cDNA Sequences for Asialoglycoprotein Receptor from Human Fetal Liver

  • The asialoglycoprotein receptor (ASGPR) was the first described mammalian lectin that mediates the specific binding and internalization of galactose/N-acetylgalactosamine-terminating glycoproteins by hepatic parenchymal cells. H1 and H2 are known as essential subunits of the functional ASGPR. There were close similarities in ASGPR H2 subunits between cultured cell line HepG2 and normal human liver cells including identical sequences at both termini. It was therefore expected that there may be some similarities between the subunits from normal liver cells and fetal liver cells. The two subunits of human fetal liver ASGPR. designated FL-H1 and FL-H2. were cloned from cDNA library by peR and the sequences were compared with the known HI and H2 sequences of HepG2, and the H1 sequence of nornal human liver cells. The results showed that FL-H1 was identical to H1 of HepG2. Whereas FL-H2 contains a 15-bp miniexon, but missing 57-bp at the near upstream from the membrane-spanning domain compared to H2 of HepG2 and normal human liver cells indicating that FL-H2 resulted from a differential splicing compared to HepG2 and normal liver cells.

  • A FRET-based high-throughput screening system was developed for the discovery of competitive smallmolecule Hsp90 inhibitors. The biarsenical fluorescein derivative FlAsH and dabcyl-conjugated Hsp90 inhibitor GM were employed as the FRET donor and quencher, respectively. The spatial proximity perturbation between FlAsH-labeled Hsp90N and GM-dabcyl upon treatment of a small molecule led to changes in the FRET-induced fluorescence, monitored in a high-throughput fashion.

  • To improve the performance of the anode-supported Solid Oxide Fuel Cell (SOFC) which can be operated at an intermediate temperature, the functional layer (FL) is introduced on a anode substrate. And the scandia-stabilized zirconia (ScSZ) and samaria-doped ceria (SDC) which have higher ionic conductivity and better chemical stability than yttria-stabilized zirconia (YSZ) are used as material for the anode FL with the Ni, The fabrication process of anode-supported single cell with the anode FL was established and the power density of those was evaluated. As a result, the sample with anode FL (Ni-YSZ) has higher power density than normal cell. The single cell which was composed of the FL (Ni-YSZ) and electrolyte (YSZ) showed about $550mW/cm^2$ of the maximum power density at $650^{\circ}C$ and $1430mW/cm^2$ at $750^{\circ}C$ respectively, In case of the single cell using the ScSZ and SDC as anode FL, the performance of samples decreased rapidly and those showed unstable voltage during long-term test. In case of using methane as a fuel, the cell performance with each FL decreased comparing with $H_2$ fuel. In the region of a high current density, there are large concentration polarizations.

Growth and Flowering Responses of Petunia to Various Artificial Light Sources with Different Light Qualities

  • This study was carried out to investigate the effect of artificial light sources with different light qualities on the growth and flowering characteristics of a herbaceous long-day plant, Petunia ${\times}$ hybrida Hort. Seedlings of petunia cultivar 'Madness Rose' were potted, acclimated for one week, and grown in a phytotron equipped with tube- and bulb-type fluorescent lamps (FL tube and bulb), tube-type white light-emitting diodes (LED tube), halogen lamps (HL), metal halide lamps (MH), and high pressure sodium lamps (HPS) for 10 weeks. The temperature, photoperiod, and photosynthetic photon flux density (PPFD) in the phytotron were $22{\pm}2^{\circ}C$, 16 h, and $25{\pm}2{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, respectively. Light sources combined with HL promoted stem elongation, and plant height and internode length decreased with increasing red to far-red (R:FR) ratio. FL tube + LED tube, HPS, and FL tube promoted branching, whereas plants grown under light sources combined with HL did not have any branches. Days to flowering (from longest to shortest) occurred as follows: FL tube + HL > FL tube + HL > MH > HPS = FL tube + FL bulb > FL tube + LED tube > LED tube > FL tube, indicating that reducing the R:FR ratio of the light sources promoted flowering. Only 20% of plants grown under an FL tube flowered, whereas under all other treatments, 100% of plants flowered. At 10 weeks after treatment, plants grown under HPS and MH had (cumulatively) 12 open flowers, and those grown under FL tube + FL bulb, LED tube, FL tube + LED tube, and HPS treatment had approximately seven flower buds. These results suggest that light sources with low R:FR ratios promote flowering and stem elongation in petunia, but they reduce its ornamental value due to overgrowth and poor branching.

Effects of Flos Lonicerae Japonicae Water Extract on Cytokine Production in RAW 264.7 Mouse Macrophages (금은화(金銀花)물추출물이 마우스 대식세포의 사이토카인 생성에 미치는 영향)

  • Flos Lonicerae Japonicae (the flower buds of Lonicera japonica Thunberg) has been used as an antibacterial and antiviral drug in Korean Medicine. The aim of this study is to evaluate the effect of Flos Lonicerae Japonicae water extract (FL) on the production of cytokines in RAW 264.7 mouse macrophages stimulated by lipopolysaccharide (LPS). After 24 h treatment, the production of various cytokines from RAW 264.7 was measured with multiplex cytokine assay using Bio-Plex 200 suspension array system. FL at concentrations of 50, 100, and 200 ㎍/mL significantly inhibited productions of tumor necrosis factor-α, macrophage inflammatory protein (MIP)-1β, and MIP-2 in LPS-stimulated RAW 264.7 cells; FL at concentrations of 100 and 200 ㎍/mL significantly inhibited productions of leukemia inhibitory factor, LIX (CXCL5), and RANTES in LPS-stimulated RAW 264.7 cells; FL at concentrations of 200 ㎍/mL significantly inhibited productions of granulocyte-macrophage colony-stimulating factor and macrophage colony-stimulating factor in LPS-stimulated RAW 264.7 cells; FL at concentrations of 50 and 100 ㎍/mL significantly increased productions of interleukin (IL)-10 in LPS-stimulated RAW 264.7 cells; FL at concentrations of 50, 100, and 200 ㎍/mL significantly increased productions of IL-6 and interferon gamma-induced protein-10 in LPS-stimulated RAW 264.7 cells; FL at concentrations of 100 and 200 ㎍/mL significantly increased productions of monocyte chemoattractant protein-1 in LPS-stimulated RAW 264.7 cells. Taken together, these data mean that FL might modulate productions of cytokines, chemokines, and growth factor in LPS-stimulated macrophages. Further study needs to verify the exact mechanism for modulatory activities of FL with macrophages.

Elution Behavior of Protein and Pullulan in Asymmetrical Flow Field-flow Fractionation (AsFlFFF)

  • An AsFlFFF channel was designed and built, and then tested for analysis of pullulans and proteins. Pullulans and proteins having various nominal molecular weights were injected at various conditions of the cross-flow rate ($F_c$) and the channel-out flow rate ($F_{out}$). The retention (measured by the retention ratio R) and the zone broadening (measured by the plate height H) were measured, and then compared with theory. When the incoming flow rate, $f_{in}$ (and thus $F_{out}$) was varied with $F_c$ fixed at 2.5 mL/min, the plate height measured for the pullulan with nominal molecular weight (M) of about 100,000 showed the trend expected by the longitudinal diffusion theory (H decreases with increasing flow rate). In contrast, when $F_{out}$ was varied with the flow rate ratio, $F_c/F_{out}$, fixed constant at 5, the plate height measured for the same sample showed the trend expected from the non-equilibrium theory (H increases with increasing flow rate). Calibration plots (log D vs. log M) obtained with pullulans and proteins were not coincide, probably due to the difference in molecular conformation, suggesting the analysis of pullulans and proteins using AsFlFFF requires independent calibration. It was found that the linearity of the protein-calibration plot was improved by using a buffer solution as the carrier.

Mean Platelet Volume Could be a Possible Biomarker for Papillary Thyroid Carcinomas

  • Thyroid cancer is the most prevalent endocrine cancer and is evident in nearly 5% of thyroid nodules. The correlation between mean platelet volume (MPV) and many other cancer types has been investigated previously. However, the correlation between papillary thyroid carcinoma (PTC) and MPV has not yet been studied in detail. The aim of this study was to examine whether MPV would be a useful inflammatory marker to differentiate PTC patients from cases of benign goiter and healthy controls. Preoperative MPV levels in patients with PTC were found to be significantly higher when compared with benign goiter patients and healthy controls ((respectively, 8.05 femtoliter (fl), 7.57 fl, 7.36 fl, p=0.001). After surgical treatment of PTC patients, a significant decrease in MPV levels was seen (8.05 fl versus 7.60 fl, p=0.005). ROC analysis suggested 7.81 as the cut-off value for MPV (AUC=0.729, sensitivity 60%, specificity 80%). In conclusion, maybe changes in MPV levels can be used as an easily available biomarker for monitoring the risk of PTC in patients with thyroid nodules, enabling early diagnosis of PTC.

Hybrid fuzzy model to predict strength and optimum compositions of natural Alumina-Silica-based geopolymers

  • This study introduces the supervised committee fuzzy model as a hybrid fuzzy model to predict compressive strength (CS) of geopolymers prepared from alumina-silica products. For this purpose, more than 50 experimental data that evaluated the effect of $Al_2O_3/SiO_2$, $Na_2O/Al_2O_3$, $Na_2O/H_2O$ and Na/[Na+K] on (CS) of geopolymers were collected from the literature. Then, three different Fuzzy Logic (FL) models (Sugeno fuzzy logic (SFL), Mamdani fuzzy logic (MFL), and Larsen fuzzy logic (LFL)) were adopted to overcome the inherent uncertainty of geochemical parameters and to predict CS. After validating the model, it was found that the SFL model is superior to MFL and LFL models, but each of the FL models has advantages to predict CS. Therefore, to achieve the optimal performance, the supervised committee fuzzy logic (SCFL) model was developed as a hybrid method to combine the benefits of individual FL models. The SCFL employs an artificial neural network (ANN) model to re-predict the CS of three FL model predictions. The results also show significant fitting improvement in comparison with individual FL models.

Correlations between Gastroscopy Results and Zone-5 Data from the 7-zone-Diagnostic System (7구역진단기의 5구역 정보와 위내시경 소견의 상관성에 관한 연구)

  • Objectives : The 7-zone-diagnostic system is a device for predetermining bodily locations by measuring the energy of the living body. The purpose of this study is to examine correlations between gastroscopy results and zone-5 data from the 7-zone-diagnostic system. Methods : This study was carried out with data from gastroscopy procedures. It involved 115 patients who had been diagnosed with gastritis, esophagitis, or gastric ulcers. These patients were divided into three groups according to the different patterns of zone-5 factors AA, FL2 and FL1. Group A was made up of patients for whom the red bar graph of zone-5 was higher than the normal range for factor AA. Group B was made up of patients for whom the red bar graph of zone-5 was within the normal range for factor AA. Group C was made up of patients for whom the red bar graph of zone-5 was lower than the normal range for factor AA. Group D E and F were made up of same way as group A B and C for factor FL2. Group G H and I were made up of same way as group A B and C for factor FL1. After the collection of gastroscopy results and data on zone-5 from the 7-zone-diagnostic system, the data was analyzed statistically. Results : 1. Group D had a higher ratio of medication than group E, and this result was statistically significant. 2. Group D G had a higher ratio of medication than group E G and group E H, and this result was statistically significant. 3. Group G had a higher ratio of esophagitis than group H, and this result was statistically significant. 4. A Group B H had a higher ratio of esophagitis than group B G, group C G, and group C H, and this result was statistically significant. 5. The medication group had a high score on the function index than the non-medication group, and this result was statistically significant. Conclusions : This study suggests that there is a slight correlation between gastroscopy results and zone-5 data from the 7-zone-diagnostic system. The research resulted in significant data that are helpful for diagnosing digestive system problems through the use of the 7-zone-diagnostic system.

Disulfide Bond Bridged Divalent Antibody-Toxin, $(Fab-PE38fl)_2$ with the Toxin PE38 Fused to the Light Chain

  • B3 antibody specifically binds the $Lewis^Y$-related carbohydrate antigen of many carcinomas, and it is used as a model antibody in this study. In a previous study, the Fab fragment of the antibody was fused to a 38 kDa truncated form of Pseudomonas exotoxin A, PE38, to make Fab-PE38, where PE38 is fused to the Fd fragment of the Fab domain. This parent monomer molecule, Fab-PE38, had no cysteine in the hinge region, and it could not make a disulfide bond to form a disulfide bond bridged homodimer. In this study, we constructed three different kinds of divalent Fab-toxin fusion homodimers where the toxin is fused to the light chain of Fab, $(Fab-PE38fl)_2$. In addition to the PE38 toxin fused to the light chain, these three molecules have different hinge sequences hi, h2, and h3 making Fabh1-, Fabh2-, and Fabh3-PE38fl monomers, respectively. These hinges contain only one cysteine on different positions of the hinge sequence. The disulfide bond between the hinge region of two monomers forms homodimers $(Fabh1-PE38fl)_2$, $(Fabh2-PE38fl)_2$, and $(Fabh3-PE38fl)_2$. The refolding yields of these dimers were 5-16-fold higher than a previously constructed dimer where the PE38 was fused to the Fd fragment $(Fabh2-PE38)_2$ [8]. Our data suggest that the steric repulsion between the two PE38s in $(Fabh1-PE38)_2$ during disulfide bridge formation is relieved by fusing it at the end of the light chain. The best cytotoxicity value of these dimers showed about 2.5-fold higher on an MCF7 cell line than that of the monovalent reference molecule in ng/ml scale, which is 15-fold higher in pM scale.
