Objective : This study aimed to compare the difference between $Angelica$$decursiva$, Peucedanum $praeruptorum$ and $Anthriscus$$sylvestris$ which have been used as herbal medicine, Jeonho (Angelicae Decursivae Radix) in Korea and provided the evidence to exclude $A.$$sylvestris$ not to use as Joenho. Methods : The similarities of original medicinal herb with samples from local market were evaluated including morphological appearance and chromatographic fingerprint. In addition, relation between original medicinal herb and local samples were analyzed using statistical clustering methods. Results : $A.$$decursiva$, $P.$$praeruptorum$ and $A.$$sylvestris$ represented different morphological appearances and chromatographic fingerprint. Several samples from China exhibited similar morphological and chromatographic appearance with $A.$$decursiva$ or $P.$$praeruptorum$. Eleven samples from Korea showed identical similarity to $A.$$sylvestris$. Conclusions : Since $A.$$sylvestris$ represented obvious differences compared to $A.$$decursiva$ and $P.$$praeruptorum$, it is required not to use $A.$$sylvestris$ as medicinal herb, Jeonho. Additionally, exact identification and quality control must be applied to $A.$$decursiva$ or $P.$$praeruptorum$ from China in order to maintain therapeutical efficacy.

All citizens over 17 year old living in Korea have to be fingerprinted to obtain a certificate of resident registration. For this reason, human identification through fingerprints has been used actively in crime scene investigation. The fingerprint is so unique that it is one of the most certain ways to identify oneself and it can differentiate between genetically identical twins. Fingerprints gained in crime scene indicate a direction of criminal investigation in conjecturing a suspect. Fingerprints help a reunion of family got scattered for a long time and make it possible to get a personal identification for missing person who met with natural calamity. We developed a new fingerprinting method using safranine O, so as to develop fingerprints on the adhesive tapes and non-porous papers in various physical environments. Results were compared to the preexisting fingerprinting method, the minutiae numbers of fingerprints were greatly increased in our newly developed safranine O fingerprinting method. This newly developed safranine O method showed a quantity and quality comparable to the preexisting fingerprinting method routinely used in these days. In our hands, the safranine O fingerprinting method is another easy and obvious choice when the forensic case sample is available for fingerprints on the adhesive tapes and non-porous papers.
Listeria monocytogenes poses an increasing health risk, which in part is due to increasing health risk, consumption of ready-to-eat food products and the introduction of increasing numbers of food products from regions with different dietary habits. L. monocytogenes can be present in meat, shellfish, vegetables, unpasteurised milk and soft cheese and poses a risk if food containing these products is stored at refrigeration temperature and is not properly heated before consumption, as L. monocytogenes is psychrophilic. Amplified-fragment length polymorphism (AFLP) analysis is the method of genotypic techinique in which adaptor oligonucleotides are ligated to restriction enzyme fragments and then used as target sites for primers in a PCR amplification. The amplified fragments are electrophoretically separated to give strain-specific band profiles. Single-enzyme approach that did not require costly equipment or reagents for the fingerprinting of strains of Listeria monocytogenes was developed. Single-enzyme amplified fragment length polymorphism (SE-AFLP) analysis was used to perform species and strain identification of Salmonella, Shigella, Yersinia and E. coli. By careful selection of AFLP primers, it was possible to obtain reproducible and sensitive identification to strain level. The AFLP patterns of L. monocytogenes are divided by the kinds of specimens in which were isolated. SE-AFLP fragments can be analyzed using standard gel electrophoresis, and can be easily scored by visual inspection, due to the low complexity of the fingerprint obtained by this method. These features make SE-AFLP suitable for use in either field or laboratory applications.
Cupredoxin-like proteins are mainly copper-binding proteins that conserve a typical rigid Greek-key arrangement consisting of an eight-stranded β-sandwich, even though they share as little as 10-15% sequence similarity. The electron transport function of the Cupredoxins is critical for respiration and photosynthesis, and the proteins have therapeutic potential. Despite their crucial biological functions, the identification of the distant Cupredoxin homologs has been a difficult task due to their low sequence identity. In this study, the overlapped conserved residue (OCR) fingerprint for the Cupredoxin superfamily, which consists of conserved residues in three aspects (i.e., the sequence, structure, and intramolecular interaction), was used to detect the novel Cupredoxin homologs in the NCBI non-redundant protein sequence database. The OCR fingerprint could identify 54 potential Cupredoxin sequences, which were validated by scanning them against the conserved Cupredoxin motif near the Cu-binding site. This study also attempted to model the 3D structures and to predict the functions of the identified potential Cupredoxins. This study suggests that the OCR-based approach can be used efficiently to detect novel homologous proteins with low sequence identity, such as Cupredoxins.
The Degradation of amur cork tree extract is investigated by GC-MS after treating the dye with three thermal degradation systems of, room temperature (RT), $4^{\circ}C$ refrigeration (LT), $100^{\circ}C$ oven (OV), and $H_2O_2$/UV/$O_2$ (PER) degradation system for 0-24 days. It was found that PER degradation system represented the highest intensity of degradation treatment followed by OV treatment among the four degradation parameters. The possible fingerprint products of amur cork tree dye, that yielded 68% (or higher) reliability in the NIST spectral match, were isobenzofuran-1,3-dione,4,5-dimethoxy- (8.37 min, PER only), 1,3-dioxolo[4,5-g]isoquinolin-5(6H)-one,7,8-dihydro (9.41 min, PER only), canthine-6-one (10.24 min, RT, LT, OV only), and dihydroberberine (15.05 min, RT, LT, OV, PER) in the order of higher to lower possibility of detection. Unknown products 7 (13.43 min) and 8 (16.35 min) are two other possible fingerprint products of amur cork tree dye that require future identification.
In the field Of individual identification in forensic Science, if the body is charred, it is sometimes impossible to identify the morphologic changes and charred tissue such as blood, muscle and bone can not be identified by forensic microbiologic method such as DNA typing. So the author used the characteristics of teeth which is relatively firm compare to other organs and stable to external environment such as heat and also possess cells needed for the DNA typing. The author conducted the experiment on teeth to detect DNA related to individual identification regarding to temperature in which other charredorgans can not be detected. The experiment was done on 64 extracted third molars consisted of unheated ones, and heated teeth to $100^{\circ}C$, $150^{\circ}C$, $200^{\circ}C$ for 45 min, 90 min, and 120 min respectively and to $250^{\circ}C$ for 45 min. DNA was extracted from each tooth and amplified fragment length polymorphism procedure(AMP-FLPs) using polymerase chain reaction(PCR) was applied and observed for the matching DNA in HumTH01 and HumCD4 locus and the followings Are the results : 1. It was able to detect matching DNA in HumTH01 and HumCD4 locus in every teeth which no heating has been done. 2. It was able to detect matching DNA in HumTH01 and HumCD4 locus in every teeth heated to $100^{\circ}C$ for 45, 90 and 120 min. 3. It was able to detect matching DNA in HumTH01 and HumCD4 locus in teeth heated to $l00^{\circ}C$, $200^{\circ}C$ for 45, 90, 120 min. 4. It was impossible to detect matching DNA in HumTH01 and HumCD4 locus in teeth heated to $250^{\circ}C$. So, it is possible to extract DNA from teeth that otherwise can not be extracted from other organs in the charred body and it can be concluded that teeth are highly reliable and applicatable as forensic odontology for individual identification.
팬티엄시스템에서 평균 정합 속도 1.55초를 기록하였으며, 0.05%의 FAR(False Acceptance Rate)에서 8.83%의 FRR(False Rejection Rate)의 오 인식율을 얻을 수 있었다.
