Comparative Study of Growth and Gonad Maturation in Diploid and Triploid Marine Medaka, Oryzias dancena

  • The marine medaka, Oryzias dancena is a suitable sample as a laboratory animal because it has a small size and clearly distinguishes between female and male. Data on the growth and maturity of the diploid and triploid sea cucurbit species suitable for laboratory animals are very useful for studying other species. Triploidy was induced in the marine medaka by cold shock treatment ($0^{\circ}C$) of fertilized eggs for 45 min, applied two minutes after fertilization. The diploid and triploid male fish were larger than their female counterparts (P<0.05), and the concentrations of thyroid stimulating hormone (TSH) and thyroxine (T4) were higher in the induced triploids over 1 year (P<0.05). In both the diploid and tri-ploid groups the concentrations of TSH and T4 were higher in the male fish than in the females (P<0.05), while the testo-sterone and estradiol-$17{\beta}$ concentrations in the induced triploids were lower than in the diploids (P<0.05). The gonadosomatic index (GSI) of the triploid fish was lower than that for the diploids, and the GSI for females in each ploidy group were higher than that for the males. For both groups the GSI was highest at 4 months of age, and decreased thereafter to 12 months. Analysis of the gonads of one-year-old triploid fish suggested that the induction of triploidy probably causes sterility in this species; this effect was more apparent in females than in males.

  • This study aimed to develop a new commercialization model for theindustrialization of head family food as a gentry families' complex food culture product. We tried to develop a head family food and propose a city sightseeing style's head family product by interviews with 'Hakindang' as the center, the head house of the Suwon Baek clan, and Injaegong group in Jeonjoo. Hakindang (in Jeonju) was confirmed as an improved model Korean-style house in the enlightenment period and the twentieth style modern head family as emerging capitalist. Hakindangsupported independence war funds in the Japanese colonial era and was widely known as gate of filial piety in Jeonju. Representative seasonal foods of Hakindang include pan-fried sweet rice cake with flower petals in the spring, hot spicy meat stew and polypus variabilis in summer, hanchae in autumn, and napa cabbage kimchi with salted flatfish seafood in the winter. When parents-in-law had a birthday, there were party noodles, sliced abalone stuffed with pine nuts, brass chafing dish, fish eggs, slices of blilde meat, and matnaji. Daily, there were mainly salted seafood, slices of raw fish, grilled short rib patties, braised fish, baked fish, syruped chestnut, grilled deodeok root, bean sprouts, radish preserved with salt, dongchimi, soy sauce-marinated horseshoe crab, butterbur, perilla seed stew, salted clams, raw bamboo shoot, agar, fried kelp, etc. The most basic virtue of the head family is filial duty, and they developed mosim dining as a representative food of Hakingdang's head family. The mosim menu is composed of 65% carbohydrates, 20% protein, and 15% fat because it is table for parents-in-law like more than 75 years old. The sensory evaluation showed a chewy texture that is easily swallowed.

  • Spawning of matured olive flounder, Paralichthys olivaceus was accelerated by using single injection of human chorionic gonadotropin (HCG 1,000 IU/kg BW) or carp pituitary extract (CPE 10 mg/kg BW). The body weight of HCG or CPE treated group slightly improved with $5.6\pm0.4\%$ at 2 days after injection or $6.4\pm0.5\%$ at 3 days after injection, Number of eggs spawned increased in the fish treated with HCG or CPE. Surface floating rates of eggs spawned increased in the fish treated with HCG or CPE. Hatching rate also increased in experimental groups compared to the control. Especially superior quality of eggs were obtained 2 and 3 days after injection from HCG treated groups and 3 and 4 days after injection from CPE treated groups at $20^{\circ}$, respectively.

Morphological Development of Embryo, Larvae and Juvenile in Yellowtail kingfish, Seriola lalandi

  • This study monitored the morphological development of embryo, larvae and juvenile yellowtail kingfish, Seriola lalandi, for their aquaculture. The fertilized eggs obtained by natural spawning were spherical shape and buoyant. Fertilized eggs were transparent and had one oil globule in the yolk, with an egg diameter of $1.35{\pm}0.04mm$ and an oil globule diameter of $0.32{\pm}0.02mm$. The fertilized eggs hatched 67-75 h after fertilization in water at $20{\pm}0.5^{\circ}C$. The total length (TL) of the hatched larvae was $3.62{\pm}0.16mm$. During hatching, the larvae, with their mouth and anus not yet opened. The yolk was completely absorbed 3 days after hatching (DAH), while the TL of post-larvae was $4.72{\pm}0.07mm$. At 40 DAH, the juveniles had grown to $30.44{\pm}4.07mm$ in TL, body depth increased, the body color changed to a black, yellow, and light gray-blue color, and 3-4 vertical stripes appeared. At 45 DAH, the juveniles were $38.67{\pm}5.65mm$ in TL and $10.10{\pm}0.94mm$ in body depth. The fish were green with a light orange color, with 7 faint green-brown stripes on the sides of their body. At 87 DAH, the juveniles had grown to 236.11 mm in TL, 217.68 mm in fork length, and 136.5 g in weight. The fish resembled their adult form, with a light yellow-green body color, loss of the pattern on the sides of their body, and a yellow coloration at the tip of the caudal fin.

Mass Production of All-Female Triploid Eggs in Rainbow Trout (Oncorhynchus mykiss) During the Fall Season by Chromosome Manipulation (염색체 조작에 의한 전 암컷 3배체 무지개송어 조기난의 대량생산)

  • For the mass production of all-female triploid rainbow trout during fall season, treatment of short-term daylength from 30th of August, 1994 and chromosome manipulation by heat shock were performed with 3-year-old fish. After treatment of short-term daylength to fish, we successfully obtained the fertilized eggs from all treated fishes. However, hatching rate were significantly lower than that of natural spawning season (P<0.05). Hormonal treatment using 3mg of $17\alpha-methyltestosterone$ per kg of diet for 55 days at $16.5^{\circ}C$ gave $100\%$ of sex-reversed male (masculinized female) population. When the fertilized eggs were treated with the various conditions of heat shocks survival rates and triploid incidencies were varied, and ranged from 15.0 to $88.2\%$ and 36.7 to $100\%$, respectively.

Proximate compositions of wild and cultured sweet smelt (Plecoglossus altivelis) muscles and eggs (천연 및 양식산 은어의 근육 및 난의 일반성분 조성)

  • Proximate compositions of muscles and eggs between wild sweet smelt which were caught in Seomjin river and Miryang river, and cultured sweet smelt which were purchased from certain cultured farms in Hadong and Miryang, were compared. Proximate compositions of wild sweet smelt muscles were $76.7\~77.5\%$ for moisture, $18.4\~19.9\%$ for protein, $2.41\~3.47\%$ for lipid, and $1.04\~1.33\%$ for ash content and there were almost no differences between the fishes from Seomjin and Miryang river. Protein and ash content in the cultured fish muscles were similar to those of the wild fish muscles, while the former contained much more lipid and carbohydrate, and less moisture than the latter, especially lipid content differed greatly according to their cultured farms. This might be the result of their different diet compositions. Lipid, protein and carbohydrate content in eggs of the wild and cultured fishes were much higher than those in their muscles. There was a negative correlation between moisture and lipid content in all samples; y= -0.2715x+24.903 (r= -0.7253, p<0.001).

  • Egg development and early life history of the Korean natural monument fish Hemibarbus mylodon (Pisces: Cyprinidae) were investigated to provide basic data on biological characteristics and ecological recovery. Adult fish were collected from nature and transferred to the laboratory. For the first time, artificial maturation of females and males succeeded after 15 months of indoor culture. Mature eggs and sperm were obtained using Ovaprim injections (0.5 mL/kg) and then the eggs were fertilized using the dry method in the laboratory. The mature eggs were adhesive, turbid, and greyish; they averaged $2.21{\pm}0.06mm$ (n=30) in diameter. The embryos began to hatch about 78 h after fertilization at a water temperature $20{\pm}1^{\circ}C$, and the newly-hatched larvae were $6.6{\pm}0.75mm$ in total length (TL). At 14 days after hatching, the post-larval individuals were $13.5{\pm}0.23mm$ (TL), and their yolk sacs were completely absorbed. Twenty one days after hatching, they entered the juvenile stage and reached $13.5{\pm}0.23mm$ (TL). At 100 days after hatching, their band patterns, external form, and a pair of barbels were similar to those of adults, and they averaged $33.0{\pm}4.25mm$ (TL). The breeding technology and characteristics of early life history obtained in this study will be very helpful for conservation of H. mylodon in nature.

Ecological Characteristics of Microphysogobio yaluensis in Dugye Stream of Geum River Basin, Korea (금강지류 두계천에 서식하는 돌마자(Microphysogobio yaluensis)의 생태 특성)

  • This study investigated the ecological characteristics of Microphysogobio yaluensis at Dugye Stream of Geum River basin from January to December 2021. The river bed structure of this species' habitat was rich in pebbles and gravel. The water depth ranged from 12 to 85 cm with an average of 23 cm, which was shallow. The stream velocity was rapid at 1.03±0.34(0.72-1.47) m/sec. The ratio of females to males was 1:0.86. The age according to the total length-frequency distribution as of summer (June, July) indicated that the group with less than 50 mm (30.4-49 mm) in total length was one year old, the group with 50-69 mm was two years old, and the group over 70-92 mm was three years old. Both male and female fish sexually matured at 50 mm long and two years old in June and July. The spawning season was from July to August, and the water temperature was between 25.8℃ and 28.2℃ during the period. The prosperous spawning season was July. The matured eggs were yellowish and spherical with a mean diameter of 0.52±0.16 (0.33-0.77) mm, and the average number of eggs found in the ovaries of mature female fish was 2,593 (1,343-4,672). As for the food of M. yaluensis, only attached algae, which grow by attaching to stones and gravel of the stream rapids, were identified. The feeding habit of M. yaluensis was reported to be herbivorous, ingesting Cyanophyta, Chlorophyta and Bacillariophya. Also, the species belonging to Bacillariophya and Chlorophyta were abundant in Dugye Stream.

Development of Eggs, Larvae and Juveniles of the Squalidus gracilis majimae from Wicheon Stream, Nakdong-River (낙동강 수계 위천에 서식하는 긴몰개 Squalidus gracilis majimae의 난발생 및 자치어 형태발달)

  • The aim of this study was to investigate the early life history of Squalidus gracilis majimae by observing the development of the eggs and morphology of juvenile fish. Squalidus gracilis majimae used in this study were captured in 2021 June from a tributary flowing into Wicheon, located in Uiseong-gun, Gyeongsangbuk-do. For artificial insemination, ovaprim was injected at 0.5 mL/kg. Egg development occurred 12 h after injection. The size of mature eggs of S. gracilis majimae was 0.80~0.85 mm (0.83±0.01 mm, n=30). Incubation time at 22±1℃ ranged from 62.5 to 69 h. The length of larvae immediately after hatching was 2.39~2.53 mm (2.44±0.04 mm, n=30). On the 18th day after hatching, the larvae had a total length of 8.51~9.84 mm (8.95±0.42 mm, n=30), and the tip of the spine was completely curved. On the 22nd day after hatching, the juvenile fish had a total length of 12.6~13.6 mm (13.0±0.36 mm, n=30), with dorsal fins at iii7 and anal fins at i7. Egg size and hatched larvae size were similar for S. gracilis majimae and S. multimaculatus, and cupulae showed a singularity as it disappeared last among related species.

Morphological Development of Eggs, Larvae and Juveniles of Trident Goby, Tridentiger brevispinis (Pisces: Gobiidae) (민물검정망둑 Tridentiger brevispinis의 난발생 및 자치어 형태발달)

  • This study aimed to examine the early life history of Trident goby (Tridentiger brevispinis) by observing their egg development and juvenile fish morphology. The average size of mature eggs was 1.13~1.41 (1.30±0.07) mm (n=30), and 0.70~0.86 (0.79±0.04) mm (n=30) in long diameter and short diameter, respectively. The incubation period at 24±1℃ ranged from 167~228 h. The newly hatched larvae measured 2.31~2.78 (2.51±0.18) mm (n=30) in total length (TL), and their mouth and anus were not yet open. At 3 days after hatching, the preflexion larvae reached 2.84~3.10 (2.98±0.10) mm in TL, where in their yolk absorption was complete and their mouths began to open. At 19 days after hatching, the larvae reached the flexion stage, measuring 4.02~4.62 (4.36±0.19) mm in TL. The tip of their notochord was bent upward. At 30 days post-hatching, the larvae reached the postflexion stage, measuring between 5.04~6.36 (5.76±0.51) mm in TL, with the tip of the caudal fin bent at 45°. After 54 days, the larvae had reached the juvenile stage, measuring between 7.43~9.84 (8.48±0.90) mm in TL, and were differentiated by their constant number of fins (6 first dorsal, 12 second dorsal, 11 anal, and 10 ventral fins). This study found that T. brevispinis had larger fertilized eggs and a greater number of myotomes in hatching larva than similar species. Additionally, the distribution of melanophores in T. brevispinis was distinct from that in other similar species, making it easy to distinguish them morphologically.
