NCBInr. In results, spot No. 28 from 2~6 mm follicle was Ig lambda chain C region, and spot No.32 and 33 from 6~10 mm was Apolipoprotein A-(APOA4). Spot No.29 and 31 were failed to analyze. These results indicate that the porcine oocyte during in vitro maturation depend on specific different expressed proteins may play an important roles in the sequence of molecular events in porcine oocyte maturation and follicular development.
