Apigenin Sensitizes Huh-7 Human Hepatocellular Carcinoma Cells to TRAIL-induced Apoptosis

  • TNF-related apoptosis-inducing ligand (TRAIL) is a promising agent for management of cancer because of its selective cytotoxicity to cancer cells. However, some cancer cells have resistance to TRAIL. Accordingly, novel treatment strategies are required to overcome TRAIL resistance. Here, we examined the synergistic apoptotic effect of apigenin in combination with TRAIL in Huh-7 cells. We found that combined treatment of TRAIL and apigenin markedly inhibited Huh-7 cell growth compared to either agent alone by inducing apoptosis. Combined treatment with apigenin and TRAIL induced chromatin condensation and the cleavage of poly (ADP-ribose) polymerase (PARP). In addition, enhanced apoptosis by TRAIL/apigenin combination was quantified by annexin V/PI flow cytometry analysis. Western blot analysis suggested that apigenin sensitizes cells to TRAIL-induced apoptosis by activating both intrinsic and extrinsic apoptotic pathway-related caspases. The augmented apoptotic effect by TRAIL/apigenin combination was accompanied by triggering mitochondria-dependent signaling pathway, as indicated by Bax/Bcl-2 ratio up-regulation. Our results demonstrate that combination of TRAIL and apigenin facilitates apoptosis in Huh-7 cells.

A Study on Heat Transfer Performances of a Heat Pipe Heat Sink for Power Control Semiconductors (전력제어 반도체용 히트파이프 냉각기의 열전달 성능 연구)

  • In this days, heat pipe heat sink has been widely applied to power controllers for railway substations to remove heat from power semiconductors(diodes or thyristors). The heat pipe heat sink consists of a aluminum heating block for mounting the thyristor, 2~3 heat pipes and large number of aluminum fins. The present study was to get fundamental informations of the structure, design parameters and heat transfer performances of heat pipe heat sink. Series of operational test for a unit with 3 heat pipes were performed and its heat flow circuit was analysed from the experimentally obtained data on wall temperature distribution. Total resistance was ranged 0.02~$0.03^{\circ}C$/W for a power range from 40W to400W. The time to get the steady state was approximately longer than 20 minutes, and overshooting was not occurred during start up operation.

Analysis on the thermal performances of air-earth direct heat exchanger in one year (공기 지중간 직접열교환시스템의 연중 열성능 해석)

  • This study is focused on the development and selection of the optimal cool tube system to maximize its thermal performance. Cool tube is devised to reduce the heating and cooling load of building by preheating or refreshing of intake air with buried pipes. Finite volume method is adopted to solve the conduction problem between the cool tube and earth. We examine the cool tube system for two operating periods, a short term(12 hours) and a long term(3 months). The results of short term operations reveal that condensation significantly influences and raises the exit air temperature. For long term operations, optimum conditions of cool tube system are obtained with variations of flow-rate, depth, length and diameter of cool tube.

  • The mechanism of the aerosol(mist) generation generally consists of spin-off, splash, and evaporation/condensation. Host researchers showed some theoretical model for predicting the particulate size and generation rate without real cutting in turning operation. These models were based on the spin-off mechanism, and verified good for modeling the process. However, in real machining, the cutting tool destroys the flow direction of the cutting fluid and generate the heat by the relative motion of between tool and workpicee, and so the mass loading of the mist is greatly increased as compared with non-cutting. In this paper, we show some experimental data that the mist formation characteristics of cutting is different from that of non-cutting.

  • Condensation of water vapor into an absorbent liquid of LiBr-water solution falling over a bank of water cooled horizontal tubes was investigated theoretically. The governing conservation equation for a re-defined physical transport phenomena were solved numerically using a finite difference method. Raw parameters were used in this study, since reliable experimental data is required prior to a dimensionless parametric study. The average values of wall heat transfer coefficient and interfacial absorption rate were defined to see the system performance. Other parameters include tube diameter, streamwise coordinate (and number of tubes in row), mass flow rate, and the wall temperature. The effects of these quantities on the absorption processes and suggestions for a rational system design have been presented.

  • Cisplatin is a chemotherapeutic drug which is widely used for cancer therapy including cervical cancer. The purpose of this study is to elucidate synergistic effect of Cisplatin and Berberine on the apoptosis of HeLa cells and to determine whether oxidants are formed as part of apoptotic process. Apoptotic death of HeLa cells by cisplatin and berberine was confirmed by chromatin condensation of HeLa cells and flow cytometric analysis of intracellular ROS(reactive oxygen species) production. In MTT assay, Cell viability was decreased and enhanced ROS generation in combination of cisplatin and berberine significantly, as compared with cisplatin only. Synergistic effect of Cisplatin and Berberine on the inhibition of cell growth by apoptosis was clearly observed and ROS may play an important role in apoptosis. This effect suggest the possibility lowering the concentration of chemotherapeutic drugs, which alleviate the side effect of drugs.

Estimation of Heat Insulation and Light Transmission Performance According to Covering Methods of Plastic Greenhouses (플라스틱온실의 피복방식에 따른 보온 및 광투과 성능 평가)

  • The objective of the present study is to provide data needed to decide covering method to be able to increase the thermal insulation and light transmittance efficiency of commercial greenhouse. The thermal insulation effect, PPF transmittance and quantity of condensation water were estimated in experimental tomato greenhouses covered with three types of coverings of single layer, air inflated and conventional double layers covering. The overall heat flow of air inflated double layers greenhouse was similar to that of conventional double layers greenhouse, but the temperature between covering material and thermal screen in air inflated double layers greenhouse was lower than that in conventional double layers greenhouse at the same outside temperature condition due to air leakage through the gap of roof vent. The overall heat transfer coefficients acquired by experiment that was performed in single layer and conventional double layers greenhouses were close to those obtained from model experiment. Even though the PPF transmittance of air inflated double layers greenhouse was lower than that of single layer greenhouse, which was greater than that of conventional double layers greenhouse. The quantity of condensation water on covering surface of single layer greenhouse was greater than that of air inflated double layers greenhouse due to lower covering surface temperature.

A Study on Finned Tube Used in Turbo Refrigerator( I ) -for Condensation Hear Transfer- (터보 냉동기용 핀튜브에 관한 연구 ( I ) - 응축 열전달에 관하여 -)

  • Through the early 1900's, the evolution of the surface condenser was closely tied to the development of steam engine and the turbine. As the chemical and petroleum industries evolved in the 1900's, the use of surface condensers in many different processes. Today, industry uses condensers in many shapes and sizes. The actual condensation process occurs on the outside surface of tubes. The nature of this surface geometry affects the condenser's heat transfer performance. The first condensers were built with plain tubes. As tube manufacturing techniques advanced, manufacturers started making tubes with integral fins. In the 1940's, fin densities were limited to about 600 to 700 fins per meter(fpm) because of manufacturing procedure. Today new manufacturing techniques allow production of tubes with fin densities ranging from 750 to 1600 fpm. The integral-fin tubes investigated in this paper are nominally 19 mm diameter. Eight tubes have been used with trapezodially shaped integral-fins having fin density from 748 to 1654 fpm and 10, 30 grooves. For comparison, tests are made using a plain tube having the same inside diameter and an outside diameter equal to that at the root of the fins for the finned tubes. Betty and Katz's theoretical modelis is used to predict the R-11 condensation coefficient on horizontal integral-fin tubes having 748, 1024 and 1299 fpm. Experiments are carried out using R-11 as working fluid. The refrigerant condensates at a saturation state of $30^{\circ}C$ on the outside tube surface cooled by coolant. The amount of noncondensable gases present in the test loop is reduced to a negligible value by repeated purging. For a given heat input to the boiler and given cooling water flow rate, all test data are taken at steady state. The observed heat transfer enhancement for the finned and grooved tubes significantly exceeded that to be expected on grounds of increased area. For the eight fin tubes and one plain tube tested, the best performance has been obtained with a tube having a fin density of 1299 fpm, and a fin bight of 1.2mm and 30 grooves.

Numerical Analysis of Single Phase Thermal Stratification in both Cold Legs and Downcomer by Emergency Core Cooling System Injection : A Study on the Necessity to Consider Buoyancy Force Term (비상노심냉각계통 주입에 따른 저온관 및 강수관에서 단상 열성층 수치해석 : 부력항 고려 필요성에 관한 연구)

  • When emergency core cooling system (ECCS) is operated during loss of coolant accident (LOCA) in a pressurized water reactor (PWR), pressurized thermal shock (PTS) phenomenon can occur as cooling water is injected into a cold leg, mixed with hot primary coolant, and then entrained into a reactor vessel. Insufficient flow mixing may cause temperature stratification and steam condensation. In addition, flow vibration may cause thermal stresses in surrounding structures. This will reduce the life of the reactor vessel. Due to the importance of PTS phenomenon, in this study, calculation was performed for Test 1 among six types of OECD/NEA ROSA tests with ANSYS CFX R.17. Predicted results were then compared to measured data. Additionally, because temperature difference between the hot coolant at the inlet of the cold leg and the cold cooling water at the inlet of the ECCS injection line is 200 K or more, buoyancy force due to density difference might have significant effect on thermal-hydraulic characteristics of flow. Therefore, in this study, the necessity to include buoyancy force term in governing equations for accurate prediction of single phase thermal stratification in both cold legs and downcomer by ECCS injection was numerically studied.

  • Liquid film flows are classified into waveless laminar, wavy laminar, and turbulent flows depending on the Reynolds number or the flow stability. Since the wavy motions of the film flows are so intricate and nonlinear, studies on them have largely been experimental. Most numerical approaches have been limited to the waveless flow regime. The various free surface-tracking schemes adopted for this problem were used to more accurately estimate the average film thickness, rather than to capture the unsteady wavy motion. In this study, the wavy motions in laminar wavy liquid film flows with Reynolds numbers of 200-1000 were simulated with various numerical schemes based on the volume of fluid (VOF) method for interface tracking. The results from each numerical scheme were compared with the experimental results in terms of the average film thickness, the wave velocity, and the wave amplitude.
