The Effect of EDTA and Fetal Cord Serum Supplementation to Ham's F-10 Culture Medium on Developmental Potential of Mouse Embryos In Vitro (Ham's F-10 배양액에 첨가된 태아제대혈청과 EDTA가 백서 수정란의 분할에 미치는 영향)

  • It is the most important to select optimal culture conditions to promote safe embryo growth in the technique of human in vitro fertilization and embryo transfer. It has been shown that the addition of biologic fluids, such as blood serum, of various origins, improved fertilization and early cleavage rates in numerous species. The purpose of this study is to attempt to measure developmental potential of mouse eggs fertilized and cleaved in Ham's F10 culture medium containing a chelating agent, EDTA and fetal cord serum. In this study, we selected 40 female mice and 20 male mice, and investigated optimal serum concentrations for mouse embryo growth. Two cell stage mouse embryos were cultured in Ham's F-10 medium, Ham's F-10 medium with various concentrations of EDTA, or Ham's F-10 medium with EDTA and 10% human cord serum. Developmental ratios to morula in Ham's F-10 medium containing various concentrations of EDTA and/or 10% fetal cord serum were significantly higher than in unsupplemented Ham's F-10 medium (p<0.05). Developmental ratios to blastocyst in Ham's F-10 containing 10% fetal cord serum and $50{\mu}M$ or $100{\mu}M$ EDTA were significanltly higher than in unsupplemented Ham's F-10 medium (p<0.05). Developmental ratios to morula in Ham's F-10 containing 10% fetal cord serum and $100{\mu}M$ EDTA were significanltly higher than in Ham's F-10 with 10% fetal cord serum used commonly in many human IVF centers(p<0.05). Developmental ratio to blastocyst in Ham's F-10 containing 10% fetal cord serum and $100{\mu}M$ EDTA was significanlty higher than in Ham's F-10 with $200{\mu}M$ EDTA(P<0.05). In summary, embryo development to morula and blastocyst was significanlty higher in the presence of human cord serum or EDTA than in the unsupplemented medium. The most significanly development to morula and blastocyst was obtained at Ham's F-10 medium with $100{\mu}M$ concentration of EDTA and 10% fetal cord serum. These results suggest that Ham's F-10 medium containing 10% fetal cord serum and optimal concentrations of EDTA significantly promoted early cleavage of mouse zygotes, and these will be useful as basic data for the selection of culture medium in human in vitro fertilization.

Correlation of Cord Serum Adiponectin and IGF-I with Fetal Growth in Healthy Term Neonates (제대혈 Adiponectin과 IGF-I이 태아성장에 미치는 영향)

  • Purpose : The purpose of this study was to evaluate the relationship between cord serum adiponectin (APN) and IGF-I concentrations and fetal growth. Methods : Umbilical cord serum APN and IGF-I concentrations were measured in healthy term singleton deliveries (n=72). The association of cord serum APN and IGF-I concentrations was evaluated in relation to birth weight, height, head circumference, gender, ponderal index, placental weight, feto-placental (F/P) weight ratio, maternal weight gain, and maternal body mass index (BMI). Results : The mean cord serum APN was 29.2${\pm}$10.46 $\mu$g/mL. The cord serum APN and birth weight demonstrated a bell-shape relationship. The cord serum APN concentration was higher in females than males (P=0.001). The cord serum APN was negatively correlated with maternal BMI (r=-0.301, P=0.027), but the mean cord serum APN concentration was not correlated with birth height, birth head circumference, ponderal index, placental weight, F/P ratio, or maternal weight gain. The mean cord serum concentrations of IGF-I was 51.26${\pm}$21.54 ng/mL. The cord serum IGF-I concentration was positively correlated with birth weight (r=0.312, P=0.009), but not birth height, ponderal index, placental weight, F/P weight ratio, or maternal BMI. Conclusion : APN demonstrated a bell-shaped relationship with birth weight in healthy term infants. IGF-I was highly correlated with fetal growth, especially birth weight.

A Study of the Cholesterol and Lipoprotein in the Maternal and Fetal Serum (산모(産母)와 태아(胎兒)의 혈청 Cholesterol 및 Lipoprotein에 관한 연구)

  • Fifteen cases of primiparas and their offsprings (fetal cord) were investigated with regard their serum total, free and esterified cholesterol by means of Liberman Buchard reaction. The serum ${\alpha}-and\;{\beta}-lipoprotein$ were analyzed by cellulose acetate electrophoresis, and the serum atherolipid numbers were calculated on the bases of the serum total cholesterol and ${\beta}-/{\alpha}-$ lipoprotein ratio, with the following conclusion. 1.Total, free and esterified cholesterol are $178.9{\pm}25.3$, $45.1{\pm}12.6$ and $133.7{\pm}20.6\;mg.%$ in the normal control women, $201.5{\pm}29.5,\;58.7{\pm}42.1$ and $157.1{\pm}26.2\;mg.%$ in the maternal blood, showing hypercholesterolemia in the latter as compared to the former. 2. The serum total, free and esterified cholesterol in the cord blood are $94.5{\pm}20.4$, $32.9{\pm}1.5$ and $61.2{\pm}18.9mg.%$, showing hypocholesterolemia as compared to the control women and maternal blood. 3. The serum ${\alpha}-$, $pre-{\beta}$, ${\beta}-lipoprotein$ and chylomicron are $24.2{\pm}4.2$, $17.3{\pm}3.4$, $51.8{\pm}4.8$ and $6.0{\pm}1.6%$ in the normal women, whereas $14.9{\pm}2.1$, $22.2{\pm}5.1$, $58.7{\pm}3.3 and 3.1{\pm}1.2%$ in the maternal serum, $32.4{\pm}8.1$, $28.8{\pm}2.4$, $25.8{\pm}7.0$ and $3.1{\pm}0.9%$ in the cord serum, showing $hyper-{\beta}-lipoproteinemia$ in the former and $hypo-{\beta}-lipoproteinemia$ in the latter. 4. The serum atherolipid number of the normal control women, maternal cord blood are $4.21{\pm}1.24$, $8.02{\pm}1.42$ and $1.12{\pm}0.37$, showing hyperlipemia in the former and hypolipemia in latter. 5. The relative ratio of the serum free and esterified cholestrol of both normal control women and maternal blood is about 1 : 3, while that of the fetal blood about 1 : 2. 6. The relative ratioes of the serum ${\alpha}-and$ ${\beta}-lipoprotein$ in the control women is about 1 : 2, that of materna blood about 1 : 3 and that of the fetal blood about equal magnitude. 7. The serum esterified cholesterol, ${\alpha}-lipoprotein,\;{\beta}-/{\alpha}-lipoprotein$ ratio and atherolipid number fluctuates are proportionally between the maternal and fetal blood, while the serum free, total cholesterol and ${\beta}-lipoprotein$ between the two vary inversely with statistically significant corelations. 8. It is apparent from the above results that the fetal nutritional demand for lipids resulted from hypocholesterolemia and hypo ${\beta}-lipoproteinemia$ seems to be met satisfactorily by maternal hypercholesterolemia and hyper ${\beta}-lipoproteinemia$, which seems to pose a significant maternal-infant nutritional relationship. A brief ciscussion was made on these conciusion in the light of biochemistry and endocrinology.

Serum Vitamin B(sub)12 Levels of Maternal-Umbilical Cord Blood and Pregnancy Outcomes (임신말 모체 및 제대혈의 비타민 B(sub)12 농도와 임신결과와의 상관성)

  • Vitamin B(sub)12(cobalamin) is an essential nutrient in human and it is particularly important during pregnancy. Nevertheless very few studies have reported, concerning vitamin B(sub)12 in relation with reproduction. This study was conducted to evaluate the vitamin B(sub)12 nutrition status of Korean pregnant women and to investigate the relationship between serum vitamin B(sub)12 levels of maternal-umbilical cord blood and pregnancy outcomes. Dietary vitamin B(sub)12 intakes of the pregnants were estimated by semiquantitative frequency questionnaire. Serum vitamin B(sub)12 levels in both maternal blood and umbilical cord blood of 30 pregnant women at delivery were measured by radioimmunoassay. Mean vitamin B(sub)12 intake was 3.3$\pm$1.4$\mu\textrm{g}$/d which was 125.8% of the Korean RDA(2.6$\mu\textrm{g}$) for vitamin B(sub)12 level of umbilical cord blood was 607.8$\pm$282.9pg/ml, more than two fold of maternal vitamin B(sub)12 level 268.6$\pm$97.8pg/ml. This finding indicates that fetal uptake of vitamin B(sub)12 in the fetus may be due to an active transport mchanism across the placenta. Umbilical cord blood vitamin B(sub)12 levels were highly correlated with maternal levels($r^2$=0.548, p<0.001), showing that fetal vitamin B(sub)12 level is affected by maternal status. However there was no significant correlation between the serum vitamin B(sub)12 levels in maternal-umbilical cord blood and the pregnancy outcomes except for the birth weight. Maternal-umbilical serum vitamin B(sub)12 levels were the highest in the group of birth weight 3.0-3.5kg, and the lowest in the group of birthweight below 3.0kg.   • The development of 2-cell mouse embryos to the blastocyst stage in vitro has been used as quality control for the culture media and supplements employed for human in vitro fertilization and embryo transfer(IVF-ET). 2-cell mouse embryos were cultured to the blastocyst stage in SECM, Medium 199-Earle's, Ham's F-10 I , Ham's F-10 II , Hoppe & Pitts, MEM and $HT_6$. The protein supplements contained in media were bovine serum albumine, fetal bovine serum and human fetal cord serum. The results were as follows; 1. The successful development was 81.3% in Medium 199-Earle’s, 91.9% in Ham’s F-10 I and 97.1% in $HT_6$. 2. 2-cell mouse embryos developed properly in all supplements but the best development was particularly noted in $HT_6$ media when HFCS was supplied as protein supplement.

  • In vitro developmental ability of early preimplantation monse embryos was shown to be depend on the embryonic stages, media and snpplements and their interaction(Experiment 1). The development of I-cell embryos were more promoted in the complex medinm(Ham's Fl0) than in the simple one(m-KRB), but that of 2-cell embryos showed the reverse effect. The bovine serum albumin(BSA) as a medium snpplement more promoted the development of I- and 2-cell embryos, compared with human fetal cord serum(HCS). On the other hand, the harmful effect of HCS was especially shown on the early cleavage in the embryonic development of the two stages. The effect of serum, in the respect of interaction between media and snpplements. was also more significantly appeared in m-KRB than Ham's Fl0. In the experiment 2, when the harmful effect of HCS was compared with that of fetal bovine serum(FBS), the former more promoted the development of l - and 2-cell embryos than the latter. The effect of HCS was more significantly shown in the development of I-cell than that of 2-cell embryos. Conclusively, as I- and 2-cell embryos were different in the requirements for the in vitro development. the optimal medium and supplement have to be selected for each embryonic stage. It is also respected to the better result if it take into consideration into the kinds of sera when serum is used for culture of early preimplantation embryos.

Quality Assay of Human Fetal-Cord Serum for Human IVF-ET with Mouse 2-Cell Embryos (생쥐 2-세포배아에 의한 시험관아기 배양용 대아제대혈청의 절적평가에 관한 연구)

  • The purpose of this study was to examine the qualitative variation of human fetal-cord sera (HCS) and to accept the sera in human lVF-ET program. One hundred and sixteenth RCS were tested with 1772 2-cell embryos of F1 (C57BL x CBA) virgin mice, Ten to sixteenth embryos were cultured in m-KRB medium with a aliquot of each serum (10%, v/v) or with bovine serum albumin(O.4%, w/v) as a control medium. Embryonic development were recorded at every 24hr for 4 days by such events as cellular compaction, cavitation, and hatching. In the control groups of eight assays, 98.1%(106/ 108) of 2-ce1l embryos developed above expanded blastocyst and the embryonic development was unified through the tests. But the developmental pattern in medium with each serum was various. Namely, the sera that supported development of 100% 2-cell embryos to above morula, early blastocyst, expanded blastocyst and hatching blastocyst was 45,7%(53/116) , 35.3%(41/116), 15.5%08/116.) and 6.9-%(8/116), respectively. And the sera that supported development of above 80% 2-cell embryos to the each embryonic stage was 92.2% (107/116), 83.6%(97/116), 63.8%(74/116) and 36.2%(42/116), respectively. Meanwhile two kinds of toxic pattern to the embryonic development were observed in some sera. The first pattern is that some sera arrested development of most embryos in pre- or post-stage of morula or blastocyst. The second pattern is that some sera promoted or arrested a part of embryos in the same dish. The ability of serum was depended on the batch of serum. Finally we could accept 69%(80/116) of the tested sera for human IVF-ET program. The base line for acceptance was the ability that supported above 80% 2-ce1l embryos to blastocyst. But some deterious sera were contained in this range. We cut off about 10% of the sera (83.6% , 97/116) that passed the baseline. This final percent of sera was similar to that of grade N of this study.

Human Cord Serum as a Fetal Bovine Serum Substitute for the Culture of Human Amnion-Derived Stem Cells (인간의 양막유래 줄기세포의 체외 배양 시 소태아혈청 대체제로서의 인간제대혈청)

  • Mesenchymal stem cells (MSC) are promising candidates for cell-based therapies. One major obstacle for their clinical use is the unsafety of fetal bovine serum (FBS), which is a crucial part of all media currently used for the culture of MSC. We investigated the effect of human cord serum (HCS) on the growth response, mRNA and protein expressions of human amnion-derived stem cells (HAM). HAM were isolated from the amnion after a Caesarean section and cultured in DMEM supplemented with 10% FBS, 5% HCS or 10% HCS. During culture, their biological characteristics at earlier and later passages were analyzed using RT-PCR and immunocytochemistry. Regardless of serum sources, HAM showed the prominent expression of Oct-4, Rex-1, SCF, FGF-5, BMP-4, nestin, GATA-4, NCAM and HLA ABC genes. The expression profile was observed even at later passages. Similarly, HAM cultured in either FBS or HCS exhibited the distinct protein expression of collagen I, II, III and XII, fibronectin, $\alpha$-smooth muscle actin, vimentin, CK18, CD54, FSP, TRA-1-60, SSEA-3, -4 and HLA ABC. However, desmin expression was only observed in HAM cultured in medium supplemented with FBS and vWF expression was only found in HAM cultured in medium supplemented with HCS. Overall pattern of gene and protein expression of HAM was typical of known adult stem cells such as bone marrow-derived MSC. In conclusion, HCS could be as effective as FBS for the culture of HAM.

  • This study was performed to investigate the effect of human follicular fluid (HFF) on development of mouse embryos, for evaluating the suitability of HFF as a substitutive material of human fetal cord serum in ART program. The various concentrations of HFF were added into the culture medium and the effects of HFF concentrations were examined to identify the optimal concentration of HFF for embryo development. The potency of HFF in improving embryo development was compared to that of other protein supplement. Collected HFFs were classified with the maturity of the containing oocytes; mature, immature, atretic, and then the effects of the classified HFFs on embryo development were examined. Also, HFF was separated into the low (<30,000 Da) and high (>30,000 Da) molecular weight fractions and the effects of the fractions on embryo development were investigated. The highest development rate was found in culture medium supplemented with 20% HFF, bnt this rate was reversely reduced at the concentrations of HFF higher than 20%. The development rates to the blastocyst, hatching blastocyst, attachment and outgrowth cultured in mature HFF was significantly higher than those in immature and atretic HFF, and mean cell number in blastocyst was higher in mature HFF than in immature and atretic HFF. The development rates of mouse embryos according to protein sources were significantly higher in HFF than in fetal cord serum (FCS), maternal serum (MS) and bovine serum albumin (BSA), and mean cell number in blastocyst cultured in HFF was higher than that in FCS, MS and BSA. The development rates of embryo and mean cell number in blastocyst cultured in high molecular weight fraction of HFF were higher than those in low molecular weight fraction, but the results of high molecular weight fraction were lower than those of whole HFF. Therefore, these results indicated that human mature follicular fluid was useful for improving the development of mouse embryos, which suggests a possibility that HFF also may be used efficiently for improving the culture condition in human ART program as a protein supplement.
