Knockdown of Ezrin by RNA Interference Reverses Malignant Behavior of Human Pancreatic Cancer Cells in Vitro

  • Background: Pancreatic cancer is one of the most aggressive tumors with a dismal prognosis. The membrane cytoskeletal crosslinker Ezrin participates in several functions including cell proliferation, adhesion, motility and survival. There is increasing evidence that Ezrin is overexpressed in vast majority of malignant tumors and regulates tumor progression. However, its roles in pancreatic cancer remain elusive. Methods: Three pairs of specific Ezrin siRNAs were designed and synthetized and screened to determine the most efficient one for construction of a hairpin RNA plasmid targeting Ezrin. After transfection into the Panc-1 pancreatic cancer cell line, real-time quantitative PCR and Western blotting were performed to examine the expression of mRNA and protein. The MTT method was applied to examine the proliferation and the drug sensibility to Gemcitabine. Flow cytometry was used to assess the cycle and apoptosis, while capacity for invasion was determined with transwell chambers. Furthermore, we detected phosphorylated-Erk1/2 protein and phosphorylated-Akt protein by Western blotting. Results: Real-time quantitative PCR and Western blotting revealed that Ezrin expression was notably down-regulated at both mRNA and protein levels by RNA interference (P< 0.01). Proliferation was inhibited and drug resistance to gemcitabine was improved (P< 0.05). Flow cytometry showed that the proportion of cells in the G1/G0 phase increased (P< 0.01), and in G2/M and S phases decreased (P< 0.05), with no apparent differences in apoptosis (P> 0.05). The capacity for invasion was markedly reduced (P< 0.01). In addition, down-regulating Ezrin expression had no effect on phosphorylated-Akt protein (P>0.05), but could decrease the level of phosphorylated-Erk1/2 protein (P< 0.05). Conclusions: RNA interference of Ezrin could inhibit its expression in the pancreatic cancer cells line Panc-1, leading to a potent suppression of malignant behavior in vitro. Assessment of potential as a target for pancreatic cancer treatment is clearly warranted.

Distributed Multi-channel Assignment Scheme Based on Hops in Wireless Mesh Networks (무선 메쉬 네트워크를 위한 홉 기반 분산형 다중 채널 할당 방안)

  • In wireless mesh networks (WMNs), the end-to-end throughput of a flow decreases drastically according to the traversed number of hops due to interference among different hops of the same flow in addition to interference between hops of different flows with different paths. This paper proposes a distributed multi-channel assignment scheme based on hops (DMASH) to improve the performance of a static WMN. The proposed DMASH is a novel distributed multi-channel assignment scheme based on hops to enhance the end-to-end throughput by reducing interference between channels when transmitting packets in the IEEE 802.11 based multi-interface environments. The DMASH assigns a channel group to each hop, which has no interference between adjacent hops from a gateway in channel assignment phase, then each node selects its channel randomly among the channel group. Since the DMASH is a distributed scheme with unmanaged and auto-configuration of channel assignment, it has a less overhead and implementation complexity in algorithm than centralized multi-channel assignment schemes. Simulation results using the NS-2 showed that the DMASH could improve remarkably the total network throughput in multi-hop environments, comparing with a random channel assignment scheme.

Moderating Effect of Self-efficacy between Work-Family Conflict and Job Burnout (직장-가정 갈등과 직무소진간 자기효능감의 조절효과)

  • Currently, enterprises are in a rapid flow of comvergence of industries in the ear of the 4th Industrial Revolution. Under this circumstances, change of economic condition, change of industrial structure, increase of women's advance into society. Based on such background of the times, the aim of this study is to examine the effect of work-home conflict on burnout and the moderating effect of self-efficacy in that relation. The research samples were obtained from nurses at hospitals in Daejeon-si, Sejong-si, Chungcheongnam-do and Chungcheongbuk-do. Total 360 copies of valid questionnaire was used for analysis. Specific analysis results from the research are as follows. Firstly, it was found that Work Interference with Family(WIF) had positive (+) impact on physical burnout. Family Interference with Work (FIW) had no impact on physical burnout. Secondly, it was found that Work Interference with Family(WIF) had positive (+) impact on emotional burnout and Family Interference with Work (FIW) had negative (-) impact on emotional burnout. Thirdly, self-efficacy had a buffer effect on the negative impact of Work Interference with Family upon emotional burnout. However, there was no moderating effect of self-efficacy in the relationship between Family Interference with Work (FIW) and emotional burnout. Therefore, it needs to consider a plan to reduce conflicts between work and family according to the causes of conflicts and it also needs to consider relevant results.

The Characteristic Investigation of the Flowfield around Two Circular Cylinders in the Tandem Arrangement Using the PIV (PIV를 이용한 직렬배열에서 2원주 주위의 유동장 특성 연구)

  • The Characteristics of the flowfield around two circular cylinders in tandem arrangement was investigated by PIV. Strouhal numbers. velocity vectors and velocity profiles were observed at centre-to-centre space ratios of P/D= 1.5. 2.0 and 2.5, and Reynolds number of $Re=3.0{\times}10^3{\sim}5.0{\times}10^3$. As the results the Strouhal numbers measured in the rear region of the cylinder of wake side were decreased with the space ratios. The flow between two cylinders was almost stagnated and the size of the stagnated region was larger in the close side than in the far side of the front cylinder. The direction of vortex between two cylinders was opposed each other with the small difference(${\alpha}\;{\pm}1.0^{\circ}$) of the attack angle ${\alpha}$.

Comparative Analysis of Flow Characteristics Using Reflected Pressure Wave at Crossing of Subway Trains in Straight Tunnel (직선터널에서 지하철 열차의 교차운행 시 반사파 간섭에 따른 유동 특성 비교분석)

  • In this study, CFD is used to compare and analyze the flow characteristics using reflected pressure wave during the intersection of two trains in straight tunnel. Two tunnels of different lengths; 600 m and 3,400 m were designed and numerical analysis of the flow characteristics of two tunnels carried out by setting the crossing state of the two trains at a constant velocity of 27 m/s form the center of the tunnel. The simulation model was designed using the actual tunnel and subway dimensions The train motion was achieved by using the moving mesh method. For the numerical analysis, $k-{\omega}$ standard turbulence model and an ideal gas were used to set the flow conditions of three-dimensional, compressible and unsteady state. In the analysis results, it was observed that the inside of the long tunnel without interference of the reflected pressure wave was maintained at a pressure lower than the atmospheric pressure and that the flow direction was determined by the pressure gradient and shear flow. On the other hand, the flow velocity in the short tunnel was faster and the pressure fluctuation was noted to have increased due to the reflected pressure wave, with more vortices formed. In addition, the flow velocity was noted to have changed more irregularly.

NUMERICAL INVESTIGATION OF SHOCK-BUFFET ON TRANSPORT AIRCRAFT WITH CHANGING THE POSITION OF NACELLE/PYLON (항공기 Nacelle/pylon 위치에 따른 Shock-Buffet 현상의 수치적 연구)

  • The shock buffet on a transonic transport aircraft are negative factors that reduce the aerodynamic performance of aircraft. The parametric studies were performed for position of nacelle/pylon to estimate the trend of flow mechanism under the wing that affects shock buffet. To generate external mesh of aircraft configuration that change the position of nacelle, snappyHexMesh provided in OpenFOAM was applied. Implicit density-based solver(ISAAC) was used for flow analysis. The change of nacelle position along horizontal direction dynamically affected the aerodynamic performance of transonic transport aircraft as comparing that of vertical direction. As a result of the parametric study of nacelle/pylon position, it was confirmed that the optimal position of nacelle can be obtained by aerodynamic design.

Effect of building proximity on external and internal pressures under tornado-like flow

  • Tornadoes are one of the world's deadliest natural phenomena. They are characterized by short life span and danger. It has been observed through post-damage surveys that localities with large numbers of buildings suffer major damage during a tornado attack resulting in huge loss of life and property. Thus,it is important to study interfering buildings exposed to tornado-like vortices. The present study focuses on external and internal pressures developed on building models exposed to translating tornado-like vortices in the presence of an interfering building model. The effects of translating speed and swirl ratio of a tornado-like vortex on external and internal pressures for a principal building in the vicinity of an interfering building are investigated. Results indicate that external and internal pressures are enhanced or reduced depending on the location of the interfering building with respect to the principal building.

Analysis of Ring Pack Lubrication

  • This paper describes a method developed for the simulation of ring pack lubrication characteristic in an internal combustion engine. In general, the quantity of oil supply for piston ring lubrication may be insufficient in filling the entire volume formed at the interference between the piston ring and the cylinder liner. Thus the oil starvation condition should be considered in analyzing piston ring lubrication. In order to reasonably estimate the amount of oil left over on the cylinder liner, the flow rate at the posterior portion of the interface should be calculated with an adequate boundary condition that confirms flow continuity condition. In this analysis, oil starvation and open-end boundary conditions are considered at the inlet and outlet of the piston rings. The lubrication characteristic of each piston ring is obtained by an iterative method with sequential steps. It is revealed that piston rings are operated under oil starvation in most operating cycles and the result under these conditions are quite different from that with the fully-flooded assumption.

NUMERICAL INVESTIGATION OF AERODYNAMIC INTERACTION OF AIR-LAUNCHED ROCKETS FROM A HELICOPTER (헬리콥터로부터 발사된 로켓의 공력 간섭 현상에 대한 수치적 연구)

  • Numerical simulation of air-launched rockets from a helicopter was conducted to investigate the aerodynamic interference between air-launched rocket and helicopter. For this purpose, a three-dimensional inviscid flow solver has been developed based on unstructured meshes. An overset mesh technique was used to describe the relative motion between rocket and rocket launcher. The flow solver was coupled with six degree-of-freedom equation to predict the trajectory of free-flight rockets. For the validation, calculations were made for the impinging jet with inclined plate. The rotor downwash of helicopter was calculated and applied to simulation of air-launched rocket. It is shown that the rotor downwash has non-negligible effect on the air-launched rocket and its plume development.

  • In this paper, we introduce a joint flow control and scheduling algorithm for multi-hop cognitive radio networks with spectrum underlay. Our proposed algorithm maximizes the total utility of secondary users while stabilizing the cognitive radio network and still satisfies the total interference from secondary users to primary network is less than an accepted level. Based on Lyapunov optimization technique, we show that our scheme is arbitrarily close to the optimal.
