Numerical Comparisons of Flow Properties Between Indivisual and Comprehensive Consideration of River Inundation and Inland Flooding (하천범람과 내수침수의 개별적·복합적 고려에 따른 흐름 특성의 수치적 비교)

  • Due to the climate change, torrential rain downpours unprecedentedly, and urban areas repeatedly suffer from the inundation damages, which cause miserable loss of property and life by flooding. Two major reasons of urban flooding are river inundation and inland submergence. However, most of previous studies ignored the comprehensive mechanism of those two factors, and showed discrepancy and inadequacy due to the linear summation of each analysis result. In this study, river inundation and inland flooding were analyzed at the same time. Petrov-stabilizing scheme was adopted to capture the shock wave accurately by which river inundation can be modularized. In addition, flux-blocking alrotithm was introduced to handle the wet and dry phenomena. Sink/source terms with EGR (Exponentially Growth Rate) concept were incorporated to the shallow water equations to consider inland flooding. Comprehensive simulation implementing inland flooding and river inundation at the same time produced satisfactory results because it can reflect the counterbalancing and superposition effects, which provided accurate prediction in flooding analysis.

Flavonoids from Orostachys Japonicus A. Berger Induces Caspase-dependent Apoptosis at Least Partly through Activation of p38 MAPK Pathway in U937 Human Leukemic Cells

  • Background: Orostachys japonicus A. Berger (A. Berger) is commonly used as a folk remedy for cancer therapy. However, the mechanisms of its anti-cancer activity are poorly investigated in human cancer cells. In this study, we investigated whether flavonoids extracted from Orostachys japonicus A. Berger (FEOJ) might have anticancer effects in human leukemia cells, focusing on cell death mechanisms. Materials and Methods: U937 human leukemic cancer cells were used. Results: FEOJ induced apoptosis in a dose-dependent manner in human U937 cancer cells. Flow cytometry revealed significant accumulation of cells with sub-G1 DNA content at the concentrations of $200{\mu}g/mL$ and $400{\mu}g/mL$. FEOJ-induced apoptosis was caspase-dependent through loss of mitochondrial membrane potential (MMP, ${\Delta}{\Psi}m$) in human U937 cancer cells, which might be associated with suppression of Bcl-2 and XIAP proteins. FEOJ induced the p38 MAPK signaling pathway, playing at least in part an important role in FEOJ-induced apoptosis. Conclusions: This study suggested that FEOJ may induce caspase-dependent apoptosis in human leukemic cells by regulating MMP (${\Delta}{\Psi}m$) through suppressing Bcl-2 and X-IAP. In addition, the results indicated that upstream p38 MAPK signaling regulates the apoptotic effect of FEOJ. This study provides evidence that FEOJ might have anti-cancer potential for human leukemic cells.

  • Purpose: Heat transfer analysis of recently developed 'slim type double-skin system window' were presented. This window system is designed for curtain wall type façade that main energy loss factor of recent elegant buildings. And the double skin system is the dual window system integrated with inner shading component, enclosed gap space made by two windows when both windows were closed and shading component effectively reflect and terminate solar radiation from outdoor. Usually double-skin system requires much more space than normal window systems but this development has limited by 270mm, facilitated for curtain wall façade buildings. In this study, we estimated thermophysical phenomena of our double-skin curtain wall system window with solar load conditions at the summer season. Method: A fully 3-Dimentional analysis adopted for flow and convective and radiative heat transfer. The commercial CFD package were used to model the surface to surface radiation for opaque solid region of windows' frame, transparent glass, fluid region at inside of double-skin and indoor/outdoor environments. Result: Steep angle of solar incident occur at solar summer conditions. And this steep solar ray cause direct heat absorption from outside of frame surface rather than transmitted through the glass. Moreover, reflection effect of shading unit inside at the double-skin window system was nearly disappeared because of solar incident angle. With this circumstances, double-skin window system effectively cuts the heat transfer from outdoor to indoor due to separation of air space between outdoor and indoor with inner space of double-skin window system.

  • Various researches on the application of polymer dispersions to the cement mortar and concrete have been practised in many countries like America, Japan and Germany and so on because of high performance and good modification effect of these. In this study, SBR, Polymer dispersion that widely used in situ is employed that the self-flowability may be induced in the cemen mortar. In order to comprehend and investigate the modification of cement mortar with self-flowability by SBR and properties and fracture mode of adhesive strength in tension of that, experimental parameter was set as SBR solid-Cement ratio(S/C) and Cement:Fine aggregate(C:F) and the experiments such as Unit weight, Flow, Consistency change, Crack resistance and Segregation that inform on the general properties have been done. In addition of that, Adhesion in tension is measured with a view to comprehending the properties and fracture mode in tensile load. Consistency change of cement mortar modified by SBR did grow better as the ratio of SBR solid-Cement increased and was much superior to that of resin based flooring such as polyurethane and epoxy which recorded the loss of consistency in 90min. after mixing. Adhesive strength in tension increased with continuity in the curing age and showed the maximum in case of C:F=1:1 and S/C=20%. As the increase of curing age, the fracture mainly happened in the concrete substrate and the interface between the specimen and concrete substrate.

  • The loss of posterior support and the abnormal jaw relation can cause pathologic findings. If deep bite patients with multiple missing teeth, can not have the stable posterior contact, the mandible moves posteriorly, and consequently the overjet and overbite get worse. And when the mandibular irregular occlusal plane is corrected, it is easier to have the bilateral balanced occlusion with the maxilla. So the treatment goal is to give proper posetrior support and establish appropriate anterior guidance, and ultimately provide improved mastication and esthetics recovery. In this case, a 68 year old man, having deep bite without posterior support was evaluated by the vertical dimesion decision flow-chart. An available prosthetic height, anterior occlusal relation such as overjet, overbite and the esthetic part such as facial height and the cephalometric analysis are the factors to be considered.

MICROSCOPIC OBSERVATIONS OF FAT TRANSLOCATION IN THE TISSUE OF YELLOW CORVENIA DURING SALTING AND DRYING ("굴비" 제조과정중의 지방의 이동에 대한 조직학적 관찰)

  • Salted and dried yellow corvenia(Pseudosciaena manchurica), so called 'Gul-bi', is one of nation-widely consuming fish foo::ls. It is suitable for a long term preservation and its pro-duce is also a great deal on sea food processing in this country. The texture of 'Gul-bi', however, have often appeared to be a delicate factor for the quality of the product. The loss or dislocation of fat in the tissue of the fish resulted by salting and drying is believed to profoundly relate to the texture of product. In this paper, the tissue of yellow corvenia and movement of fat were microscopically observed before salting, immediately after salting, and after drying and the results observed in the tissues dry salted, brine salted, and brine salted with the addition of BHA were compared. The cross section of yellow corvenia muscle showed that a distinctive border by connective tissue between white and red muscle could not be seen in general, and red muscle was surrounded by hypodermic fatty tissues. In the tissue of fresh yellow corvenia, the fat was mainly distributed in hypodermic fat layer which located under the corium while rarely distributed in white muscle. It was found that some parts of the fat in the tissue were permeated into intermuscular tissue passing through the connective tissues during salting. The result Was the same in both dry-salting and brine-salting tissue. However, the fat translocated into intermuscular tissues disappeared during drying process in the salted without BHA tissues whereas in BHA added tissue. This result suggested that BHA may take a role of multiple effect in translocation of fat in tissues as well as in retarding oxidation. In an advanced stage of salted and dehydration, the muscle fibers were ajoined together and then limits between muscle fibers already became indistinguishable. And the migrated fat into intermuscular tissue aggregated around the connective tissue and are apt to gradually to flow out from the muscular system through these tissues.

Evaluation of Sediment Yield Prediction and Estimation of Sediment Yield under Various Slope Scenarios at Jawoon-ri using WEPP Watershed Model (WEPP Watershed Version을 이용한 홍천군 자운리 농경지 토양유실 예측 및 경사도에 따른 토양유실량 평가)

  • To evaluate the soil erosion best management practices, many computer models has been utilized over the years. Among those, the USLE and SWAT models have been widely used. These models estimate the soil erosion from the field using empirically-based USLE/MULSE in it. However, these models are not good enough to estimate soil erosion from highland agricultural watershed where severe storm events are causing soil erosion and muddy water issues at the receiving watersheds. Thus, physically-based WEPP watershed version was applied to a watershed, located at Jawoon-ri, Gangwon with very detailed rainfall data, rather than daily rainfall data. Then it was validated with measured sediment data collected at the sediment settling ponds and through overland flow. In this study, very detailed rainfall data, crop management data, soil data reflecting soil reconditioned for higher crop production were used in the WEPP runs. The $R^2$ and the EI for runoff comparisons were 0.88 and 0.91, respectively. For sediment comparisons, the $R^2$ and the EI values were 0.95 and 0.91. Since the WEPP provides higher accuracies in predicting runoff and sediment yield from the study watershed, various slope scenarios (2%, 3%, 5.5%, 8%, 10%, 13%, 15%, 18%, 20%, 23%, 25%, 28%, 30%) were made and simulated sediment yield values were analyzed to develop appropriate soil erosion management practices. It was found that soil erosion increase linearly with increase in slope of the field in the watershed. However, the soil erosion increases dramatically with the slope of 20% or greater. Therefore special care should be taken for the agricultural field with slope greater than 20%. As shown in this study, the WEPP watershed version is suitable model to predict soil erosion where torrential rainfall events are causing significant amount of soil loss from the field and it can also be used to develop site-specific best management practices.

Application of Cold Weather Concreting with Accelerator for Freeze Protection to Full Scale Structures (내한촉진제를 사용한 한중콘크리트의 실구조물 적용에 관한 연구)

  • In this study, the results of applying cold weather concreting mixed with Accelerator for Freeze Protection(AFP) to full scale structures are presented. Since the determination of W/C and amount of AFP significantly have an effect on strength gain and protection of frost damage in early, a full investigation is needed to determine these values at stage of nux design. The flowability of fresh cold weather concreting with AFP was similar to the same W/C. Lower loss of workability and initial slump flow of concrete using superplasticizer of polycarboxylic ester than that of melamine sulphonate showed that polycarboxylic ester was more effective on elapsed time. Temperature histories of specimens located in insulation boxes at the site was similar to that of structures. Thus, it is cleared that simple adiabatic curing method is effective for evaluating in-place concrete strength than specimens cured by sealing method. The investigation results of development of compressive strength of cold weather concreting included AFP with curing methods by logistic curves indicated that AFP can be effective to gain strength at lower temperature than normal curing temperature. In field testing, vinyl sheets were placed over the concrete sections and AFP enabled concrete to gain $5N/{mm}^2$ to protect frost damage in early ages and specified compressive strength of concrete at 28 days under average temperature of $-2^{\circ}C$ (lowest temperature was $-12^{\circ}C$) during site application.

  • Recently, ground pits that have been occurring frequently in urban areas are hindering traffic flow and causing property damages and loss of human life, acting as factors that are threatening the safety of citizens. Therefore, sunken ground must be quickly restored and provisions must be made for additional damage but current domestic detailed standards regarding ground pits and accurate definitions regarding causes and measures to be taken for reoccurrences are lacking. Restoration methods of sunken ground include backfilling by reusing sunken soil or other fill material and paving the road and while this is the most often used method, this only prevents ground from sinking temporarily and can not serve as a fundamental solution. Also, additional ground pits can occur on ground that is reinforced using this method due to faulty backfill material or faulty hardening. This study used Eco-friendly High-Strength Material (EHSM) as restoration material that can be used in the restoration of underground cavities that have occurred due to ground subsidence to analyze the engineered characteristics of modified dredging clay and test pieces made from changed ratios of EHSM and weathered granite soil were uniaxial compression tests were conducted and freezing-thawing tests were conducted to study strength properties according to environmental changes of restoration material, and after tests were concluded by each level, uniaxial compression tests and dynamic elasticity tests were conducted for intensity analysis. Also, to evaluate strength characteristics of the restored ground, dynamic plate load tests were conducted to verify the improvement effectiveness of the restored ground.

Anti-metastatic Effects on B16F10 Melanoma Cells of Extracts and Two Prenylated Xanthones Isolated from Maclura amboinensis Bl. Roots

  • Inhibitory effects of Maclura amboinenesis Bl, one plant used traditionally for the treatment of cancers, on metastatic potential of highly metastatic B16F10 melanoma cells were investigated in vitro. Cell proliferation was assessed using the MTT colorimetric assay. Details of metastatic capabilities including invasion, migration and adhesion of B16F10 melanoma cells were examined by Boyden Chamber invasion and migration, scratch motility and cell attachment assays, respectively. The results demonstrated that n-hexane and chloroform extracts exhibited potent anti-proliferative effects (p<0.01), whereas the methanol and aqueous extracts had less pronounced effects after 24 h exposure. Bioactivity-guided chromatographic fractionation of both active n-hexane and chloroform extracts led to the isolation of two main prenylated xanthones and characterization as macluraxanthone and gerontoxanthone-I, respectively, their structures being identified by comparison with the spectral data. Interestingly, both exhibited potent effective effects. At non-toxic effective doses, n-hexane and chloroform extracts (10 and $30{\mu}g/ml$) as well as macluraxanthone and gerontoxanthone-I (3 and $10{\mu}M$) significantly inhibited B16F10 cell invasion, to a greater extent than $10{\mu}m$ doxorubicin, while reducing migration of cancer cells without cellular cytotoxicity. Moreover, exposure of B16F10 melanoma cells to high concentrations of chloroform ($30{\mu}g/ml$) and geratoxanthone-I ($20{\mu}M$) for 24 h resulted in delayed adhesion and retarded colonization. As insights into mechanisms of action, typical morphological changes of apoptotic cells e.g. membrane blebbing, chromatin condensation, nuclear fragmentation, apoptotic bodies and loss of adhesion as well as cell cycle arrest in the G1 phase with increase of sub-G1 cell proportions, detected by Hoechst 33342 staining and flow cytometry were observed, suggesting DNA damage and subsequent apoptotic cell death. Taken together, our findings indicate for the first time that active n-hexane and chloroform extracts as well as macluraxanthone and gerontoxanthone-I isolated from Maclura amboinensis Bl. roots affect multistep of cancer metastasis processes including proliferation, adhesion, invasion and migration, possibly through induction of apoptosis of highly metastatic B16F10 melanoma cells. Based on these data, M. amboinensis Bl. represents a potential candidate novel chemopreventive and/or chemotherapeutic agent. Additionally, they also support its ethno-medicinal usage for cancer prevention and/or chemotherapy.
