Breeding for Potato Virus Y Resistant Male-sterile $F_1$ Hybrid KB 109 in Nicotiana tabacum L. (연초의 감자바이러스 Y 저항성 웅성불임 일대잡종 KB 109 육성)

  • Potato Virus Y(PVY), vein necrosis strain, in Korea causes severe symptoms on burley tobacco(Nicotiana tabacum L.). As the results, programs to incorporate PVY resistance into commercial cultivars were initiatEd. But the development of the homozygous fertile line resistant to PVY is time consumming. This study was conducted whether the Fl hybrid could be used to reduce the yield losses caused by PVY. Four F1 hybrids were made between male - sterile(ms) NC 107 and KB 107 as maternal parent, and TC 612 and TC 613 as Pollen donor, respectively, and were evaluated for their PVY resistance and negatively associated traits. (ms NC 107 X TC 612) F1, named as KB 109, Ivas applied to yield trial and compared with commercial cultivars for the level of disease resistance, agronomic characteristics, chemical contents and physical properties. All Fl hybrid could be used commercially as the PVY resistant cultivar. Especially KB 109 have the resistance against PVY, tobacco mosaic virus and black shank(Phytophthora parasitica var. nicotianae). It had wider leaves, flowered one day later, and yield of acceptable quality was higher than that of Burley 21, standard cultivar in Korea.

STUDIES ON THE DIMORPHISM OF THE PERSICARIA SENTICOSA NAKAI (Persicaria senticosa Nakai의 Dimorphism에 관한 연구)

  • HARN, Chang Yawl (Chonnam U. Kwangju, Korea): Siudies on the dimorpism of the Perisicaria senticosa Nakai-Kor. jour. Bot. 3(1) 16-25 During his researches regarding the morphological and physiological properties of Polygonecenae, the author has found that the species, Persicaria senticosa, aiso, besides the heterostylous plants of Polygonum family, Fagopyrum esculentum, and Persicaria japonica which was recently verified by the author as dimorphic, shows the typical floral structure of heterostylism, the description of which is not found in taxonomical works. Further research on this species have revealed that this plant, despite pressessing characteristic structural dimorphism, does not exhibit even the slight signs of heterostlylous properties physiologically. This is a deviation from the usual behavior of normal heterostylous plants. What is interesting is the fact that the physiological behavior of this species is quite contrary to that of P. japonica which is considered to be the most highly specialized dimorphic plant. Thus it is assurred that if some species of this family had taken a heterostylic form in its course of development from autogamy to allogamy, P. seoticosa would be the least differentiated type of dimorphic forms among the three heterostylous plants, including buckwheat, of this family. The results obtained in this experiment are summarized as follows: 1) P. senticosa has two forms of flower, one, long style-short stamened; the other, short style-long stamened. Not only conspicuous is this primary difference, but the secondary difference, such as pollen grain size, is noticeable between long and short styled individuals, thus expressing structurally the definite trait of a dimorphic plant. 2) Structural alteration of floral parts towards dimorphism has preceeded far less in comparison with those of the P. japonica and F. esculentum. 3) Elaborate studies on fertility reveal that this species does not show the slightest sign of the physiological characteristics of dimorphic plants. In other words, regardless of the modes of combinations, legitimate and illegitimate, fertilization and fruit setting flourish unimpaired. 4) Growth of pollen tubes apparently parallels the results in the fertility, tubes reaching ovary approximately 30 minutes after pollination both in legitimate and illegitimate combinations. Pollen tube penetration appears to be comparatively rapid. 5) A slight difference in the growth of pollen tube seems to exist between legitimate and illegitimate combinations, legitimate union giving slightly faster tube penetration. 6) In the present experiment it was clarified that P. senticosa, known to possess one form of flower in taxonomy, is in reality dimorphic plant having two of flowers. Although this species is definitely heterostylous in floral structure, physiological evidence and pollen tube behavior show that the differentiation of this plant toward the dimorphism has apparently proceeded slightly except for some parts of floral organ. In ordinary heterostylous plants it is a matter of common occurrence that when illegitimately cmbined, there is poor or no fertility, Contrary to the universal property of heterostylous plants, no difference is observed in the fertility and pollen tube growth between the legitimate and illegitimate combinations in the case of P. senticosa. Compared to the P. japonica and F. esculentum, which are supposed to have undergone high degree of dimorphic differentiation, it is an unavoidable conclusion that P. senticosa has not yet developed as a heterostyle plant except for some of its floral parts. If P. japonica is assumed to be the most differentiated type of hetenostylous plant, thenthe P. senticosa would be regarded as the primitive, retaining still the self-fertile nature so common to the Polygonum genus. In nature, however, this plant has a better chance to be pollinated legitimately owing to the two forms of flowers than to be pollinated illegitimately. Characteristics of Flowering and Leaf Emergence in Lycoris Species (상사화류의 개화와 출엽 특성)

  • This study was conducted to examine the pattern and characteristics of flowering and leaf emergence for twenty two kinds of the genus of Lycoris sp. One species of Lycoris was flowered on July to August, twelve species on August, and nine species on September. Shape of flowers were as follows : Nine species belong to L. radiara-shaped flower, seven species L. squamigera-shaped flower, and seven species the intermediate type. Flower color of three kind were an order of descent red and pink. seven kind white, four kind yellow, three kind orange, and two kind purple colors. Ten Lycoris species were good fertile, and two kinds were poor fertile. The average length of pollen grain was ranged from $48.53\;to\;88.62{\mu}m$, and the width from $22.87\;to\;33.67{\mu}m$. Eleven kinds were spring-leaf emergence type, and ten kind autumn-leaf emergence types. Hunter values in leaves were as follows : $L^*$ value was ranged from 31.6 to 45.2; $a^*$ values from -6.74 to -17.46; $b^*$ values from 6.19 to 21.89 Leaf width was ranged from 0.90 to 2.35cm, and its shape was oblong. The length of epidermal cells was 0.33 to 0.75mm, and the width was $38.53\;to\;90.00{\mu}m$. Most of stomata were distributed in the back side of leaves. The length of stomata was ranged from $67.12\;to\;104.89{\mu}m$, and its width was from $14.90\;to\;71.52{\mu}m$.

Crossability, Variation in Chromosome Number, and Agronomic Characters in Hybrid of Hexaploid Triticale with Tetraploid Rye (6배체 트리티케일과 4배체호밀 교잡에서의 교잡율, 염색체수 변이 및 작물학적 특성)

  • This experiment was carried out to investigate seed set, variation in chromosome number, and agronomic characteristics of the progeny in the cross between hexaploid triticale variety, Sinkihomil(P$_1$) and tetraploid rye variety, Dooroohomil(P$_2$). Seed set rate obtained was 30.5% in the cross of Sinkihomil with Dooroohomil, whereas 3.26% in reciprocal cross using Dooroohomil as female. Alsoseed set was 8.75% in F$_1$/P$_1$, 7.20% in F$_1$/P$_2$, and 1.53% in F$_2$(=F$_1$ /F$_1$, respectively. Germination rate of crossed seed was 37% in cross of P$_1$ with P$_2$, 39.0% in F$_1$/P$_1$(BC$_1$), 50% in F$_1$/P$_2$(BC$_2$) and 43.0% in F$_1$/F$_1$(F$_2$), and 1,000 grain wight was 20.7g in the cross of P$_1$ with P$_2$, which have 41.9g and 47.7g, respectively, 24.5g in F$_1$/P$_1$, 23.6g in F$_1$/P$_2$, and 24.5g in F$_1$/F$_1$, respectively. In pollen fertility of F$_1$ plant, 69.8% turned out to be abnormal or sterile pollen grains, whereas 30.2% was fertile or normal. In meiosis of pollen mother cell of F$_1$ plant, 13.5 univalents, 8.89 bivalent and 1.24 trivalent were appeared. Somatic chromosome number of 35 in F$_1$, both 32 to 33 and 35 to 36 in F$_2$, 35 to 39 in BC$_1$ and 28 to 36 in BC$_2$ which mean producing female gamate was 14 to 18 chromosome in PMC of F$_1$ plant. Rate of fertile plant turned out to be 100% in F$_1$, 4.5% in F$_2$, 42.9% in BC$_1$, and 50.0% in BC$_2$, respectively. Number of seed set per spike appeared to be 1.17 in F$_1$ plant, 13.3 in F$_2$, 2.36 in BC$_1$, and 3.75 in BC$_2$, respectively. Days to heading of F$_1$ was intermediate, but F$_2$ was later than both parents. Plant height of F$_1$ , BC$_1$ ,and BC$_2$ was shorter than both parent, but F$_2$, longer than both parents.

  • Selection procedures for breeding lines resistant to both bacterial wilt caused by Ralstonia solanacearum and Phytophthora blight caused by P. capsici were executed to generations from $F_2$ and $BC_1F_1$ to $F_4$ and $BC_1F_3$ of crosses between a Phytophthora resistant breed, 'Chilbok No. 1' and bacterial wilt resistant accessions introduced from Vietnam during 2009 and 2010. The breeding populations or lines were tested for resistance to P. capsici and resistant plants were selected. The resistant selections were inoculated with R. solanacearum to discard susceptible plants. Resistance to P. capsici was conspicuously improved by selection from $F_2$ and $BC_1F_1$ and the final selections showed a similar level of resistance to P. capsici as a commercial Phytophthora resistant cultivar, Muhanjilju. A few $BC_1F_2$ selections were crossed to a cytoplasmic male sterile line, Chilbok-A, to identify their nuclear genotype interacting with male sterile cytoplasm. Majority of them was fixed to maintainer (Nrfrf) and only two resulted segregating into male sterile and male fertile plants indicating that the pollen parents were heterozygous in the fertility-restoring gene.

Enhanced Seed Development in the Progeny from the Interspecific Backcross (Fagopyrum esculentum ${\times}$ F. homotropicum) ${\times}$ F. esculentum

  • To facilitate the introgression of F. esculentum into the traits of F. homotropicum, several accessions of the hybrids between these two species were pollinated with F. esculentum as the recurrent parent. The embryo in vitro rescue was performed to increase the recovery of backcross progenies. The $F_{2}$ generation was more amenable than $F_{1}$ hybrids to produce backcross progenies. The $F_{1}$ hybrids were backcrossed twice with common buckwheat (pin-type F. esculentum) (recurrent backcrossing). Also, alternate backcrosses with common buckwheat and F. homotropicum (congruity backcrossing) were carried out. Pollen tube growth of BC$F_{1}$ ${\times}$ F. esculentum (thrum) and F. homotropicum ${\times}$ BC$F_{1}$ was the disturbed penetration exceeded for all initial interspecific hybrids, and its requirement was proportionally lower when the common buckwheat was used as the recurrent parent and as the last parent of congruity hybrids. Effects of both common buckwheat and F. homotropicum on seed success rate for hybridization were observed. Growth of hybrid embryos before rescue, regeneration of mature hybrids all increased recurrent and congruity backcrosses and inter-crosses between $F_{1}$ plants and selected fertile plants of the second congruity backcrosses.

Application of the Molecular Marker in Linkage Disequilibrium with Ms, a Restorer-of-fertility Locus, for Improvement of Onion Breeding Efficiency

  • To analyze the linkage relationships among molecular markers recently reported to be linked to onion (Allium cepa L.) Ms, a restorer-of-fertility locus, in onion (Allium cepa L.), three single nucleotide polymorphism markers were converted into cleaved amplified polymorphic sequence (CAPS) markers based on onion transcriptome sequences and the rice genome database. Analysis of the recombinants selected from 4,273 segregating plants using CAPS and other linked markers demonstrated the jnurf13 and jnurf610 markers to perfectly co-segregate with the Ms locus. In contrast to jnurf13, the jnurf610 marker was not in perfect linkage disequilibrium with the Ms locus in diverse breeding lines. Thus, the jnurf13 marker and the marker for identification of cytoplasm types were utilized to enhance the efficiency of onion breeding through four applications. First, 89 maintainer lines containing the normal cytoplasm and homozygous recessive Ms genotypes were successfully identified from 100 breeding lines. Second, these two molecular markers were used to analyze the main sources of male-fertile contaminants frequently found in the male-sterile parental lines during F1 hybrid seed production. The majority of the contaminants contained heterozygous Ms genotypes, indicating that pollen grains harboring the dominant Ms genotype may have been introduced during propagation of the maintainer lines. Therefore, the genetic purity of the two maintainer lines was analyzed in the third application, and the results showed that both maintainer lines contained 13-21% off-types. Finally, the two markers were used to increase the seed yield potentials of two open-pollinated varieties containing sterile cytoplasms by removing the plants harboring homozygous recessive and heterozygous Ms genotypes.

Studies on the Taxonomical Characters in Populus tomentiglandulosa and P. glandulosa (Populus tomentiglandulosa와 P. glandulosa의 분류학적(分類學的) 연구(硏究))

  • Populus glandulosa and Populus tomentiglandulosa, which were known to be natural hybrids, were examined for morphological, physiological and karyological traits to illucidate its hybridity and taxonomical importance. The results abtained were as follows; 1. Survival rate in rooting of cuttings and grafting was different between the hybrids and their rooting abilities showed incomplete dominance. 2. Their leaf openings showed incomplete dominance. The leaf longevities of P. alba ${\times}$ glandulosa and P. tomentiglandulosa were stronger than the other hybrids. 3. There were differences in resistance to toxicity of $KClO_3$ between the hybrids. 4. Many external leaf characters of the hybrids also showed incomplete dominance. P. tomentiglandulosa was similar in those characters to P. alba ${\times}$ glandulosa while P. glandulosa was similar to hybrids crossed, reciprocally crossed or back-crossed between P. davidiana and P. alba. 5. Their numbers of male flower showed incomplete dominance or hybrid vigor. The numbers of P. tomentiglandulosa were similar to thosa of P. alba ${\times}$ glandulosa while those of P. glandulosa to those of P. alba ${\times}$ davidiana or P. davidiana ${\times}$ alba. 6. Morphology and band color of male catkin bract showed incomplete dominance. Those of P. glandulosa were similar to those of P. alba ${\times}$ davidiana while those of P. tomentiglandulosa to those of. P. alba ${\times}$ glandulosa. 7. There were differences in vascular bundle number and arrangement of petiole between the hybrids. 8. Differences in the anatomical traits of stem did not exist between the hybrids but those in wood fiber size existed. 9. The chromosomes of artificial hybrids, P. glandulosa and P. tomentiglandulosa showed irregular behavior in metaphase I and II. 10. All hybrids including P. glandulosa and P. tomentiglandulosa showed small number of P.M.C. with 19 II but many univalent chromosomes were exhibited in metaphase I. 11. All hybrids including P. glandulosa and P. tomentiglandulosa showed a little abnormal nuclear plates as laggard chromosome and chromosome bridge in anaphase I and II. 12. The frequency of pollen tetrad and fertile pollen was low in most of the hybrids including P. glandulosa and P. tomentiglandulosa.

Mid-late Maturing Male Sterile Line of Onion (Allium cepa L.) 'Wonye 30001' (양파 중생계 웅성불임 중간모본 '원예 30001')

  • A new intermediate parent 'Wonye 30001' (Allium cepa L.) was developed by Bioenergy Crop Research Center, NICS in 2009. As a male sterile line, 'Wonye 30001' could be used for hybrid seed production by crossing with pollen parent. The first cross was conducted in 2000. Male sterile plants were selected from 'Ginque' breeding lines and fertile plants were selected from YG-1-1. The male sterile line 'Wonye 30001' has circular bulb and bulb weight of 260 g. As mid-late maturing type, lodging date is around May 20. Plant height and stem diameter are 57 cm and 15.9 mm, respectively. In seed harvesting characteristics, number of flower stalks and the length are 6 and 135cm, respectively. Flowering date of 'Wonye' is May 23 and is completely male sterile. 'Wonye 30001' is a promising male sterile line for hybrid bulb onion seed production.

  • The sterile phenomenon is frequently found in the inter-species hybrids of ginseng as in other plants. It is known that among the hybrids between Panax Ginseng (PG) and Panax Quinquefolium (PQ), and between Panax Ginseng and Paxax Japonicus (PI), PG${\times}$PI is fertile only very rarely, while PG ${\times}$ PQ is always sterile. Therefore, in order to clarify the relationship between this sterility phenomenon and the metabolism of free amino acids, the changes of free amino acids through the formation of the flower organs and seeds of two hybrids, PG ${\times}$ PQ and PG ${\times}$ PI were investigated by thin layer chromatography. The results are summarized as follows: 1. Distinct differences in the quantity and number of free amino acids were recognized between PG ${\times}$ PQ, PG ${\times}$ PI and their parent plants. From the hybrid PG ${\times}$ PQ, 19 kinds of ninhyrin sensitive substances were detected in all. They were (1) 17 amino acids: alanine, valine, leucine, phenylalanine, proline, hydroxy-proline, serine, threonine, tyrosine, aspartic acid, glutamic acid, lysine, arginine, ${\gamma}$-amino butyric acid, ${\beta}$-alanine, cysteic acid and tryptophan, and (2) two amides: asparagine and glutamine. From the hybrid PG ${\times}$ PI, in addition to the above 19 substances, methionine and one unknown substance were detected. 2. Generally, alanine, as partie acid, glutamic acid, cysteic acid and asparagine were detected in large amounts in the two hybrids as in PG, PG and PJ but it was a noticeable fact concerning these two hybrids that the largest quantity of asparagine was found at microspore satge and pollen mature stage. 3. The decrease of cysteic acid in the two hybrids at the red ripened stage was the same as in PQ and PJ but opposite to the change in PG. The detection of methionine in PG ${\times}$ PJ was worthy of notice. 4. The change of proline was conspicuously different from that in their parent plants. It was detected as a trace of color at the micros pore stage while asparagine was detected in the greatest amount at that time. It is well known that the quantity of proline is closely related to the sterility of plant. This fact was also found true in the formation of ginseng seeds. It was reported as well that asparagine accumulated when proline decreased. 5. The deficiency of proline seemed to be closely related with the sterility of hybrids and with the degradation of pollen in anther. 6. The difference in the changes of free amino acids between the selfed lines of PG, PQ and PJ, and their hybrids seemed to be caused by the transformation of gene-action system by hybridization. On these phenomena along with proline metabolim and its physiological role in seed formation further studies are required.
