Ultrafast Excited State Intramolecular Proton Transfer Dynamics of 1-Hydroxyanthraquinone in Solution

  • Proton transfer reaction is one of the most fundamental processes in chemistry and life science. Excited state intramolecular proton transfer (ESIPT) has been studied as a model system of the proton transfer, since it can be conveniently initiated by light. We report ESIPT reaction dynamic of 1-hydroxy-anthraquione (1-HAQ) in solution by highly time-resolved fluorescence. ESIPT time of 1-HAQ is determined to be $45{\pm}10$ fs directly from decay of the reactant fluorescence and rise of the product fluorescence. High time resolution allows observation of the coherent vibrational wave packet motion in the excited state of the reaction product tautomer. The coherently excited vibrational mode involves large displacement of the atoms, which shortens the distance between the proton donor and the acceptor. With the theoretical analysis, we propose that the ESIPT of 1-HAQ proceeds barrierlessly with assistance of the skeletal vibration, which in turn becomes excited coherently by the ESIPT reaction.

Opticsal Characteristics of Bismuth-doped Aluminosilicate Glass Codoped with Li and Ge (Bi 첨가 알루미노실리케이트 유리에서 Li 및 Ge 공첨가가 광 특성에 미치는 영향)

  • The possibility of improving amplification characteristics and lowering the melting point of bismuth-doped aluminosilicate glass as a new amplification material, which has broadband near-infrared emission at 1300 nm regions, was investigated. Spectroscopic analysis of bismuth-doped aluminosilicate glass shows that the addition of an alkali metal oxide, $Li_{2}O$ increases FWHM of fluorescence spectrum but decreases fluorescence intensity, while $GeO_{2}$ composition increases both FWHM of fluorescence spectrum and fluorescence intensity. Also, excellent optical amplification gain characteristics in a $GeO_{2}$-added sample were observed.

Analysis of O-J-I-P Transients from Four Subtropical Plant Species for Screening of Stress Indicators under Low Temperature (저온스트레스 지표 선발을 위한 아열대성 식물 4종의 O-J-I-P곡선 분석)

  • The changes of O-J-I-P transients were investigated using leaves of four subtropical plant species (Crinum asiaticum var. japonicum, Osmanthus insularis, Chloranthus glaber and Asplenium antiquum) under the natural conditions in winter, in order to select the stress indicators for diagnosing physiological states of plants under low temperature. In the O-J-I-P transients of these species, the fluorescence intensity was found to be higher in O-step and lower in P-step in winter than in summer. Particularly, the fluorescence intensity of the P-step in Crinum asiaticum var. japonicum was lower than those of other three plant species, indicating that Crinum asiaticum var. japonicum is the most sensitive to low temperature. Of the chlorophyll fluorescence parameters derived from O-J-I-P transients of four subtropical plants, Fm, Fv/Fo, ABS/CS, TRo/CS, $\phi_{po}$ and $\phi_{po}/(1-\phi_{po})$ decreased significantly with the increase of Fo, Sm, N, EToICS, ETo/RC and $\psi_o/(1-\psi_o)$ depending on temperature drop in winter. Therefore, these parameters could be used as indicators for estimating low temperature stress and diagnosing physiological states of plants under the natural conditions in winter.

Absorption and Fluorescence of Sm(III) Complexes with some Terdentate Ligands

  • Spectroscopic measurements and theoretical calculations are performed for the four 1 : 3 Sm(III) : ligand solutions. The ligands included in this study are oxidiacetate, iminodiacetate, methyliminodiacetate and dipicolinate. The oscillator strengths for the $4f{\to}4f$ multiplet-to-multiplet transitions are empirically determined from the absorption spectra. The intensity parameters ${\Omega}_{\lambda}\;({\lambda}=2,\;4,\;6)$ of $Sm^{3+}$(aquo) and ${SmL_3}^{3-}$ complexes are also evaluated by applying the Judd-Ofelt theorem to the observed oscillator strengths. The values of the intensity parameters are compared and discussed in term of structural properties of the complexes. In addition, the fluorescence spectra are reported for the Sm(III) complex systems under mild alkaline condition. The excitation from the $^6H_{5/2}$ ground state to any excited states lying above the emitting energy level $(^4G_{5/2})$ produces four fluorescence bands, following nonradiative transitions from a certain excited state to the $^4G_{5/2}$ state. The ratios of oscillator strengths of ${SmL_3}^{3-}$ complexes to that of $Sm^{3+}$(aquo) are also evaluated from the fluorescence spectra and compared to the results obtained from the absorption bands.

Finding Needles in a Haystack with Light: Resolving the Microcircuitry of the Brain with Fluorescence Microscopy

  • To understand the microcircuitry of the brain, the anatomical and functional connectivity among neurons must be resolved. One of the technical hurdles to achieving this goal is that the anatomical connections, or synapses, are often smaller than the diffraction limit of light and thus are difficult to resolve by conventional microscopy, while the microcircuitry of the brain is on the scale of 1 mm or larger. To date, the gold standard method for microcircuit reconstruction has been electron microscopy (EM). However, despite its rapid development, EM has clear shortcomings as a method for microcircuit reconstruction. The greatest weakness of this method is arguably its incompatibility with functional and molecular analysis. Fluorescence microscopy, on the other hand, is readily compatible with numerous physiological and molecular analyses. We believe that recent advances in various fluorescence microscopy techniques offer a new possibility for reliable synapse detection in large volumes of neural circuits. In this minireview, we summarize recent advances in fluorescence-based microcircuit reconstruction. In the same vein as these studies, we introduce our recent efforts to analyze the long-range connectivity among brain areas and the subcellular distribution of synapses of interest in relatively large volumes of cortical tissue with array tomography and superresolution microscopy.

Basic Study on the Application of a Computational Technique to Behavior Characteristics Analysis of the Evaporative Diesel Spray (증발디젤분무의 거동특성해석을 위한 계산기법 적용에 관한 기초 연구)

  • In this study, an analysis of evaporative diesel spray and an usefulness of a general-purpose program, ANSYS CFX release 11.0, are investigated through the comparison and investigation of the experimental results carried out under an evaporative field, in which there is phase transition, by an exciplex fluorescence method and the results analyzed by the CFX program. The diesel fuel called n-Tridecane, $C_{13}H_{28}$, is injected from a single-hole nozzle (l/d=1.0mm/0.2mm) into a constant volume chamber under a high temperature and pressure. In the same condition as the experimental condition, the analysis was carried out. Both results of the spray tip penetration were almost coincident at each time. The results have validated the usefulness of this analysis. As a result, if the ambient pressure is high, the spray tip penetration will be shortened and move toward the nozzle exit.

  • : A DNA analysis system based on fluorescence analysis has to have a DNA amplifying thermal cycle system. DNA amplification is executed by the temperature control. Accuracy of fluorescence analysis is influenced by the temperature control technology. For that reason, the temperature control is core technology in developing the DNA analysis system. Therefore, the objective of this paper is to develop the hardware to apply thermoelectric module to the DNA amplifying thermal cycle system. In order to verify the developed hardware for controlling the temperature of thermoelectric module, a DNA amplifying thermal cycle test was performed. From the test, the developed hardware controlled the temperature of thermoelectric module successfully. Therefore, it is expected that the developed hardware can be applied to the DNA amplifying thermal cycle system.

The Effects of ozone on the greening of barley (Hordeum vulgare L.) seedling (보리 (Hordeum vulgare L.) 유식물의 녹화에 미치는 오존의 영향)

  • The effects of 0.2 ppm ozone on the developing chloroplast of barley (Hordeum vulgare L.) seedling during greening were examined by chlorophyll contents, photosynthetic electron tiansport activity, Fo, Fv and fluorescence quenching coefficient. Chlorophyll contents of seedling treated with ozone were not changed in comparison with the control during the 96 h greening experiment, but PS II activity of the chloroplasts of seedlings treated with ozone was decreases by- l5%. Fo was slightly decreased but Fv was decreases by 5% in comparison with the control, In fluorescence quenching analysis, qP and qE were decreased by 11% and 9%, respectively, in comparison with the control. These results suggest that oxidation site of PS II is the site affected mostly and PQ pool is also affected slightly by 0.2 ppm ozone.

Cometabolism in the Biodegradation of Benzene, Toluene, and ${\rho}-xylene$ Mixture by Isolated Pseudomonas fluorescence BE103

  • A microorganism showing degradative activity towards benzene, toluene and ${\rho}-xylene$ (BTX) was isolated from an activated sewage sludge and was tentatively identified as Pseudomonas fluorescence BE103. This strain was found to utilize benzene and toluene as growth substrates, but to degrade ${\rho}-xylene$ in the obligate presence of a growth substrate. The metabolic product resulted from the cometabolism of ${\rho}-xylene$ was identified as 3, 6-dimethylpyrocatechol by LC/MS analysis, and the metabolic pathway was analyzed to be similar to the tod pathway. From the kinetic studies done regarding BTX biodegradation using Pseudomonas fluorescence BE103, it was revealed that the cometabolism of ${\rho}-xylene$ is significantly affected by the ratio of growth substrate concentration to biomass concentration, and that the cometabolism of ${\rho}-xylene$ initiates only when this ratio was about 0.03.

Chicken Disease Characterization by Fluorescence Spectroscopy

  • Fluorescence spectroscopy was used to characterize chicken carcass diseases. Spectral signatures of three different disease categories of poultry carcasses (airsacculitis, cadaver and septicemia) were obtained from fluorescence emission measurements in the wavelength range of 360 to 600 nm with 330 nm excitation. Principal Component Analysis (PCA) was used to select the most significant wavelengths for the classification of poultry carcasses. These wavelengths were analyzed for pathologic correlation of poultry diseases. Using a Soft Independent Modeling of Class Analogy (SIMCA) of principal components with a Mahalanobis distance metric, poultry carcasses were individually classified into different classes with $97.9\%$ accuracy.
