육상 양식장 인접해역의 해양학적 환경특성 (Oceanographic Environment Characteristics in Waters Adjacent to Fish Farm on Land)

  • Field observations for research of oceanographic characteristics in waters adjacent to fish farm on land and water quality at area where water is supplied to fish farm and area where water is discharged from fish farm are carried out in coastal waters of Pyoson located in the southeastern region of Jeju Island in March, May, August and November of 2000. Semidiurnal tides to the northeast and southwest dominate. The mean velocities of current around neap tides and spring tides are 8.52cmㆍs$^{-1}$, 28.63cmㆍs$^{-1}$, respectively. The difference of temperature between area where water is supplied to fish farm and area where water is discharged from fish farm is about 0.6$^{\circ}C$. Seasonal variation range for temperature and salinity is 13.5$^{\circ}C$ and 3.5psu, individually. Concentrations of nitrate(NO$_3$$^{-}$), phosphate(PO$_4$$^{3-}$ ), silicate(SiO$_2$) and COD(chemical oxygen demand) are high in autumn. Particularly, concentrations of nitrate and COD in winter and spring at station where water is supplied to fish farm are higher than these at station where water is dischared from fish farm. However, concentration of phosphate and silicate at station where is supplied to fish farm in spring, summer and autumn is higher than that at station where water is dischared from fish farm. Water quality in the study area is grade 1 and this level is kept up in the whole year.

제주도 연안 해역의 오염지표세균의 분포 (Distribution of Indicator Bacteria in Seawater off the Coast of Jeju Island)

  • We examined correlations of the density of fish farms with the distributions of indicator bacteria (Escherichia coli, fecal streptococci) and a bacterial fish pathogen (Streptococcus parauberis) off the coastline of Jeju Island. Seawater samples were collected at four coastal sites on the Island [Aewol (control), Gujwa, Pyoseon and Daejeong] in June, August and October 2016. The indicator bacteria were generally more frequently isolated from samples taken in August when water temperatures and human activities on nearby beaches were highest. Although fish farms were least common at Daejeong, the numbers of isolated fecal indicator bacteria were highest in the seawater and effluent water collected from this site. Hence, fish farms were not likely major contributors of indicator bacteria at Daejeong. We found discrepancies between the isolated bacterial counts and the predicted bacterial copy numbers deduced from our qPCR results, indicating that this pathogen may exist in a viable but non-culturable (VBNC) state in seawater. Thus, livestock wastewater and chemical fertilizer loading off Jeju Island may negatively impact seawater quality more than the effluent released from fish farms does.

  • Despite a variety of development in fish farming during the last decades, fish diseases by bacteria, virus, and parasites are still major problems in aquaculture. Aquaculture of Hounder fish is widely performed around Korea as well as Jeju island, due to relatively stable seed production, short farming period, and a higher value in market. However, intensive feeding and environmental pollution in aquacutural farm act as a great limiting factor in economic aspect. (omitted)

  • In 2017, total coliform (TC), fecal coliform (FC) and fecal Streptococci (FS) bacteria were examined in seawater samples collected at coastal sites on Jeju Island (Aewol as a control, Namwon, and Daejeong) and at Pohang (Yeongil as a control, Guryongpo-North, and Guryongpo-South) to examine the correlations between the density of fish farms and distributions of the indicator bacteria. Only a few TC, FC, and FS colonies were detected in all of the samples obtained from Jeju Island. Of note, 2,000 and 1,000 CFU of FS $100mL^{-1}$ were detected in samples from Guryongpo-South in June and August, respectively. Although the total area of approved fish farms located within 5 km of the sampling point at Guryongpo-South is 5-16 times smaller than in other regions, the number of indicator bacteria was highest in this region. Therefore, microbiological pollution in the Guryongpo-South region might be due to sources other than the effluent released from nearby fish farms.

제주도 어류양식장에 대한 미생물학적 오염도 조사 (Survey of the level of Microbial Contamination in Fish Farms on the Jeju-Island)

  • 본 연구는 2006년 3월부터 8월까지 제주도에서 성업 중인 육상 수조식 양식장 21개소를 대상으로 하여 미생물학적 위해요소를 양어 용수, 양어 사료, 양식 넙치에서 조사하였다. 식품 병원성 미생물 중 일반세균과 총 대장균군, 그리고 Salmonella spp., Escherichia coli O157:H7, Staphylococcus aureus, Bacillus cereus, Vibrio parahaemolyticus를 조사하였다. 이번 연구를 진행하는 동안 특이할만한 점은 여름에 들어서면서 집중적이면서도 빈번하게 비가 내렸다. 양어용수를 검사한 결과 S. aureus, B. cereus and Salmonella spp.는 각각 $0-3.3{\times}10^2 CFU/ml,\;0-2.2{\times}10^2CFU/ml\;and\;0-5.1{\times}10^1 CFU/ml$ 검출이 되었다. 또한 사료 내 미생물을 조사하여 본 결과 조사한 MP사료 85%에서 S. aureus, B. cereus and total coliforms이 검출이 되었다 반면 양식 넙치를 조사한 결과 Escherichia coli O157:H7, total coliforms은 검출이 되지 않았다. 추후 이와 관련하여 좀더 체계적인 연구 시스템을 양식현장에 적용한다면 근래 들어 문제가 되고 있는 식품위생상 수산물이 야기하는 문제점들이 해결이 되어 안전한 식품으로서의 가치를 인정받을 수 있을 것 같다.

제주도지역 넙치양식장의 스쿠티카증 발생동향 (Scuticociliatosis in flounder farms of Jeju island)

  • The prevalence of the scuticociliatosis in flounder farms of Jeju island was surveyed for ten years from 1995 to 2004. The occurrence ratio had maintained as less than 10% until mid 1990s, and shown increasing trends to year 1998. The ratio was equivalent to over 40% of overall disease occurrences in flounder farms since year 2000. The monthly infection rates by the scuticociliates indicated relatively higher levels from May to September, and mixed infection phenomena with bacterial disease of vibriosis were common.

양식장 넙치 폐사어를 이용한 단백질 소재의 개발에 관한 연구(2) -산업화공정 연구- (A Study on Development of Protein Materials using Dead Flatfish from Fish Farms(2) -Industrial Process-)

  • In manufacturing of flatfish skin collagen peptide (FSCP) and flatfish protein hydrolysate (FPH) by reuse of dead flatfish from fish farm in Jeju island, the industrial process was optimized with the laboratory scale research and the on-field process. Segmented unit processes from raw material incoming to shipment were established to produce commercial product of FSCP and FPH. Total plate counts of FSCP were twenty five times of FPH, but food poisoning bacteria were not detected in two samples. FSCP and FPH were safe from heavy metal such as Pb(II), Cd(II) and Hg(II). The residual contents of antibiotics and disinfection matter in FSCP and FPH were not detected. The optimized process for mass production made the one-third of the running time and two times of the yield. From economic analysis, the production cost was estimated to 22,000 and 12,000 won/kg for FSCP and FPH, respectively. Therefore the product from the reuse of dead flatfish was expected to have a considerable competitive price and high added-value functional food material compared with other commercially available fish products.

  • 2000년도 2회에 걸쳐 제주도 일원 넙치 양식장을 대상으로 감염어에서 분리된 연쇄구균에 대한 특성시험 결과를 요약하면 다음과 같다. 감염어로부터 분리된 연쇄구균은 형태적으로 연쇄상의 gram 양성의 구균이었고, catalase와 oxidase에 음성이었으며, 용혈성 시험에서는 $\beta$-용혈성을 나타내었다. 감염어에서 각 장기별 생균수에서는 환부, 신장, 혈액 그리고 뇌순으로 연쇄구균이 검출되었다. 약제 감수성에서는 ampicillin, ciprofloxicin 둥에 감수성이 있었지만 erythromycin 등에는 감수성이 없었다. 복강에 주사하여 병원성 시험을 한 결과 10일 이내에 전량 폐사하는 강한 병원성을 보였다. 감염균의 용혈성에 대한 특성은 $50^{\circ}C$ 30분의 배양에서 용혈활성이 마르게 증가되어 최대값에 도달되었고 또한 $pH 5.5\~6.5$에서 높은 활성값을 보였다.
