Telomeres Distribution and Telomerase Activity During Chick Embryonic and Developmental Stages

  • Telomeres are the end of chromosomes and consist of a tandem repeat sequence of (TTAGGG)n and associated proteins. Telomerase is a ribonucleoprotein which act as a template for the synthesis of telomeric DNA. Telomeres are essential for chromosome stability and are related with cell senescence, apoptosis and cancer. Even though telomeres and telomerase have been studied extensively, very little is known about telomere dynamics in embryonic cells. This study was carried out to analyze the telomeres distribution and telomerase activity of chicken cells during embryonic and developmental stages. The target cells for analysing were sperms, ovulated ova, early embryonic cells and the cells from brain, heart, liver, kidney and germinal tissue in fetus. Telomeres distribution on target cells was analyzed by Q-FISH (Quantitation-Fluorescence in situ Hybridization) techniques using a chicken telomere repeat probe. Telomerase activity was performed by TRAP assay (Telomeric repeat Amplification Protocol) with target DNA. In results, the telomeres of chicken were found at the ends of all chromosomes. In addition, chicken had interstitial telomeres on chromosomes 1, 2 and 3. Telomerase activity was highly detectable in early embryonic cells, germinal tissues and kidney cells. Whereas telomerase activity was gradually down-regulated when the organs, including brain, heart, and liver, were developed from embryos. In the distribution of telomeric DNA on the embryonic and developmental stages, most of the cells was gradually decreased in telomere quantity during ontogenesis.

Molecular Cloning, Characterization and Expression Analysis of an ILF2 Homologue from Tetraodon nigroviridis

  • Interleukin-2 enhancer binding factor 2 (ILF2) was reported to regulate transcription of interleukin-2 (IL-2), a central cytokine in the regulation of T-cell responses. This property of ILF2 was well characterized in human and mammals, but little is known in bony fish. In this paper, an ILF2 homologue was cloned and well characterized from Tetraodon nigrovirid is for the further investigation of the function of ILF2 in bony fish. The full-length Tetraodon ILF2 cDNA was 1380 bp in size and contained an open reading frame (ORF) of 1164 bp that translates into a 387 amino-acid peptide with a molecular weight of 42.9 kDa, a 5' untranslated region (UTR) of 57 bp, and a 3' UTR of 159 bp containing a poly A tail. The deduced peptide of Tetraodon ILF2 shared an overall identity of 58%~93% with other known ILF2 sequences, and contained two N-glycosylation sites, two N-myristoylation sites, one RGD cell attachment sequence, six protein kinase C phosphorylation sites, one amino-terminal RGG-rich single-stranded RNA-binding domain, and a DZF zinc-finger nucleic acid binding domain, most of which were highly conserved through species compared. Constitutive expression of Tetraodon ILF2 was observed in all tissues examined, including gill, gut, head kidney, spleen, liver, brain and heart. The highest expression was detected in heart, followed by liver, head kidney and brain. Stimulation with LPS did not significantly alter the expression of Tetraodon ILF2. Gene organization analysis showed that the Tetraodon ILF2 gene have fifteen exons, one more than other known ILF2 genes in human and mouse. Genes up- and down-stream from the Tetraodon ILF2 were Rpa12, Peroxin-11b, Smad4, Snapap and Txnip homologue, which were different from that in human and mouse.

  • To keep relics excavated in the era of the Japanese Colonist's rule in the Korean Peninsula in order, the National Museum of Korea selected some of horse strap pendants (Haengyeob) that were excavated from Seobongchong, Sikrichong and Geumryeongchong, and it investigated conservation and production technique. The horse strap pendants selected were classified into three, that is to say, Oval and fish-tail shape, heart shape and atypical: At the results of microscopic observation, XRF and SEM-EDS analysis, base plate was made of iron, and top plate as well as pattern plate was made of iron, copper and silver. Both Oval and fish-tail shape and heart shape horse strap pendants had horse strap pendants having same production technique and sizes at all of three ancient tombs were made of iron and copper. The gold layer of horse strap pendants except the atypical one having no gold layer were made by amalgam gilding.

Egg Development and Larvae and Juveniles Morphology of Carp, Cyprinus carpio in Korean

  • This study was conducted to observe egg and larvae morphological development of carp to obtain basic data for resource conservation and taxonomic research. Brood carp used in the research (total length 67.3-75.5 cm, average $71.0{\pm}3.45cm$) were bred in a circular rearing aquarium ($600{\times}300{\times}100cm$) using a running water system from January to July, 2015. Breeding water temperature was maintained at $23.0-25.0^{\circ}C$(average $24.0^{\circ}C$). Fertilized carp eggs were translucent and globular, and their size was 1.75-1.89 mm (average $1.82{\pm}0.06mm$). Blastoderms formed 10 min after fertilization and reached the two-cell stage 30 min after fertilization. Then, the embryo turned dark and exhibited melanophores, and blood started flowing from the heart across the egg yolk at 42 hrs and 50 min after fertilization. Hatching began 70 hrs and 26 min after fertilization larvae emerged through the egg membrane, starting from the head. The length of prelarvae immediately after hatching was 5.23-5.38 mm (average $5.31{\pm}0.11mm$) the mouth and anus were closed, and the pectoral fin was formed. Postlarvae at 18 days after hatching had a total length of 11.9-13.9 mm (average $12.9{\pm}1.40mm$), separate anal fin and back membranes, and fin ray. Juveniles fish at 35 days after hatching had a total length of 29.9-30.2 mm (average $30.1{\pm}0.13mm$), with the body covered with scales, and the same number of fin rays, color, and shape as their broodstork.

Surgical Outcome of Achilles Reconstruction Using Allotendon and a Calcaneal Tunneling Technique in Patients with Chronic Achilles Rupture (동종건과 종골 터널을 이용한 만성 아킬레스건 파열 환자의 아킬레스건 재건술의 수술적 결과)

  • Purpose: Achilles tendon rupture is a frequently encountered ankle pathology associated with a substantial burden of intense pain and functional deficits. Chronic Achilles tendon ruptures with considerable defects pose intricate repair challenges that are often marred by complications such as re-rupture and persistent pain. Various treatment methods, including allograft transplantation, have been proposed, but the literature on this technique is limited. In this study, we propose a surgical approach utilizing allotendon transplantation and a calcaneal tunneling technique and provide clinical evaluation details. Materials and Methods: Fifteen patients with chronic Achilles tendon ruptures treated with allotendon between 2020 and 2022 were included in the study. Patients were evaluated at 1, 3, and 6 months postoperatively using Visual Analog Scale (VAS) scores and Achilles Tendon Total Rupture Scores (ATRSs). Complications were assessed postoperatively. Results: The average VAS score was 7 before surgery, 7.3 immediately after surgery, and 4.3 at 1 month, 2.5 at 3 months, and 1.3 at 6 months after surgery. Because the sample was limited to 15 individuals and distributions were non-normal, the analysis was conducted using the non-parametric Wilcoxon's signed-rank test, and statistical significance was accepted for p-values<0.05. Results showed a significant improvement in ATRS and VAS scores versus preoperative and immediate postoperative values. VAS scores showed a decreasing trend after surgery, whereas average ATRS scores increased from 14 before surgery, 33.8 at 1 month, 82.7 at 3 months, and 93.9 at 6 months. Conclusion: This study suggests that allograft transplantation using the described calcaneal tunnel technique provides an effective treatment for chronic Achilles tendon ruptures. However, extensive research and long-term clinical trials are required to validate and better understand the technique's efficacy.

The Study of Biochemical Changes Induced by Fish Oil Diet in Rat ( II ) - Changes in Lipoprotein Lipase Activity and Mitochondrial Respiration and Structure - (어유(魚油)식이에 의한 흰쥐체내의 생화학적 변화연구(II) - Lipoprotein Lipase 활성과 미토콘드리아 호흡계의 변화 -)

  • The effect of dietary fish oil ( mackerel oil : MO, eel oil : EO) on energy utilization in rats was studied with measurements of various tissue lipoprotein lipase( LPL ) and live and heart mitochondrial respiration. Fatty acid composition of mitrochondrial inner membrane matrix was also investigated. Dietary fat level was 10%( w/w) and reference groups were fed soybean oil (SO), repeseed oil ( RO) and beef tallow( BT ). Activity of LPL was about 60% higher in post-heparin plasma and 2 to 3 times higher in adipose tissue of BT group than fish oil or vegetable oil group. But there was no significant difference between fish oil and vegetable oil groups. Inclusion of EO above 2% (w/w) in dietary fat with fille oil of BT, markedly reduced both post -heparin plasma and adipose tissue LPL. Effects of MO and EO were not different in adipose tissue LPL, but EO was more effective than MO, in reducing post -heparin plasma LPL when mixed fat with varying amount of fish oil was used. Hepatic mitochondria isolated from fish oil-fed group showed the lowest rate of respiration but had P/O ratio comparable to SO and BT groups. On the other hand, cardiac mitochondria of fish oil group showed no difference in all the mitochondrial respiration parameters observed RO group had lowest P/O ratio both in hepatic and cardiac mitochondria. Fatty acid compositions of mitochondrial lipid differ between SO, RO, BT and MO groups, notably in the content of $C_{22:1}$ fatty acid.

Cardiorespiratory Regulations in the Japanese Amberjack (Seriola quinqueradiata) Exposed to Acute Hypoxia (저산소환경이 방어(Seriola quinqueradiata) 순환계의 산소운반 기능에 미치는 영향)

  • We studied the cardio-respiratory properties in the Japanese amberjack (Seriola quinqueradiata) during acute hypoxia exposure. Fish were exposed to three levels of hypoxia (80, 60 or 50 mmHg) for 60 min at $25^{\circ}C$. Cardiovascular parameters (cardiac output; Q, heart rate; HR, stroke volume; SV, blood pressure; $P_{DA}$) changed little from pre-exposure values during both 80 and 60 mmHg of hypoxia. During 50 mmHg of hypoxia, the fish showed a bradycardia which significantly affected Q, whereas no change in SV. $P_{DA}$ increased transiently. Arterial oxygen partial pressure ($PaO_2$) immediately reduced along with a decrease of the water oxygen partial pressure ($P_WO_2$). Arterial $O_2$ content ($CaO_2$) decreased significantly only after 60 min of 50 mmHg of hypoxia. Arterial pH (pHa) and hematocrit value (Hct) did not change significantly. Comparing the effects of different levels of hypoxia, oxygen delivery to the tissues ($Q\;{\times}\;CaO_2$) should be maintained a constant over a broad range of $P_WO_2$, however, severely depressed below 50 mmHg of hypoxia.

A Case report of Cyprinid Herpesvirus 2(CyHV-2) in Imported Ornamental Goldfish Carassius auratus (수입산 관상 금붕어(Carassius auratus)의 CyHV-2 감염 증례)

  • In this study, imported ornamental goldfish (Oranda and black moor) with high mortality was investigated with histopathological analysis. Fishes showed clinical symptoms characterized by gill necrosis and cumulative mortality ranging from 60% to 90% within a month after import from Thailand and China in 2021. CyHV-2 was detected in gill, kidney, spleen and liver of goldfishes. Histopathological analysis revealed epithelial necrosis, intranuclear inclusion bodies associated with viral symptoms in epithelial cell, macrophage infiltration and vacuolation in secondary lamella. In addition, disseminated granuloma formation was observed in various organs such as kidney, heart and mesenchymal membranes. These lesions refer to coinfections with CyHV-2 and other pathogens that cause chronic inflammation, which leads to mortality. Therefore, early disease monitoring needs to reduce the mortality when ornamental goldfish is imported.

A Study on the Sex Bias in the Nutrition Knowledge, Food Preference and Food Roles in the Family (영양지식, 식품기호 그리고 식행동에 나타난 남, 녀 차이에 관한 연구)

  • This research was attempted to investigate the sex bias in nutrition knowledge, food preference and food roles in the family. 152 couples living in Seoul, Cheonan in the 205∼605 were selected for this research as subjects from October 1 to December 20, 1991. The data analysis was made by way of ire quency, percentage, X2-test using SPSS package. The summarized results are as follows. 1. The survey on the sex bias in nutrition knowledge related heart disease on the subjects showed that there were no significant differences between men and women. But women had a relatively good nutrition knowledge than men. Especially women had a good nutrition knowledge that low sodium diet and exercise are efficient on preventing heart disease. 2. On the food preference, the subject showed significant difference between men and women. Men showed a relatively highest preference for protein food just as pork, fish, and tofu. Women showed a relatively highest preference for bread, fruit and vegetable. 3. On the food roles, the subjects showed that gender difference in household labor regarding food preparation still exit. Food-related activities are clearly still the domain of women in most families. Husbands and wives in younger families tended to think the husbands showed increase their efforts in food related activities.

Methylmercury Exposure and Health Effects

  • Methylmercury is a hazardous substance that is of interest with regard to environmental health, as inorganic mercury circulating in the general environment is dissolved into freshwater and seawater, condensed through the food chain, ingested by humans, and consequently affects human health. Recently, there has been much interest and discussion regarding the toxicity of methylmercury, the correlation with fish and shellfish intake, and methods of long-term management of the human health effects of methylmercury. What effects chronic exposure to a low concentration of methylmercury has on human health remains controversial. Although the possibility of methylmercury poisoning the heart and blood vessel system, the reproductive system, and the immune system is continuously raised and discussed, and the carcinogenicity of methylmercury is also under discussion, a clear conclusion regarding the human health effects according to exposure level has not yet been drawn. The Joint FAO/WHO Expert Committee on Food Additives proposed to prepare additional fish and shellfish intake recommendations for consumers based on the quantified evaluation of the hazardousness of methylmercury contained in fish and shellfish, methylmercury management in the Korea has not yet caught up with this international trend. Currently, the methylmercury exposure level of Koreans is known to be very high. The starting point of methylmercury exposure management is inorganic mercury in the general environment, but food intake through methylation is the main exposure source. Along with efforts to reduce mercury in the general environment, food intake management should be undertaken to reduce the human exposure to methylmercury in Korea.
