FeO/Fe(II) 시스템에서 TCE의 제거 특성 (Characteristics of the TCE removal in FeO/Fe(II) System)

  • 철의 환원 특성에 관한 연구는 이미 널리 수행되었으며 특히 미네랄과 2가철의 반응 메커니즘은 2가철의 흡착이나 바운드를 통해 Fe(II)-Fe(III) (hydr)oxides를 생성하여 2가철이 3가철로 산화됨으로써 물질을 환원시키는 것으로 받아들여지고 있다. 그러나 2가철로 개질된 재강슬래그를 이용한 DS/S 실험과정에서 이러한 메커니즘으로 설명하기 힘든 현상을 발견하였다. 재강슬래그의 주요 성분중의 하나인 FeO와 Fe(II)만을 이용하여 TCE의 분해과정을 실험해 본 결과 초기 TCE의 분해가 이루어지지 않다가 급속히 분해되는 현상을 보였으며 이러한 시스템에서 TCE의 분해는 예상치 못한 결과였다. FeO/Fe(II) 시스템은 3가철이 존재하지 않기 때문에 기존의 Fe(II)-Fe(III) (hydr)oxides를 형성하는 환원 메커니즘으로는 설명할 수 없었다. 따라서 본 연구에서는 TCE의 분해실험과 분해 부산물의 측정, 2가철과 3가철을 확인함으로써 FeO/Fe(II) 시스템의 환원특성을 확인해 보고자 하였다. 실험 결과 2가철이 FeO에 흡착 또는 바운드 되는 것을 확인 할 수 는 있었으나 기존의 메커니즘으로 설명하기에는 부족한 부분이 있었다. 분해부산물들을 통해 환원으로 인한 TCE의 분해는 의심의 여지가 없었으나 FeO/Fe(II) 시스템이 새로운 species를 형성하는지, 혹은 FeO에 Fe(II)가 흡착 또는 바운드 되어 이제껏 알려지지 않은 형태의 새로운 미네랄 상을 형성하는지는 좀 더 상세한 연구가 필요하다.

Pseudomonas synxantha A3가 생성하는 siderophore A3에 관한 연구 (Biochemical studies of the siderophore A3 produced by pseudomonas synxantha A3)

  • A yellow-green, fluorescent siderophore A3 was extracellularly produced under iron-limited growth conditions from Pseudomonas synxantha A3. The physicochemical and biological properties of siderophore A3 were examined. The approxiamte molecular weights of the Fe(III)-siderophore A3-1 complex and Fe(III)-siderophore A3-2 complex were estimated to be about 1,300 and 1,100, respectively, by Bio-gel P2 gel exclusion chromatography. The molar ratio between the siderophore and the Fe(III)was 1.08 mole. The molecular weight of the complex could be calculated with this ratio and the new values were 1,150 and 960, respectively. The binding constant(K) between thesiderophore A3 and Fe(III) that determined by displacing the iron from the Fe(III)-siderophore complex with EDTA was 4.12*10$^{18}$ at pH 5.0. Siderophore A3 appeared to have antibacterial activity on several bacterial strains, however, ferric siderophore Ae complex did not show that activity. The cytotoxicity of siderophore A3 was obtained from Human Chronic Myelogenous Leudemia K562 cells. Inhibition concentration (50%)($IC_{50}$ ) was $0.17\mu$\{g/ml}.

Dual positional substrate specificity of rice allene oxide synthase-1: insight into mechanism of inhibition by type II ligand imidazole

  • Phylogenetic and amino acid sequence analysis indicated that rice allene oxide synthase-1 (OsAOS1) is CYP74, and is clearly distinct from CYP74B, C and D subfamilies. Regio- and stereo-chemical analysis revealed the dual substrate specificity of OsAOS1 for (cis,trans)-configurational isomers of 13(S)- and 9(S)-hydroperoxyoctadecadienoic acid. GC-MS analysis showed that OsAOS1 converts 13(S)- and 9(S)-hydroperoxyoctadecadi(tri)enoic acid into their corresponding allene oxide. UV-Visible spectral analysis of native OsAOS1 revealed a Soret maximum at 393 nm, which shifted to 424 nm with several clean isobestic points upon binding of OsAOS1 to imidazole. The spectral shift induced by imidazole correlated with inhibition of OsAOS1 activity, implying that imidazole may coordinate to ferric heme iron, triggering a heme-iron transition from high spin state to low spin state. The implications and significance of a putative type II ligand-induced spin state transition in OsAOS1 are discussed.

일상적인 음식 섭취 후 발생한 급성 메트헤모글로빈혈증 (Acute methemoglobinemia after a normal diet)

  • Methemoglobin is generated by the oxidation of ferrous iron to ferric iron within a hemoglobin molecule. Methemoglobin is unable to bind and transport oxygen, resulting in methemoglobinemia, which can lead to fatal tissue hypoxia. The most common cause of methemoglobinemia is poisoning by oxidizing agents such as dapsone, benzocaine, and primaquine. However, methemoglobinemia can also be caused by normal dietary sources. We present two cases of methemoglobinemia that developed after a normal diet in two male patients. In this case report, the patients suddenly developed dyspnea and cyanosis after eating the same meal. They had no history of suspected poisoning, such as the use of drugs, exposure to chemicals, or gas inhalation. Their symptoms did not improve even after a high dose of oxygen was administered; further, an abnormal 'oxygen saturation gap' was observed. Because of CO-oximetry, the methemoglobin levels of the patients were 50.0% and 46.6%, respectively. We administered methylene blue (1 mg/kg), and the patients recovered completely without any complications. Emergency physicians should, therefore, be aware that methemoglobinemia can also be caused by normal dietary sources. In addition, if the source and route of contamination are unclear, an epidemiological investigation should be conducted.

구충증(鉤蟲症)에 관(關)한 연구(硏究) 제2편(第2編) 감염경중(感染輕重)에 따른 구충증(鉤蟲症)의 임상증상(臨床症狀), 빈혈상(貧血像) 및 약물(藥物) 특(特)히 철제제(鐵製劑)의 효과(效果) (Studies on Ancylostomiasis II. Clinical Symptomatology, Effects of Drugs, especially of Iron Compounds, with Relation to the Severity of Artificial Infection)

  • This study was aimed; firstly to observe various clinical symptomatology with reiation to the number of Ancylostoma duodenale larvae orally given to the human beings, secondly to evaluate the effects of some drugs like steroid hormones, antihistaminic3 and antitussives, and, thirdly to study the influences of some iron compounds in prevention and treatment of anemia of such origin. Ten healthy volunteers free from the previous history of hookworm infection were divided into 4 groups, to whom various numbers of actively moving filariform Ancylostoma duodenale larvae were orally given; 500 to 4 cases, 250 to 3 cases and 100 to 3 cases. Following were the results: 1. Clinical symptomatology 1. The most frequently encountered symptoms and signs were general malaise, cough and hoar seness. The tracheal itching and pain low back pain, arthralgia, sputum and salivation, acid belching, loss of appetite, abdominal pain and vomiting were also noted. 2. If the larger number of the larvae was given, the clinical symtomatology was more severe. 3. Prednisolone medication caused some improvement of such symptomatology, while the antihistaminics and antitussives like codeine or ephedrine were ineffective. 4. In volunteers whose nutritional conditions were rather poor appeared to show more severe symptomatology. 2. Effects of iron compounds 1. The oral administration of ferrous fumarate induced a slight increase of serum iron levels in the initial stage of the infection, then a decrease from $15{\sim}20$ days later and a recovery after 2 months. 2. The intravenous administration of saccharated ferric oxide induced a steady upkeep of the serum iron levels. 3. The hemoglobin contents also showed the upkeeps after either the oral or intravenous administration of the iron compounds. 4. The iron compounds, therefore, are considered to have the preventive as well as the therapeutic effects on hookworm anemia, which may strongly suggest that hookworm anemia is essentially the iron deficiency type. 3. Hematological changes 1. The severity of hookworm anemia generally correlated with the number of larvae given. 2. The moderate leucocytosis was observed in all cases regardless of the number of larvae given, which reached to a peak in $25{\sim}35$ days. 3. Eosinophilia was observed in all cases, but was more severe in cases given larger number of larvae, which was slightly less evident after the medication of prednisolone. 4. Red cell survival time The red cell survival time determined by $^{51}Cr$ was generally in the normal ranges except for the severe anemia patients.

Rat에 있어서 탈지사료 및 철분투여가 다불포화 지방산, vitamin E 및 그 유도체에 미치는 영향 (Effect of administration with fat-free diet and dietary iron on polyunsaturated fatty acid, vitamin E and their derivatives in male rats)

  • This study was designed to elucidate the effects of administration with dietary iron and fat-free diet on the contents of unsaturated fatty acid in phospholipid molecules, vitamin E contents and malondialdehyde contents in liver, kidney, muscle and testis of the male rats. The rats were divided into 3 experimental groups, namely, control, iron injection and fat-free diet administration groups. The control group was fed with normal diet, iron injection group injected intraperitoneally 20mg of ferric hydroxide/100g of body weight 20 times every 3 days and fat-free diet group administered lipid extraction diet with hexane in normal diet. All experimental groups were maintained for 60 days with feeding on the respective ration. The results obtained were summarized as follows: 1. In the mean contents of unsaturated fatty acid in phospholipid of liver, kidney, muscle and testis among groups, control group was 21.31mg/g, 19.38mg/g, 1.67mg/g, 13.68mg/g, iron injection group was 13.83mg/g, 16.53mg/g, 0.71mg/g, 10.11mg/g and fat-free diet group was 21.07mg/g, 19.38mg/g, 1.49mg/g and 13.40mg/g, respectively. 2. In the mean contents of vitamin E in liver, kidney, muscle and testis among groups, control group was 6.77mg/g, 1.93mg/g, 0.12mg/g, 0.17mg/g, iron injection group was 3.16mg/g, 0.86mg/g, 0.07mg/g, 0.09mg/g and fat-free diet group was 7.41mg/g, 1.50mg/g, 0.11mg/g and 0.16mg/g, respectively. 3. In the mean contents of malondialdehyde in liver, kidney, muscle, testis and serum among groups, control group was 11.29nM/0.1g, 23.25nM/0.1g, 42.47nM/0.1g, 7.01nM/0.1g, 4.33nM/ml, iron injection group was 34.98nM/0.1g, 40.55nM/0.1g, 72.21nM/0.1g, 12.26nM/0.1g, 11.27nM/ml and fat-free diet group was 8.07nM/0.1g, 20.63nM/0.1g, 39.92nM/0.1g, 6.95nM/0.1g and 4.27nM/ml, respectively.

전기영동법으로 알루미늄에 침적된 영가 철 나노입자에 의한 질산성 질소의 환원 (Reduction of Nitrate-Nitrogen by Zero-valent Iron Nanoparticles Deposited on Aluminum yin Electrophoretic Method)

  • 최근 주요 수질오염 물질로 대두되고 있는 질산성 질소의 제거를 목적으로 영가 철 나노입자에 의한 질산성 질소의 환원반응성을 평가하였다. 영가 철 나노입자의 제조방법에 따른 반응성 차이를 규명하기 위해 유기용매 상에 계면활성제를 첨가하여 나노미터 크기 수준의 수용액 분산상에서 입자를 합성하는 마이크로에멀젼 방법과, 수용액 상의 철 이온을 환원시켜 입자를 합성하는 두 가지 방법으로 영가 철 나노입자를 합성하였다. 또한 전기영동법으로 알루미늄에 침적시킨 영가 철 나노입자에 의한 질산성 질소 제거속도를 측정하고, 고정화되지 않은 나노 철 입자에 의한 반응속도와 비교하였다. 환원반응을 질산성 질소에 대한 1차 반응으로 가정하여 수용액 방법 및 마이크로에멀전 방법으로 제조된 영가 철 나노입자의 반응성을 평가한 결과, 반응속도상수는 각각 $1.40{\times}10^{-2}min^{-1}$$3.49{\times}10^{-2}min^{-1}$ 로서 비표면적에 비례하여 증가하였다. 알루미늄에 침적된 나노입자는 현탁된 나노입자의 반응과 비교하여 약 30% 감소된 반응속도를 보였으나, 과량의 질산성 질소가 존재하는 경우 나노 철의 단위 질량당 질산성 질소의 제거효율 면에서 더 우수한 특성을 보였다. 나노철 입자의 현탁액은 반응시간 30분 이내에 반응속도가 감소하는 경향을 보였으나, 알루미늄에 침적된 나노철 입자는 3시간 이상 활성을 유지하였으며, 최종 생성물로 기체 질소를 발생시키는 것을 확인하였다.

Grignard 시약과 브로모벤질과의 반응에서 첨가전이 금속이온의 영향 연구 (Effect of Transition Metal Ion on the Reaction of Benzylbromide with Grignard Reagent)

  • Grignard 시약,CH_3$MgI와 브로모벤질과의 반응은 전자전달과정(SET)과 라디칼 이온쌍 중간체를 거쳐 생성되는 동종짝지음 생성물 bibenzyl을 주생성물로 이온성 공격에 의해 생성되는 교차짝지음 생성물, 에틸벤젠을 부생성물로 생성시키며 전이금속 불순물이 거의 제거된 순수 금속 Mg으로 CH_3$MgI을 제조하였을 때 그 비는 78:22이었다. 그러나 실험실용 금속 Mg을 사용하였을 때 그 비는 감소하여 67:33이었으며 이 비는 촉매량으로 첨가시킨 $FeCl_3$ 양의 증가로 더욱 감소되어 최고 20:80 비로 Fe 이온은 이온성 교차생성물, 에틸벤젠을 주생성물로 유도하였으며 이러한 반응결과는 좋은 수소원자주게로 디이소프로필 에테르를 디에틸 에테르 대신 용매를 사용하였을 때에도 큰 변화가 관찰되지 않았다. 이러한 관찰은 Grignard 시약과 유기할로겐 화합물이나 카르보닐 화합물과의 반응에서 촉매 양의 전이금속염이 전자전달과정 속도를 증가시켜 동종짝지음 이합체 생성물의 증가를 일으키는 보고와 반대의 경향을 보인다. 이러한 촉매량의 $FeCl_3$ 첨가가 CH_3$MgI의 이온성 공격의 증가와 그에 따른 교차짝지음 생성물의 증가로 유도하는 것은 브로모벤질과 반응 중 생성된 Fe 종과의 착화합물 형성에 의해 벤질탄소-브롬 결합이 CH_3$MgI의 탄소음이온 공격에 대해 반응성이 증가되는 것으로 설명하였다. 이러한 설명은 2-페닐-1-브로모에탄의 CH_3$MgI와의 반응연구에서 관찰된 반응성 결여가 뒷받침하고 있다.

탄소/산화철 나노복합재료의 Brilliant Green 흡착에 대한 반응속도론적, 열역학적 연구 (Kinetic and Thermodynamic Studies of Brilliant Green Adsorption onto Carbon/Iron Oxide Nanocomposite)

  • 본 연구에서는 수용액 중에서 유해한 brilliant green (BG) 제거를 위한 탄소와 산화철 나노복합재료의 흡착특성을 살펴보았다. 탄소와 산화철 나노복합재료는 화학침전법과 $750^{\circ}C$에서 질산 철과 탄소를 열처리 함으로서 합성하였다. 그 생성물은 TEM, XRD, 그리고 TGA를 이용하여 확인하였다. 나노복합재료에 대한 BG의 흡착 연구는 열역학적 인자와 반응속도론적인자들을 이용하여 수행하였다. 흡착 속도식은 준 이차 속도식이 준 일차 속도식에 비해 잘 들어맞는다고 보여준다. 실험 결과는 Langmuir 과 Freundlich 흡착 등온선을 이용하여 분석하였다. 평형 데이터는 Langmuir모델에 잘 들어맞으며 최대 단일 층 흡착 용량 64.1 mg/g 을 갖는다. 열역학적 인자들은 나노복합재료에 BG의 흡착 으로부터 유도하며 흡착은 자발적이며 흡열 과정임을 확인하였다.

Separation of Phosvitin from Egg Yolk without Using Organic Solvents

  • The objective of this study was to develop a new method to separate phosvitin from egg yolk without using organic solvents. Phosvitin was extracted from yolk granules using 10% NaCl or 10% $(NH_4)_2SO_4$ (final concentration) and then treated with heat to precipitate the lipoproteins from the extracted solution. The optimal pH for the phosvitin extraction from yolk granules was determined, and the iron-binding ability of the extracted phosvitin (final product) was tested. Adding 10% $(NH_4)_2SO_4$ disrupted the granules, and the subsequent thermal treatment at $90^{\circ}C$ for 1 h precipitated low density and high density lipoproteins, which enabled separation of phosvitin by centrifugation. The phosvitin concentration in the extract was significantly higher when the pH of the solution was adjusted to pH ${\geq}9$. The purity and recovery rate of phosvitin at the end of the separation process were approximately 78% and 56%, respectively. The separated phosvitin was confirmed to have ferrous and ferric iron binding ability. The advantages of this new method compared with the traditional methods include no organic solvents and high-priced equipment are needed for the separation. Also, this method is more environment and consumer friendly than that of the traditional methods.
