Textural Changes in Rat Tissues by Carbofuran and Its Suppression by Phenobarbital Sodium and 3-Methylcholanthrene (Carbofuran이 쥐의 조직에 미치는 형태적 변화와 Phenobarbital Sodium 및 3- Methylcholanthrene에 의한 억제효과)

  • This study was carried out to investigate the toxicological effects of carbofuran on the histological and fine structures in the kidney, liver, and brain of rat and also to clarify compensatory effects of phenobarbital sodium (PB) and 3-methylcholanthrene(3-MC) on the carbofuran toxicity. SPF albino rats were treated with carbofuran(3.8mg/kg), PB(60mg/kg), 3-MC(60mg/kg), carbofuran+PB, carbofuran+3-MC and subjected to the light microscopic study. In the kidney of rat, hemorrhage and extremely atropic change of renal corpuscles were frequently observed at 48 hrs after carbofuran treatment. Combination treatment groups of carbofuran and PB or 3-MC showed atrophic changes were largely recovered at 6 hrs, and the tissue findings of the kidney became similar to those of control group at 48 hrs after treatment. In the liver of rat treated only carbofuran, the degenerative and necrotic changes of hepatic lobules were frequently observed at 48 hrs after carbofuran treatment. Combination treatment of carbofuran and PB or 3-MC showed the hepatic lobules were similar to those of control groups at 6 hrs after the combination treatment. In the brain of rat treated with carbofuran alone, degenerative changes and dilation of capillary vessel of cerebral cortexes were observed at 48hrs after treatment. Combination treatment of carbofuran and PB or carbofuran and 3-MC showed the cerebral cortexes were similar to those of control groups at 6 hrs after the treatment. These results suggest that PB and 3-MC could regenerate the toxicity of carbofuran to the tissue of kidney, liver and brain of rat.

  • The present study was carried out light and electron microscopic analysis of the structures of the tritocerebral neurons in butterfly (Pieris rapae Linne) from Insecta. We have summarized our morphological analyses as follows: In insecta, tritocerebrum of butterfly shows tentatively classified 6 neuronal types without any arrangement of cell layers. Type I cells with long oval or spindle shape are the largest out of 6 kinds. The nucleus contains a small amount of chromatin. In the large volume of cytoplasm there contained rich cell organelles except granular endoplasmic reticulum. Type II cells are smaller than type I neurons, but their ultrastructural features are similar to type I neuron. They have well-developed granular endoplasmic reticulum. Type III cells are spherical or triangular and smaller than type I neuron with spherical nuclei. Cell organelles are mostly prominent, esp., mitochondria, ribosomes and fine fibrils which are arranged in periphery of the cytoplasm.

Effects of the cis-Dichlorodiammineplatinum on the Fine Structures of the Interalveolar Septum in the Mouse (cis-Dichlorodiammineplatinum (II) 이 생쥐 폐포간중격의 미세구조에 미치는 영향)

  • cis-Dichlorodiammineplatinum (II) (cis-Platin), a metallic compound, has widely been used as an effective anticancer chemotherapeutic agent. The precise mechanism of action of this agent is still unknown, but it is postulated that cis-Platin may act on the cancer cell like bifunctional alkylating agents. Although this agent is very beneficial to the patients with cervical cancer, germinoma of testis, neuroblastoma and others, it may also damage to the normal cell so that many side effects; severe hemorrhagic enterocolitis, bone marrow depression, renal damage and liver damage will develope. This experiment has been undertaken to pursue the cytotoxic effects of the cis-Platin on the ultrastructures of the interalveolar septum in the mouse lung. A total of 55 healthy male mice of ICR strain were used as experimental animals and divided into 5 mice of normal control group and 50 mice of cis-Platin treated group. The mice of cis-Platin treated group were sacrificed by carotid exsanguination at 6, 12, 24 hours, 3 days and 7 days after intraperitoneal injection of 6.0 mg of cis-Platin ($Abiplatin^R$ Abic Co. Ltd.) per kg of mouse body weight. The specimen obtained from the lower lobe of left lung were sliced into $1mm^3$ and prefixed with 2% glutaraldehyde -2.5% paraformaldehyde solution prepared with Millonig's phosphatae buffer solution (pH 7.4) at $4^{\circ}C$ for 3-4 hours. After postfixation with 1% osmium tetroxide solution all specimens were embedded in Epon 812. Ultrathin sections about $600-800{\AA}$ in thickness were stained with uranyl acetate and lead citrate and observed with Hitachi-600 electron microscope. The results obtained were as follows: 1. Local swellings with increase of electron density and number of pinocytic vesicles in the cytoplasms of the type I pneumocyte and endothelial cell of the blood air barrier in interalveolar septum of cis-platin treated mice were observed. 2. Cisternae of rough endoplasmic reticulum were dilated and sacculated in association with detachment of membrane bound ribosomes of the type II pneumocyte in interalveolar septum of cis-Platin treated mice. 3. Swollon mitochondria with uneven electron density of their matrix were observed in the type II pneumocyte of interalveolar septum in the cis-Platin treated mice. 4. The lamellae of lammelar bodies in type II pneumocyte of interalveolar septum in cis-Platin treated mice were devoided or transformed into homogeneous electron dense material. It is consequently suggested that cis-Platin would induce the cellular edema of type I pneumocyte and endothelial cell, and degenerative changes of cytoplasmic organelles of the type II pneumocyte in the interalveolar septum of the mouse lung.

Role of Sand Shoal in the Intertidal Flat Sedimentation, Gomso Bay, Southwestern Korea (서해 곰소만 조간대 퇴적작용에서 모래톱의 역할)

  • A sand shoal (1300 m long and 400 m wide) with an orientation of north-south is formed on the lower tidal flat of Gomso Bay, southwestern coast of Korea. Surface bedforms, sedimentary structures, sedimentation rate, grain size distribution and can-corer sediments have been measured and analysed along the sand shoal proper zone B and its offshore zone A and onshore zone C during the period of 14 months. These three zones can be differentiated based on sedimentological characteristics: A zone - fine sand (3${\varphi}$ mean), linguoid-type ripples, 70 mm/month in sedimentation rate and no bioturbation, B zone - medium sand (2.5${\varphi}$ mean), dunes (4${\sim}$5 m in wavelength), 30 mm/month in sedimentation rate and no bioturbation, and C zone - coarse silt (5${\varphi}$ mean), sinuous-type ripples, 10 mm/month in sedimentation rate and well-developed bioturbation. These characteristics indicate that the zone C represents a relatively low-energy regime environment whereas the zone A corresponds to a relatively high-energy environment. The zone B would play an important role for a barrier to dissipate the approaching wave energy, resulting in maintaining of low-energy conditions in the inner part of Gomso-Bay intertidal flat behind.

Soil Characteristics on the Fluvial Surface in the Basin of Kyeongan-cheon (Stream) (경안천 유역 하성면에 발달한 토양 특성)

  • The purpose of this study is to clarify the relict landform development of fluvial terrace and the soil characteristics occurring on the fluvial deposits. The physico-chemical properties of soil that are developed on terrace deposits and X-ray diffraction analysis of clay were investigated specifically. The horizon of $A_1$ consists of silt loam with reddish-brown color (5YR4/3). Its soil structures is a weak, fine, subangular, and blocky, breaking to granular. The horizon of $B_{1t}\;and\;B_{2t}$ are silt clay with either a yellowish red (5YR5/6), bright red (2.5YR4/6) color. This soil structure is weak, subangular, and blocky, with thin discontinuous bright red (2.5YR4/6) clay cutans and soft manganese concretions. This red soil structure is made on heavy-textures. It is packed compactly with parent materials of high fluvial surface sediments, and usually has a $A_1-B_{1t}-B_{2t}-C$ profile, from top to bottom. In most cases, clay accumulation in the B-horizon and clay cutans on ped surfaces are observed, which means the argillic horizon has formed. The soils derived from fluvial surface deposits are associated with soils. The soils on the high fluvial surface are considered to be a kind of paleo-red soil which were developed by strong desilicification and rubefaction, and strong leaching of bases under warmer bio-climatic condition during the old Pleistocene period. According to these morphological and anlaytical characteristics,geomorphological features and bio-climatic conditions under which the soil have developed on the high terrace sediment indicate that the soil should be classified as paleo-red soils.

Pore Structure and Physical Properties of Heterogeneous Bonding Materials of Recycled Aggregate according to Carbonation Reforming (순환 골재 부착 이질재의 탄산화 개질에 따른 공극구조 및 물리적 특성)

  • At present, about 40 million tons of concrete is dismantled each year, which accounts for the largest portion of the total amount of construction waste with 60.8%. It is known about 97.5% of it is recycled. However, most of the usage of waste concrete is limited to lower value-added business areas, and considering the increasing amount of waste concrete generated due to the deterioration of structures, the need for converting waste concrete to structural concrete is urgent. Therefore, this study aims at estimating the period for the optimum carbonation reforming to improve the quality of recycled aggregate, by making use of the method of accelerated carbonation reforming of the bonding heterogeneous (cement paste and mortar) for the purpose of converting recycled aggregate to structural concrete. Based on the period appropriate for the heterogeneous thickness and each bonding thickness of recycled aggregate which was drawn from previous studies, the changes in the characteristics and physical properties of pore structure according to progress of accelerated carbonation were analyzed. The result shows that with the progress of carbonation, the pore volume and the percentage of water absorption of the bonding heterogeneous decreased and the density increased, which indicates improvement of the product quality. But after certain age, the tendency was reversed and the product quality deteriorated. Synthesizing the results of previous studies and those of the present study, this study proposed 4 days and 14 days respectively for the period for the optimum carbonation reforming of recycled fine aggregate and recycled coarse aggregate.

Origin and Characteristics of Sand Ridges in the western Continental Shelf of Korean Peninsula (한반도 서부대륙붕에 발달한 사퇴의 발생기원과 특성)

  • From northeast to southwest, discontinuous sand ridges distribute on the western continental shelf of Korean Peninsular. The dimension of sand ridges is 3 to 21 m high, 3.1 to 6.8 km wavelength and 9-64 km long with 0.5 steep slope. they are probably originated and reformed by the intensity of tidal current according to the sea level rise. The characteristics of sand ridges revealed in study area are summarized as follows: (1) The sand ridges line up with the long axes of the tidal current ellipses, indicating a tidal control. (2) these are composed of two sedimentary sequences on the 3.5 kHz seismic profiles and core sediments. The upper sequence characterized by prolonged type is covered with thin veneer of massive fine sand(Mz, 2-3$\phi$) with Olive Gray(5Y 5/2). The lower sequence is characterized by internal reflector type with parallel and discontinuous. It consists of sandy mud or muddy sand(Mz, 5-7$\phi$) with laminar structures. the parallel internal reflectors are truncated on the slope of sand ridges. (3) Asymmetrical sand waves are superimposed on the sand ridges, and facing to the crest. However, symmetrical sand waves lie on the crest. Sand ridges having characteristics above is originated by scouring of tidal current, covered with coarase relict sediments, and modified by sadware.

Fine Structures of the Enteroendocrine Cells in the Duodenal Mucosa of the Hedgehog, Erinaceus koreanus (고슴도치 십이지장 점막의 장내분비세포의 미세구조)

  • In order to discriminate the enteroendocrine cell types in the mucosal epithelium of the normal duodenum of the Korean hedgehog (Erinaceus koreanus). The tissues were fixed in the mixture of 1% paraformaldehyde and 1% glutaraldehyde in phosphate buffer (pH 7.2), and postfixed in 2% osmium tetroxide (phosphate buffer, pH 7.2). They were embedded in Araldite, and the ultrathin sections were made by LKB-V ultratome following the inspection of semithin sections stained with toluidine blue-borax solutions. Ultrathin sections contrasted with uranyl acetate and lead citrate were observed with JEM 100B electron microscope. At least six types of enteroendocrine cells distributed in the mucosal epithelium of the duodenum were identified according to their morphological characteristics mainly based on the size, shape, number and electron density of the secretory granules. Type I cells had moderately developed organelles. The secretory granules were pleomorphic ($370X510nm$), and the granule cores with high electron density were enveloped in limiting membrane and characterized by a narrow halo. Type II cells contained an indented nucleus and well-developed organelles. The secretory granules were round (350 nm) and classified in two kinds by electron density, moderate and high. Both granules were surrounded by limiting membrane and those with high electron density showed often a wide halo. Type III cells had an indented nucleus. The secretory granules with various electron density were round (220 nm) in shape. The granules with high electron density were enveloped in limiting membrane and characterized by a narrow halo, but those with low or moderate electron density had not been observed the limiting membrane. Type IV cells contained an indented nucleus and moderately developed organelles. The secretory granules were round (180 nm) in shape, and the granule cores with high electron density were enveloped in limiting membrane and showed often a wide halo. Type V cells had a large amount of rough endoplasmic reticulum. Secretory granules with low or moderate electron density were round (230 nm) in shape, and surrounded by limiting membrane and showed a narrow halo. Type VI cells contained an oval nucleus and well-developed organelles, especially Golgi complex. The secretory granules with high electron density were round (210 nm) in shape. The granules were enveloped in limiting membrane and showed often a wide halo.

The Effect of Lithospermum erythrorhizon Extracts in UVB-Irradiated Mouse Skin (자초(Lithospermum erythrorhizon) 추출물이 UVB로 조사된 생쥐 피부에 미치는 영향)

  • This study was intended to identify the effectiveness of Lithospermum erythrorhizon in the UVB-irradiated mouse skin. The C57BL mice were divided into three groups; the control group, the UVB irradiated group(UVB group), and the group treated with Lithospermum erythrorhizon extracts after UVB irradiation(UVB+Le group). 10 mouses were collected and sacrificed at 24 hrs, 48 hrs, 72 hrs, 120 hrs, and 168 hrs, respectively. In the result, the transepidermal water loss (TEWL) was decreased the UVB+Le group than UVB groups by time. At the 168 hrs group was significantly lower(p<0.05). In the result, the melanin value was decreased in the UVB+Le group than UVB group, but meaningless(p>0.05). In the result of erythema index, the UVB+Le group was meaningfully lower at 24 hrs, 48 hrs, and 72 hrs group than UVB group(p<0.05). In the result of scanning electron micrograph observation, the UVB+Le group was allevited swelling than UVB group at the 24 hrs, formation of the scab at the 48 hrs, regular plate shap at the 72 hrs, new keratin observated at the 120 hrs partially, and fine fiber covered epidermis surface at the 168 hrs. In the result of transmission electron micrograph observation, the UVB+Le group was facilitation of increased lamellar bodies and reformation lamellar bodies than UVB group at the all groups. Almost all the structures were recovered at the 160 hrs group. In conclusion, Lithospermum erythrorhizon extracts may recovery on the UVB-irradiated mouse skin.

Scanning electron microscopy on proliferative forms of Toxoplasma gondii and Sarcocystis species (Toxoplasma gondii와 Sarcocystis 원충(原蟲)의 증식형(增殖型)에 대(對)한 주사전자현미경적(走査電子顯微鏡的) 관찰(觀察))

  • For the comparison of surface fine structures in the proliferative forms of two major protoroan parasites, Toxoplasma gondii and Sarcocystis species in mammalian hosts, isolated from the artificially infected mice and from the naturally infected cattle, respectively, an SEM(Hitachi S-570) was applied to the fixed, dried and coated with gold ion on the microslide glasses. The tachyzoites of T gondii from the peritoneal cavity of the mouse showed the crescent-like feature and measured as $5.57{\mu}m$ in length and $2.33{\mu}m$ in width, while the bradyzoites of Sarcocystis species from the heart muscle of slaughtered cattle was banana-shaped and measured as $14.18{\mu}m$ in length and $2.85{\mu}m$ in width. On the surface of Sarcocystis species bradyzoite, a distinct elliptical micropore was identified in the high magnification observation of 60,000X, and it measured as $0.35{\mu}m$ in length and $0.18{\mu}m$ in width.
