Continuous Production of Lactosucrose by Immobilized Sterigmatomyces elviae Mutant

  • In this study, in order to develop a continuous production process of lactosucrose in a packed-bed reactor, Sterigmatomyces elviae ATCC 18894 was selected and mutated. The mutant strain of S. elviae showed 54.3% higher lactosucrose production than the wild type. Reaction conditions such as temperature, pH, substrate concentration and flow rate were also optimized. Under optimized reaction conditions ($50^{\circ}C$, pH 6.0, 25% sucrose and 25% lactose as substrate, flow rate 1.2 ml/min), the maximum concentration of lactosucrose (192 g/l) was obtained. In a packed-bed reactor, continuous production of lactosucrose was performed using S. elviae mutant immobilized in calcium alginate, and about 180 g/l of lactosucrose production was achieved for 48 days.

  • <나탈리>(2010)와 <7광구>(2011)를 통해 한국 3D 영화제작의 가능성이 확인됐다. 그러나 동시에 한국 3D 영화제작 환경에서의 다양한 기술적, 제작과정상의 문제점이 도출되었다. 본 연구는 <7광구>의 3D 영화제작 사례를 분석한 결과이다. 분석은 주로 기술적인 오류와 제작 공정상의 파이프라인에서 드러난 문제점에 주목하여 이루어 졌다. <7광구>를 통해 얻은 제작 경험과 시행착오를 분석함으로써 향후 제작될 한국형 3D 영화의 제작진들에게 실무적인 연구결과를 전달하는 것이 본연구의 목적이다. 분석결과 <7광구>는 기술적인 완성도뿐만 아니라 효율적인 제작 파이프라인 구축에도 실패한 것으로 드러났다. 그럼에도 불구하고 <7광구>는 한국적인 3D 제작공정을 능동적으로 구축하려했다는 점에서 긍정적으로 평가 받는다.

유동안정성과 생산량을 고려한 2단 생산튜빙 디자인 (Tapered production tubing design considering flow stability and production rate)

  • 생산량 증대를 위한 방법으로 다른 두 내경의 2단 튜빙을 이용한 기법이 제안되고 있다. 본 연구에서는 다양한 2단 튜빙조합을 고려하여 각 목적에 부합하는 조합들을 제시하였다. 기존연구에서 고려한 생산량뿐만 아니라 유동안정성을 확보하는 결과를 제시하였다. 유동안정성은 tubing pressure traverse, 액체부피비, 운영조건의 안정성을 통해 분석되었다. 본 연구에서 분석한 가상유전의 경우 순현재가치가 가장 큰 2단 튜빙은 하단이 4.5 in 내경에 길이가 2000 ft이며 상단은 5.5 in 내경에 길이가 8000 ft인 조합이다. 2단 튜빙간 마찰압력손실 차이는 크지 않았기 때문에 단일튜빙과 비교했을 때보다 다양한 조합이 가능하였다. 튜빙하단에선 liquid loading을 방지하기 위해 작은 액체부피비를 유지하는 것이 중요한데, 이는 2단 튜빙 하단에서 작은 내경을 사용함으로써 가능하였다. 생산운영의 분석결과로부터 다양한 튜빙 디자인이 가능함을 확인하였다.

플러그 흐름 소화기 속에서 Grain Dust의 혐기성 소화에 의한 메탄가스 생산 (Methane Production by Anaerobic Digestion of Grain Dust in a Plug Flow Digester)

  • 3리터의 실험실 크기의 플러그 흐름의 혐기성 소화기를 사용하여 grain dust로부터 메탄가스를 생산하였다. 이 실험에서 사용한 소화기는 온도(35, 45, 55$^{\circ}C$), 체류 시간(6, 12일), 초기농도(7.8, 9.0% Total Solids)의 함수로 10가지 조건에서 실험이 행하여 졌는데, 실험 결과 55$^{\circ}C$, 6일 HRT, 7.8%TS 에서 가장 높은 메탄가스를 생산하였으며, 실험결과를 토대로 메탄가스생산 관계식을 제시하였다. 또한, 위의 실험의 조건에서는 thermophilic 의 경우가 mesophilic 경우보다 메탄가스 생성이 좋은 것으로 나타났다.

죽력(竹瀝)과 대나무 추출액의 혼합물이 뇌혈류 및 혈압에 미치는 작용 기전 (Mechanism of Mixture of Bambusae Caulis in Liquamen and Bamboo Extract on the Cerebral Blood Flow and Blood Pressure in Rats)

  • This Study was designed to investigate the mechanism of Mixture of Bambusae Caulis in Liquamen and Bamboo Extract on the change of regional cerebral blood flow (rCBF) and blood pressure (BP) in normal rats, and further to investigate cytokines production in serum of cerebral ischemic rats. Mixture were as follows ; Bamboo Extract extracted with distilled water at 98 $^{\circ}C$ for 3 hrs, Mixture of Bambusae Caulis in Liquamen and bamboo Extracts (MLE) mixed at the ratio 1 to 100 (MLE100), 1 to 50 (MLE50), 1 to 20 (MLE20), 1 to 10 (MLE10), 1 to 5 (MLE5). The results were as follows ; The MLE-induced increase in rCBF was significantly inhibited by pretreatment with indomethacin (1 mg/kg, I.p.), an inhibitor of cyclooxygenase as well as methylene blue (10 $^{\mu}g/kg$, I.p.), an inhibitor of guanylate cyclase. The MLE-induced increase in BP was significantly inhibited by pretreatment with methylene blue. In cytokines production in the serum drawn from femoral arterial 1 hr after middle cerebral artery occlusion, MLE5 significantly increased production of TGF-${\beta}$ and increased production of IL-10, but significantly decreased production of TGF-${\alpha}$ compared with control group. In cytokines production in the serum drawn from femoral arterial 1 hr after reperfusion, MLE5 significantly increased production of TGF-${\beta}$ and IL-10, but significantly decreased production of TGF-${\alpha}$ compared with control group. AS results above. And MLE5 had anti-ischemic effect by inhibiting TGF-${\alpha}$ production, and by accelerating IL-10 and TGF-${\beta}$ production.

다품종 소량생산 설비의 총괄생산계획에 관한 사례 연구: 시스템다이내믹스 시뮬레이션 모델링을 중심으로 (A Case Study on the Aggregate Planning of Multi-product Small-batch Production Facilities: Focusing on System Dynamics Simulation Modeling)

  • Purpose: The purpose of this study is to guide the operation managers who plan daily production of large mass-processing facility that services multi-customers with multi-product, small-batch item characteristics by providing the practical best production quantity and the inventory allowed to build. Methods: Close observation of a subcontract paint-shop operator captured the daily decision process which was reflected in the subcontractor-unique mathematical model and the system dynamics simulation model. Multiple simulations were run to find the practical best production quantity and the maximum allowable stock level of inventory that did not undermine the profit from practical best daily production. Actual data and a few constant values were obtained from the firm under study. Results: While the inventory holding cost for the customer-owned material harms the total profit of the subcontractor, the running cost of the processing facility hinders production in small batches. This balances the maximum possible productions and results in practical best daily production which can be found through simulation runs with actual data. The maximum level of stocked inventory is deduced from the practical best daily production. Conclusion: To build a large volume that enables economy-of-scale production, operators should deal with multi-product small-batch items from multiple customers. When the planned schedule of the time and amount of material in-flow tend not to be reliable, operators can find it practical to execute level production across the planning horizon instead of adjusting to day-to-day in-flow fluctuations.

  • Chemical Mechanical Polishing(CMP) in semiconductor production is characterized its output property by Removal Rate(RR) and Non-Uniformity(NU). Some Previous works shows that RR is determined by production of pressure and velocity and NC is also largely affected by velocity of flow-field during CMP. This study is about the direct measurement of velocity of slurry during CMP and reconstruction whole flow-field by Particle Image Velocimetry(PIV) Techniques. Typical PIV system is tuned adequately for inspecting CMP and Slurry Flow-field is measured by changing both Pad RPM and Carrier RPM. The results show that velocity is majorly determined not by Carrier RPM, but by Pad RPM.

생산계획 및 우선순위규칙에 따른 Order Release 정책의 유효성에 관한 연구 (The Effectiveness of Order Release Strategies considering Production Plan and Dispatching Rules)

  • Order Review/Release (ORR) System is the linkage between planning system and actual production. Reduction of the waiting time on the machines, work in process and lead time variation may be achieved by adopting ORR strategies. But researchers on the ORR do not agree on the effectiveness of ORR. Some say that the overall system flow time may be increased if ORR is adopted, but others say that ORR can reduce work in process, flow time and variation of flow time. The objective of this research is to clarify under what environments order release strategy is effective. Simulation study was conducted in a hypothetical job shop. The experimental results show that dispatching rule is much more important than ORR is in controlling the shop floor. But the results indicate that ORR can reduce mean shop flow time, average work in process and variation of shop flow time under such environments where utilization level is high and planned order is weekly released to the order pool. And the results also show that the effect of plan smoothing on the ORR is insignificant, which is inconsistent with the results of the previous researches.

  • In this study, the qualitative prediction and evaluation of clean room being utilized for mass production of electrronic components have been performed with the help of flow simulation. Compared to the experimental analysis based on measurements of the number of particles, concentration of contaminants and flow characteristics, the numerical analysis used in this study is much cost-effective.

산소부화 압축착화기관을 이용한 메탄으로부터 수소 생산 (Hydrogen Gas Production from Methane Reforming Using Oxygen Enriched Compression Ignition Engine)

  • The purpose of this paper is to investigate the reforming characteristics and maximum operating condition for the hydrogen production by methane reforming using the compression ignition engine induced partial oxidation. An dedicated compression engine used for methane reforming was decided operating range. The partial oxidation reforming was investigated with oxygen enrichment which can improve hydrogen production, compared to general reforming. Parametric screening studies were achieved as $O_2/CH_4$ ratio, total flow rate, and intake temperature. When the variations of $O_2/CH_4$ ratio, total flow rate, and intake temperature were 1.24, 208.4 L/min, and $400^{\circ}C$, respectively, the maximum operating conditions were produced hydrogen and carbon monoxide. Under the condition mentioned above, synthetic gas were $H_2\;22.77{\sim}29.22%,\;CO\;21.11{\sim}23.59%$.
