A New-Generation Fluorescent-Based Metal Sensor - iLOV Protein

  • The iLOV protein belongs to a family of blue-light photoreceptor proteins containing a light-oxygen-voltage sensing domain with a noncovalently bound flavin mononucleotide (FMN) as its chromophore. Owing to advantages such as its small size, oxygen-independent nature, and pH stability, iLOV is an ideal candidate over other reporter fluorescent proteins such as GFP and DsRed. Here, for the first time, we describe the feasibility of applying LOV domain-based fluorescent iLOV as a metal sensor by measuring the fluorescence quenching of a protein with respect to the concentration of metal ions. In the present study, we demonstrated the inherent copper sensing property of the iLOV protein and identified the possible amino acids responsible for metal binding. The fluorescence quenching upon exposure to Cu2+ was highly sensitive and exhibited reversibility upon the addition of the metal chelator EDTA. The copper binding constant was found to be 4.72 ± 0.84 µM. In addition, Cu2+-bound iLOV showed high fluorescence quenching at near physiological pH. Further computational analysis yielded a better insight into understanding the possible amino acids responsible for Cu2+ binding with the iLOV protein.

  • The difference of whiteness, brightness and lightness was clearly explained in this paper through the use of a tinting dye and a fluorescent whitening agent which are commercially wide]y used to make paper look whiter. Other optical properties such as tint, color shade, and color difference were also discussed. It was suggested that in comparing two tinting dyes, lightness $(L^*)$ is the most important property to be compared, while whiteness data should be used in caution in order not to surpass its significant range, and $a^*,\;b^*$ values can also be used to see the change of color shades together with ${\Delta}E$ as color difference. In comparing two fluorescent whitening agents, whiteness or brightness values are most important to be compared, but lightness values are not suitable fur this purpose; $a^*,\;b^*$ and color difference ${\Delta}E$ can also be referred, but with less significance.

  • Two new 3D coordination complexes [Cd(3-bpcd)(pht)] (1) and [Zn(3-bpcd)(pht)] (2) [3-bpcd = N,N'-bis(pyridin-3-yl)cyclohexane-1,4-dicarboxamide, $H_2pht$ = phthalic acid] have been hydrothermally synthesized. X-ray diffraction analysis reveals that the complexes 1 and 2 represent a 4-connected diamondoid topology with a 3-fold interpenetrating feature. Moreover, the fluorescent properties of complexes 1 and 2 are studied.

  • This study addresses a microfluidic method to uniformly form diacetylene (DA) liposomes and control their size. DA liposomes are biochemical sensor materials with a unique property such that when they are polymerized to polydiacetylene (PDA) they exhibit non-fluorescent blue to fluorescent red phase transition upon chemical or thermal stress. The liposome size and distribution are important because they significantly affect the phase transition. So far, DA Liposomes, have been prepared by mixing of bulk phases leading to heterogeneous, polydisperse distribution in size. Therefore, additional post-processes are required such as sonication or membrane extrusion to obtain an appropriate size of liposomes. Here, we report a novel strategy using a microfluidic chip and hydrodynamic focusing to form DA liposomes and control their size. Preliminary results obtained by scanning electron microscope (SEM) and dynamic light scattering (DLS) show that the microfluidic strategy generates more monodispersed liposomes than a bulk method.

  • Novel fluoroionophore of dibenzothiazolyl-dibenzo-crown ethers were synthesized from diformal-dibenzo 18-crown-6 (24-crown-8) with 2-aminothiophenol, and they were characterizated by $^1H$-NMR, $^{13}C$-NMR, IR spectrum, Mass spectrum, elemental analyses, respectively. The fluorescent properties of the newly synthesized crown ether were examined with $Li^+$, $Na^+$, $K^+$, $Rb^+$, $Cs^+$, $NH_4^+$ and $CF_3COOH$, respectively. With protonation using $CF_3COOH$, the absorption bands of the new crown ethers are further blue shifted, the maximum emission wavelengths further red shifted.

  • An external electrode fluorescent lamps (EEFLs) have the advantage of a long lifetime in the early stages of the study on plasma discharge, interest in the lamp continues. Studies on the operation of external electrode fluorescent lamps have focused mainly on its use of a type of high frequency (MHz). By performing high brightness using a square wave operation method with the low frequency below 100kHz, which is applied to a narrowed tube type lamp that has several mm of lamp diameter. To solve these problems of CCFL, EEFL (External Electrode Fluorescent Lamp) is introduced. Because electrode of EEFL is on the outer surface of discharge tube, the electrode is perfectly prevented from the sputtering by accelerated ions. And it is possible to drive the many CCFLs at the same time, because EEFL shows the positively resistant characteristic. But EEFL has the large non-radiative power loss in sheath. In this study the novel electrode structure was introduced in order to reduce non-radiative power loss in sheath of EEFL. The novel electrode structure comes from the idea to combine conceptually capacitive discharge with inductive discharge. Thus, this study verifies the change in the optical characteristics according to the change in electrode structure through a Maxwell's electromagnetic field simulation and examines the relationship between the change in the EEFL electrode structure and brightness by measuring the optical characteristics.

  • A novel TPC-PEG-TPC with active end-groups was obtained from the end-groups of polyethylene glycol (PEG) modified by terephthaloyl chloride (TPC). These active end-groups can link up with a rare earth ion, which is a luminescent center of a rare earth fluorescent complex. Complexes of Eu-PEG with novel ligands (TPC-PEG-PTC) were synthesized by the coordination of the active reactant (as the first ligand) and phenanthroline (as the second ligand) with $Eu^{3+}$.IR, $^1H$-NMR, element analysis, DSC, WAXD, fluorescent spectroscopy, TGA, and SEM were used to characterize the structure and properties of these complexes. The results showed that this type of complex is a heat storage material with the phase change character of polyethylene glycol (PEG) and the luminescent properties of europium. There was no thermal decomposition of the complex of Eu-PEG until $300^{\circ}C$. SEM showed that the complex of Eu-PEG can be dispersed in PE.

The discharge characteristic of Li ion doped MgO film in a flat fluorescent lamp structure

  • This paper investigates how various concentrations of lithium ion influence on crystallization of MgO in thin films formed by spin coating and an the discharge characteristic in a flat fluorescent lamp structure. The XRD results indicate $Li^+$ ion enhances the growth of MgO crystal in a spin coated thin film. The discharge property with the $Li^+$ ion doped MgO films show the lithium ion in MgO film clearly reduce the initial discharge voltages of test devices. Interestingly, the test panels with various doped MgO film have somewhat higher static memory margin of than that of pure-MgO owing probably to the pore structure of spin coated MgO films. The CL spectra, which confirm that the doping creates defects energy levels in the band gap of MgO, show the $F^+$ center is the main defects in doped MgO films.

Study on full color RGB LED source lighting for general lighting and Improvement of CRI (Color Rendering Index)

  • The purpose of this study is to check if LED lighting can be used as general lighting and examine the color rendering property of full color RGB LED lighting. CRI is one of the important properties of evaluating lighting. However the present CRI does not fully evaluate LED lightings. Firstly, the performance of a simple task was compared other than comparing CRI values for different lighting. For experimental preparation three types of lightings were used; standard D65 fluorescent tube, general household fluorescent tube, and RGB LED lighting. All three lightings show high error for Purple-Red. All three lightings show similar error for all hues and prove that color discrimination is not affected by the lighting. This proves that LED could be used as general lighting. Secondly, problems of the conventional CIE CRI method are considered and new models are suggested for the new lighting source. Each of the models was evaluated with visual experiment results obtained by the white light matching experiment. The suggested model is based on the CIE CRI method but replaces the color space model by CIELAB, color difference model by CIEDE2000, and chromatic adaptation model by CAT02.

  • ${\alpha}-Tocopherol$ was found to be a fluorescent probe in determination of the phasetransition temperature of liposome. Since this was a discovery of a new aspect of ${\alpha}-tocopherol$ as an important biochemical molecule, its molecular spectroscopic characterization was carried out in order to obtain some informations on its spectral and, structural properties in various media, anticipating that the compound may entertain a wide applications in biochemical systems as a spectroscopic probe. Two species of α${\alpha}-tocopherol$, monomer and dimer, were found to exist in organic media, especially in solvents of nonhydrogen bonding ability. Monomer with maximum UV-absorption around $(291{\sim}294nm)$ is highly fluorescent, while dimer which is formed by intermolecular hydrogen bonds and absorbes with spectral peak at 298nm is nonfluorescent. ${\alpha}-tocopherol$ incorporated to liposome exhibits emission property quite different from that in various organic media showing broad and red-shifted fluorescence excitation and emission spectra. This spectral abnormality is to be interpreted to arise from chromanolate-type ion, H-dissociated ${\alpha}-tocopherol$.
