Effects of Salinity and Standard Toxic Metals (Cu, Cd) on Fertilization and Embryo Development Rates in the Sea Urchin (Strongylocentrotus nudus) (염분과 표준 독성물질(Cu, Cd)이 둥근성게(Strongylocentrotus nudus)의 수정 및 배 발생률에 미치는 영향)

  • Effects of salinity and standard toxic metals on fertilization and embryo development rates were investigated in the sea urchin, Strongylocentrotus nudus. Spawning was induced by injecting 1ml of 0.5 M KCl into the coelomic cavity. The fertilization and embryo development rates were below 20% when salinity was 25 psu or lower, but were above 90% when salinity was between 30 and 35 psu. The fertilization and embryo development rates in the control condition (not including Cu and Cd) were greater than 90%, but decreased with a high negative correlation (r) of 0.89 and 0.91 with the increasing of Cu and Cd concentrations, respectively. These results suggest that salinity concentrations for successful fertilization and normal embryogenesis of S. nudus are between 30 and 35 psu, and the biological assays of fertilization and embryo development rates using S. nudus are useful methods for the ecological toxicity test of marine pollution elements.

Studies on the Fertilization Rates using Intracytoplasmic Sperm Injection with In Vitro Matured Porcine Oocytes (돼지 체외성숙 난자의 세포질내 정자주입에 의한 수정에 관한 연구)

  • This study was carried out to investigate on the improvement of fertilizing ability of in vitro matured oocytes from sperm density and motility by intracytoplasmic sperm injection(ICSI) into the porcine oocytes. 1. The in vitro fertilization and cleavage rates of oocytes from 1.0, 2.0, 3.0, 5.0 ($\times$10$^{6}$ $m\ell$) sperm concentration by IVF and ICSI of porcine oocytes were 46.7%~75.0%, 60.0%~85.7% and 10.6%~25.0%, 20.0%~64.3%, respectively. 2. The in vitro fertilization and cleavage rates of oocytes from 20, 40, 60, 80% of sperm mortilty by IVF and ICSI of porcine oocytes were 46.4%~71.4%, 67.9%~85.7% and 7.1%~21.4%, 28.6%~60.7%, respectively. 3. The in vitro fertilization and developmental rates of oocytes by IVF and ICSI methods were 55.6%~60.0%, 77.8%~80.0% and 17.8%~24.0%, 42.2%~56.0%, respectively. This ICSI method was improved high fertilization rates of porcine oocytes.

  • Micromanipulation procedures have been used to improve fertilization rates in patients with male factor or with unexplained infertility. Partial zona disseetion(PZD), a method using mechanical force to open the zona pellucida increase the chances of fertilization. The purpose of this study is to increase rates of fertilization and pregnacy in the ART program by using PZD. The influence of PZD on the fertilization rate was investigated in 57 couples with semen defects, antisperm antibodies(ASA), or unknown factors. PZD directly performed in 35 couples with a history of fertilization failure in previous cycle (Group 1), and PZD applied in 22 couples with the failure of initial fertilization in the same cycle (Group 2). The fertilization rates of the male facor, ASA positive factor and unknown factor in Group 1 were 37.6%, 20.0% and 59.2%, respectively. The rates of fertilization of male factor, ASA positive factor and unknown factor in Group 2 were 34.8%, 20.0% and 26.5%, respectively. The incidences of polyspermy in Group 1 and Group 2 were 5.9% and 9.0%, respectively. Among 35 patients of Group 1, one patient was pregnant and successfully delivered, whereas 1 of 22 patients of Group 2 became pregnant, but aborted at 7 weeks.

Fertilization and Tree Density Effects on Cellulose Decomposition in a Larix leptolepis Plantation

  • Cellulose mass loss by cellulose filter papers was measured for 3 time (35 days, 70 days, 105 days) incubation during the growing season (from May to September 2002) with different tree density and after fertilization in a Japanese larch (Larix leptolepis) plantation. Cellulose mass loss rates were significantly different between tree density types and fertilization treatments during the study periods. After 105 day incubation of cellulose filter paper, cellulose mass loss rates were significantly higher in the low tree density (70.1 $\%$) than in the high tree density (49.9$\%$). Cellulose mass loss rates averaged 62.8$\%$ in the fertilization and 58.9% in the unfertilization treatments during the same periods. However, cellulose mass loss was not significantly different between the forest floor and the mineral soil layer except for 35 day incubation. The results indicate that cellulose decomposition rates are a useful index to express differences in organic matter decomposition activity in different tree density and after fertilizer treatments.

Studies on the In Vitro Fertilization and In Vitro Development of Porcine Embryos (돼지 난포란의 체외수정 및 체외발달에 관한 연구)

  • To improve the efficiency of in vitro production of embryos with follicular oocytes in pig, the recovery rates, in vitro fertilization and development. The results obtained were as fellows: The number of oocytes recovered 37 ovary was 1,365 by aspiration, 1,884 by slicing and 3,830 aspiration post slicing, per ovary was averaged 103.5 aspiration post slicing than 30.7 by aspiration and 50.8 by slicing (P<0.05). The percentage of grade I and II oocytes recovered was 0.05∼0.2% and 1.7∼2.3% respectively(p<0.05). The fertilization rates of ejaculate and epididymis sperm was 83.0 and 83.1%. And cleavaged rate was 60.8 and 69.0% respectively(P<0.05). However, there were no significant differences between sperm sources. The clevage rates of fertilized oocyte was significantly(P<0.05) higher as B.O(92.8%) than TALP (90.1%) or mTBM (80.1%). And in vitro developed to blastocyst rates of mTBM media used for fertilization was significantly (P<0.05) higher as 12.4%, compared with the results using the media of TALP(1.6%) or B.O (0.0%). The embryos developed 2-cell stage after in vitro fertilization were co-cultured with or without POEC and BOEC in NCSU-23 and TCM-199 media. In vitro developed to blastocyst rates was NCSU-23 with POEC(2.3%) or BOEC(1.2%), but in vitro cultured in TCM-199 medium with POEC or BOEC was not developed to blastocyst. The percentage of embryos that developed to morula stage in 0, 50, 100, 200 and 250uM was 16.6, 22.0, 13.5, 19.0 and 22.0%, respectively.

Effects of Long-Term Fertilization for Cassava Production on Soil Nutrient Availability as Measured by Ion Exchange Membrane Probe and by Corn and Canola Nutrient Uptake

  • The effects of long-term fertilization on soil properties and nutrient availability are not well documented for cassava cultivation in Vietnam. In 1990, a field research plots were established with 12 treatments to test the effect of different rates of nitrogen (N), phosphorus (P) and potassium (K) on soil properties in Acrisols at Thai Nguyen University in Northern Vietnam. In 1999, composite soil samples (0 to 20cm depth) were collected from eight selected plots for measurements of nutrient supply rates by ion exchange membrane probes and for growing corn and canola in a growth chamber with and without added lime. Generally, long-term nitrogen (N) fertilization increased available N supply rates but decreased available potassium (K) and magnesium (Mg). Long-term phosphorus(P) applications increased canola N, calcium (Ca) and Mg uptake. Canola P uptake increased with increased P rates only when lime was added. Long-term K applications increased canola N, K, Ca, Mg uptake but only significantly increased corn N uptake. Liming significantly increased uptake of N, P, K, Ca, Mg and S for both corn and canola. However, N $H_{4-}$N, K and Mg soil supply rates were reduced when lime was added, due to competition between Ca from the added lime and other nutrients.

Effects of hCG on Sexual Maturation in Korean Loach (hCG가 한국산 미꾸라지(Misgurnus mizolepis)의 성성숙에 미치는 영향)

  • This stduy was carried out to obtain basic information available on ovulation, spawning, fertilization rate, hatching rate, and deformity rate after hCG injection in cyprinid loach, Misgurnus mizolepis. The results obtained in these experiments were as follows: 1. Ovulation and spawning occurred simultaneously and spawing was completed within 1 hour after ovulation. 2. More than 80% of fertilization rates appeared within 12 hours at 21$^{\circ}C$, 8 hours at $25^{\circ}C$, and 4 hours at 29$^{\circ}C$, respectively, following the onset of spawing. Afterwards, the fertilization rates of released eggs sharply decreased in three different water temperatures. 3. More than 70% of hatching rates appeared within 8 hours at 21$^{\circ}C$, 6 hours at $25^{\circ}C$, and 2 hours at 29$^{\circ}C$, respectively, following the onset of spawing. Afterwards, hatching rates of spawned eggs abruptly decreased in three different water temperatures. 4. The deformity rates of hatched larvae were high at $25^{\circ}C$, 8 hours following the onset of spawing. 5. Based on the developmental ability of oocytes, the optimum time of fertilization was 4 hours (stage 5) following the onset of spawing.

Effect of LH Bioactivity on Fertilization and Cleavage Rates of Mature Oocytes in Hyperstimulation Cycles for IVF-ET (과배란유도시 난포기 LH의 생물학적 활성이 성숙난자의 수정율 및 난할율에 미치는 영향에 관한 연구)

  • To investigate the factors that affect the fertilization and cleavage rates of mature oocytes, 44 patients undergoing controlled ovarian hyperstimulation(COH) with FSH/hMG/hCG regimen for IVF - ET were analyzed. During follicular phase, serum LH levels were measured by radioimmunoassay and bioassay. Based on the mean follicular immunoactive LH(i-LH) and bioactive LH(b-LH) levels, patients were divided into 3 groups, respectively. There were no significant differences in basal serum FSH levels on menstrual cycle day 3, serum estradiol($E_2$) and progesterone ($P_4$) levels on the day of hCG administration, and the numbers of follicles aspirated and oocytes retrieved among groups. In relation to the mean follicular i-LH levels, the fertilization and cleavage rates of mature oocytes did not show a significant difference among groups. However, in groups with higher mean follicular b-LH levels, the fertilization and cleavage rates were reduced significantly. During late follicular phase, day-to-day variance in b-LH levels was not significant, but there was a significant difference among groups. There was no significant correlation between serum P. and b-LH levels. These data suggest that the fertilization and cleavage rates of mature oocytes are adversely affected by the raised mean follicular b-LH levels, and monitoring of serum b-LH levels is more useful in COH when compared with i-LH. It appears that the reduced rates are not due to the attenuated endogenous LH surge.

  • This experiment was carried out to study the effect of pure nitrogen fertilization on botanical composition of grassland in 1987~ 1988 under practical conditions at the " Federal Institute for Agriculture in the Alps" in Austria. The application rates were 0, 30, 60, 90 and 120kg/ha/cut, the cutting regimes 3-, 4 , 5- and 6-cuts/year. The results were as follows: 1. Only PK fertilization resulted in higher botanical composition of Trifolium repens among legumes in grassland, which was increased by cutting frequency .2. Nitrogen fertilization on three-cut areas resulted in higher existence of generally valuable tall grasses such as Arrhenotherum elatius Trisetum flavescens and Dactylis glomerata . On the other hand, nitrogen fertilization on four-, five- and six-cut areas showed mainly Doctylis glomerato and Poa pratensis appearance. 3.For some cases, appearance of less valuable grasses, herbs and weeds such as Agropyron repens, Poa trivialis, Poa annua, Setoria viridis, Aegopodium podagra rio, Men landrium rubrum, Taraxacum officlnale, Achillea millefolium, Rorippa sylvestris and Polygonum ocleulare was *Bundesanstalt fur alpenlandische Landwirtschaft Gumpenstein(A-8952 Irdning, 6sterreich) increased on medium and high rates of N fertilized areas. 4.Reduction of sward density may also diminish the advantages of nitrogen fertilization and may be threatened by mainly high dressings of nitrogen combined with too late utilization of the sward. 5.Location altered the efficiency of nitrogen fertilization on botanical composition.mposition.

The Recovery, In Vitro Maturation and Fertilization of Nili-Ravi Buffalo Follicular Oocytes

  • Four types of serum supplements viz. estrus cow serum (ECS), estrus buffalo serum (EBS), pro-estrus buffalo serum (PrBS) and post-estrus buffalo serum (PtBS), added to TCM-199, were evaluated for in vitro maturation and fertilization of buffalo follicular oocytes. The oocytes were recovered from buffalo ovaries after slaughter, using either aspiration or scoring (multiple incisions) method. The recovered oocytes were categorized as A, B and C based on their cumulus investment and ooplasm homogeneity and cultured in four media. The in vitro matured oocytes were inseminated with $1{\times}10^6$ spermatozoa washed in 2.9% sodium citrate solution. The scoring method yielded greater number of morphologically good oocytes than the aspiration method (3.85 vs 1.76 per ovary, p < 0.01). The maturation rates of three categories of oocytes did not differ from one another. The maturation rates of 80.00, 82.08, 78.77 and 66.23%, while the fertilization rates of 54.54, 55.38, 52.80 and 36.76% were recorded for media containing ECS, EBS, PrBS, and PtBS, respectively. The medium containing PtBS gave lower maturation, as well as fertilization, rates than the other three media (p < 0.05). Thus, the scoring method was better than the aspiration method for the recovery of follicular oocytes. The oocytes categorized A, B and C had similar maturation capabilities. The TCM-199 containing buffalo/cow serum collected at pro-estrus or estrus appeared better for in vitro maturation and fertilization of buffalo follicular oocytes than that containing serum collected at post estrus.
