Comparison of Experimental and Radiation Therapy Planning (RTP) Dose Distributions on Air Cavity (공동(air cavity)의 존재 시 실험적 선량분포와 치료계획상의 선량분포 비교)

  • This study is compared that the dose distribution by experimentation and radiation therapy planning (RTP) when the air cavity region was treated high energy photon. The dose measurements were performed with a 6 MV photon beam of linear accelerator. The polystyrene and self made acyl phantom were similar to tissue density of the human body. A parallel plate chamber was connected to an electrometer. The measurement setup was SCD (Source Chamber Distance) 100 cm and the distance of surface from air cavity was 3 cm. Absorbed dose of interface were measured by area and height. The percent depth dose were measured presence and absence of air cavity, depth according to a ratio of field size and air cavity size. The dose distribution on planning was expressed to do the inhomogeneity correction. As the area of air cavity was increased, the absorbed dose were gradually reduced. It was slightly increased, when the height of air cavity was changed from 0 cm to 0.5 cm. After the point, dose was decreased. In case of presence of air cavity, dose after distal air cavity interface was more great than absence of air cavity. The rebuild up by field size and area of air cavity occurred for field size, $4{\times}4\;cm^2$, $5{\times}5\;cm^2$ and $6{\times}6\;cm^2$, with fixed on area of air cavity, $5{\times}5\;cm^2$. But it didn't occur at $10{\times}10\;cm^2$ field size. On the contrary, the field size was fixed on $5{\times}5\;cm^2$, rebuild up occurred in area of air cavity, $4{\times}4\;cm^2$, $5{\times}5\;cm^2$. but, it did not occur for air cavity, $2{\times}2\;cm^2$, $3{\times}3\;cm^2$. All of the radiation therapy planning were not occurred rebuild up. It was required to pay attention to treat tumor in air cavity because the dose distribution of planning was different from the dose distribution of patient.

  • The distribution of the secretory ducts, fine structures of the secretory epithelial cells, and the ingredients of the metaplasmic inclusions were studied at light and electron microscopical levels in seeds, stems, leaves, and roots of ginseng. The secretory ducts occurred in the hypocotyl of the embryo, in the cortex of the roots, and also both inside and outside of each vascular bundle in the stems and leaves. Especially, it is considered that the circular layers of the secretory ducts in roots may represent their ages. The epithelial cell has well developed nucleolus, mitochondria and smooth endoplasmic reticulum. Sudanophyl and osmiophilic inclusions were found in the epithelial cytoplasm and duct lumen. But these inclusions were not observed when extracted with pyridin or alcohol. In contrast to the lumen with red color, the epithelial cells were blue in color as stained with nile blue, suggesting that the former inclusions are neutral lipid while the latter are acidic lipid. The electron density of the cell inclusions was quite high as fixed with osmium tetroxide, indicating that most of these secretory materials seem to be unsaturated lipid. Therefore, since ginseng secretory ducts are closely associated with the lipid metabolism, it should be called lipid canal or lipid duct.

Methods for Coating the Killed Whole Cell Antigens of Salmonella typhi in Enzyme-linked Immunosorbent Assay (효소면역측정법을 위한 장티푸스 균체항원의 부착방법)

  • The advantages of enzyme-linked immunosorbent assay(ELISA) are its senstivity and simplicity in detecting IgG, IgM and IgA antibody. To apply ELISA to diagnosis of typhoid fever, antigen such as lipopolysaccharide of Salmonella typhi or killed whole cell must be coated on solid phase. It is easy to coat lipopolysaccharide on ELISA plate but troublesome to purify it. As it is easy to obtain the killed whole cells, the development of the appropriate method by which those antigens of S. typhi are optimally coated on solid phase is needed. To establish the appropriate method, carbonate buffer, methanol or poly-L-lysine was applied as binding substance on polystyrene or polyvinylchloride plate as solid phase when the killed whole cell antigens of S. typhi varided as follows: $10^6$, $10^7$, $10^8$ and $10^9\;cell/ml$. The criteria of the optimal method were determined as follows: 1. The optical density of positive sera is above 1.0(0.6 in IgM) at 1:10 serum dilution and is 0.3(0.2 in IgM) higher than that of negative sera: 2. The O.D. of sera is flat or lowering according to serum dilution: 3. It must be that the O.D. of negative sera is lower than 0.2 at the point of serum dilution where the O.D. of positive sera is higher than 1.0(0.5 in IgM). The results obtained were summarized as follows: 1. The methods which fitted the above criteria were to use poly-L-lysine as binding substance, polyvinylchloride plate as solid phase and $10^7\;cell/ml$ as antigen concentration of S. typhi(poly-L-lysine/polyvinylchloride/$10^7$) and poly-L-lysine/polyvinylchloride/$10^8$ in detecting IgG antibody, methanol/polystyrene/$10^9$, poly-L-lysine/polyvinylchloride/$10^8$ and poly-L-lysine/polyvinylchloride/$10^9$ in IgM and carbonate buffer/polystyrene/$10^8$, carbonate buffer/polystyrene/$10^9$, methanol/polystyrene/$10^8$, methanol/polyvinylchloride/$10^8$, methanol/polyvinylchloride/$10^9$, poly-L-lysine/polyvinylchloride/$10^8$ and poly-L-lysine/polyvinylchloride/$10^9$ in IgA. 2. The coaling method using poly-L-lysine, polyvinylchloride plate and $10^8\;cell/ml$ was best to assay IgG, IgM and IgA antibody all in one. By this method, to assay the each immunoglobulin calss with an appropriate fixed serum dilution, 1:320 dilution was best.

Genome-wide association study of carcass weight in commercial Hanwoo cattle

  • Objective: The objective of the present study was to validate genes and genomic regions associated with carcass weight using a low-density single nucleotide polymorphism (SNP) Chip in Hanwoo cattle breed. Methods: Commercial Hanwoo steers (n = 220) were genotyped with 20K GeneSeek genomic profiler BeadChip. After applying the quality control of criteria of a call rate ${\geq}90%$ and minor allele frequency (MAF) ${\geq}0.01$, a total of 15,235 autosomal SNPs were left for genome-wide association (GWA) analysis. The GWA tests were performed using single-locus mixed linear model. Age at slaughter was fitted as fixed effect and sire included as a covariate. The level of genome-wide significance was set at $3.28{\times}10^{-6}$ (0.05/15,235), corresponding to Bonferroni correction for 15,235 multiple independent tests. Results: By employing EMMAX approach which is based on a mixed linear model and accounts for population stratification and relatedness, we identified 17 and 16 loci significantly (p<0.001) associated with carcass weight for the additive and dominant models, respectively. The second most significant (p = 0.000049) SNP (ARS-BFGL-NGS-28234) on bovine chromosome 4 (BTA4) at 21 Mb had an allele substitution effect of 43.45 kg. Some of the identified regions on BTA2, 6, 14, 22, and 24 were previously reported to be associated with quantitative trait loci for carcass weight in several beef cattle breeds. Conclusion: This is the first genome-wide association study using SNP chips on commercial Hanwoo steers, and some of the loci newly identified in this study may help to better DNA markers that determine increased beef production in commercial Hanwoo cattle. Further studies using a larger sample size will allow confirmation of the candidates identified in this study.

FOUR-YEAR SURVIVAL RATE OF RBM SURFACE INTERNAL CONNECTION NON-SUBMERGED IMPLANTS AND THE CHANGE OF THE PERI-IMPLANT CRESTAL BONE (RBM 표면처리 내부연결형 비매립 임플란트의 4년 생존율과 주변골 흡수에 관한 임상 및 방사선학적 연구)

  • Implant-supported fixed and removable prostheses provide a proper treatment modality with reliable success. The SS $II^{(R)}$ Implants is a one-stage nonsubmerged threaded titanium implants with Resorbable Blasting Media (RBM) surface developed by Osstem company (Busan, Korea) in October of 2002. This study is to evaluate the survival rate of the SS $II^{(R)}$ Implants for 4 years using radiographic parameters and to review the retrieved implants by the cytotoxicity tests. Since September 2003, 439 SS $II^{(R)}$ implants had been used for 173 patients at Ewha Womans University Medical Center in Korea. Patients consisted of 91 females (52.6 %) and 82 males (47.4 %). The patients' mean age was $42\;{\pm}16$ years, ranging from 21 to 83 years. The follow-up period ranged from 9 to 46 months (mean F/U $24.2\;{\pm}\;10.2$ months). The results are as follows; 1. Of 439 implants, 17 implants were removed and 4-year cumulative survival rate was 96.1%. 2. 82.3% of 17 failed implants were founded during healing phase, and 94.1% of failed fixtures were removed within 5 months after implantation. 3. Crestal bone around the implants was resorbed to 1 mm in 89.0%, to 1 - 2 mm loss of the marginal bone in 8.3%, and the bone loss over 2 mm was occurred in 2.7%. 4. Microscopic examination of the retrieved implants disclosed Grade 0 cytotoxicity in 4 and Grade 1 cytotoxicity in 2 of 6 groups divided according to LOT numbers. Inhibition rate with optical density was acceptable as low as ISO standard.

A Study on the etching mechanism of $CeO_2$ thin film by high density plasma (고밀도 플라즈마에 의한 $CeO_2$ 박막의 식각 메커니즘 연구)

  • Cerium oxide ($CeO_2$) thin film has been proposed as a buffer layer between the ferroelectric thin film and the Si substrate in Metal-Ferroelectric-Insulator-Silicon (MFIS) structures for ferroelectric random access memory (FRAM) applications. In this study, $CeO_2$ thin films were etched with $Cl_2$/Ar gas mixture in an inductively coupled plasma (ICP). Etch properties were measured for different gas mixing ratio of $Cl_2$($Cl_2$+Ar) while the other process conditions were fixed at RF power (600 W), dc bias voltage (-200 V), and chamber pressure (15 mTorr). The highest etch rate of $CeO_2$ thin film was 230 ${\AA}$/min and the selectivity of $CeO_2$ to $YMnO_3$ was 1.83 at $Cl_2$($Cl_2$+Ar gas mixing ratio of 0.2. The surface reaction of the etched $CeO_2$ thin films was investigated using x-ray photoelectron spectroscopy (XPS) analysis. There is a Ce-Cl bonding by chemical reaction between Ce and Cl. The results of secondary ion mass spectrometer (SIMS) analysis were compared with the results of XPS analysis and the Ce-Cl bonding was monitored at 176.15 (a.m.u). These results confirm that Ce atoms of $CeO_2$ thin films react with chlorine and a compound such as CeCl remains on the surface of etched $CeO_2$ thin films. These products can be removed by Ar ion bombardment.

EFFECTS OF CARCINOGENICITY AND GROWTH RAGULATORY FACTORS IN HUMAN EPITHELIAL CELLS EXPOSED WITH TOBACCO-SPECIFIC N-NITROSAMINE (흡연특이성 N-Nitrosamine이 인체상피세포의 발암화와 성장조절인자에 미치는 영향)

  • Since NNK is one of the most abundant tobacco-specific alkaloids and a strong carcinogenic nitrosamine, it has been used for evaluating a potential of carcinogenicity in the animal models. The present study has attempted to examine the potential of carcinogenicity of NNK in human epithelial cells, from which the cell type the most of cancers including oral cancer and nasal cavity cancer are originated. The cellular model used for the study is a human keratinocyte cell system immortalized by Ad12-SV40 hybrid virus. The cellular system has successfully been used for the carcinogenicity studies because of its limitless life span, epithelial morphology and nontumorigenicity. When cells were treated with a variety of NNK concentrations, levels of saturation density and soft agar colony formation were increased in a dose-dependent fashion. Colonies of large cell aggregates were above 5 at the higher doses. The results indicate that exposure of human cells with NNK induced loss of contact inhibition and increases of anchorage independence and cellular adhesion, which are typical characteristics of the neoplatically transformed cells. When cells were exposed with 100uM NNK for 2hr, mRNA levels of IL-1 and PAI-2 were increased in a dose-dependent manner, but expression of TGF- 1 was not affected. While expression of growth regulatory factors were altered with a short-term exposure, there was no alteration of these factors in the NNK-transformed cells. However, mRNA levels of fibronectin were increased both in the short-term treatment and in the transformation. The results suggest that altered expression of extracellular matrix such as fibronectin following short-term exposure might be fixed in the genome and these altered properties be continuously transfered throughout the cell division. Western blot analysis showed a translocation of PKC- from cytosolic fraction to the particulate fraction, indicating a possible role of NNK in the signal transduction pathway. The present study provided an evidence that NNK in the smoking may be associated with epithelial origin cancer such as oral and nasal cavity cancers. In addition, this study suggested that altered expression of extracellular matrix and PKC may play an important role in the carcinogenic mechanism of NNK.

  • AlN thin films were fabricated by reactive sputtering for the application of MIS devices with Al/AlN/Si structure. It has investigated the surface morphology change, I-V characteristics, C-V characteristics, and chemical composition of AlN films with the intriducing time of hydrogen on the fixed deposition condition(RF power: 150W, sputtering pressure: 5mTorr, flow rate ratio of $Ar/N_2=1$, hydrogen concentration: 5%). By addition of the hydrogen the deposition rate decreased drastically whereas the surface morphology changed little. It has been found from the analysis of I-V and C-V characteristics curves that the films deposited with hydrogen addition in initial stage had lower leakage current density, lower flat band voltage and hystersis profile when compared with those with hydrogen addition in last stage. The oxygen concentration in AlN films decreased with addition of hydrogen gas, which suggesting a profitable role in the insulation and C-V characteristics of AlN films.

Recommendation of Nitrogen Topdressing Rates at Panicle Initiation Stage of Rice Using Canopy Reflectance

  • The response of grain yield(GY) and milled-rice protein content(PC) to crop growth status and nitrogen(N) rates at panicle initiation stage(PIS) is critical information for prescribing topdress N rate at PIS(Npi) for target GY and PC. Three split-split-plot experiments including various N treatments and rice cultivars were conducted in Experimental Farm, Seoul National University, Korea in 2003-2005. Shoot N density(SND, g N in shoot $m^{-2}$) and canopy reflectance were measured before N application at PIS, and GY, PC, and SND were measured at harvest. Data from the first two years(2003-2004) were used for calibrating the predictive models for GY, PC, and SND accumulated from PIS to harvest using SND at PIS and Npi by multiple stepwise regression. After that the calibrated models were used for calculating N requirement at PIS for each of nine plots based on the target PC of 6.8% and the values of SND at PIS that was estimated by canopy reflectance method in the 2005 experiment. The result showed that SND at PIS in combination with Npi were successful to predict GY, PC, and SND from PIS to harvest in the calibration dataset with the coefficients of determination ($R^2$) of 0.87, 0.73, and 0.82 and the relative errors in prediction(REP, %) of 5.5, 4.3, and 21.1%, respectively. In general, the calibrated model equations showed a little lower performance in calculating GY, PC, and SND in the validation dataset(data from 2005) but REP ranging from 3.3% for PC and 13.9% for SND accumulated from PIS to harvest was acceptable. Nitrogen rate prescription treatment(PRT) for the target PC of 6.8% reduced the coefficient of variation in PC from 4.6% in the fixed rate treatment(FRT, 3.6g N $m^{-2}$) to 2.4% in PRT and the average PC of PRT was 6.78%, being very close to the target PC of 6.8%. In addition, PRT increased GY by 42.1 $gm^{-2}$ while Npi increased by 0.63 $gm^{-2}$ compared to the FRT, resulting in high agronomic N-use efficiency of 68.8 kg grain from additional kg N. The high agronomic N-use efficiency might have resulted from the higher response of grain yield to the applied N in the prescribed N rate treatment because N rate was prescribed based on the crop growth and N status of each plot.

An Adaptive Contention Windows Adjustment Scheme Based on the Access Category for OnBord-Unit in IEEE 802.11p (IEEE 802.11p에서 차량단말기간에 혼잡상황 해결을 위한 동적 충돌 윈도우 향상 기법)

  • The study aims at offering a solution to the problems of transmission delay and data throughput decrease as the number of contending On-Board Units (OBU) increases by applying CSMA medium access control protocol based upon IEEE 802.11p. In a competition-based medium, contention probability becomes high as OBU increases. In order to improve the performance of this medium access layer, the author proposes EDCA which a adaptive adjustment of the Contention Windows (CW) considering traffic density and data type. EDCA applies fixed values of Minimum Contention Window (CWmin) and Maximum Contention Window (CWmax) for each of four kinds of Access Categories (AC) for channel-specific service differentiation. EDCA does not guarantee the channel-specific features and network state whereas it guarantees inter-AC differentiation by classifying into traffic features. Thus it is not possible to actively respond to a contention caused by network congestion occurring in a short moment in channel. As a solution, CWminAS(CWmin Adaptation Scheme) and ACATICT(Adaptive Contention window Adjustment Technique based on Individual Class Traffic) are proposed as active CW control techniques. In previous researches, the contention probabilities for each value of AC were not examined or a single channel based AC value was considered. And the channel-specific demands of IEEE 802.11p and the corresponding contention probabilities were not reflected in the studies. The study considers the collision number of a previous service section and the current network congestion proposes a dynamic control technique ACCW(Adaptive Control of Contention windows in considering the WAVE situation) for CW of the next channel.
